Objective To characterize the human cell mediated immune response to Treponema pallidum(TP), and to determine the effect of TP infection on the various lymphocyte subpopulations and natural killer cells in the peripheral blood of the patients. Methods Twenty four untreated patients with early syphilis were analyzed, including 3 groups of 8 patients with disseminated syphilides, palmoplantar syphilides and latent syphilis each. Monoclonal antibodies of CD labeled with either fluorescein isothiocyanate or phycoerythrin were used on a flow cytometry to detect the changing of peripheral blood lymphocyte immunophenotypes. Results The results showed that the percentages of panT,T H,NK cells and the ratio of T H/T S were significantly lower (P0.05). Conclusion It suggests that there is prominent suppression of cellular immunity in the untreated patients suffering from early syphilis. Meanwhile, the difference of 3 groups in skin lesions was due to the different dermatobarrier function against the systemic invasion of TP, rather than attributed to the alteration of the lymphocyte immunophenotypes in the peripheral blood.

OBJECTIVE:To systematically review the efficacy, safety and economy of acarbose versus metformin in the treat-ment of type 2 diabetes, and provide evidence-based reference for the clinical treatment. METHODS:Retrieved from PubMed, Medline,CJFD,Wanfang database,VIP database, randomized controlled trails (RCT) about acarbose (test group) versus metfor-min(control group)in the treatment of type 2 diabetes were collected. Meta-analysis was performed by using Rev Man 5.2 statis-tics software, and the decision tree model was used to do the cost-effectiveness analysis by using TreeAge Pro 2011.1.0.12.1 soft-ware. RESULTS:A total of 8 RCT,involving 418 patients. Results of Meta-analysis showed 2 h postprandial blood glucose (2 h PG)in test group [MD=-2.21,95%CI(-2.92,-1.51),P<0.001] was lower than that of control group, there was no signifi-cant diffcrencc in the glycated hemoglobin levels[MD=0.02,95%CI(-0.38,0.34),P=0.91],fasting blood glucose level[MD=0.05,95%CI(0.91,1.01),P=0.92] and incidence of adverse reactions [OR=1.84,95%CI(0.80,4.24),P=0.92] between 2 groups. Results of decision tree analysis showed the cost-effectiveness ratio in test group and control group was 847.15 and 272.53,respec-tively;and incremental cost-effectiveness ratio was 13 776. CONCLUSIONS:Acarbose shows an obvious advantage on decreasing the 2 h PG of type 2 diabetes, however, pharmacoeconomics shows metformin has higher economic effects. Due to the limit of methodological quality,large-scale and high quality RCT are required for further validation of the conclusions.

Objective To construct and identify the recombinant retroviral vectors expressing siRNA for MDR1 gene knock-down,sequentially to detect the reversing effect of RNAi on the multidrug resistant phenotype in MCF-7/ADR cells. Methods The target sequences of siRNAs for MDR1 knock-down were designed by IDTdna online. The DNA sequences containing a palindrome structure were synthesized and annealed to form double-stands which were inserted into linear pSUPER vectors later. The recombinant vectors were identified by agarose gel electrophoresis after cleaving of double enzymes and by sequencing, then were transfected into 293T packaging cells which produced retroviral particles in supernatant, the retrovirus soup was used to infect MCF-7/ADR cells. The expression level of MDR1 was detected from both mRNA level and protein level by RT-PCR and Western blotting, respectively. Results The recombinant pSUPER vectors expressing siRNA were confirmed by agarose gel electrophoresis after cleaving of double enzymes and by sequence analysis. The inserted oligonucleatide sequences were identical to the original sequences and in corresponding position. Compared with control group, the inhibitory effects of pSUPER.retro-MDR1 and pSUPER.retro-MDR2 were significant. Conclusion pSUPER recombinant retroviral vectors expressing siRNA for MDR1 knock-down is construted successfully, and it has inhibitory effect on the expression of MDR1 gene in MCF-7/ADR cells.

Virologica Sinica 2008;23(5):369-377
The envelope gene gp85 of ev/J,a new family of endogenous avian retroviral sequences identified recently, has the most extensive nucleotide sequence identity ever described with ALV-J avian ieukosis virus. This report described expression of ev/J envelope gene gp85 derived from commercial meat-type chicken using the Invitrogen Bac-to-Bac baculovirus expression system. The antigenicity and immunoreactivity of the recombinant endogenous gp85 gene product (SU) were analyzed by indirect immunofluorescence, Western blot, indirect and blocking Enzyme-Linked ImmunoSorbent Assay (ELISA) using JE9 monoclonal antibody (MAb) against the envelope protein of ALV-J (ADOL-4817), positive mouse antiserum against the ev/J gp85 SU and sera from chicken naturally infected with ALV-J. The results showed that the ev/J gp85 SU can bind specifically to JE9 MAb and antiserum from chicken naturally infected with ALV-J, and the binding reactivity between exogenous ALV-J gp85 SU and natural positive chicken serum against exogenous ALV-J can be blocked by positive mouse serum against the ev/J gp85 SU. It is concluded that recombinant endogenous gp85 gene product (SU) has close immunological relatedness to the envelope protein of exogenous ALV-J (ADOL-4817 and IMC<,10200> strain).
