3.Loss of multi-sites allde heterozygosity on chromosome 9 in esophageal carcinoma

Objective To investigate the gene variation and the dependability and to evaluate the possible tumor suppressor genes on chromosome 9 in the development and progression of EC. Methods LOH was detected in normal esophageal mucosa, high-grade squamous dysplasia and esophageal squamous cell carcinoma by microdissection, polymerase chain reaction, denaturing polyacrylamide gel eleetrophoresis and silver nitrate staining technology. The changes of LOH at six microsatellite markers and the relationship between LOH rate were analyzed. Results In the informative cases, total frequency of LOH was 17.2 % in high-grade squamous dysplasia and 24.9 % in esophageal squamous cell carcinoma. In high grade squamous dysplasia and squamous cell carcinoma, LOH was detected at marker D9S162 (20.8 %, 36.7 %), D9S171 (33.3 %, 36 %), D9S753(34.8 %, 46.2 %), D9S1748(4.2 %, 13.8 %), D9S242(14.3 %, 21.2 %), D9S43(0, 0). The frequency of LOH showed significant difference among the six microsatellite markers (X2=17.26, P< 0.005; X2=22.66,P<0.005). Conclusion The progression from normal squamous epithelium to high-grade Squamous dysplasia and subsequently to squamous cell carcinoma of the esophagus is associated with accumulation of chromosomal change. The situs of D9S171, D9S162, D9S242, D9S753 exist higher LOH and all exceed 20 %. Possible tumor suppressor genes at or near D9S171, D9S162, D9S242, D9S753 may be related to the progression of esophageal squamous cell carcinoma.
4.Determination of hydrochlorothiazide and rutin in Zhenjujiangya Tablets by HPLC

AIM: To establish a method for determination of hydrochlorothiazide and rutin in Zhenjujiangya Tablets(Flos Chrysanthemi Indici, hydrochlorothiazide, rutin, etc.) by HPLC. METHODS: HPLC was used with Spherisorb C 18 (4.6mm?250mm 5?m) column 0.05mol?L -1 citric acid acetonitrile(80∶20) (adjusted the mixture with triethylaminum to pH 4.1?0.1) was used as a mobile phase. The flow rate was 1.0mL?min -1 . The detection wavelength was at 270nm. RESULTS: The linear response range lay in 10~50?g?mL -1 of hydrochlorothiazide( r =0.9994 n =5) and 40~200?g?mL -1 of rutin( r =0.999 n =5), respectively. The average recoveries of hydrochlorothiazide and rutin were 99.8% and 99.5%, respectively. CONCLUSION: This method is rapid, accurate and suitable for the determination of the two constitutents in the compound tablets.

Objective To investigate the dose-response and its mechanisms of preventive effect of 103 Pd radioactive stent on restenosis in rabbits after balloon injury. Methods The abdominal arteries of the rabbits randomized into common or radioactive stent groups (including 5 Gy, 15 Gy, 25 Gy, 35 Gy groups) were balloon injured, and then stented. Intravascular ultrasound and angiography on abdominal arteries, immunohistochemical study of PCNA, and TUNEL assay were performed 8 weeks after stenting. Results Both the minimal lumen diameters and in-stent lumen areas in 103 Pd radioactive stent groups [(1.94?0.21) mm and (5.99?0.43) mm2, (2.17?0.12) mm and (7.01?0.93) mm2, (2.43?0.22) mm and (8.09?0.89) mm2, (2.56?0.27) mm and (8.49?0.54) mm2 in turn in 5 Gy, 15 Gy, 25 Gy, 35 Gy groups, respectively]were significantly improved with increases of the doses at the end of 8 weeks,revealing a dose-response effect. 7.VEGF enhances serum levels of HGF and IL-6 in rats following 50 percent partial liver transplantation

Objective To determine the effects of exogenous VEGF on the serum levels of HGF and IL-6 in rats following partial liver transplantation.Methods Totally 180 SD rats were subjected to 50 percent partial liver transplantation,then each 60 rats were treated either with VEGF,anti-VEGF or normal saline.HGF,IL-6 and hepatic biochemical markers were assessed at 0,12,24,72,168 h after operation.Results In the VEGF group,the serum levels of HGF and IL-6 were significantly increased as compared with those of anti-VEGF group or control group.Blockage of endogenous VEGF led to a marked increase of ALT and AST.Conclusion VEGF can markedly increase the secretion of HGF and IL-6,which may be related to its enhancing liver regeneration and improving liver function.
8. Effects of angiotensin II inhibitor on HIF-1α expression in the renal tissue of 5/6th kidney ablation/infarction rats

