1.Application of in vitro bionic digestion and biomembrane extraction for metal speciation analysis, bioavailability and risk assessment in lianhua qingwen capsule.

One of the causes of the high cost of pharmaceuticals and the major obstacles to rapidly assessing the bioavailability and risk of a chemical is the lack of experimental model systems. A new pre-treatment technology, in vitro bionic digestion was designed for metal analysis in Lianhua Qingwen capsule. The capsule was digested on 37 degrees C under the acidity of the stomach or intestine, and with the inorganic and organic compounds (including digestive enzymes) found in the stomach or intestine, and then the chyme was obtained. Being similar to the biomembrane between the gastrointestinal tract and blood vessels, monolayer liposome was used as biomembrane model Affinity-monolayer liposome metals (AMLMs) and water-soluble metals were used for metal speciation analysis in the capsule. Based on the concentration of AMLMs, the main absorption site of trace metals was proposed. The metal total contents or the concentration of AMLMs in the capsule were compared to the nutritional requirements, daily permissible dose and heavy metal total contents from the "import and export of medicinal plants and preparation of green industry state standards". The metal concentrations in the capsule were within the safety baseline levels for human consumption. After in vitro bionic digestion, most of trace metals were absorbed mainly in intestine. The concentration of As, Cd, Pb was 0.38, 0.07, 1.60 mg x kg(-1), respectively, far less than the permissible dose from the "import and export of medicinal plants and preparation of green industry state standards".

2.Identification of Chaetomium sp. CQ31 and Optimization of Xylanase Production in Solid State Fermentation

A fungus CQ31 isolated from soil samples was identified as Chaetomium sp.. This strain produced effectively xylanases utilizing several liguocellulosic materials in the solid-state fermentation (SSF), and corn straw was the best carbon source. The results of single-factor-experiment showed that the corn straw as the carbon source, tryptone as the nitrogen source, initial moisture content of 80% and initial pH 9.0 were the optimal conditions for xylanase production. Under the optimized conditions, it produced xylanase which was 4897 U/g dry substrate while mannanase was 803 U/g dry substrate after 7 days of cultivation. Therefore, xylanase and mannanase production by Chaetomium sp. CQ31 in SSF possess potential for commercial applications.
3.Estabishment of Identification Method for Longdan Xiegan Pills(Honey Pills) by TLC

Objective:To research the TLC identification method for Longdan Xiegan pills( honey pills) . Methods:TLC was used in the identification. The samples were extracted by 70% methanol with a heating reflux method, and then extracted by the agents with dif-ferent polarity, including petroleum ether, ethyl acetate and butanol. The petroleum ether part was detected by fluorescence at 365nm for Angelica sinensis, and 1% vanillin-sulfuric acid color reaction was used to detect Alisma orientale. The ethyl acetate part was determined by fluorescence at 365nm for Scutellaria baicalensis, and the butanol part was detected with chloroform-methanol-water (30∶12∶3) as the developing solvent for Bupleurum and Glycyrrhiza, and with acetone-ethyl acetate-water (6∶6∶1) as the developing solvent for gentiopi-croside, geniposide and liquiritin. Results:The developed TLC spots were clear with good separation, high specificity and promising re-producibility. Conclusion:The method can be exactly used in the qualitative identification and quality control of Longdan Xiegan pills ( honey pills) .
4.MRI and Pathological Features of Ovarian Endometrioid Adenocarcinoma

