1.Bifunctional Fluorescence Molecular Probes for Detection of Aluminum and pH

A fluorescent molecular probe R6G-Flu was prepared by modifying fluorescein onto Rhodamine 6G.The probe could be used to recognize Al3+ specifically, and the detection limit could reach as low as 10-8 mol/L.After addition of Al3+ (10 μmol/L) to the probe, the solution showed a color change from colorless to pink, and green fluorescence was observed under the UV irradiation, which could be perceived by the naked eye.By measuring the fluorescence emission intensity of R6G-Flu at different pH, the probe could also be used to determine pH in acidic pH range (3.00-6.00) and basic pH range (8.00-10.50).The detection results of Al3+ and pH indicated that the R6G-Flu was a dual-functional fluorescent molecular probe.
2.Antibiotic-Associated Diarrhea in COPD Patients:Analysis of 82 Cases

OBJECTIVE:To probe into the incidence,latency,clinical features and prevention of the antibiotic-associated diarrhea (AAD). METHODS:968 patients with chronic obstructive pulmonary diseases (COPD) treated with antibiotics in the respiratory department of our hospital were prospectively followed for 30 days immediately after administration of antibiotics. RESULTS:The incidence of AAD was 8.47% (82/968) and the latency of AAD was (8.35?5.19) days. 84.15%(69/82)of the AAD cases manifested as simple diarrhea. The AAD was chiefly induced by cephalosporins (42.68%) and penicillins (23.17%). The incidence of AAD increased markedly if the length of the antibiotic treatment was ≥7 days or if at least 3 kinds of the antibiotics were used concomitantly. After discontinuation or alteration of the antibiotics or administration of microbial ecological agents,metronidazole or vancomycin,71(86.59%) of the total 82 AAD cases were cured,10(12.20%) improved and 1 (1.21%) showed no response. CONCLUSION:AAD is a frequent complication of antibiotic therapy. A powerful measure to prevent AAD is rational use of antibiotics.
4.Cloning and characterization of promoter region of cell division autoantigen 1

Objective To clone different lengths cell division autoantigen1(CDA1) promoters(CDA1P) and measure their activities in order to provide basis for fumctional study of CDA1P.Methods Different lengths CDA1P were amplified by PCR using mouse liver genomic as template and cloned into BglⅡ digested pGL3-basic plasmid to produce recombinant plasmid pGL3-basicmCDA1P,then they were transfected into Lewis lung cell line(LLC) and RAW 264.7 cells differently,these cells were collected after transfected for 48 h,finally luciferase activities of pGL3-basic-mCDA1P were measured.Results ①Different lengths CDA1P were cloned and characterized by restriction enzymes.②Different lengths CDA1P were ligated into pGL3basic vector,and were proved with PCR and DNA sequencing .③Activities of pGL3-basic-mCDA1A were different in the two kinds of Leuwis and mononuclear macrophage cells(RAW264.7 cells).Conclusion The different activities of CDA1P may be related with different cellular types.
5.THE EFFECT OF bFGF ON THE CALICUM INFLUX IN SPIRAL GANGLION CELLS IN STREPTOMYCIN RELATED DEAFNESS