Objective To investigate the influences of Angiotensin II inhibitor on renal expression of HIF-1α mRNA and protein in the chronic renal failure (CRF) rats (induced by 5/6th kidney ablation/infarction \[5/6A/I\]) and the related mechanism. Methods SD rats were made into CRF model by means of 5/6th kidney ablation/infarction. The study was divided into sham operation group (Group A), model group (Group B) and Angiotensin II inhibitor(Cozaar-with-Monopril) group (Group C), each group containing 15 rats. The biochemical parameters of the rats were examined 60 days later in the 3 groups. The expression of HIF-1α mRNA and protein in the remnant renal cortex and medulla were assessed by Real-time PCR and Western blotting analysis. Results Compared with group A, group B had significantly increased SCr and BUN levels and significantly decreased CCr level(P<0.01). Compared with group B, group C had significantly decreased SCr and BUN levels and significantly increased CCr level(P<0.05). The expression of HIF-1α mRNA and protein in the renal cortex and medulla in group B was significantly higher than that in group A(P<0.05), and that in group C was significantly lower than that in group B (P<0.05). Conclusion Angiotensin II inhibitor can improve the renal function in CRF rats induced by 5/6(A/I), which might be associated with HIF-1α signaling pathway.
9.Clinical experience on 5 patients with delayed complications after iatrogenic bile duct injuries

Objective To evaluate the therapeutic methods and the key points in the management of delayed complications of bile duct stenosis after iatrogenic bile duct injuries.Methods Five patients with bile duct injuries developed delayed complications of bile duct stenosis after cholecystectomy were retrospectively studied.All clinical information were derived from the medical data of these 5 patients treated in our department from June 2002 to July 2006.Results Of the 5 patients with delayed complications of iatrogenic bile duct injuries,2 patients developed common bile duct occlusion,2 patients developed hilar bile duct stenosis,and 1 patient developed anastomotic stenosis after cholangiojejunostomy.Bile duct injuries occurred in 3 patients after laparoscopic cholecystectomy,and in another 2 patients after mini-laparotomy-cholecystectomy.The locations of the bile duct injuries,according to Strasberg classification,were type E1 (n=1),type E2 (n=1),type E3,(n=1),and type E5 (n=2).All 5 patients received PTCD initially to relieve obstructive jaundice and then they received surgical exploration and biliary reconstruction.These patients received cholangioplasty at the porta hepatis,and hepaticojejunostomy using a Roux-en-Y anastomosis.Two patients received Roux-en-Y anastomosis of the common hepatic duct to the jejunum.All these patients had good recovery and were discharged well postoperatively.All patients had been followed-up for 7 years or more,with no evidence of obstructive jaundice or biliary tract infection.Conclusions For patients who present with delayed complications of bile duct stenosis after iatrogenic bile duct injuries,primary PTCD is carried out to relieve jaundice and to control infection.Meticulous dissection of the porta hepatis,reliable anastomosis of the proximal healthy bile duct tissues to a jejunal loop,and adequate size of anastomosis guarantee success of surgery.
10.MRI and MRCP Diagnosis of Primary Gallbladder Carcinoma

Objective To analyse MRI and MRCP features of primary gallbladder carcinoma.Methods MRI and MRCP data of 87 cases with gallbladder carcinoma confirmed by operation and pathology were studied retrospectively.Results Four patterns of gallbladder carcinomas were found by MRI:wall thickening (35 cases),papillary nodular(17 cases),mass-type (25 cases) and mixed (wall thickening and nodular) (10 cases).Pathology showed the tumors in combination with chronic cholecystitis and cholecystolithiasis in 69 cases(79.31%).In comparison with that of operations,the accurate diagnostic rates were 46/50(92%) with MR imaging,13/19(68.4%) with MRCP and 80/82(97.6%) in combination with both MR imaging and MRCP.The other features included:hepatic invasion directly by gallbladder cancer in 25/32(78.12%),liver metastases in 17/19(89.47%),lymph node metastases in 28/34(82.35%),intra-adbominal distant metastases in 13/13(100%) and invasion of bile ducts in 25/26(96.15%).Conclusion MRI combined with MRCP is of significant value in diagnosing gallbladder carcinoma.
