1.Study on Susceptibility of Ureaplasma Urealyticum to Antimicrobial Agents and tetM Gene Detection

Objective To determine the susceptibility of Ureaplasma urealyticum(Uu) to 7 kinds of antimicrobial agents, to compare the result with tetracycline resistant determinant(tetM) detected by PCR technique.Methods Forty two Uu isolates were subcultured for 3 generations and then cultured with drugs with a series of concentrations from 64?g/ml ~ 0 06?g/ml. MIC was determined when the color of culture medium did not change from yellow to red in 72h culture. The tetM determinant in the isolates was detected by PCR with specific primers, and PCR products were digested by Taq 1 restrictive endonuclease. Results The antimicrobial activity of tetracyclines to Uu was low. No Uu strains resistant simultaneously to 3 groups of antibiotics were found in the study. Detection rate of tetM was 71 4% with correspondent digested fragments. Conclusion The susceptibility of Uu isolates to tetracycline group was low, but higher to macrolides and fluoroquinolones. Uu strains carrying tetM determinant might become drug resistant ones which should be monitored and followed up with MIC determination.

3.The value of transvaginal color doppler sonography combined with CA125 detection in the the diagnosis of ovarian of benign and malignant

Objective To investigate the applicative value of transvaginal color doppler sonography com-bined with CA125 which is tumor detection maker in the the diagnosis of ovarian of benign and malignant tumor . Methods We analyzed sonographic features ,blood flow of transvaginal color doppler sonography and serum CA 125 level in 102 ovarian tumor cases and compared with pathological findings and study the sensitivity ,specificity and accuracy of diagnosis.Results The sensitivity,specificity and accuracy of the diagnosis of ovarian of benign and malignant tumor detected by transvaginal color doppler sonography were respectively 80.6%,95.8%and 91.2%. The sensitivity ,specificity and accuracy of the diagnosis of ovarian of benign and malignant tumor detected by tumor makers CA125 were 87.1%,87.3%and 87.3%respectively.The sensitivity,specificity and accuracy of the diag-nosis of ovarian of benign and malignant detected by transvaginal color doppler sonography combined with tumor makers CA125 were 93.5%,97.2%and 96.1%respectively.The sensitivity,specificity and accuracy of results de-tected by the combination of both methods were higher than the single one .Conclusion The combination among the sonographic features ,blood flow of transvaginal color doppler sonography and serum tumor markers CA 125 im-proves the efficiency of ovarian benign and malignant tumor diagnosis ,and therefore shows the great clinical value .
4.Protective Effect of Yulangsan Polysaccharide on Acute Alcohol Hepatic Injury in Mice

OBJECTIVE:To study the protective effect of Yulangsan polysaccharide(YLSPS) on acute alcoholic hepatic injury in mice.METHODS:50% alcohol at a dose of 10 mL?kg-1 was administered (ig) to mice to establish acute alcoholic hepatic injury model.The activities of serum transaminase and superoxide dismutase(SOD) and the triglyeride(TGL) level were determined and a pathohistological study was performed. RESULTS:The activity of serum transaminase and the content of triglyceride in mice were significantly down-regulated by different doses of YLSPS,but the activity of superoxide dismutase in YLSPS-treated groups was significantly up-regulated and the number of fat droplets was reduced. Fatty degeneration in acute alcoholic hepatic injury model mice was significantly abated by high-dose and middle-dose YLSPS.CONCLUSION:The protective effect of YLSPS on alcohol-induced acute hepatic injury in mice was attributed to its action in lessening the effect of alcohol on lipometabolism.
5.Effects of Millettia pulchra Extracts on TNF-?、PGE_2、NO in Pleuritis Model Rats

OBJECTIVE: To investigate the effect of Millettia pulchra extracts on tumor necrosis factor ? (TNF-?), prostaglandin E2(PGE2) and nitric oxide (NO) in pleuritis model rats. METHODS: 56 Wistar male rats were randomly divided into control group, model group, dexamethasone (DEX) group, water extract of M.pulchra (TYLS) group (high dose and low dose) and total flavonoids of M.pulchra(FYLS) group (high dose and low dose). After preoperational intragastric administration for 7 days, the pleuritis model was induced by injecting carrageenan into pleural cavity in 30 minutes after the last medication. The amounts of pleurorrhea, leucocyte, TNF-?,PGE2 and NO in the pleurorrhea were measured at 8 hours after modeling. RESULTS: As compared with the model group, in TYLS and FYLS group the pleurorrhea volume, leukocyte amount, contents of TNF-? and PGE2 reduced markedly, but the synthesis of NO had little change.CONCLUSION:M.pulchra Extracts show a marked inhibitive effect on pleuritis. Their anti-inflammation effects may be related to inhibiting the increase of TNF-? and the infiltration and transmigration of leucocytes, but not associate with the synthesis of NO.
6.Preliminary Study on Molecular Subtyping of Treponema pallidum in Hen gyang and Jiangmen Regions

Objective To investigate the distribution of subtypes of T. pallidum (TP) in Hengyang and Jiangmen regions. Methods Eighty-five specime ns taken from patients with suspected chancre collected in Hengyang and Jiangmen from February 2002 to January 2004 were screened by a PCR targeted TP polA gene , and then the arp gene and tpr gene were amplified from TP positive specimens. The PCR products of tpr gene were digested by restriction endonuclease MSe I. Th e sizes of the arp gene and the restriction fragment length polymorphism (RFLP) of the tpr gene were analyzed for subtyping. Results Of 69 TP-positive specime ns, 57 could be subtyped, and 10 subtypes were found. Among them, 26 (45.6%) wer e subtype 14d, and other subtypes included 10d(1), 12a(3), 12g(2), 13d(6), 14a(5 ), 14b(2), 14f(6), 15d(5) and 16d(1). No significant difference of the distribut ion of TP subtypes between Hengyang and Jiangmen was found. Conclusion Multipl e T.pallidum subtypes have been prevalent in Hengyang and Jiangmen, although the predominant subtype is 14d, there is no significant geographic heterogeneity be tween these two regions.
7.Evaluation of in vitro cytotoxicity and drug release property on pharmaceutical dressing

