1.Effects of Cilostazol on microvascular complications of diabetic patients

Objecfive To evaluate the effects of cilostazol on the prevention of microvascular complications in diabetic patients.Methods 60 diabetic patients with microvascular complications were orally given cilostazol for 1 month.Changes of Mean platelet volume (MPV),plateletcrit (PCT),platelet distribution width (PDW) and platelet count (PLT) were studied.Results With administration of cilostazol,MPV and PDW decreased significantly. Conclusion Cilostazol improves platelet parameters,suggesting that it could prevent the progression of microvascular diseases.
2.Application of a 3D printed trabecular block cage in treatment of patients with basilar invagination and atlantoaxial dislocation

Objective:To explore the short-term outcomes of a 3D printed trabecular block cage to assist posterior internal fixation for the treatment of patients with basilar invagination and atlantoaxial dislocation.Methods:Between June 2017 and February 2019, 12 patients with basilar invagination and atlantoaxial dislocation underwent atlantoaxial distraction and posterior internal fixation at Department of Orthopedics, The First Affiliated Hospital to Zhengzhou University. They were 5 males and 7 females, aged from 34 to 62 years (average, 45.6 years). 3D printed cages were inserted intraoperatively between the joints of the atlantoaxial lateral mass. The atlanto-dental interval interval (ADI), cervico-medullary angle (CMA) and distance from tip of the odontoid process to Chamberlain's line (DOCL) and the Japanese Orthopedic Association (JOA) scale were compared between preoperation and 12 months postoperation to observe the fusion of the joints of the atlantoaxial lateral mass.Results:Operation went on uneventfully in all the 12 patients. Operation time averaged 116.5 min (from 85 to 190 min), fluoroscopy frequency 9.4 times (from 6 to 21 times), and intraoperative bleeding 82.3 mL (from 50 to 210 mL). No such postoperative complications occurred as cerebrospinal leak, cerebral infarction, or breakage, displacement or loosening of implants. All patients were followed up for 18 to 42 months (mean, 26.3 months). Their preoperative JOA, ADI, CMA and DOCL [8.33±0.98, (8.66±1.64) mm, 119.63°±4.15° and (9.66±2.15) mm] were significantly improved to 14.17±1.03, (2.63±0.59) mm, 153.76°±7.88° and (2.07±0.69) mm ( P<0.05) at 12 months postoperation. Bony fusion was achieved in all the operative segments. Conclusion:In the treatment of patients with basilar invagination and atlantoaxial dislocation, a 3D-printed trabecular block cage can be used to assist posterior internal fixation to achieve satisfactory reduction and maintain the height of joint space, leading to satisfactory short-term outcomes.
3.Association between lncRNA IFNG-AS1 SNP and Hashimoto's thyroiditis

Objective To assess the association between three single nucleotide polymorphisms( SNPs) ( rs10878724、 rs7980829 and rs11177020 ) of lnc IFNG-AS1 and Hashimoto's thyroiditis ( HT) susceptibility. Methods TaqMan probe technology was used to genotype the selected SNPs in a total of 179 subjects, including 70 HT cases, and 109 controls. The expression levels of lnc IFNG-AS1 and IFNG were detected by SYBR-Green qRT-PCR. Results Compared with control, not only the A allele and AA genotype frequencies of rs10878724 were significantly different in group HT ( P=0. 01, P=0. 003), but also the T allele and TT genotype frequencies of rs7980829 were significantly high in group HT. Haplotype analysis showed that the G-G-A decreased the risk of HT (P=0.04), while the haplotype of A-T-T incresed the risk of HT( P=0.01). The relative expression of both IFNG mRNA and lnc IFNG-AS1 were higher in group HT than in control( P=0. 001,P=0. 013). In HT patients, IFNG mRNA relative expression in both rs7980829-TT and rs1087872-TT were significantly higher than those of other genotypes(P=0.017,P=0.009). Conclusion The SNPs of Inc IFNG-AS1 were correlated with the expression levels of IFNG and lncRNA IFNG-AS1. Noncoding genes should be further screened as potential biomarkers in prediction of HT susceptibility.
4.FAN Yongsheng's Experience in Treating Gouty Arthritis by Using Simiao Powder

