1.Primary Evaluation of the Detection Effect for Vibrio parahaemolyticus Chromogenic Medium

Vibrio parahaemolyticus is an important food-borne pathogenic bacterium that widely exists in all kinds of seafood.As the traditional media and method were very costly-time and money,a new chromogenic medium(HKC vibrio)was designed in this assay.Through detecting the artificially contaminated samples and natural samples,the sensitivity,specificity and detection effect of HKC vibrio were studied and compared with CHROMagar vibrio and TCBS.The results showed that HKC vibrio had the same sensitivity as CHROMagar vibrio and TCBS,also had highly specificity.In conclusion,HKC vibrio was an invaluable medium which may improve the detection efficiency of Vibrio parahaemolyticus dramatictly.
2.Alternation and rehabilitation of proprioceptive sense following anterior cruciate ligament reconstruction

Anterior cruciate ligament (ACL) has two important functional roles, including biomechanics and proprioceptive sense. A groat propr!oceptive sense is a basis to exactly obtain highly functional exercise. ACL injury may block perception and conduction information and seriously influence motion and daily activities. A complete rehabilitation of ACL function depends on rehabilitation of both mechanical structure and proprioceptive sense. Therefore, reconstruction of joint function following ACL injury is focus on reconstruction of both joint biomechanical stability and proprioceptive sense. This study was designed by distribution and function of proprioceptive sense to analyze the altemation of propdoceptive sense following ACE injury, and to investigate the related factors and ways for rehabilitation of propriocaptive sense following ACL reconstruction.
3.Reversal of multidrug resistance by neferine in human gastric carcinoma cell line SGC7901/VCR

Purpose:To study the reversal of multidrug resistance (MDR) and its mechanism by neferine which is a new calcium channel blocker in human gastric carcinoma cell line SGC7901/VCR. Methods:The cytotixic effect was tested by MTT assay. The expression of multidrug-resistance-associated protein in human gastric carcinoma cells was examined by SP immunocytochemical and flow cytometry. Results:Neferine at the concentration 10 ?mol/L was not of significant cytotoxicity to SGC7901 and SGC7901/VCR cells. Neferine at the concentration of 2.5,5,10 ?mol/L decreased the IC_(50)value of VCR to SGC7901/VCR cells from 2.32 ?g/ml to 0.340,0.128 and 0.053 ?g/ml, respectively and with the increase by 6.8-,18.1-and 43.8-fold in the chemosensitivity, respectively. It had more potent reversal action on SGC7901/VCR cells than Verapamil at the concentration of 10 ?mol/L(P
4.Comparison of the Efficacy and Safety of Alfacalcidol and Strontium Ranelate in the Treatment of Senile Menopausal Osteoporosis

OBJECTIVE:To compare the efficacy and safety of alfacalcidol and strontium ranelate in the treatment of senile menopausal osteoporosis. METHODS:158 elderly menopausal patients with osteoporosis were randomly divided into control group (79 cases)and observation group(79 cases). All patients were orally given 1 Calcium carbonate D3 tablet,once a day. Based on it, control group was given 2 Alfacalcidol soft capsules,once a day. Observation group was additionally given 1 bag of Strontium ranelate dry suspension,orally before bedtime,once a day. The treatment course for both groups was 12 months. L2-4 and BMD of femoral neck,osteocalcin,TPINP and VAS and ADR before and after 6 and 12 months in 2 groups were observed. RESULTS:Before treatment,there were significant differences in the BMD of L2-4 and femoral neck,osteocalcin,TPINP and VAS scores be-tween 2 groups(P>0.05). BMD of L2-4 and femoral neck in control group after 12 months and in observation group after 6 and 12 months were significantly higher than before,and it gradually increased be time in observation,observation group was higher than control group;TPINP and VAS scores in control group after 12 months and in observation group after 6 and 12 months were significantly lower than before,and it gradually decreased be time,observation group was lower than control group,the differenc-es were statistically significant(P<0.05);osteocalcin levels in 2 groups were significantly higher than before,and it gradually in-creased be time,the difference was statistically significant (P<0.05),but there was no significant difference between 2 groups (P>0.05). There were no ADR during treatment. CONCLUSIONS:Based on the conventional treatment,the efficacy of alfacal-cidol and strontium ranelate is superior to alfacalcidol alone in the treatment of senile menopausal osteoporosis,with similar safety.
5.Significance of determination of serum thyroid hormonens of stable coronary heart disease patients in different glucose metabolic statuses

Objective To evaluate the importance of serum thyroid hormonens level of patients with stable coronary heart disease(CAD)in different glucose metablic statuses.Methods 173 cases of CAD patients in different glucose metablic statuses and 58 cases of healthy people were enrolled in the study.The levels of thyroid hormonens and serum insulin was measured by electro-chemiluminescence,and the evaluation of glucose metablic statuses and insulin resistance index was by means of 75 g oral glucose tolerance test.Results There was no significant difference in the levels of systolic pressure(SBP),diastolic pressure(DBP),insulin resistence index and serum thyroid hormonens among CAD patients with different glucose metablic statuses(P >0.05).The levels of fasting plasma glucose,2 hour postprandial plasma glucose(PG2h)and glycosylated hemoglobin(HbA1c)in CAD complicated with type 2 diabetes group were significantly higher than those in CAD complicated with impaired glucose tolerance group and CAD complicated with normal glucose tolerance group (P < 0.05 ).And the PG2h level of CAD complicated with impaired glucose tolerance group was significantly higher than both of CAD complicated with normal glucose tolerance group and normal control group s(P <0.05).Conclusion It indicates that serum thyroid hormonens level may not affect glycometabolism in CAD patients.
6.Study of Human Cytomegalovirus mRNA Transcription Profile at Different Infection Stages in Infants

