Objective To investigate whether pharmacological serum from the rats taking radix salviae miltiorrhizae (RSM) can inhibit increase of intracellular free calcium level in hepatic stellate cells (HSCs). Methods A total of 32 healthy SD rats were divided into 4 groups. Two groups were modeled into hepatic fibrosis by 40% CCl 4 injection subcutaneously for 9 weeks and RSM were filled into the stomachs of one group (group A) but the same volume of saline into the other group (group B) twice a day for 6 d. Another two groups were only filled with RSM (group C) and saline (group D) for 6 d without modeling into hepatic fibrosis. The pharmacological serum was drawn from the inferior vena cava at 2 h after the last time of RSM or saline and used to culture HSCs at the concentration of 10% for 24 h. After HSCs were loaded with Fluo-3/AM, the effects of RSM on [Ca 2+ ] i with laser scanning confocal microscopy were examined before and after angiotensin Ⅱ treatment. Results In the activated HSCs of groups A and C, [Ca 2+ ] i decreased significantly as compared with groups B and D (P

AIM: To observe whether metallothionein plays a role in cardiac protective effect of basic fibroblast growth factor on anoxic/reperfusion (A/R) injury in cultured cardiomyocytes and study the possible mechanism of cardiac protection by bFGF.METHODS: The present study made the model of myocyte A/R injury after having a 24 h incubation by bFGF( 10-10、10-9、10-s mol/L) and bFGF( 10-9 mol/L) + PD098059 respectively. We measured the levels of MT and MDA in myocytes, and the changes of LDH and protein in cultured medium. We also counted the number of viable cell in groups. RESULTS: The contents of myocardial MT were significantly increased after treatment by bFGF. The levels of MT in I0-l0 mol/L、10-9 mol/L and 10-8 mol/L bFGF treated groups increased 54 %、 62%、 76% respectively, compared with the A/R group, and the number of viable cell were also greatly increased, LDH and protein leakage in cultured medium and MDA contents in myocyte were dramatically decreased in bFGF treated groups. All the protection were completely disappeared with the inhibition of MT production with PD098059, theinhibitor of mitogen- activated protein kinase(MAPK). CONCLUSION: MT involves in the protection of bFGF in cultured cardiomyocytes. It might be related with activation of MAPKase.

Objective To analyze the clinical features in multiple myeloma(MM)patients, and the summaries the treatment effect of different therapies. Methods The clinical initial symptoms, laboratory data, clinical efficacy of treatment with MP, M2, VAD, BD,T hal + DEX and other chemotherapy in 120 patients newly diagnosed with multiple myeloma were retrospectively analyzed. Results Among 120 MM cases, the incidence peak was situated in 40 - 60 years age group, the mean age at oneset was 54 years, which indicated that the onset age were getting younger in China. Bone pain, anemia, infection were the most common initial symptoms of MM. In terms of clinical efficacy:①Significant difference between MP and VAD programs(χ2 = 10. 626,P <0. 05=,but no significant difference between MP and M2 programs(χ2 = 2. 439, P > 0. 05), MP and Thal + DEX programs(χ2 = 5. 870, P >0. 05. ②Significant difference between M2 and VAD programs(χ2 = 8. 307, P < 0. 05=; No significant difference between M2 and Thal + DEX programs(χ2 = 2. 592, P > 0. 05). ③ No significant difference between VAD and Thal +DEX programs(χ2 =0. 916,P> 0. 05). ④BD program is the most efficient program, but this discovery need further confirmation because of the small number of cases observed. Conclusions At present, chemotherapy is the main treatment for MM, most scheme like MP, M2, VAD, BD, etc. , have certain effect depend on different patient groups but remain improving. The choice of appropriate chemotherapy and autologous or allogeneic hematopoietic stem cell transplantation will help to improve the prognosis of the disease.

Objective To observe the transport of hepatitis B virus (HBV)-infected peripheral blood mononuclear cells (PBMC)through placental barrier set up by choriocarcinoma trophoblast cells (Bewo cells),and to explore the biological role of PBMC as a carrier for HBV transport. Methods Bewo cells and PBMC were cultured and their proliferation and activity were detected by cell counting kit (CCK)-8.One hundred μL serum containing 5 ×10 6 copy/mL HBV DNA was used to infect PBMC,and cells infected with HBV were labeled by fluorescent dye carboxyfluorescein diacetate succinimidyl ester (CFSE).A co-culture model of Bewo cells and HBV-infected PBMC was set up by transwell chamber. The migration of HBV-infected PBMC was detected by flow cytometry.Realtime fluorescence quantitative polymerase chain reaction method was used to detect HBV DNA contents of PBMC under transwell chamber. Results PBMC and Bewo cells proliferated at around 24 h and entered into growth stagnation at around 120 h.The contents of PBMC labeled by green fluorescent at 0,12,24 and 48 h during co-culture under chamber were (0.445 ±0.021)%,(21 .180 ± 4.653 )%,(34.830 ± 7.156 )% and (64.185 ± 3.161)%,respectively.The amount of PBMC marked green fluorescence increased over prolonged incubation time (F =68.983,P =0.001 ).PBMC HBV DNA contents at 24 and 48 h of co-culture under chamber were (1.925±0.431)×103 copy/mL and (2.565 ±0.361)×103 copy/mL,respectively,indicating that PBMC under chamber were infected with HBV.Conclusions PBMC may be a target for HBV infection in extrahepatic tissues.Placental trophoblastic barrier built by transwell chambers may provide new ideas to investigate HBV transmission across the placenta in vitro .