To evaluate the effect of bFGF on the calcium influx in SGCs and its antagonistical effect on streptomycin. The SGCs of guinea pig were isolated using an enzyme machine methods and loaded with 10?mol/L Fluo 3/AM for 30 min at 37℃,then cultured 60min at room temperature. Individual Fluo 3 loaded SGCs were examined with a confocal microscope (ACAS Ultima, USA) using a 20 x objective lens and linear scan mean.The level of SGCs[Ca 2+ ] i was steady within the process of normal extracellular liquid perfusion. SGCs[Ca 2+ ] i was increased when SGCs were perfused with 150mmol/L high potassium media(10/10) and normal media containing 1nmol/L bFGF(8/9), but the level of SGCs[Ca 2+ ] i did not change in high potassium free calcium media(9/9) and free calcium media containing 1nmol/L bFGF(10/11). After the treatment with 1 mmol/L streptomycin, the level of SGCs[Ca 2+ ] i was increased 1/11 and 5/12, respectively, when perfused high potassium media and bFGF media. And after being treated with 1nmol/L bFGF, high potassium media, SGCs[Ca 2+ ] i was increased more obviously and persisted for a longer time. After being treated with 1 mmol/L streptomycin, 0 1nmol/L, 1nmol/L and 10nmol/L bFGF, respectively, and 14/14 the level of SGCs[Ca 2+ ] i was increased 5/11, 9/12, and 14/14 when perfused high potassium media. High potassium media and bFGF perfusion could result in an increase of SGCs[Ca 2+ ] i ,and SGCs[Ca 2+ ] i increase was the result of calcium influx, and there was a synergic effect between high potassium and bFGF. Streptomycin could block the process of calcium influx induced by high potassium media, the blocking effect could be antagonized by bFGF, and the antagonistic effect was bFGF concentration dependent.
6.Effect of continuous obstetric-pediatric cooperative nursing on anxiety, depression and the parenting sense of competence in NICU premature mothers

Objective:To explore the effect of continuous obstetric-pediatric cooperative nursing on anxiety, depression and the parenting sense of competence in NICU premature mothers.Methods:Totally 140 mothers of NICU preterm infants in hospital from January 2018 to December 2018 were selected and randomly divided into the control group and intervention group according to the method of random number table, and each group 70 cases. The control group received the routine nursing mode in obstetrics and paediatrics and the intervention group received the continuous obstetric-pediatric cooperative nursing based on routine nursing. To compare the anxiety level, depression level, the score of parenting sense of competence, hospitalization days of preterm infants, the outpatient rate and the readmission rate of preterm infants within 1 months of discharge between two groups.Results:The levels of anxiety and depression of the mothers of premature infants in the intervention group during hospitalization ,1 week after discharge and 1 month after discharge were (43.17±6.89), (8.96±2.67), (34.65±6.87), (7.65±1.92), (34.97±6.06), (6.29±1.54) points, lower than control group (47.03±8.06), (10.97±3.34), (45.44±7.72), (9.43±2.61), (41.46±7.26), (8.28±2.23) points, the differences between the two groups showed statistical significance ( t value was 2.331-6.097, P<0.05). The two dimensions of parenting efficacy and parenting satisfaction were (31.47±4.97), (43.38±4.21) points, which were higher than (25.91±5.42), (39.43±5.91) points of the control group, there were significant statistical differences among the two groups ( t value was -6.314 and -4.528, P<0.01). The hospitalization days of premature infants, outpatient and readmission rate within 1 month of discharge were (13.25±3.27) days, 8.57% (6/70), 2.86% (2/70), which were shorter or lower than (15.59±2.79) days, 24.29% (17/70), 12.86% (9/70) in the control group, the differences were statistically significant ( t value was 2.909, χ2 values were 6.295, 4.834, P<0.05). There were significant statistical differences among interaction effect, time effect and intergroup effect ( F values were 4.921-99.310, P<0.05). Conclusion:Obstetrical-pediatric cooperative nursing can effectively to lower the anxiety and depression level of preterm mothers, improve the parental competence, promote the mental health of preterm mothers, and ultimately shorten hospital days and decrease the rate of outpatient and readmission of premature infants, which is conducive to preterm infants′ rehabilitation.
7.Clinic curative effects of low-dose mifepristone in treating recurrent endometriosis and changes in ovarian function and serum cortisol level

