Objective To study the association of peptidylarginine deiminase 4 (PADI4) (padi4_94 and padi4_104) genetic single nucleotide polymorphism (SNP) and rheumatoid arthritis (RA) susceptibility in Han population of Hebei province. Methods This hospital-based case-control study included 115 RA patients and 106 healthy controls. All the individuals were recruited from Han Hebei residents and were randomly selected. The genotype and allele frequencies of PADI4 gene polymorphisms (padi4_94 and padi4_104) were analyzed by PCR-DNA sequencing method. Results The distribution of padi4_94 and padi4_104 genotypes between the two groups was not significantly deviated from that expected by Hardy-Weinberg equilibrium (P>0.05). The combined effect of padi4_94 and padi4_104 SNPs was analyzed and three haplotypos (AA, AG and GG) were found but without GA haplotype. The genotype and allele distribution of padi4_94 and padi4_104 in the patients with RA was not significantly different from that in healthy controls, and the analysis of haplotypes revealed that no haplotypes were risk factors for RA. Conclusion In Han population of Hebei province, the SNPs of PADI4 (padi4_94 or/and padi4_104) is no associated with RA susceptibility. Therefore, it should not be regarded as a genetic risk factor for RA.

Objective To study the effect of age and sex on physiological and biochemical indexes of SJ5-SPF chickens at the age of different weeks. Methods Blood biochemical indexes of SJ5-SPF chickens at the age of different weeks were measured using an automatic blood analyzer, including 19 items: ALT, ALP, GGT, AST, LDH, TP, GLB, ALB, TB, DB, IBIL, GLU, TG, TCH, BUN, CRE, K+, Na+, and Ca2+. At the same time, the body temperature (T), respiratory frequency (R), heart rate (HR), diastolic pressure (DBP) and systolic pressure (SBP) of the SJ5-SPF chickens were measured using a RM6240C multi-channel physiological signal acquisition and processing system. Results (1) Among the physiological parameters, the body temperature, respiratory rate, heart rate, diastolic blood pressure and systolic blood pressure showed significant differences between the 4-week old chickens and the chickens at ages of 20, 25, and 25 weeks (P < 0. 05), but there were significant differences between the males and females at the same weeks of age except body temperature. The diastolic blood pressure was only significantly different in the 40-week old male and female chickens (P < 0. 01). The heart rates showed significant differences between the male and female chickens at 4, 20 and 25 weeks of age (P < 0. 01). The respiratory rate and systolic blood pressure in the 4-week old male and female chickens were significantly different (P < 0. 05). (2) The 19 blood biochemical indexes showed significant differences between the male and female chickens: the GGT, ALT, AST, TB, DB, IBIL, GLU, BUN and Na+at an week-age (P< 0. 05), the LDH, TP, GLB, ALB and CRE at two different week-ages (P< 0. 05), the ALP, TG and Ca2+at three different week-ages (P< 0. 05), and the TCH and K+at four different week-ages (P< 0. 05). Conclusions The above results can provide a scientific basis for diagnosis of SJ5-SPF chicken disease, animal quarantine and other related research.

Objective:
To investigate the clinical value of gallbladder-preserving cholelithotomy by natural orifice transumbilical endoscopic surgery on patients with cholecystolithiasis.
Methods:
A retrospective study was performed on data of 15 patients with cholecystolithiasis, who underwent gallbladder-preserving cholelithotomy by natural orifice transumbilical endoscopic surgery from April 2018 to July 2018. The operative data, including situation of operation, operative time, intraoperative hemorrhage, and postoperative complications were recorded.
Results:
The procedure was performed successfully in all patients with a mean operative time of 108±12 min (ranged from 92-129 min). The intraoperative hemorrhage was 10-30 mL. Eight patients suffered from slight right upper abdominal pain, and 7 patients felt slight pain in umbilical a week after surgery. No fever, incision infection, umbilical hemia, peritonitis, and ascites were reported. The clear-liquid diet was recommended for one day after operation, and postoperative activity was allowed since the second day after operation. All patients were discharged on the fourth or fifth day, and all recovered to their normal life at one week after discharge. Follow-up showed that the scar was small and hidden in umbilical without visible incision after one month. Ultrasonic examination results showed that gallbladder contractile function worked perfectly in four patients and no gallbladder stone was found after three months.
Conclusion
Gallbladder-preserving cholelithotomy by natural orifice transumbilical endoscopic surgery is a safe and effective option for patients with cholecystolithiasis, provides excellent cosmetic outcomes, and can be appropriately carried out under the strict control of surgical indications.
