Objective To observe the effect of Exendin-4 on the apoptosis of cardiomyocyte induced by H2O2,and approach the relationship between GLP-1 signal pathway and the injury of cardiomyocyte. Methods Cardiomyocytes were isolated and cultured, and were divided into 5 groups.Intercelluar ROS (reactive oxygen species) was measured, and cell apoptosis rate was evaluated by Flow cytometry in different groups. Also expressions of apoptosis-associated proteins (caspase-3) and PI3 K/AKT were evaluated by western blot. Results Compared with H2O2 group,Ex-4 co-incubation decreased the production of intercelluar ROS levels, also improved the cardiomyocyte apoptosis. At the same time,Ex-4 resulted in the alterations in expressions of the caspase-3 and p-AKT/AKT proteins. However, these effects of Exendin-4 were counteracted significantly by the co-incubation of LY294002.Conclusion The interventions of GLP-1 signal pathway can improve cardiomyocyte apoptosis induced by H2O2 incubation, and the mechanisms might partly attribute to the PI3K/AKT system.

Clinical Medicine of China 2008;24(6):554-556
Objective To study the clinical characteristics of acute non-ST segment elevation myocardial infarction(NSTEMI).Methods 211 patients of NSTEMI and STEMI underwent coronary artery angiography and echocardiogram.Patients'history and symptome were collected and the data of coronary artery angiography and echocardiogram were analyzed. Results Compared with STEMI,NSTEMI patients had more risk factors and postinfarction angina pectoris;severe coronary artery disease and three coronary vessel disease. But NSTEMI had relatively little effects on cardiac function. Conclusion NSTEMI always has more severe coronary artery disease and postinfarction ischemic effects. So more attention should be paid to its standard therapy.

Objective To observe the changes in expressions of spleen regulatory T cells (Tregs)and the related factor forkhead box protein-3 (Foxp3) after irradiation with different doses of X-ray in mice at different times, and to elaborate the effects of X-rays on regulatory T cells and Foxp3.Methods 112male ICR mice were randomly divided into 2 groups and irradiated by X-rays at the doses of 0.075 and 2 Gy,respectively. The mice were killed at 0,4,8,16,24,48,and 72 h post-irradiation and the spleens removed.Flow cytometry was used to detect the percentage of CD4 + CD25 + Treg and protein expression of (Foxp3),and RT-PCR was used to exmiamine the mRNA expression of Fox3.Results Compared with those before irradiation, the CD4 + CD25 + Treg positive rates began to increase and peaked at 8 h post-irradiation with 0.075 Gy at 8,16,24,72 h(t = 8.73,10.55,4.21,4.65 ,P < 0.05) and 2 Gy at 8,16,48,72 h(t = 4.65,4.28,3.71,2.88,P < 0.05),and then slightly decreased, but still remained at high levels. The mRNA protein levels of Fox3 did not change significantly after exposure to the dose of 0.075 Gy,but began to significantly increase at 8 h after exposure to the dose of 2 Gy.However, the Foxp3 protein level began to increase 4 h post-irradiation,peaked at 16 h,and then slightly decreased, but still ramained at high levels (t =2.59,3.37,3.70,3.20,P<0.05).Conclusions The changes in expressions of Tregs and Foxp3 after high- and low-dose X-ray irradiation may be used to explain the differences in immune effects induced by ionizing radiation at different doses.

Objective To observe the features of juvenile obesity and its effect on glycometabolism and lipid metabolism. Methods A total of 194 students aged from 12 to 18 were selected for our study. The assessment included serum lipid concentration, a 75 g OGTT and insulin releasing test. Blood glucose, insulin and the blood lipids were measured. Results Compared with the control group, blood pressure, serum Fins (fasting insulin) and Pins 2h (2-hour postprandial insulin) in the obesity group were higher and were statistically distinguished by the t-test (P

OBJECTIVE:To study the compatible stability of puerarin injection with 10 commonly-used drugs.METHODS:The changes in appearance,pH,particulate and puerarin content after mixing with 10 drugs were observed and assayed.RESU_LTS & CONCLUSION:Puerarin injection mixing with adenosine triphosphate, coenzyme A and ribavirin showed significant change in pH,which indicats that they are incompatable each other, however, when puerarin injection was mixed with vitamin B6,potassium chloride, etamsylate, cimetidine, gentamycin, amikacin and erythromycin,no significant changes in pH,particulate and puerarin content were found, which indicates their compatibility.

Objective To explore colloidal gold method used to detect fecal occult blood tests(FOB)detection capability and establish the laboratory standard operation of detecting FOB limit of blank(LOB),limit of detection (LOD)and quantifica-tion limit (LOQ)according to the CLSI document《Evaluation of Detection Capability for Clinical Laboratory Measurement Procedures;Approved Guideline-Second Edition》(EP17-A2),in order to reduce the false negative rate of the weakly positive samples, and to provide a way of quantitative detection for qualitative detection of colloidal gold method. Methods Detected series of solution of hemoglobin made of dissolved fresh whole blood with the ELISA kit of human free hemoglobin, and es-tablished the standard curve of detection of FOB with colloidal gold method.Detected the blank samples and a series of low concentration samples with the colloidal gold test strip of FOB and measured the color bands by the Nato Checker710.The quantitative results obtained were statistically analysised by SPSS 1 9.0 and calculated blank limit, detection limit and quanti-fication limit. Results The LOB,LOD and LOD were 99.01,340.48 and 354.9 ng/ml according to the methods in CLSI EP1 7-A2 ducument. Conclusion The detection limits established by CLSI EP1 7-A2 document was more scientific in j udge-ment positive or negative to FOB than which used naked eye and can meet the clinical laboratory and clinical doctor require-ment better.Clinical laboratories should be strictly in detection limits of reagents in order to ensure their effectiveness, and should be generaly to other tests based on colloidal gold method.

Objective To study the clinical significance of end-tidal carbon dioxide partial pressure (PetCO2 ) during cardiopulmonary resuscitation (CPR) and seek the fixed value according which to decide if we should and when to give up.Methods This was a prospective, observational study.A total of 124 patients with cardiac arrest in or out-of-hospital from may 2003 to March 2009 in emergency department of our hospital were selected. All of them had definite etiological factors.Changes of PetCO2 in 124 cardiac arrest patients during CPR were tracked. Results The gender, age, rescue time in seventy-one patients with the return of spontaneous circulation (ROSC) after endotracheal intubation have a significant difference with that in fifty-three patients without ROSC (P <0.01 ).The PetCO2 of the survival were higher than that of patients without ROSC or with ROSC,but finally died (P <0.01 ).A fixed point 14.4 mmHg of PetCO2 after 20 minutes' CPR can be used as a reference value to guide CPR or predict prognosis. Conclusions Monitoring PetCO2 during CPR has a predictive value on the success of resuscitation.