Objective To evaluate the cellular toxic and release of pharmaceutical dressing.Method Following the State standard GB/T14233.2-2005.the L929 cellular merphology was observed by inveded microscopy after 72h and proliferation of the cells was examined using mitochondrial function(MTT)assay.Relative growth rate (RGR)was calculated and cytotoxicity grade was evaluated by absorbency(OD)data.With PBS7.4 as dissolution media,and(32±0.5)℃as dissolution temperature,the release rate was determined with UV method with the determination wavelength of 288 nm and the dissolved liquid in 1/6,1/2,1,3,16,24,36 and 48 h.Results The average cell RGR of the pharmaceutical dressing is 91.25%and reaches 1 cytotoxicity grade.L929 cellular morphology is normal.Pharmaceutical dressing release accord to Higuchi equation,and the simulated equation is M_t/M_∞=0.3271t~(0.239).Conclusions Biologic compatibility of the pharmaceutical dressing is good,and the release of levofloxacin from the pharmaceutical dressing is sustained in vitro.
8.Efficacy of wound dressing with microspheres containing levofloxacin on burns treatment.

This research was aimed to find the skin irritation and burns treatment effect of wound dressing with mi- crospheres containing levofloxacin. We used reference GB/T16886. 10-2005 to evaluate the dressing skin irritation. We prepared rabbit models divided into three groups. The control group was rapped with Vaseline gauze bandage, while the positive control group was rapped with the wounds of nano-silver paste bandage. The experimental sample group was rapped with wound dressing with microspheres containing levofloxacin. We measured the wound without healing area and the hydroxyproline content at the ends of 3 d, 6 d, 9 d, 14 d, 21 d, 28 d. and meanwhile performed histopathological examination. The experimental results showed that the dressing primary irritation index was 0. The nonhealing wound area of theexperimental sample group and positive control group at the ends of 6 d, 9 d, 14 d, 21 d were less than that of the control group (P<0. 05). The nonhealing wound area of the experimental sample group at the ends of 9 d and 14 d was significantly lower than that of the positive control group (P < 0.05). The hydroxyproline content of the experimental sample group at the ends of 6 d, 9 d and 14 d was significantly higher than that of the positive control group and blank control group (P<0. 05). The pathology observed of the experimental sample group at 21 d were the earliest appendages. The wound dressing with microspheres containing levofloxacin has minimal skin irritation, effectively promote wound healing of burn.

9.Analysis of risk factors of late preterm infants with hyperbilirubinemia

Objective To explore the risk factors of hyperbilirubinemia in late preterm infants. Methods Clinical data of 211 cases of late preterm infants with hyperbilirubinemia and 246 cases of late preterm infants without hyperbilirubinemia were retro-spectively analyzed between 2011 and 2012. The risk factors of hyperbilirubinemia were filtered. Results Twenty-seven cases of late premature infants with hyperbilirubinemia were severe. Hospital stay less than 3 days, birth asphyxia history, small for gestatio-nal age, head hematoma, delivery injury, hypoalbuminemia, polycythemia, infection, hemolytic disease, feeding intolerance, and fe-tal excretion delay were associated with hyperbilirubinemia (P<0.05). Rural origin, pregnancy-induced hypertension syndrome and premature rupture of membrane were also associated with hyperbilirubinemia (P<0.05). Multivariate logistic regression analysis showed the history of birth asphyxia , fetal excretion delay, hypoalbuminemia, pregnancy-induced hypertension syndrome were risk factors of hyperbilirubinemia in late preterm infants (OR=2.35-4.05). Pregnancy-induced hypertension syndrome and hemolytic dis-ease were risk factors of severe hyperbilirubinemia in late preterm infants (OR=5.74, 73.64). Conclusions Neonatal asphyxia, fetal excretion delay, hypoalbuminemia and pregnancy-induced hypertension syndrome are risk factors of hyperbilirubinemia in late pre-term infants. Strengthening the management of pregnancy-induced hypertension syndrome and the treatment of newborn hemolytic disease can reduce the occurrence of severe hyperbilirubinemia in late preterm infants.
10.Effect of airway remodeling on airway responsiveness in asthmatic guinea pigs

AIM: To establish a guinea pig asthma model and to evaluate the effect of airway remodeling on airway responsiveness. METHODS: The guinea pig asthma model was established by ovalbumin (OVA) sensitization and challenge repeatedly. Bronchial provocation tests were conducted through intravenous injection of acetylcholine. The airway morphologic parameters were measured by computer image analysis system. White blood cells and the differential count in bronchoalveolar lavage fluid (BALF) were examined. RESULTS: The resistance of airway was increased significantly after 4 weeks of OVA exposure, but the increase disappeared upon prolonged exposure. After 8 weeks of OVA exposure, fiber tissue in large airway was increased, and the thickness of smooth muscle layer of small airway was enlarged, as compared with that in control animals. CONCLUSION: Airway responsiveness has changed after prolonged OVA exposure in guinea pigs. This change is related to airway remodeling. [
