1.Factors responsible for nursing personnel's occupational injuries and maintenance of their health

It is pointed out that nursing personnel at the clinical forefront are a high-risk group for occupational injuries. The risk factors include: lack of knowledge on occupational injuries and their prevention in elementary and continuing education; various factors affecting nursing personnel's physical health, such as drug-dissolving facilities for the special purpose of chemotherapy, imperfect clean work station or relatively hermetical drug-dissolving environment ; increase of diseases that could be easily contracted in the hospital, such as hepatitis B, with the stabbing of nursing personnel by sharp medical instruments infected by blood or saliva being the main route of their occupational infection. The disadvantageous impact of occupation on their health puts nursing personnel under great psychological pressure. In order to safeguard their physical and mental health, it is imperative to put knowledge on hospital infection, occupational injuries and health maintenance on the curriculum of medical colleges and universities, reinforce nursing personnel's training in occupational safety, and take measures to prevent occupational injuries, such as using disposable gloves when dispensing medicines in chemotherapeutic drug centers.
2.The curative effect of triple lumen feeding tube on upper gastrointestinal tract perforation after en- doscopy

Objective To explore the curative effect of triple lumen feeding tube on upper gastroin-testinal tract perforation after endoscopic operation.Methods A total of 1 12 patients with upper gastrointes-tinal perforation after endoscopic operation were involved in this prospective control test.The remission rate of abdominal pain and abdominal distension,gastric pH,lesion healing time,length of stay and the success rate of conservative treatment were compared between the control group (indwelling traditional gastric tube and intravenous use of omeprazole injection)and the experimental group (intestinal administration of PPI through triple lumen feeding tube).Results The remission rate of abdominal pain of the experimental group was 61. 3% at 6 hours,and 83. 2% at 12 h,which were significantly higher than those of the control group (P<0. 05 ).The lesion healing time(of cases where lesions were not clipped)and length of hospitalization of the experimental group were significantly shorter than those of the control group(P<0. 05 ).The gastric pH and success rate of conservative treatment of the two groups had no significant difference(P>0. 05 ).Con-clusion The curative effect of triple lumen feeding tube on upper gastrointestinal tract perforation after endo-scopic operation is ideal.
3.Construction of recombinant adenovirus vector for human matrix metalloproteinase-1 gene and detection of collagen type III degradation in vitro

BACKGROUND:Matrix metaloproteinase-1 can degrade extracelular matrix, which is mainly colagen type I, and has the potential to reverse fibrosis tissue. OBJECTIVE:To construct the recombinant adenovirus vector containing human matrix metaloproteinase-1 (hMMP-1) gene with GatewayTM Clone Technology, and observe the capacity of degrading colagen type IIIin vitro. METHODS: The gene hMMP-1 was amplified by using PCR from the pcDNA3.1 plasmid and was cut down by the double endonuclease. The linear gene fragment was connected to the entry vector pENTERTM 1A. Then the entry clone and the destination vectors pJTI? R4 Dest CMV-N-EmGFP pA Vector recombined using the LR reaction to form the expression clone pAd-hMMP-1-eGFP. The linear pAd-hMMP-1-eGFP cut down by endonucleasePac I was transfected into HEK293A cels to packaging the Ad-hMMP-1-eGFP. The transfected situation was observed under a fluorescence microscope, the target protein expression was detected by western-blot assay and RT-PCR. Cels can be divided into three groups: blank control group: HEK293A cels, AD-EGFP group: HEK293A cels were infected by Ad-eGFP, AD-HMMP1-EGF group: HEK293A cels were infected by Ad-hMMP1-eGFP and colagen type III. The content of colagen type III was detected by ELISA kits after 24, 48 and 72 hours. RESULTS AND CONCLUSION: It was confirmed that the entry vector and the destination vector both contained hMMP-1 target gene by restriction analysis and sequencing. The green fluorescent protein was observed in the 293A cels transfected by the Ad-hMMP-1-eGFP at 4 days. The fluorescence intensity was the highest at 10 days. The virus was colected at 12 days, the viral titer was determined as 4.84 × 1010 PFU/mL, the target protein was efficient expressionvia western-blot assay. Blank control group and AD-EGFP group had no obvious change of colagen content with the extension of time. The rate of colagen degradation in AD-HMMP1-EGFP group was 24%, 56% and 81% respectively at 24, 48, 72 hours. AD-HMMP1-EGFP group degraded colagen significantly compared with the other two groups (P < 0.01). The recombinant adenovirus vector containing hMMP-1 was successfuly constructed by using the Gateway technology, this method was more efficient and specific than with the traditional methods. The hMMP1 degraded colagen type III significantlyin vitro.
4.The study of ghrelin and obestatin system in patients with essential hypertension

