1.ISOLATION AND CULTURE OF NEURAL STEM CELLS FROM BRAIN OF RAT

Objective To investigate the effect of optimized conditions on proliferation and differentiation of the neural stem cells(NSCs) from different age brain of rat in vitro. Methods The brain tissues from embryonic(E12~E15d),newborn(1~2?d) hippocampus and olfactory bulb from adult of rat were isolated.After proliferation in conditional culture medium including growth factor and induced differentiation,the cell type specific antibodies were used for the immune fluorescence staining to identify the cells. Results A lager scale NSCs were obtained from brain tissue and could proliferate in conditional culture in vitro. They could self renew and generate both nerve and glia.Conclusion\ The NSCs got from rat brain can be used for basic and clinical research.\;[
2.Effects of Exercise and Estrogen Replacement on the Content of Nitric Oxide and Expression of eNOS and iNOS in Liver of ovariectomized Rats

Objective In this study,the effects of moderate treadmill exercise and estrogen replacement treatment on the production of liver nitric oxide(NO)and the expression of endothelial nitrie oxide synthase(eNOS)and inducible nitric oxide synthase(iNOS)in ovariectomized(OVX)rats were observed.Methods Female mature SD rats were randomly divided into six groups according to their body weight:sham group(group S),sham exercise group (group S+EX),group OVX,OVX+exercise group (group OVX+EX),OVX+estrogen group(group OVX+E_2),and OVX+exercise+E_2 group(group OVX+EX+E_2).The rats ran on the treadmill at a speed of 18m/min for 60min,5 times per week.50μg/kg of E_2 was given subcutaneously three times per week to groups OVX+E_2 and OVX+EX+E_2.After 14 weeks,NO production in the liver was determined by nitrate reductase method.Protein expression of eNOS and iNOS was determined by immunohistochemistry.Results The NO content of group S was significantly lower than that of group S+EX,but significantly higher than that of group OVX.The NO content of groups OVX+EX,E_2,and OVX+EX+E_2 were significantly higher than that of group OVX.The positive reaction of eNOS mainly located in the cytoplasm of hepatocytes and endothelial cells of the hepatic sinusoids in all groups.The changing tendency after exercise or/and estrogen replacement was almost similar with that of the content of NO.The positive reaction of iNOS mainly located in the cytoplasm of hepatocytes in groups S,S+EX,E_2,and OVX+EX+E_2,while it located in the nuclei in both groups OVX and OVX+EX.Conclusion Moderate exercise and estrogen supplementation both had the same effect of upregnlated synthesis of NO in liver of ovariectomized rats.
3.Design of Portable Medical Measure System on Palm

Objective: This item presents a feasible scheme of home monitoring device which is a low power and miniaturization equipment for human signal detection, based on single-chip technology. Methods: Taking MSP430F149 as core, the realized functions of system contain real time data acquisition, data processing, data storage, LCD and alarming, keyboard as well as com-munication, replay and reproeessing by host computer. Results: The physiological and biochemical parameters are acquired by system and transmitted to host computer. Conclusions: This design provides a platform for multi-parameters portable detection just as dynamic ecg, ambulatory blood pressure, and so on. It has a good future for application.
4.Research on carrying situation of Clostridium difficile toxin gene cdtA/B in stool of patients with ulcerative colitis

Objective To detection the carrying situation of Clostridium difficile toxin gene A/B in stool specimens of the pa-tients with ulcerative colitis (UC) and persons undergoing physical examination and to research the relationship between Clostridium difficile toxin gene carrying and UC.Methods The stool specimens were collected from 53 cases of UC(32 cases of active stage and 21 cases of resting stage) and 45 persons undergoing physical examination.Total DNA was extracted from stool specimens.The Clostridium difficile toxin gene cdtA and cdtB were detected by real-time PCR ,then the PCR products were amplified and performed the agarose gel electrophoresis and gene sequencing for conducting the amplification products verification.Results 7 cases of cdtA and 4 cases of cdtB were checked out in the UC group ,in which 5 cases of cdtA and 2 cases of cdtB were in the UC active stage group ,2 cases of cdtA and 2 cases of cdtB were in the UC resting stage group.In the healthy control group ,5 cases of cdtA and 2 cases of cdtB were checked out.The detection rate of cdtA and cdtB had no statistically significant difference between the UC group and healthy control group(P>0.05).There was no statistically significant difference between UC active group and inactive group (P>0.05).Conclusion There is no appearant correlation between the carrying situation of cdtA and cdtB in stool with UC onset and UC stage.
5.A clinical analysis of primary Sj(o)gren's syndrome with anticentromere antibodies