[Objective] To summarize the experience of professor FAN Yongsheng who uses the herbal prescription"modified Simiao powder"to treat gouty arthritis, so as to direct our clinical practice better. [Method] By learning from professor FAN as well as collating his medical cases that applicating "modified Simiao powder" to the treatment of gouty arthritis, clearing and definiting how teacher FAN thinks etiologies and pathogenesis of gouty arthritis and analyzing the prescription of"Simiao powder"and giving two clinical cases, so that we can learn and discuss. [Result] Teacher FAN believes that gout pathogenesis for the body fluid metabolic abnormalities, and body fluid metabolism and liver and spleen and kidney three organs are closely related. Patients usually irritable or liver qi stagnation, or like to eat greasy meat, temper deficiency, or lack of kidney essence, gasification weakness, resulting in wet endogenous, wet evil staying in the joints, the emergence of local fever, phlegm, blood stasis, poisonous evil, into gouty arthritis. Acute onset of the pathogenesis of "hot and humid", the course of time for the "hot and humid phlegm stagnant each other", treatment should follow the law of "clear away heat-dampness", according to the cause, changes in the pathogenesis of flexible application of four wonderful addition and subtraction; And to Qufeng, detoxification, blood circulation, dredge meridian, so as to achieve dialectical treatment, with the card addition and subtraction. [Conclusion] Professor FAN Yongsheng uses"modified Simiao powder"to treat gouty arthritis and has achieved good clinical efficacy, worthy of clinical study and reference.
5.Determination of Hesperidin in Jinshuiliujun Decoction by RP-HPLC

Objective: The content of hesperidin in Jinshuiliujun Decoction (JSLJD) was determined by RP HPLC. Methods: The chromatographic column was Hyperisl BDS C 8(10?). The mobile phase was acetonitrile water containing 8% acetic acid (17∶83). The flow rate was 1.0 mL/min. The column temperature was 40 ?C and the detection wavelength was 284nm. The external standard and peak area method were used to determine hesperidin content. Results: There was a very good linear relationship between hesperidin and peak area when the sample injection was from 0.02 to 2.00?g. The average recovery of adding sample was 100.5% and RSD was 1.85%. Conclusions: This method is simple and believable. It can be effectively used for the quality control of JSLJD.
6.Chain mediating effect of experience avoidance and emotional eating between intolerable uncertainty and life satisfaction in college students

Objective:To explore the mediating effect of experiential avoidance and emotional eating between intolerable uncertainty and life satisfaction of college students.Methods:From October to December 2021, a total of 890 college students in the Beijing-Tianjin-Hebei region were tested by the intolerable uncertainty scale (simplified version), the acceptance action scale second edition, the Dutch eating behavior questionnaire and the life satisfaction scale. All data processing and analysis were preformed by SPSS 22.0 software, and the mediating effect was tested by Pearson correlation analysis and Bootstrap analysis.Results:The intolerable uncertainty score was (26.43±7.64), the experiential avoidance score was (22.63±9.67), the emotional eating score was (26.43 ±11.83), and the life satisfaction score was (16.74±6.03). Intolerable uncertainty was significantly and negatively correlated with life satisfaction( r=-0.76, P<0.01), and was significantly and positively correlated with experience avoidance and emotional eating( r=0.66, 0.78, both P<0.01). Experience avoidance was significantly and positively correlated with emotional eating( r=0.70, P<0.01), and was significantly and negatively correlated with life satisfaction( r=-0.62, P<0.01). Emotional eating was significantly and negatively correlated with life satisfaction( r=-0.68, P<0.01). Intolerable uncertainty affects life satisfaction through four paths.The direct effect value of intolerable uncertainty on life satisfaction was -0.53, accounting for 70.23% of the total effect. The separate mediating effect value of experience avoidance was -0.11, accounting for 50.00% of the total indirect effect.The separate mediating effect value of emotional eating was -0.08, accounting for 36.36% of the total indirect effect, and the chain mediating effect value of experience avoidance and emotional eating was -0.03, accounting for 13.64% of the total indirect effect. Conclusion:Intolerable uncertainty can directly affect life satisfaction of college students and indirectly through experiencing avoidance and emotional eating.
7.Preliminary clinical effects of total knee arthroplasty with iASSIST navigation system

