1.Determination of Atmospheric Krypton and Xenon by Gas Chromatography-Mass Spectrometry in Direct Injection Mode

Volume concentration determination for atmospheric krypton and xenon is very important for krypton-85 and radioactive xenon isotopes monitoring. An injection setup integrated adjustable quantity sample injection and quantitative dilution function was designed. The effects of EI source parameters on the sensitivity of MS detector were studied. The optimized values were as following: ionization energy of 70 eV, emission current of 40 mA, cathode voltage of 27 mV, focus voltage of 85 mV and lens compensation of 20 V, respectively. A GC-MS method for the determination of krypton and xenon in atmosphere without of sample pretreatment was developed. The minimal detected concentrations for krypton and xenon were 3. 3×10-8(V/V) and 2. 6×10-9(V/V). Moreover, the krypton and xenon concentrations in the ground level air around our laboratory were measured with the results of 1 . 1 × 10-6 ( V/V ) and 9 . 3 × 10-8 ( V/V ) . The related combined standard uncertainties for krypton and xenon results were 2. 38% and 3. 15%, respectively.
2.Clinical Observation of Chronic Congestive Heart Failure Treated with Buzhong Yiqi Wuling Decoction Com-bined with Western Medicine

OBJECTIVE:To observe the clinical efficacy and safety of Buzhong yiqi wuling decoction combined with western medicine in the treatment of chronic congestive heart failure(CHF). METHODS:120 CHF patients were divided into observation group and control group by random number table method,with 60 cases in each group. Control group received conventional western medicine treatment as rest,low salt diet and diuretics. Observation group was additionally given Buzhong yiqi wuling decoction,one dose a day,at twice,on the basis of control group. Treatment course of 2 groups lasted for 2 weeks. Average TCM symptom score, level of plasma NT-proBNP,6 min walk test(6MWT)distance before and after treatment,clinical efficacy and the occurrence of ADR were compared between 2 groups. RESULTS:Before treatment,there was no statistical significance in average TCM symptom score,level of plasma NT-proBNP and 6MWT distance between 2 groups(P>0.05). After treatment,average TCM symptom score and level of plasma NT-proBNP of 2 groups were decreased significantly,and the observation group was more significant than the control group,with statistical significance(P<0.05);6MWT distance of 2 groups were improved significantly compared to before treatment,and the observation group was significantly longer than the control group,with statistical significance(P<0.05). After treatment,total effective rate of observation group(93.33%)was significantly higher than that of control group(83.33%),with sta-tistical significance(P<0.05). No obvious ADR was found in 2 groups. CONCLUSIONS:Buzhong yiqi wuling decoction is an ef-fective prescription to treat CHF,and can relieve clinical symptoms,improve the cardiac function,decrease NT-proBNP level and en-hance the patient exercise tolerance with good safety.
3.Clinical research of Zhuanggu-Huoxue-Buyang decoction combined with negative pressure seal flushing drainage treatment for traumatic osteomyelitis

Objective To investigate the clinical curative effect of Zhuanggu-Huoxue-Buyang decoction combined with negative pressure seal flushing drainage treatment for traumatic osteomyelitis. Methods According to the random number method, 132 patients with traumatic marrow from June 2015 to June 2016 in our hospital were divided into the control group and research group with 66 cases in each group. The patients of control group were treated with negative pressure seal flushing drainage, and the patients of research group were given to Zhuanggu-Huoxue-Buyang decoction on the basis of control group.Both groups were treated for 15 days. The clinical effective rate and clinically relevant indicators were counted, which included fracture healing time, granulation, covering time, average length of hospital stay, antibiotic use time, the negative pressure drainage time and positive rate of bacteria. At the same time patients complications were observed during the treatment. In addition, the recurrence rate of two groups was in the following half a year.Results The clinical effective rate of patients in the research group [95.5% (63/66) vs. 83.3% (55/66), χ2=5.114]was significantly higher than control group(P<0.05).The fracture healing time of patients in research group(5.3 ±1.1 months vs.7.6 ±1.9 months,t=-8.705)was significantly lower than the control group(P<0.05). The granulation covering time of patients in research group (27.8 ± 5.6 d vs.35.1 ± 6.4 d,t=-6.961) was significantly lower than the control group (P<0.05). The average length of hospital stay of patients in research group (31.2 ± 4.3 d vs.39.2 ± 5.1 d,t=-9.870) was significantly lower than the control group (P<0.05). The antibiotic use time of patients in research group(8.9 ±2.0 d vs.13.3 ±3.9 d,t=-8.152)was significantly lower than the control group (P<0.05). The negative pressure drainage time of patients in research group (13.6 ± 3.3 d vs.19.2 ± 4.2 d,t=8.744)was significantly lower than the control group(P<0.05).The culture positive rate of patients in research group[6.1%(4/66)d vs.18.2%(12/66),χ 2=4.552]was significantly lower than the control group(P<0.05).The rate of complications of patients in control group[18.2%(12/66)vs.4.6%(3/66),χ 2=6.092] was significantly higher than the research group (P<0.05). The relapse rate of patients in control group [21.8% (12/55)vs.7.9%(5/63),χ 2=4.589]was significantly higher than the research group(P<0.05).Conclusions The Zhuanggu-Huoxue-Buyang decoction combined with negative pressure seal flushing drainage treatment improves clinical indicators and reduces the incidence of complications and the recurrence rate.
4.Death and prediction analysis of adult hematological malignancies in Shenzhen