Objective To investigate the clinical manifestations, immunological features and prognosis of primary Sj(o)gren's syndrome(pSS) with anticentromere antibodies(ACA). Methods Sixty pSS patients with ACA in our hospital between 1985 and 2006 were screened retrospectively and compared with those without ACA. Results The mean age at the onset of pSS with ACA was higher than that of those without ACA[(48±11)yr vs(41±12)yr,P=0.000].There was no difference in sex ratio,dry mouth.dry eyes and positive salivary gland biopsy between the two groups(P>0.05).Compared with those without ACA,patients with ACA presented a higher prevalence of liver involvement(68.3%vs 37.0%,P=0.000),while a lower prevalence of renal involvement(13.3%vs 30.9%,P=0.009), neuropathy(1.7% vs 11.5%,P=0.025)and hypergammaglobulinemia(20.8%vs 45.7%,P=0.002). The difference was not significant between the two groups in Raynaud's phenomenon,articular involvement,myositis,hematologic involvement,lung involvement,and thyroiditis. While both groups showed the same prevalence of antinuclear antibody(ANA),the patterns of ANA-IF were different and the discrete speckled pattern was the most freqent in patients with ACA and occurred in 61.7%. Different from those without ACA,patients with ACA presented a lower prevalence of anti-SSA,anti-SSB, rheumatoid factor, and antiU1 RNP,while showed a higher prevalence of antimitochondrial antibodies(AMA) and AMA-M2. The most frequent cause of death was the complications associated with cirrhosis,notably bleeding varices(3/5 cases). Conclusion Patients with ACA present a high risk of liver involvement. Because of the remarkable difference in the mean age of disease onset and also differences in systemic damage, immunological and antibody features, pSS with ACA may be a special subtype of pSS.
6.BIOTRANSFORMATION OF (+)-AND (-)-CLAUSENAMIDE IN RATS

AIM To study the metabolic pathway of chiral clausenamide in the rat and understand its stereoselectivity. METHODS The urine, feces and blood of rat were gathered after the drug was administered, the known metabolites were analyzed by HPLC-DAD and one unknown metabolite was elucidated by using LC-MS analysis. Metabolic stereoselectivity was determined by comparing the metabolic results of (+)- and (-)-clausenamide. RESULTS Six known metabolites were determined and one unknown metabolite was elucidated as N-demethylclausenamide. The metabolic stereoselectivity was shown distinctly. CONCLUSION Chiral clausenamide was mainly metabolized by hydroxylation in liver and the biotransformation exhibited pronounced substrate stereoselectivity.
7.Observation on the effect of continuous infusion of dopamine and furosemide in treatment of 76 elderly patients with refractory heart failure

Objective To explore the clinical effect of continuous infusion of dopamine and furosemide on elderly patients with refractory heart failure.Methods 76 elderly patients with heart failure were selected as the observation subjects,they were randomly divided into observation group and control group,38 cases in each group.The control group was given oxygen inhalation,the expansion of vascular,cardiac and other conventional treatment.The observation group was given conventional treatment and continuous infusion of dopamine and furosemide.Clinical efficacy and cardiac function improvement were compared between the two groups.Restlts The effective rate of observation group was 92.1%,which was higher than that of the control group (73.7%) (x2 =10.29,P < 0.05).After treatment,the improvement of cardiac function between the two groups had significant difference (t =15.94,10.18,all P <0.05).Conclusion Continuous infusion of dopamine and furosemide in treatment of elderly patients with refractory heart failure could improve heart function,enhance curative effect.
8.Lung cancer complicated with syndrome of inappropriate secretion of antidiuretic hormone:A report of 2 cases and literature review