Objective To measure the fasting plasma ghrelin and obestatin concentrations in essential hyper-tension (EH)patients with obesity,and to observe their relationship with blood pressure,obesity index,insulin resist-ance (IR),blood glucose and blood lipid.Methods 68 hypertensive obese patients,60 normotensive obese patients and 65 healthy controls were included in the study.The fasting plasma obestatin and ghrelin were measured using a radioimmunoassay method.The ghrelin/obestatin ratio was calculated.Results Hypertensive obese patients had lower plasma ghrelin and obestatin compared with normotensive obese patients (t =3.771,P <0.01;t =4.373,P <0.01) and controls (t =16.451,P <0.01;t =17.862,P <0.01).Normotensive obese patients had lower plasma ghrelin and obestatin compared with controls (t =13.121,P <0.01;t =13.686,P <0.01 ).Hypertensive obese patients had higher ghrelin/obestatin ratio than controls (t =7.720,P <0.01).The ghrelin/obestatin ratio in normotensive obese patients was higher than controls (t =4.587,P <0.01).In hypertensive obese group,the plasma ghrelin and obestatin were negatively associated with BMI (r =-0.882,P <0.01;r =-0.806,P <0.01),SBP (r =-0.787,P <0.01;r =-0.837,P <0.01),DBP (r =-0.769,P <0.01;r =-0.810,P <0.01),and the HOMA-IR (r =-0.800, P <0.01;r =-0.810,P <0.01).In normotensive obese group,ghrelin and obestatin were all negatively associated with BMI (r =-0.577,P <0.01;r =-0.372,P <0.01)and HOMA-IR (r =-0.866,P <0.01;r =-0.662,P <0.01).The ghrelin/obestatin ratio was positively associated with BMI (r =0.460,P <0.01)and HOMA-IR (r =0.420,P <0.01).Conclusion The peripheral blood ghrelin,obestatin and ghrelin/obestatin ratio were significantly correlated with EH,obesity and IR.No correlation was observed between ghrelin/obestatin system and blood lipid or blood glucose.
5.Detection of Pathogens of Urogenital Infections and Their Drug-resistant Types by a DNA Chip

Conclusions This DNA chip combined with multiplex PCR is a rapid diagnostic assay with high specificity and sensitivity for the detection of Neisseria gonorrhoeae, Chlamydia trachomatis and Ureaplasma Urealyticum and their drug-resistance, and may be applied in the diagnosis of urogenital infections.
6.Therapeutic effect of BMSCs with over-expressed MMP1 on liver fibrosis.

OBJECTIVE:
To investigate the function of bone marrow mesenchymal stem cells (BMSCs) with over-expressed matrix metalloproteinase 1 (MMP1) on liver fibrosis.
METHODS:
Fifty SD male rats were randomly divided into 4 groups: recombinant adenovirus Adhuman MMP-1(hMMP-1)-enhanced green fluorescent protein (EGFP) transfected BMSCs group (Group A, n=10), Ad-EGFP transfected BMSCs group (Group B, n=10), liver fibrosis group (Group C, n=15), and a normal group (Group D, n=15). The liver fibrosis model was formed by subcutaneous injection of the mixed liquor of carbon tetrachloride (CCL4) and vegetable oil. After 10 weeks, the model of liver fibrosis was formed. Group A and B were administered the transfected BMSCs via the tail veins, while Group C and D were administered normal saline. After 3 weeks, the rats were sacrificed. The body weight, liver weight, liver function, liver fibrosis indexes and liver pathological changes were tested.
RESULTS:
Compared with the control group, the rats administered BMSCs with over-expressed MMP1 showed a significant improvement in the body weight, liver weight and plasma albumin (ALB) (P<0.05), and a significant reduction in the plasma alanine aminotransferase, total bilirubin, hyaluronic acid, laminin and procollagen III (P<0.05). Hematoxylin-eosin staining confirmed that the degree of liver fibrosis was significantly ameliorated under average visual fields (P<0.05).
CONCLUSION
The repair ability of BMSCs on liver fibrosis can be enhanced by over-expression of hMMP-1.

