1.Evaluation of left atrial function serial change after circumferential pulmonary vein ablation for paroxysmal atrial fibrillation by tissue Doppler echocardiography

Objective To evaluate left atrial function serial change after circumferential pulmonary vein ablation(CPVA)for paroxysmal atrial fibrillation using tissue Doppler echocardiography.Methods One hundred and eight patients with paroxysmal atrial fibrillation underwent CPVA guided by CARTO.Tissue Doppler imaging and conventional echocardiography were underwent 48 h before CPVA,48 h,1 month,3 month and 6 month after CPVA.Results One hundred and six patients with paroxysmal atrial fibrillation finished CPVA successfully.Compared tO 48 h pre-CPVA,left atrial diameter and volume decreased 48 h,1 month,3 month,6 month after CPVA,significant change were found at 3 month,6 month after CPVA(P<0.05).Left ventricular end-diastolic diameter(LVEDD),left ventricular end-systolic diameter(LVESD).left ventricular ejection fraction(LVEF),mitral flow E revealed no significant change(P>0.05).Mitral flow A decreased significantly 48 h after CPVA(P<0.05),restored significantly after 3 month.Tissue Doppler parameter S'and E'revealed no significant change(P>0.05)through the period,A'decreased significantly 48 h after CPVA(P<0.05),restored significantlv after 1 month.Conclusions Left atrial diameter and volume decreased after CPVA.Left atrial active contraction function decreased shortly after CPVA(LA stunning),and restored progressively.
2.Correlation of P-gp,bcl-2,STAT3 and the resistance of lymphoma cells to chemotherapeutic agents

Objective To investigate the correlation of P-gP,bel-2,STAT3,IL-6,IL-10 and the resistance lymphoma cells to chemotherapeutic agents.Methods There were 28 patients,among them were divided into three groups:refractory lymphoma,lymphoma which is sensitive to agents and lymphodenitis.P-glycoprotein(P-gp)on lymphoma cell membrane,STAT3,bcl-2,IL-6 and IL-10 level into lymphonla cell were detected using FCM,and the correlation between them and chemotherapy efficacy were analysed. Results In refractory lymphoma patients,P-gp and hcl-2 are significant higher than that of the group which is sensitive to agents(P:0.01,P=0.039),but STAT3,IL-6 and IL-10 were not significant different between these two groups(P>o.05).P-gp is significant higher in lymphoma than in lymphodenitis(P=0.01).STAT3 in lymphoma is significant lower than that of in lymphodenitis (P=0.04).The level bcl-2,IL-6 and IL-10 between lymphoma and lymphodenitis are not significant different(P>0.05).Conclusion The expression level of bel-2 and p-gP is correlated to resistance to the chemotherapeutic agents in lymphoma cells.STAT3 play a role in lymphoma cell signal transduction,but it is not certain in lymphoma cell muhidrug resistance.
3.The influence of umbilical cord milking on important clinical outcomes of preterm infants

Placental blood transfusion can be achieved by delayed cord clamping and umbilical cord milking.The World Health Organization has proposed delayed cord clamping as the standard care for newborns that do not require resuscitation.In recent years, umbilical cord milking has become an alternative to delayed cord clamping due to the short duration, quick recovery of the fetus after delivery, and similar effect on placental blood transfusion to that of the delayed cord clamping.This study aims to summarize the research results of the two intervention methods, and their potential benefits and risks, aiming to provide references for the optimal umbilical cord cutting.
4.Effect of E54 mutation of human secreted phospholipase A2 GIIE on substrate selectivity.

Human secreted phospholipase A2 GIIE (hGIIE) is involved in inflammation and lipid metabolism due to its ability of hydrolyzing phospholipids. To reveal the mechanism of substrate head-group selectivity, we analyzed the effect of mutation of hGIIE on its activity and selectivity. hGIIE structural analysis showed that E54 might be related to its substrate head-group selectivity. According to the sequence alignment, E54 was mutated to alanine, phenylalanine, and lysine. Mutated genes were cloned and expressed in Pichia pastoris X33, and the enzymes with mutations were purified with 90% purity by ion exchange and molecular size exclusion chromatography. The enzymatic activities were determined by isothermal microthermal titration method. The Km of mutant E54K towards 1,2-dihexyl phosphate glycerol decreased by 0.39-fold compared with that of wild type hGIIE (WT), and the Km of E54F towards 1,2-dihexanoyl-sn-glycero-3-phosphocholine increased by 1.93-fold than that of WT. The affinity of mutant proteins with phospholipid substrate was significantly changed, indicating that E54 plays an important role in the substrate head-group selectivity of hGIIE.