0.05).After treatment,blood serum E2 level in group M was (74.18?22.32) pmol?L-1,whereas that in group D was (24.96?3.94)pmol?L-1,there was significant difference(P0.05).Conclusion Clinical symptoms of patients with recurrent endometriosis in two groups are improved.Serum E2 level in group M remains in the follicular phase,but group D declines to postmenopausal range.Mifepristone does not result in lack of estrogen,ovulation recovers rapidly after treatment in group M compared with group D.Mifepristone and danazol have not obvious impact on blood serum cortisol.
8.Effect of comprehensive nursing intervention on rehabilitation of long-term hospitalized patients with schizophrenia

Objective To explore the effect of comprehensive nursing intervention on the rehabilitation of patients with schizophrenia,and to provide reference for clinical nursing.Methods Sixty-seven long-term hospitalized patients with schizophrenia received 6 months comprehensive nursing intervention.The instruments of the nurse observation scale for inpatient evaluation(NOSIE),insight and treatment attitude questionnaire (ITAQ),scale of social function in psychosis inpatients (SSPI) were administered both at the start and the end of intervention.Result After 6 months of treatment,the scores on mental disorder,recovery of social ability and treatment compliance and recovery or selfconcicousness increased as compared with pre-intervention (all P<0.001).Conclusion The comprehensive nursing intervention can improve the mental symptoms and social function,alleviate their depression and promote mental rehabilitation of the patients with schizophrenia.
9.Establishment and evaluation of rat model of acute stress during chronic heart failure

Objective To establish the rat model of acute stress during chronic heart failure and evaluate it.Methods Male Wistar rats were injected adrimycin(ADR)via abdominal cavity to induce heart failure.Cardiovascular dynamics and heart pathology were measured.Rats with heart failure received bacterial lipopolysaccharide(LPS)0.2 mg/kg by peritoneal injection to induce acute stress,and we measured blood pressure,heart rate and the expression of HO-1 after LPS injection.Results ADR peritoneal injection led to heart failure successfully.Compared with that in normal group,left ventricular function(?LVdp/dtmax)of rats in model group reduced significantly,and myocardial fibrosis and hypertrophy occurred,too.LPS peritoneal injection led to acute stress in rats with heart failure.Heart rate and blood pressure began to rise 0.5 h after LPS injection and reached at 1 h after LPS injection.The expression of myocardium HO-1 increased at 6 h and was significantly higher at 12 h after LPS injection.The expression of HO-1 was higher in model group than in normal group.Conclusion LPS peritoneal injection successfully leads to acute stress in rats with heart failure,providing a reliable model for study of decompensated heart failure.
10.Specific inhibitory protein Dkk-1 blocking Wnt/β-catenin signaling pathway improve protectives effect on the extracellular matrix.

The present study examined the role of Wnt/β-catenin signaling pathway in the degeneration of nucleus pulposus cells and the protective effect of DKK1 on nucleus pulposus cells. The model of nucleus pulposus cell degeneration was induced by intra-disc injection of TNF-α, and the expression of β-catenin protein was detected by Western blotting. The cultured rabbit nucleus pulposus cells were divided into 4 groups. In group A, the cells were cultured with normal medium and served as control group. In group B, the cells were cultured with TNF-α and acted as degeneration group. In group C, the cells were cultured with TNF-α and transfected with Adv-eGFP and was used as fluorescence control group. In group D, the cells were cultured with TNF-α and transfected with Adv-hDKK1-eGFP, serving as intervention group. The expression of type II collagen, proteoglycan, β-catenin, and MMP-13 in each group was detected by immunocytochemistry and RT-PCR. The result showed that TNF-α increased the expression of β-catenin and MMP-13, and significantly inhibited the synthesis of type II collagen and proteoglycan, which resulted in the degeneration of nucleus pulposus cells. This effect could be obviously reversed by DKK1. We are led to concluded that TNF-α could activate the Wnt/β-catenin signaling pathway, and increase the expression of MMP-13, thereby resulting in disc degeneration. Specifically blocking Wnt/β-catenin signaling pathway by DKK-1 could protect the normal metabolism of intervertebral disc tissue. The Wnt pathway plays an important role in the progression of the intervertebral disc degeneration.