7.Effect of Anchusa Italica Retz on Cerebral Ischemia-reperfusion Injury in Rats Based on Disease and Active Compound Target Network

Objective To study the effect and mechanism of Anchusa italica Retz on cerebral ischemia-reperfusion inju-ry in rats based on the target network of active compounds in Anchusa italica Retz.Methods The rat model of cerebral ische-mia-reperfusion injury was established by the thread occlusion method.After performing ischemia for 1.5 h and then reperfusion for 24 h,the neurological function of rats was scored and the volume of cerebral infarction was measured by the 2,3,5-triphenyltet-razolium chloride staining method.The molecular network analysis technique of network pharmacology,protein-protein interaction network,gene ontology(GO)enrichment analysis,KEGG signal pathway analysis,and molecular docking was used to study the mechanism of Anchusa italica Retz in the treatment of cerebral ischemia-reperfusion injury.Results The administration of An-chusa italica Retz could significantly improve the neurobehavioral dysfunction caused by cerebral ischemia-reperfusion injury and reduce the pathological injury of brain tissue.Anchusa italica Retz could regulate inflammation,apoptosis,protein phosphorylation,and other biological processes through 143 ischemic stroke-related targets,and interfere with the TNF signal pathway,VEGF signal pathway,HIF-1 signal pathway,and other pathways.Conclusion Network pharmacology and experimental verification had shown that Anchusa italica Retz could effectively reduce brain injury and protect neurological function through multi-target,multi-mechanism,and holistic treatment of cerebral ischemia-reperfusion injury.
8.Dynamic changes of lymphocytes and the disease progression in patients with coronavirus disease 2019 in Xi’an

【Objective】 To analyze the clinical characteristics of patients with coronavirus disease 2019 (COVID-19) in Xi’an so as to investigate the relationship between the dynamic changes of lymphocytes and the disease progression. 【Methods】 We retrospectively analyzed the clinical data of 15 patients with COVID-19 in The First Affiliated Hospital of Xi’an Jiaotong University from January 22 to February 16, 2020. 【Results】 Among the 15 patients with COVID-19, 8 were males and 7 were females, aged from 22 to 89 years. There were 12 ordinary cases (80%), 1 severe case (6.67%), and 2 critical cases(13.33%). There were 6 groups of family clusters.Most of the patients (14/15, 93.3%) had fever of different degrees. The average time from illness onset to admission was 2.80±1.66 days, and the average time from illness onset to diagnosis was 2.83±2.29 days. The main accompanying symptoms were dry cough (8/15, 53.33%) and shortness of breath (4/15, 26.67%). Nine patients (60%) who had low lymphocyte counts at admission, including of all of the critically ill patients (1 severe case and 2 critical cases) and 6 (6/12, 50%) ordinary patients. Lymphocyte counts in the ordinary cases increased gradually, but fluctuated in the severely ill patients. They were always at low level, or even decreased overall in critical cases. 【Conclusion】 In Xi’an City, COVID-19 mostly occurred in family clusters. Lymphocyte counts were reduced in most patients, especially in critically ill (severe and critical) ones. The lymphocyte count at admission and its kinetics during therapy may be an important predictor for the severity and prognosis of the disease.
9.Clinical data analysis of 2491 acupuncture inpatient consultations: a report from Qilu Hospital of Shandong University.

OBJECTIVETo summarize the 2491 acupuncture inpatient consultations in Qilu Hospital of Shandong University during 2014, and analyze the application of acupuncture in comprehensive hospital, hoping to provide direction and methods for clinical practice and scientific research of acupuncture.
METHODSThe consultation record and hospitalization information during 2014 in Department of Acupuncture-Moxibustion and Tuina, Qilu Hospital of Shandong University, were collected and summarized through Inpatient Information Management System. The distribution of consultation discipline and disease were analyzed.
RESULTSAmong all the consultations, 15 second-class disciplines were involved, and the number of neurology and surgery was 1399, accounting for 59.00% of all consultation. 111 types of diseases were found in consultation, mainly postoperative complications, cerebrovascular disease and arthropathy. The primary disease of consultations involved all the systems, mainly nervous system disease, musculoskeletal system disease, connective tissue diseases and genitourinary system diseases, accounting for 73.89% of all consultations. .
CONCLUSIONAcupuncture is closely related with clinical disciplines of neurology and surgery. The cooperation between acupuncture and pediatrics and oncology should be strengthened. Acupuncture has an advantage over treating postoperative complications and neuromuscular diseases.