Objective Human cytomegalovirus (HCMV) can induce infection in multiple organs, including genitouri-nary system, central nervous system and liver. It can even lead to serious defects and death. China, as a high-incidence area of HCMV, need employ early treatment. However, the sensitivity of present HCMV diagnosis is not satisfied. Methods In the present study, we focused on transcription level of HCMV IE and pp67 mRNA using real-time quantitative PCR. Results IE mRNA was positive even in a portion of patients who were regarded as negative by traditional DNA diagnosis method. On the other hand, pp67 mRNA that were normally expressed at the late stage of HCMV infection was 100%posi-tive in the first-visit patients who were regarded as positive using traditional DNA diagnosis method. Conclusion The pres-ent study indicated that mRNA detection based on PCR method was more sensitive compared with DNA detection for assess-ing HCMV virus infection. Finally, different treatment should be performed upon mRNA transcription profile at different in-fection stages.
7.Determination of lidocaine and its metabolites by HPLC to evaluate CYP3A activity of human liver microsomes

AIMTo evaluate CYP3A activity of hum an liver microsomes by the ratio of lidocaine(LDC) to its metabolites. M ETHODSIn 1 g?L -1 microsomal protein lidocaine was incubated at 37 ℃ for 1 hour. Lidocaine, monoethylglycinexylidide(MEGX) and glycinexylidide(GX) were determined by HPLC. RESULTSThe linear regress equations fo r LDC, MEGX and GX were = 0 293 4 X- 0 005 661 (r= 0 9 99 7 )、= 0 791 3 X- 0 008 916 (r= 0 999 3 ) and = 0 679 9 X- 0 007 770 (r= 0 998 5 ). The best conditio n for the incubation test in vitro was 2 0 mg?L -1 LDC plus 1 0 g ?L -1 microsomal protein for 60 min. The mean (MEGX+GX)/LDC ratio was 3 2 8. CONCLUSIONThe (MEGX+GX)/LDC ratio can be used to evaluate C YP3A activity of human liver microsomes.
8.Nerve growth factor attenuates cochlear injury induced by brain ischemia and reperfusion in rats

AIM: To investigate the effect of nerve growth factor (NGF) on auditory and cochlear damage induced by brain ischemia and reperfusion. METHODS: The rats with brain ischemia and reperfusion were divided into two groups at random. In the experimental group, the rats were injected intramuscularly with NGF . In control group, the animals were injected with normal saline instead of NGF. Then the hearing loss and cochlear structural changes of rats in both groups were compared. A total of 96 patients with Graves'disease(GD)were followed for one and half years to observe the effect of antithyroid drugs(ATD)treatment.Serum TRAb,total iodine concentration and CD80 mRNA expression of peripheral blood monouclear ceils were measured.Logistics regression analysis was conducted with the combination of above parameters.Those GD patients with high level of TRAb,positive family history of GD, increased expression of CD80 and early age of onset were more inclined to relapse after ATD treatment.
10.Effect of Concurrent Chemoradiotherapy on Patients'Serum CD44v6 and VEGF Levels and Its Clinical Significance

Objective: To investigate the changes in serum CD44v6/v EGF(sCD44v6/sVEGF)levels after concurrent chemoradiotherapy in patients with inoperable non-small cell lung cancer(NSCLC)patients and to analyze the correlation of serum markers with patients'prognosis. Methods: The sCD44v6/v EGF levels were detected by ELISA in 50 inoperable NSCLC patients before and after concurrent chemorediotherapy. Results: The sCD44v6 and sVEGF levels before treatment was significantly higher than that in the control group.The pre-treatment sVEGF level was closely related to cTNM stage.metastasis,cell differentiation and primary tumor size,but was not correlated with histological classification,lymph node status,age or gender.The pre-treatment sCD44v6 level was closely correlated with cTNM stage,metastasis,cell differentiation,and primary tumor size,but was not correlated with histological classification,age or gender.Patients'sCD44v6 before treatment(570.89±63.30 ng/L)was significantly higher than that after treatment(281.44±74.28 ng/L).Patients'sVEGF before treatment(241.09±85.96 ng/L)was significantly higher than that after treatment(133.64±67.69 ng/L).PD patients had the highest level of sCD44v6 and sVEGF and CR patients had the lowest sCD44v6 and sVEGF level.The sVEGF level was highly correlated with sCD44v6(r=0.291,P<0.05).Conclusion:The sCD44v6 and sVEGF levels might be helpful for evaluation of the biological behavior and prognosis of NSCLC.