Objective:To investigate the characteristics of death, tendency and the prediction of Shenzhen residents with adult hematological malignancies from 2017 to 2020.Methods:The surveillance data of hematological malignancies from 2017 to 2020 and the demographic data in Shenzhen were collected from Shenzhen death cause monitoring system and Shenzhen Center for Disease Control and Prevention, respectively. The data of the 7th national demographic data in 2020 were set as the standardized population data. Crude mortality rate (CMR), standardized mortality rate (SMR) and annual percentage change (APC) of mortality were calculated by using Joinpoint software. The grey model GM(1,1) was built to predict the mortality of adult hematological malignancies in Shenzhen between 2021 and 2025.Results:From 2017 to 2022, the male CMR of hematological malignancies was 1.15/100 000 to 1.85/100 000, and the SMR was 2.24/100 000 to 2.44/100 000; the female CMR of hematological malignancies was 0.81/100 000 to 1.75/100 000, and the SMR was 1.67/100 000 to 1.90/100 000. There were no statistically significant differences in the annual CMR and SMR between male and female hematological malignancies (all P > 0.05), and the annual change trend of CMR and SMR was not significant. The APC of male and female CMR was 27.28% and 12.70%, respectively (χ 2 = 0.01, P = 0.939); the APC of male and female SMR was 1.12% and 4.77%, respectively (χ 2 = 0.91, P = 0.318). The death causes of hematological malignancies were successively acute myeloid leukemia (AML), lymphoma, multiple myeloma, acute lymphoblastic leukemia (ALL), myelodysplastic syndrome (MDS) plus chronic myelomonocytic leukemia (CMML), chronic lymphoblastic leukemia (CLL) plus chronic myelogenous leukemia (CML). The CMR of patients with hematological malignancies aged 18-40 years was low, the CMR began to rise in patients above 40 years, especially the rapid increase at the age of 60 years, reaching the peak at the age of 80 years or above. The shortest median time of all kinds of hematological malignancies from the onset of disease to the death was found in AML group (8 months, range 0.1-168 months), the longest time was in CLL+CML group (24 months, range 0.1-300 months). Infection was the most direct cause of death, followed by single organ failure. GM(1,1) model had the better predictive effects and the total SMR would increase from 2021 to 2025 (4.52/100 000, 4.76/100 000, 5.01/100 000, 5.28/100 000 and 5.57/100 000, respectively). Conclusions:The incidence of hematological malignancies in Shenzhen residents over 40 years old is on the increase. The trend of adult hematological malignancies in Shenzhen will rise predicted by GM (1,1) grey model.
5.Effect of miR-758-3p on invasion and proliferation of gastric cancer cell line MGC803 by targeting MDM2

Objective To observe the regulation of microRNA (miRNA,miR)-758-3p on the expression of murine double microsomal gene 2 (MDM2) and its effect on invasion and proliferation of gastric cancer cell line MGC803.Methods The bioinformatics software was used to predict MDM2 as target gene of miR-758-3p.The wild type MDM2 gene 3'untranslated region luciferase reporter gene vector and miR-758-3p target sequence mutated vector and the corresponding miRNA were transfected into gastric cancer cells MGC803 by lipofectamine.Dual luciferase reporter system detects luciferase activity.The miR-758-3p mimics were transfected into gastric cancer cell MGC803 by lipofectamine.Real-time PCR was used to detect the transfection efficiency.Real-time PCR and Western blot were used to detect miR-MDM2 expression level in cells after transfection.Transwell assay and CCK-8 assay were used to detect cell invasion and proliferation.SPSS 20.0 was used to conduct the statistical analysis.Results Dual luciferase reporter assay confirmed that miR-758-3p could target MDM2 gene(P < 0.05).The expression level of miR-758-3p in MGC803 cells transfected with miR-758-3p mimics was significantly higher than that in miR-NC cells [(6.68 ±0.53) vs (0.84 t0.12),P <0.01].Compared with miR-NC group,MDM2 expression was down-regulated in MGC803 cells transfected with miR-758-3p mimics (P < 0.05).The number of invasive cells in miR-NC group and miR-758-3p group were (136.00 ± 16.62) and (79.49 ± 6.42).After knockdown MDM2,the invasiveness of cells was significantly decreased (P < 0.05).The results of CCK-8 showed that the proliferation of MGC803 cells transfected with miR-758-3p group was significantly lower than that of miR-NC group (P < 0.01).Conclusion miR-758-3p can reduce the invasion and proliferation of MGC803 cells by targeting MDM2.