PurposeTo study the MRI and pathological features of ovarian endometrioid adenocarcinoma (OEC) in order to evaluate the value of MRI in diagnosing OEC. Materials and Methods The MRI imaging features of 8 patients confirmed by surgery and pathology were analyzed retrospectively and were compared with the results of surgery and pathology. The MRI outcome and the related histological findings were further analyzed.Results Six out of the eight patients had unilateral tumor, 3 tumors in the left and the other 3 in the right; 2 patients had masses in the both ovaries. The total number of masses was ten.The diameters of the tumors ranged from 3.5 to 16.5 cm, with the average size of (10.5±4.1) cm. The border of 2 tumors was partially fuzzy and that of the other 6 was clear. The MRI scans showed that 8 tumors were cystic-solid and the other 2 were solid with heterogeneous signals. The solid components mainly presented slightly short T1 signals and long T2 signals; the cystic ones revealed long T1 and T2 signals. The enhanced scanning showed that the solid components of 8 tumors were patchy and obviously enhanced and the other 2 had mild or moderate enhancement. The cystic components were not hyper-intense. The enhanced MRI scans of two cases of primary endometrial carcinoma with metastases to the ovaries showed thickened endometrium and mild hyper-intense.Conclusion MRI can reveal the pathological features of OEC and clearly presents the forms, components and the relationship with its surroundings of tumors. Therefore, MRI is of great importance to the clinical diagnosis of OEC.
5.Apparent Diffusion Coefficient in Assessing Early Response to Tyrosine Kinase Inhibitors in Non-small Cell Lung Cancer

[Abstact] Purpose To investigate the value of apparent diffusion coefficient (ADC) of diffusion weighted imaging (DW1) in the evaluation of the efficacy of treatment of nonsmall cell lung cancer (NSCLC) with tyrosine kinase inhibitor (TKI).Materials and Methods Nineteen patients with advanced NSCLC who treated with TKIs from May 2014 to December 2015 were recruited prospectively.All patients underwent CT and MRI scans three times before targeted therapy,after 1 week and 4 weeks of treatment.The tumor maximal diameter and mean ADC value at different time points were compared.The correlations between mean ADC value and tumor maximal diameter and maximum diameter change rate at different time points were analyzed.Bland-Altman analysis was performed to confirm the reproducibility of measurements.Results The ADC value after 1 week of treatment was significantly increased compared with that before treatment (t=-6.329,P<0.05),but the change of tumor maximal diameter was not significant (P>0.05).The ADC value was significantly increased after 4 weeks of treatment compared with that before treatment (t=-4.878,P<0.05),and the change of tumor maximal diameter was also significant (t=7.054,P<0.05).The negative correlation was detected between the ADC before treatment and the maximal diameter change ratio after 4 weeks of treatment (r2=-0.474,P<0.05).In the group of responders,there was significant difference between the ADC value before treatment and 1 week after treatment (P<0.05).There was significant difference in ADC value between the groups of responders and non-responders after 4 weeks of treatment (P<0.05).The Bland-Altman analysis showed good reproducibility among the operators.Conclusion The ADC value after 1 week of targeted therapy is more sensitive to the change of tumor than the maximal diameter,and the ADC value before treatment has a certain value in predicting the maximum diameters of the target after 4 weeks of targeted therapy with repeatability.
6.The promotion of nerve regeneration in peripheral nerve by progesterone after terminolateral neurorrhaphy: an experiment study

Objective To evaluate the effect of progesterone (PROG) on promoting peripheral nerve collateral growth and nerve regeneration after terminolateral neurorrhaphy (TLN). Methods Thirty-six female SD rats were from nine to ten weeks old and weighed from 250 to 270 grams. They were randomly divided into two groups by different drugs they were administrated, the PROG group and the control group. Both groups were subdivide into three groups respectively, PROG four weeks group, PROG eight weeks group, PROG twelve weeks group, control four weeks group, control eight weeks group and control twelve weeks group. There were six subjects in every subgroups. First, both groups were performed ovariectomy. Then, two weeks later, TLN was operated. In the PROG group, the left peroneal nerve was transected, then an 1mm epineukral window was created on the neighbouring. The distal end of peroneal nerve was sutured to the windowed tibial nerve by means of end-to-side attachment. In the control group, nerve TLN was performed as in the PROG group, 0.2ml (10mg/ml) PROG and tea-oil (the solvent of PROG) was injected by subcutaneous injection for days in PROG group and control group respectively. Electrophysiological, histolgicial and morphological examinations were measured at 4,8, and 12 weeks after the operation. Results Regeneration of peroneal nerve occurred in both groups. 8.Analysis on the arsenic speciation in urine of rats treated with sodium arsenite and sodium arsenate

