Objective To study the expression of activator protein-1(AP-1)and connexin 43(Cx43)in uterine smooth muscle of term pregnancy and its relationship with preterm delivery. Methods Immuno-histochemistry was applied for 15 uterine smooth muscle samples of term pregnancy without labor(Group Ⅰ),15 of term pregnancy in labor(group Ⅱ)and 10 of preterm delivery in labor(group Ⅲ)to investigate the expression of two subunits of AP-1(c-Jun and c-Fos)and Cx 43. Results (1)The expression of Cx43 in group Ⅰ and Ⅱ(4.33±0.51 and 4.20±0.42)were significantly higher than that in group Ⅲ(3.15±0.41,P<0.01).Lable index of c-Jun protein in group Ⅲ,Ⅱ and Ⅰ was(52.34±4.18)%,(45.25±5.24)%and(34.14±4.26)%,respectively (P<0.01),and the lable index of c-Fos protein was(53.48±4.36)%,(43.32±6.21)%and(31.29±3.34)%,respectively(P<0.01).Positive correlation was found between the expression of Cx43and c-Jun,c-Fos in pregnancy uterine smooth muscle(r=0.65,0.63,P<0.01). ConclusionsThe Cx43 plays an important role in the onset of labor. The expressions of Cx43 is positively related with the expression of AP-1 in pregnancy uterine smooth muscle.

Objective To investigate the effects of SLB elements presented at 5' UTR of dengue virus genome on viral translation and RNA replication. Methods The 5 end of RNA secondary structures of dengue virus genome were predicted using mfold 3.2,and three mutants with different modifications in SLB elements were designed:SLB1,part deletion of the 5' UAR sequence(82-87nt);SLB2,mutation of the sequence on stem(Mut 77G/C);SLB3,mutation of the sequence on stem(M77-78AG/GA),respectively.The mutants described above were constructed by OL-PCR based on DEN-R.luc2A-RP,respectively.Replicon RNAs corresponding to DEN-R.luc2A-RP,DEN-R.luc2AGDD-RP and the 3 mutants mentioned above were in vitro transcribed and equal amounts of RNA were transfected into BHK cells with Lipofectamine 2000.After RNA transfection, the replicons were detected and characterized by RT-PCR,IFA,Renilla Luciferase assay system and real time RT-PCR,respectively.Results It was shown that SLB1 mutant did not significantly affect the translation of the input RNA,but seriously compromised RNA synthesis;SLB2 mutant did not significantly affect the translation of the input RNA either, but its RNA replication was abolished;both the translation and replication of SLB3 mutant were abolished. Conclusion Both the nucleotide sequence and the RNA secondary structure of the second loop and short stem of SLB element are highly conserved.SLB element may play an essential role on viral translation and RNA replication. The findings of present study may set a foundation for elucidating the role of SLB during viral translation and replication.

Objective To construct human apolipoprotein O (apolipoprotein O, ApoO) expression vector and obtain recombinant fusion protein thioredoxin (Trx)-ApoO by pET prokaryotic expression system. Methods The ApoO gene fragment from the human liver cDNA library was amplified by PCR. The resulting product was cloned into pET-32a(+) vector and sequenced. The confirmed cDNA was cloned into plasmid E.coli DH10B and then transformed into E.coli BL 21 (DE3) where it was induced to express protein by isopropyl β-D-1-thiogalactopyranoside (IPTG).The fusion protein was purified by Ni-NTA resin. Results The ApoO gene was cloned by PCR and a 519 bp DNA fragment was shown on the agarose electrophoresis. The cloned gene was sequenced and demonstrated to have the same sequence as that of human ApoO gene in GenBank which justified a successful construction of recombinant plasmid. ApoO cDNA gene fragment was induced by IPTG, and a 34 kD recombinant fusion protein Trx-ApoO was tested on sodium dodecyl sulfate polyacrylamide (SDS-PAGE). Conclusion Human ApoO gene is successfully cloned and its recombinant fusion protein Trx-ApoO is expressed.

Objective To evaluate hepatectomy for liver metastasis in patients of gastric carcinoma. Methods Clinical data of 32 gastric cancer cases undergoing hepatectomy for hepatic metastatic tumor were reviewed retrospectively from 2006 to 2012.16 cases underwent radical gastrectomy and synchronous hepatectomy for liver metastasis, the remaining 16 cases underwent radical resection of gastric cancer and liver resection heterochronously. The relationship between prognosis and clinicopathology was analyzed. Results The overall survival rates were 84%,50% and 37% in 1 year,3 years and 5 years. The median survival time was 32 months.Gastric cancer invasion depth, intravascular tumor thrombi, lymphatic metastasis and intraoperative blood transfusion was related to poor prognosis by single factor analysis, while gastric serosal invasion, tumor thrombus and liver metastasis tumor > 5 cm related to poor prognosis by multiple factors analysis. Conclusions Gastric cancer patients with liver metastasis who underwent hepatic resection can achieve good prognosis if hepatic metastatic tumor < 5cm or the primary gastric cancer does not invade the serosa and without tumor thrombus.

Jaundice is frequent clinic symptoms with high incidence especially among the critical patients. The sepsis complicated with jaundice has drawn more and more consideration. However, the pathogenesis about sepsis complicated with jaundice is not confirmed and it is sometimes identified incorrectly as cholestatic jaundice. This article summarises the pathogenesis and elucidate the management about sepsis complicated with jaundice.

Objective To investigate the function of apoptosis in the renal injury by observing change of renal pathology and ultrastructure in the rat with obstructive jaundice(OJ) and if salvia miltiorrhiza(SM) can lighten the renal dysfunction of obstructive jaundice. Methods A total of 100 adult Sprague-Dawley rats were studied.Among them, the 80 models of obstructive jaundice were established by ligating bile duct(BDL),then divided into two groups: the OJ rats administered daily abdomen injections of SM(1.7g per rat) after operation of bile duct ligation(the SM group,n= 40);the OJ rats receiving only the same normal saline(the OJ group,n=40).The other rats with sham operation receiving only normal saline(the control group,n=20).Three groups of rats were sacrificed in groups at postoperation 1,2,3 and 4 week, respectively. Then serum BUN & Cr were tested and renal change of histopathology and ultrastructure were observed;Apoptosis of renal tissue were assayed by TUNEL method. Results With the time of BDL extending, the value of serum BUN & Cr increased;Apoptotic cells increased in renal tissue. After treatment with SM,the injury degree of renal function and histopathologic changes decreased. Conclusion Obstructive jaundice can lead to renal injury.Apoptosis had an important effect on renal function injury in the rat with obstructive jaundice.Salvia miltiorrhiza can ease the degree of renal function injury.