Objective To explore the application value of iASSIST portable navigation in total knee arthroplasty.Methods Seventy-four patients with knee osteoarthritis from April 2016 to April 2017 were retrospectively recruited.Thirty-seven patients (37 knees) underwent TKA with iASSIST navigation,while 37 patients (37 knees) underwent conventional TKA.Five parameters were measured on the weight-bearing radiographs at six months after TKA,including mechanical axis (MA),mechanical lateral distal femoral angle (mLDFA),mechanical medial proximal tibial angle (mMPTA),sagittal femoral component angle (sFCA) and sagittal tibial component angle (sTCA).Duration of operation,blood loss volume,postoperative hospital day,Western Ontario and McMaster Universities (WOMAC) osteoarthritis index,Knee Society Score (KSS) clinical score and functional score at 6 weeks,12 weeks and 24 weeks after surgery were also recorded.Results The accuracy of MA (180.85°±0.88° versus 182.23°±1.09° in the conventional group,P<0.05),mLDFA (90.52° ±0.78° versus 91.09° ±0.96° in the conventional group,P<0.05),and mMPTA (90.34°± 1.25° versus 91.13°± 1.46° in the conventional group,P<0.05) was improved significantly in navigation group.The WOMAC osteoarthritis index at 6 weeks postoperatively in the navigation group and in the conventional group were 58.54±1.45 and 56.54± 1.77 respectively.The KSS clinical score in the navigation group and in the conventional group were 53.14± 1.13 and 49.35± 1.11 respectively.The KSS functional score in two groups were 61.24± 1.30 and 59.81 ± 1.29 respectively (P<0.05).The WOMAC osteoarthritis index at 12 weeks postoperatively in the navigation group and in the conventional group were 43.54± 1.19 and 41.92± 1.42 respectively.At 12 weeks,the KSS clinical score in two groups were 67.11 ± 1.51 and 62.08± 1.46 respectively.The KSS functional score in two groups were 68.14±1.11 and 66.38±1.26 respectively (P<0.05).The blood loss volume in the navigation group and in the conventional group were 113.11±57.29 ml and 147.57±68.77 ml respectively (P<0.05).There were no significant difference in the duration of operation,postoperative hospital day,WOMAC osteoarthritis index,knee clinical score and functional score at 24 weeks postoperatively between two groups (P>0.05).Conclusion More accurate restoration in mechanical axis and optimal implantation can be achieved with the help of iASSIST navigation.This navigation system can also achieve better knee function in the early stage after TKA.
8. Clinical research of extensive alveolar cleft treatment with computer assistant distraction osteogenesis

Objective:
To evaluate the feasibility and outcome of computer assisted distraction osteogenesis in the treatment of extensive alveolar cleft.
Methods:
Four patients [1 male and 3 females, aged (15.5±3.7) years] received treatment in the Department of Oral-Maxillofacial Surgery and Plastic Surgery, School of Stomatology, China Medical University from June 2016 to April 2018 were involved in this study. All the patients with extensive alveolar cleft [cleft width (7.64±1.29) mm] were performed orthodontic treatment to expand the dental arch and interdental space between the first molar and premolars. Three-dimensional (3D) model of the maxilla and the osteotomy guider were printed according to the CT data. The fix wings of the distractor were pre-shaped according to the 3D model. The osteotomy was performed at the interdental space and horizontal plate of palate to dissociate the alveolar bone segment. The distractor was fixed on the predetermined position. Distraction (0.4-0.8 mm/day) was performed in 7 days later and stopped when the incision connected with the canine. The distractor was removed after six months.
Results:
The distraction period was (10.8±2.5) d in four cases. The cleft was completely closed with interdental bone anchored distraction in four cases. The imaging examination in six months showed good new bone structure in the distraction zone and bone connection of the cleft.
Conclusions
Computer assisted distraction osteogenesis was effective and feasible to close the extensive alveolar cleft and provide sufficient new bone tissue.
9.An optimized method for embedding undecalcified mouse tibias in plastic blocks.