Objective To analyze the levels and speciation of arsenic metabolites in urine of rats treated with sodium arsenite and sodium arsenate in order to investigate the different aspects of metabolism between sodium arsenite and sodium arsenate,thus to understand further the basic data about relationship between it's metabolism and mechanism of toxicity. Methods Seventy Wistar rats,weighting 80-120 g,were divided into 7 groups of 10 each,such as normal control group,high,middle and low sodium arsenite group and high,middle and low sodium arsenate group. After the animals were fed for one month,the urine was collected by metabolic cage in 12 hours. Applying the high efficiency liquid chromatography and hydride genesis atomic fluorescence spectroscopy (HPLC-HGAFS),the levels and speciation of arsenic metabolites were determined in urine of rats. Meanwhile,the recovery rate of dimethyl arsinic acid(DMA) would be determined to estimate the degree of accuracy of results. Results The levels of iAs~(3+),iAs~(5+) and DMA in middle sodium arsenite group[(121.66±1.26),(10.26±2.68),(200.91±0.56) μg/L]were higher than the high sodium arsenite group[(113.20±0.75),(5.16±1.32),(147.70±μ0.77)μg/L,all P < 0.05]and low sodium arsenite group[(79.35±2.12),(5.13±2.25),(56.35±1.23)μg/L,all P < 0.05]. The levels of iAs~(3+) and DMA in middle sodium arsenate group[(315.81±1.69),(245.12±1.18)μg/L]were higher than the high sodium arsenate group[(85.03±0.56),(110.34±1.04)μg/L,all P< 0.05]and low sodium arsenate group[(22.97±2.67),(15.75±2.15)μg/L,all P < 0.05]. Compared with sodium arsenate group,the levels of iAs~(3+) and DMA in high and low sodium arsenite group were higher(all P < 0.05) ; and the levels of iAs~(3+) and DMA in middle sodium arsenite group were lower(all P < 0.05). Meanwhile,the average urinary recovery rate of DMA of rats in different sodium arsenite group were 94.80%-102.70%,and the average urinary recovery rate of DMA of rats in different sodium arsenate group were 95.33%-108.40%. Conclusion The speciation and levels of arsenic are influenced by the external exposure dose,and some distinction appeared in the metabolism and metabolic path between sodium arsenite and sodium arsenate in urine in vivo.
10.Optimization of extraction technology from Paeoniae Radix Alba using response surface methodology.

To ensure the stability of chemistry components and the convenience of operation, ultrasound method was chosen to study in this investigation. As the total common peaks area in chromatograms was set to be evaluation index, the influence on the technology caused by extraction time, ethanol concentration and liquid-to-solid ratio was studied by using single factor methodology, and the extraction technology of Paeoniae Radix Alba was optimized by using response surface methodology. The results showed that the extracting results were most affected by ethanol concentration; liquid-to-solid ratio came the second and extraction time thirdly. The optimum ultrasonic-assisted extraction conditions were as follow: the ultrasonic extraction time was 20.06 min, the ethanol concentration in solvent was 72.04%, and the liquid-to-solid ratio was 53.38 mL · g(-1), the predicted value of total common peaks area was 2.1608 x 10(8). Under the extraction conditions after optimization, the total common peaks area was 2.1422 x 10(8), and the relative deviation between the measured and predicted value was 0.86%, so the optimized extraction technology for Paeoniae Radix Alba is suitable and feasible. Besides, for the purpose of extracting more sufficiently and completely, the optimized extraction technology had more advantages than the extraction method recorded in the monogragh of Paeoniae Radix Alba in Chinese Pharmacopoeia, which will come true the assessment and utilization comprehensively.