Objective:To analyze the clinical materials of 2 patients with lung cancer complicated with syndrome of inappropriate secretion of antidiuretic hormone (SIADH),and to improve the clinical awareness and management.Methods:One patient was treated with lung cancer chemotherapy alone for 7 courses.The other patient received the strictly limited for liquid intake and tolvaptan by oral against hyponatremia without chemotherapy.Results:With the relapsing and advancing of lung cancer,the hyponatremia and its relevant symptoms of the first patient were poorly relieved.While the level of serum sodium of the second patient returned to the normal level 3 d after administration of tolvaptan.Conclusion:About one-third of the patients with hyponatremia have SIADH.The etiological treatment and symptomatic therapy of hyponatremia should be given to the patients in clinical treatment.
9.THE EXPERIMENTAL STUDY ON THE ABSORPTIOM OF IRON IN ~(59)Fe-ENRICHED NUTRIENT YEAST

The absorption of 59 Fe incorporated into yeast was studied in rats. Yeast was cultured in a nutrient solution to which radioactive 59Fe was added as an intrinsic tag. The 59Fe-enriched yeast was separated from the solution by centrifuge and given to rats by a stomach tube. The rats of the control groups were given the radioactive " Fe together -with ordina-ly yeast plus ferrous sulfate or only with ferrous sulfate separately.All feces were collected until less than 0.5 per cent of the administered dose was present in a 24-hour specimen of feces. The radioactivity in the feces was then measured daily for three successive days. The absorption of iron was calculated by the difference between intake and fecal loss.The result revealed that the absorption of iron in "Fe-enriched yeast (38.7%) was similar to that of ferrous sulfate (38.3%). It indicated that the iron incorporated into yeast was easily to be absorbed.This study afforded the scientific data for researching and popularizing fortified food with iron-enriched yeast.
10.Correlation among serum prealbumin level, red cell distribution width and severity of heart failure in advanced aged male patients

Objective: To explore the correlation among serum prealbumin (PAB) level, red cell distribution width (RDW) and cardiac function in advanced aged male patients with chronic heart failure (CHF). Methods: Research objects included 60 patients with heart failure(28 cases with NYHA class Ⅱ and 32 cases with NYHA class Ⅲ-Ⅳ, heart failure group)and 30 cases with normal cardiac function admitted in the same period(normal control group). Serum PAB, hemoglobin (Hb) concentration and RDW, red blood cell (RBC) count were measured. Patients all received echocardiography examination. Changes of all indexes were compared among patients with different cardiac function class. Results: ① Compared with normal control group, there was significant decrease in serum PAB level [(252±49) mg/L vs. (185±36) mg/L] and significant increase in RDW [(13.84±0.60) % vs. (15.79±1.33) %] in heart failure group, and compared with class Ⅱ group, there was significant decrease in serum level of PAB and significant increase in RDW in class Ⅲ-Ⅳ group, P<0.01 all; ② Compared with normal control group, there were significant decrease in E/A [(1.02±0.36) vs. (0.75±0.18)] and left ventricular ejection fraction [LVEF, (59±9) % vs. (49±11) %] in heart failure group, and the worse cardiac function was, the lower these two indexes were, P<0.01 all; ③ Pearson linear correlation regression analysis indicated that PAB was positively correlated with E/A and LVEF (r=0.451, P<0.05; r=0.596, P<0.05), and RDW was negatively correlated with E/A and LVEF (r=-0.391, P<0.05; r=-0.574, P<0.05) in heart failure group. Conclusion: Prealbumin and red cell distribution width are closely correlated to cardiac function class in advanced aged male patients. Both of them can indirectly reflect cardiac function in advanced aged patients with chronic heart failure.
