1.Effect of Broad-spectrum Antibiotics on Coagulation Function in Patients with Cerebrovascular Disease

OBJECTIVE:To study the effect of broad-spectrum antibiotics on coagulation function in patients with cerebrovascular disease. METHODS:10 cases of coagulation dysfunction in patients treated with broad-spectrum antibiotics were colleted and analyzed in respects of PLT amount,PT,INR and clinical hemorrhage manifestations before and after treatment. RESULTS:After treatment,10 cases showed significant decreased amount of PLT;3 cases showed prolongation of PT,INR. CONCLUSION:The effect of antibiotics on coagulation function in patients with cerebrovascular disease partly associates with many factors e.g. drug combination and characterize with individual difference and sensitivity.
2.Relationship between velocity of middle cerebral artery,severity of disease and watershed infarction in patients with extracranial internal carotid artery stenosis or occlusion

3.Expression and preparation of polyclonal antibodies against human apolipoprotein A5 and tissue distribution

Objective To prepare the recombination protein and polyclonal antibodies against human apolipoprotein A5(ApoA5)and detect the expression of ApoA5 in human tissues with the antibody,then further study the fuction and serum level of ApoA5.Methods With PCR and gene recombinated techniques,the prokaryotic ApoA5 expression vector was constructed.The His-ApoA5 fusion protein was expressed in E.ColiM15,and the fusion protein was injected into New Zealand rabbits to prepare polyclonal antibody.The antibody was tested for their specificity with ELISA and Western-blot,then it was used to detect the tissue distribution of ApoA5.Results It was found that there was a specific brand at 40 ku of the recombination ApoA5(rApoA5);the polyclonal antibody had high immunoreactivity and specificity.ApoA5 was expressed in human sera and hepatic tissue,not in other tissues(heart,vessel,spleen,intestine and lung),regardless of the lipid level.Conclusion The pQE30-ApoA5 is constructed successfully,the rApoA5 can be expressed in E.Coli M15;ApoA5 antibody has highly specifity.It can be used in clinical test.There is ApoA5 in human serum,and ApoA5 is expressed in liver which may be significant in lipid metabolism.

Objective:In order to study of diagnose anaphylactic shock in forensic field,a new in vitro testing sysytem were established.Methods:The RBL 2H3 were cultured and sensitized,the release of histamine and stained for Annexin V were testing by fluorescent spectrometer and flow cytometry respectively.Results:The degree of RBL 2H3 degranulation after sensitived serum treated was higher than that after control serum treated,and the degranulation degree increased dose dependently.Conclusion:RBL 2H3 are helpful to diagnose anaphylactic shock,and the release of histamine and the positive staining for Annexin V may be the ideal indices.
7.Cognitive Emotion Regulation Questionnaire in hypertensive patients

Objective To examine the reliability,validity,and practicability of Cognitive Emotion Regulation Questionnaire (CERQ) in hypertensive patients in China.Methods Altogether 434 hypertensive patients and 462 healthy subjects were recruited. All the subjects were assessed with the CERQ-Chinese version (CERQ-C), Dysfunctional Attitude Scale (DAS), Mood and Anxiety Symptom Questionnaire-Short Form (MASQ-SF), and Center for Epidemiologic Studies Depression Scale (CES-D). We calculated the mean inter-item correlations for the total CERQ and for each of the subscales. Cronbach's alpha coefficient was used to analyze the inter-correlation and reliability, and confirmatory factor analysis was used to examine the 9-factor model. Results 1) Hypertension group reported significantly higher score than that of healthy ones on rumination (12.19±2.51 vs. 11.51±2.60, P<0.001), catastrophizing(8.82±2.19 vs.8.11±2.70,P<0.001),and blaming others(10.76±2.11 vs. 9.88±2.48,P<0.001), and had significantly lower score than that of healthy ones on positive reappraisal(13.80±3.55 vs.14.71±4.11,P<0.001).2)Reliability:In the hypertension group the Cronbach's alpha for the total CERQ was 0.80, and that for the 9 subscales ranged from 0.71 (self-blame) to 0.90 (rumination). In the healthy group the Cronbach's alpha for the total CERQ was 0.79, and that for the 9 subscales ranged from 0.71 (positive reappraisal) to 0.90 (rumination). The mean inter-item correlation coefficient for the 9 subscales was 0.21-0.42(the hypertension group)/0.19-0.32 (the healthy group). In the hypertension group,the test-retest reliability of the total scale was 0.82, the test-retest reliability of the 9 subscales ranged from 0.73 to 0.92. The confirmatory factor analysis showed that the 9 first-order factor data fitted both 2 samples well. Conclusion CERQ meets the psychometric standard and it is reliable and valid for cognitive emotion regulation strategies, which may be regarded as an appropriate assessment tool.

9.Activation and apoptosis of spleen lymphocytes in patients with viral myocarditis

AIM: To explore the role of activation and apoptosis of splenic lymphocytes in the development of viral myocarditis (VMC). METHODS: Apotosis and MHC II antigen of splenic lymphocytes were detected in the VMC group (VMC, death, 8 cases) and control group (non-cardiac death, 4 cases) with TUNEL and immunohistochemistry methods.RESULTS: Compared with the control group, the increased expression of MHC II antigen and apoptosis were found in the splenic lymphocytes in the VMC group. CONCLUSION: These results suggest that the abnormality of the apoptosis/activation of splenic lymphocytes may be involved in the pathogenesis of VMC.
