1.Effect of Aerobic Exercise on the Maximal Oxygen Uptake in Sedentary Healthy Adults: A Meta Analysis

Objective Through this meta analysis of controlled clinical trials of the effect of aerobic exercise on the maximal oxygen uptake(VO_2max)in sedentary healthy adults,we attempt to provide evidence for exercise prescription.Methods MEDLINE search for controlled trials that involved in the effect of aerobic exercise on the VO_2max from 1976 to 2008 was conducted.The literatures were selected according to inclusion criteria and exclusion criteria.Review Manager 5.0 software provided a systematic evaluation for calculating VO_2 max Weighted Mean Difference with random effects model.Assessment of the sub-group of aerobic exercise intensity and literature publication bias were conducted.Results(1)Thirty five literatures were included in this review;involving 889 adults met the inclusion criteria.VO_2max Weighted Mean Difference is 3.61ml/kg/min,95%confidence interval 2.96~4.25ml/kg/min,with an increase of 13.2%,comparing with the average VO_2max values(27.26 ml/kg/min)before aerobic exercise.(2)The most effective exercise intensity was 70~80%VO_2max,71~80%HRmax and 61~70% HRR according to the sub.group of aerobic exercise intensity meta analysis.Conclusion VO_2max increased significantly due to participating in aerobic exercise in sedentary healthy adults.The results will provide a theoretical basis for me development of aerobic exercise prescription with appropriate intensity.
2.Impact of NBP on expression of HSP70 and TLR4 in rat with ischemia-reperfu-sion

Objective:To analyze the impact of NBP on the expression of HSP 70 and TLR4 in the rat with ischemia-reperfusion.Methods:The 144 rats were divided into 3 groups,Sham group,IR group and NBP group.The 3 groups were divided into 4 groups again according to the time of reperfusion (6h,12h,24h and 48h).The intraluminal ischemia-reperfusion model was made by improved Longa.When the model was made successfully ,the rat in NBP group was gave oral NBP immediately while the IR and Sham group was gave oral NS.The neurological deficit scores , number of apoptotic cells , expression of HSP70 and TLR4 were compared during the 3 groups.Results:The neurological function ,apoptosis,the expression of HSP70 and TLR4 in NBP group at any reperfusion time were statistical lower than in IR group.In all the infusion periods,apoptotic cells in IR and NBP group were significantly higher than that in the Sham group.In all the infusion periods ,the positive cells in IR and NBP group were significantly lower than that in the Sham group.Conclusion:NBP can protect the function of neuron by improving the inhibition of HSP 70 and TLR4.

4.Epigenetic regulatory effects of the methylation of CpG island shore on the expression of lag3 gene in various human T cell lines

Objective To investigate the effects of DNA methylation on the expression of lympho-cyte activation gene 3 (lag3) in different human T cell lines.Methods A quantitative PCR and a flow cy-tometry analysis were performed to measure the expression of lag3 gene in various T cell lines at mRNA and protein levels.The distribution of CpG sites within the promoter and body of lag3 gene was detected to locate the potential regulatory region(s) (CpG island and CpG island shore).The levels of DNA methylation in each cell line were analyzed.The T cell lines were demethylated with 5-Aza-2′-deoxycytidine (5-Aza-2′-dc) for further investigation on the changes of lag3 gene expression and DNA methylation.Results Jurkat E6-1 cells showed the highest expression level of lag3 gene as compared with J.CaM1.6 and CEM cells.Hyperm-ethylated CpG islands were detected in cells of each cell line.The methylation levels of CpG island shore in J.CaM1.6 and CEM cells were higher than that in Jurkat E6-1 cells.Treatment of J.CaM1.6 and CEM cells with 5-Aza-2′-dc significantly promoted the expression of lag3 gene at mRNA and protein levels as well as the demethylation of CpG island shore.No significant differences with the expression of lag3 gene and the methylation of CpG island were observed in Jurkat E6-1 cells with or without 5-Aza-2′-dc stimulation.Con-clusion Methylation and demethylation of CpG island shore played important roles in regulating the tran-scription of lag3 gene.
5.Clinical effect on microwave coagulo-necrotic therapy combined with laparoscopic splenectomy in treatment of primary hepatocellular carcinoma complicated by hypersplenism

Objective To investigate the feasibility and safety of microwave coagulo-necrotic therapy (MCN) combined with laparoscopic splenectomy (LS) in the treatment of primary hepatocellular carcinoma (PHC) complicated by hypersplenism. Methods A retrospective analysis was performed for the clinical data of 22 PHC patients with hypersplenism who were admitted to Baoji Municipal People' s Hospital from January 2008 to December 2012 and underwent MCN combined with LS. Their clinical data were collected, and postoperative complications and survival were observed. The t-test was used for comparison of continuous data, and the Kaplan-Meier method was used to plot cumulative survival curves. Results In MCN therapy, 5 patients underwent thoracotomy, 15 underwent laparotomy, and 2 underwent laparoscopic surgery; in LS treatment, 17 underwent hand-assisted LS and 5 underwent simple LS. The mean time of operation was (303. 24 ±56. 02) min, the mean blood loss volume was (146. 92 ± 60. 72) ml, and the mean weight of the resected spleen was (670. 42 ± 204. 54) g. Of all patients, 6 (27. 27% ) experienced postoperative recurrence, 5 (22. 73% ) experienced portal vein thrombosis, and 2 (9. 09% ) experienced pleural effusion. There was a significant increase in platelet count at 1 month after surgery [ (15. 72 ± 5. 47) ×104/μl vs (4. 43 ±1. 03) ×104/μl, t = 6. 83, P < 0. 001]. The 1-, 3-, and 5-year overall survival rates after surgery were 90. 91%, 68. 18%, and 59. 09%, respectively, and the 1-, 3-, and 5-year disease-free survival rates were 59. 09%, 13. 64%, and 13. 64%, respectively.Conclusion MCN combined with LS is safe, effective, and feasible in the treatment of PHC complicated by hypersplenism.

cFos is one of the most widely-studied genes in the field of neuroscience. Currently, there is no systematic database focusing on cFos in neuroscience. We developed a curated database—cFos-ANAB—a cFos-based web tool for exploring activated neurons and associated behaviors in rats and mice, comprising 398 brain nuclei and sub-nuclei, and five associated behaviors: pain, fear, feeding, aggression, and sexual behavior. Direct relationships among behaviors and nuclei (even cell types) under specific stimulating conditions were constructed based on cFos expression profiles extracted from original publications. Moreover, overlapping nuclei and sub-nuclei with potentially complex functions among different associated behaviors were emphasized, leading to results serving as important clues to the development of valid hypotheses for exploring as yet unknown circuits. Using the analysis function of cFos-ANAB, multi-layered pictures of networks and their relationships can quickly be explored depending on users’ purposes. These features provide a useful tool and good reference for early exploration in neuroscience. The cFos-ANAB database is available at www.cfos-db.net.