OBJECTIVE:
To optimize the method for embedding multiple undecalcified mouse tibias in plastic blocks, improve the efficiency and stability of plastic embedding and reduce the detachment rate of plastic slides.
METHODS:
Thirty undecalcified tibias from 15 B6 mice were used for plastic embedding after calcein labeling, fixation, dehydration and infiltration. The tibias were embedded in cylindrical plastic blocks with a diameter of 4 mm. For each bone, the 1/4 proximal tibia was cut off, and the remaining 3/4 was used for re-embedding. Five bones were embedded in a single block with each bone standing closely on the surface of a flat plate. The samples were randomized into control and experimental groups in all the processes of embedding, sectioning and staining. In the 3 groups with modified embedment, flowing CO was added into the embedding solution, embedding solution was applied to the section surface, and the slides were heated at 95 ℃ for 15 min. The polymerization time, slide detachment rate, bone formation and osteoblast parameters were analyzed.
RESULTS:
We prepared 6 plastic blocks, each containing 5 tibias, whose cross sections were on the same plane. The blocks were completely polymerized and suitable for sectioning. Flowing CO into the embedding solution reduced the polymerization time and increased the rate of complete polymerization. Application of the embedding solution on the section surface significantly reduced the detachment rate of the sections ( < 0.05) without affecting bone formation analysis ( > 0.05). Heating the slides significantly lowered the detachment rate of the sections ( < 0.05) without affecting osteoblast analysis ( > 0.05).
CONCLUSIONS
The optimized method allows effective embedding of multiple undecalcified mice tibias in the same block and can be an ideal method for histological analysis of undecalcified bones.

10.Observation of dendrite osteocytes of mice at different developmental stages using Ploton silver staining and phalloidin staining.

OBJECTIVE:
To assess the value of Ploton silver staining and phalloidin-iFlour 488 staining in observation of the morphology of osteocyte dendrites of mice at different developmental stages.
METHODS:
The humerus and femurs were harvested from mice at 0 (P0), 5 (P5), 15 (P15), 21 (P21), 28 (P28), and 35 days (P35) after birth to prepare cryo-sections and paraffin sections. HE staining of P35 mouse femur sections served as a reference for observing osteocytes in the trabecular bone and cortical bone. The humeral sections at different developmental stages were stained with Ploton silver staining to observe the morphology of osteocytes and canaliculi, and the canalicular lengths in the cortical and trabecular bones of the humerus of the mice in each developmental stage were recorded. The cryo-sections of the humerus from P10 and P15 mice were stained with phalloidin iFlour-488 to observe the morphology of osteocytes and measurement of the length of osteocyte dendrites in the cortical bone.
RESULTS:
In the trabecular bone of the humerus of P0-P15 mice, Ploton silver staining only visualized the outline of the osteocytes, and the morphology of the canaliculi was poorly defined. In P21 or older mice, Ploton silver staining revealed the morphology of the trabecular bone osteocytes and the canaliculi, which were neatly arranged and whose lengths increased significantly with age (P21
CONCLUSIONS
Mouse osteocyte dendrites elongate progressively and their arrangement gradually becomes regular with age. Ploton silver staining can clearly visualize the morphology of the osteocytes and the canaliculi in adult mice but not in mice in early stages of development. Phalloidin iFlour-488 staining for labeling the cytoskeleton can be applied for mouse osteocytes at all developmental stages and allows morphological observation of mouse osteocytes in early developmental stages.
