1.Association of serum homocysteine levels with metabolic syndrome and its components in adults

Objective To explore the relationship of serum homocysteine ( Hcy) level with metabolic syndrome (MS) and its components.Methods A cross-sectional study was conducted in 594 adults consec-utively sampled from people visiting Fuwai Hospital for physical examination between September and Decem-ber 2012, including 160 MS participants (MS group) and 434 without MS participancs (without MS group). Serum Hcy levels, serum biochemical parameters, and anthropometric measurements of all the participants were collected.Binary logistic regression was used to assess the association of serum Hcy levels with MS and its components.Results No significant difference in Hcy levels was observed comparing the MS group and the without MS group [(12.2 ±7.0) μmol/L vs.(12.6 ±8.4) μmol/L, P>0.05].Between participants with and without hyperhomocysteinemia (≥15 μmol/L vs.<15 μmol/L) , no statistically significant differ-ences were observed in prevalence of MS and its metabolic disorders ( MS 27.3% vs.26.9%; high blood pressure 27.3% vs.19.6%; high triglycerides 50.0% vs.48.3%; high-density lipoprotein cholesterol 39.8%vs.36.6%; dysglycemia 30.7% vs.32.6%; abdominal obesity 31.8% vs.31.2%; all P >0.05).Among participants having 0 to 5 metabolic abnormalities, there were no significant differences in ser-um Hcy levels [ (12.3 ±7.4) μmol/L; (12.4 ±6.7) μmol/L; (13.2 ±11.2) μmol/L; (12.5 ± 7.8) μmol/L; (11.2 ±2.7) μmol/L; (12.4 ±4.3) μmol/L; all P>0.05].In a binary logistic re-gression model adjusted for age, gender, body mass index, total cholesterol, low-density lipoprotein choles-terol, and high-sensitivity C-reactive protein, neither MS nor its components were independent predictors of serum Hcy levels ( all P>0.05 ) .Conclusions Elevated serum Hcy levels are not associated with MS or its components in people without evident cardiovascular diseases .MS and elevated serum Hcy levels may cause cardiovascular diseases via different mechanisms .
2.PRELIMINARY STUDY ON MONOCLONAL ANTIBODIES AGAINST CYSTICERCUS CELLULOSAE

In order to produce valuable monoclonal antibodies for immunodiagnosis of cysticerco-sis, hybridoma technique was adopted using the fusion of splenic lymphocytes from BALB/c mice immunized with Cysticercus cellulosae antigens and the myeloma cell line NS-1. Through screening and cloning, three monoclonal antibody cell lines have been established. Im- munoglobulin subclasses of 3 lines were determined to be IgGl. These monoclonal antibodies were found to be highly specific and without cross reaction with Echinococcus granulosus hy-datid cyst and Cysticercus fasciolasis. Antigen fractions specifically reacting to monoclonal antibodies were demonstrated with Western blot method. The results showed that specific reaction bands of SCW located in 25 kDa and 17 kDa regions.

Objective To better understand the regulation of the soluble receptor of vascular endothelial growth factor (VEGF), s-VEGFR-1 (or soluble fms-like tyrosine kinase, SFlt-1), in angiogenetic process in endometriosis. Methods Levels of free VEGF and s-VEGFR-1 were measured by enzyme-linked immunosorbent assay (ELISA) in peritoneal fluid from 28 subjects with surgically confirmed endometriosis, and 10 controls with no clinical evidence of the disease and other diseases. Meanwhile, we calculated a VEGF activity index by means of the ratio VEGF/s-VEGFR-1. Results We found higher VEGF concentration in endometriotic lesions than controls (P0.05). VEGF activity index in controls, stage Ⅰ-Ⅱ and stages Ⅲ-Ⅳ of endometriosis was 0.310, 0.276 and 0.273, respectively. 7.Dobutamine stress echocardiography combined with Isoket detecting viable myocardium in patients with coronary artery disease

Objective To observe the effect of Isoket on standard dobutamine stress echocardiography(DSE).Methods Thirty-seven patients were divided into three groups according to the results of quantitative coronary angiography(QCA):the control group(CON) including 12 patiens,the coronary artery disease group 1(CAD1) including 15 patients,and the coronary artery disease group 2(CAD2) including 10 patients.Among the three group,the DSE+Isoket was done in CAD2,while the standard DSE was done in the other two groups.The improvement of segments of left ventricular after percutaneous transluminal intervention(PCI) was looked as the standard evaluating DSE.Results Isoket could increase heart rate and decrease systolic blood pressure,but could hardly affect diastolic blood pressure.The rate of biphysic response was 62.5% in group CAD1,and the sensitivity of detecting viable myocardium was 61%,while the specificity was 76%.In group CAD2,the rate of biaphysic response was 72.2% ,and the sensitivity of detecting viable myocardium was 82%,while the specificity was 88%.The rate detecting viable myocardium and sensitivity was improved significantly in group CAD2.Conclusions DSE combined with Isoket was beneficial for detecting viable myocardium.
8.Clinical observation of azithromycin in treatment of mycoplasma pneumonia in 90 children

Objective To evaluate the effect of azithromycin on mycoplasmal pneumonia (MP).Methods We divided 90 MP cases into azithromycin and erythromycin treatment groups. In azithromycinThe pyretolysis time, cough improvement time, the disappearing time and the mean length of hospitalization of azithromycin group were shorter than that of erythrornycin group. The local ache, stomach and intestinal tract adverse reaction, and damage of hepar function were less than these in erythromycin group. ConclusionAzithromycin is an effective and safe drug to MP.
9.Study of Human Cytomegalovirus mRNA Transcription Profile at Different Infection Stages in Infants

Objective Human cytomegalovirus (HCMV) can induce infection in multiple organs, including genitouri-nary system, central nervous system and liver. It can even lead to serious defects and death. China, as a high-incidence area of HCMV, need employ early treatment. However, the sensitivity of present HCMV diagnosis is not satisfied. Methods In the present study, we focused on transcription level of HCMV IE and pp67 mRNA using real-time quantitative PCR. Results IE mRNA was positive even in a portion of patients who were regarded as negative by traditional DNA diagnosis method. On the other hand, pp67 mRNA that were normally expressed at the late stage of HCMV infection was 100%posi-tive in the first-visit patients who were regarded as positive using traditional DNA diagnosis method. Conclusion The pres-ent study indicated that mRNA detection based on PCR method was more sensitive compared with DNA detection for assess-ing HCMV virus infection. Finally, different treatment should be performed upon mRNA transcription profile at different in-fection stages.
10.Antitumor effect of corosolic acid and it’s impact on CAM and YSM angiogenesis

Objective To evaluate the antitumor effect of corosolic acid and its impact on CAM and YSM angiogenesis.Methods The effects of CRA on A549 proliferation was studied by MTT method in vitro.Tumor-bruden nude mice model were established by injecting A549 lung cancer cell marked with bioluminescent to nude mice,and the growth of tumor in mice were detected by IVIS small animal in vivo imaging system.Experiment of chicken embryos eggs are used to observe the role of drugs on CAMand YSMblood vessels. Results The value of IC50 of CRA for A549 in vitro was 26.8μg/mL.A tumor-burdened animal experiment results showed that the CRA to A549 solid tumor has a certain therapeutic effect.CAM and YSM blood vessels of chicken embryos eggs treated with CRA were decreased significantly than negative control group.Conclusion CRA has certain therapeutic effect for A549,which may be related to the inhibition of angiogenesis in tumor tissues.
