ObjectiveTo explore the clinical efficacy of different treatment programs for patients with acute organophosphate poisoning (AOPP).MethodsOne hundred and thirty patients with AOPP were divided into treatment group (80 cases) and control group (50 cases) by table of random digit.The treatment group was given atropine intravenous injection combined with pralidoxime chloride ladder intramuscular injection on the basis of the conventional treatment.The control group was given atropine intravenous injection combined with pralidoxime chloride intravenous drip on the basis of conventional treatment.The healing rate,creatine kinase(CK),aspartate aminotransferase(AST),amylase and C-reactive protein (CRP) were compared between two groups.ResultsThe healing rates of patients with mild,moderate and severe poisoning patients in treatment group were respectively significantly higher than those in control group [ 100.00％(19/19) vs.75.00％(9/12),95.35％(41/43) vs.64.29％(18/28) and 88.89％(16/18) vs.60.00％ (6/10),P ＜0.05].There was no significant difference in CK,AST,amylase and CRP before treatment between two groups(P ＞ 0.05).The CK,AST,amylase and CRP after treatment in treatment group were significantly lower than those in control group [ ( 152.3 ± 23.2) U/L vs.(258.5 ± 22.2) U/L,(37.5 ± 11.4) U/L vs.(44.5± 12.4) U/L,(114.2±43.8) U/Lvs.(147.3 ±61.4) U/L,(5.7±4.1)mg/Lvs.(9.8±5.2)mg/L,P ＜0.05].Conclusions The use of atropine intravenous injection combined with pralidoxime chloride ladder intramuscular injection for mild,moderate and severe AOPP patients is excellent in therapeutic effects,and the clinical cure rates and blood biochemical parameters are more desirable.It is worthy of clinical application.

Objective:To detect expression of SNCG and CXCR4 mRNA in gastric adenocarcinoma and evaluate its function in carcinogenesis, progression, infiltration, and metastasis, as well as the correlation of these proteins. Methods:The expression of SNCG and CXCR4 mRNAs was detected using reverse transcription-polymerase chain reaction in gastric adenocarcinoma and normal gastric mucosa. Results:The expression of SNCG and CXCR4 mRNAs was obviously higher in gastric adenocarcinoma than in normal gastric mucosa (P<0.01). The expression of SNCG mRNA was closely correlated with tumor infiltration depth (P<0.05) and lymph node metas-tasis (P<0.01). The expression of CXCR4 mRNA closely correlated with lymph node metastasis (P<0.01). The expression of SNCG and CXCR4 mRNAs positively correlated in 29 cases of gastric adenocarcinoma (r=0.346, P<0.05). Conclusion:SNCG and CXCR4 may be overexpressed in tumor tissue and may have important functions in tumorigenesis, progression, infiltration, and metastasis of gastric adenocarcinoma.

Objective To compare the characteristics between two kinds of 40Hz auditory evoked potentials and provide suitable index for clinical auditory test of low-frequency. Methods The potentials including 40Hz auditory event related potential of middle latency response(40Hz AERP-MLR) and 40Hz auditory event related potential of early latency response(40Hz AERP-ELR) from window and vertex were recorded in guinea pigs. Results The common characteristics of the potentials were good frequency response,easy identification and lower responsive thresholds.Amplitude of 40Hz AERP-ELR was higher and threshold was lower than 40Hz AERP-MLR,especially 0.5kHz tone burst,which was (14.15?6.06)dBnHL.In vertex,the threshold of 40Hz AERP-MLR was (43.50?9.65)dBnHL,which was higher than that in round window(P

Objective:To explore the way to separate and culture eutopic and ectopic endometrial glandular cells and their stromal cells,providing an in vitro cell model of endometriosis for study of its mechanism. Methods:Digestion,filtration and sedimentation were used to isolate and culture the eutopic and ectopic endometrial glandular cells and their stromal cells. The estrogen level was imatated to study the way of promoting cell growth. Morphological characters of eutopic and ectopic endometrial cells were examined using optical microscope. Results:The success rate of separation and culture of normal control endometrial glandular cells and its stromal cells was 91.7%(11/12);of eutopic endometrial glandular cells and its stromal cells of endometriosis was 93.8%(15/16);of etopic endometrial glandular cells and its stromal cells of endometriosis was 75.0%(12/16) . Conclusion:The cultured eutopic and ectopic endometrial cells is more like human body feature than the endometriosis model of animals. So the isolation and culture of eutopic and ectopic endometrial glandular cells and their stromal cells may serve as an in vitro experimental model.

Objective:To investigate the expression of C-X-C chemokine ligand-14 (CXCL14) and epidermal growth factor receptor (EG-FR) in human gastric cancer and to analyze the relationship of CXCL14 with clinicopathological features. Methods:The expression of CXCL14 and EGFR was detected by Immunohistochemical SP method in 121 cases of gastric cancer tissue, 62 cases of the adjacent non-tumor gastric mucosa, and 60 cases of allogeneic non-tumor gastric mucosa. Results:The positive rates of CXCL14 and EGFR expres-sion were 80.17%and 48.76%in gastric cancer, respectively, and both were significantly higher in the adjacent non-tumor gastric mu-cosa and allogeneic non-tumor gastric mucosa. The differences were significant (P<0.01). Overexpression of CXCL14 was closely corre-lated with the depth of cancer invasion, differentiation, and clinical stage, and the differences were significant (P<0.05). Overexpres-sion of EGFR was correlated with cancer differentiation, lymph node metastasis, and clinical stage. The differences were significant (P<0.05). Based on the Spearman correlation analysis, the expression of CXCL14 and EGFR in gastric cancer was positively correlated (rs=0.195, P<0.05). Conclusion:Abnormal CXCL14 expression in gastric cancer may be associated with the occurrence and development of gastric cancer. CXCL14 expression is positively correlated with EGFR expression, suggesting that the two have a synergistic effect in gas-tric cancer development.

OBJECTIVE: To explore the method of primary culture for endometriotic cells and to find out the differences in morphological manifestations among endometriotic cells and eutopic endometrial cells sampled from patients with endometriosis and endometriosis-free women. METHODS: Endometriotic and eutopic endometrial cells were cultured by modified method of primary culture. The endometriotic cell types were observed and differentiated under optical and electron microscopes. RESULTS: The success rates for culture of eutopic endometrial cells from endometriosis-free women and patients with endometriosis were 91.67% and 93.75% respectively. The success rate for culture of endometriotic cells was 75.00%. The size of endometriotic glandular cells was similar to those of eutopic endometrial glandular cells from endometriosis-free women and patients with endometriosis. The chromatin was manifold and the nucleus was augmented in the endometriotic glandular cells. The endometriotic stromal cells were smaller than the eutopic endometrial stromal cells from endometriosis-free women and patients with endometriosis. Many tiny villi and protuberances on plasma membrane could be seen in the endometriotic stromal cells. CONCLUSION: The success rate for culture of endometriotic cells can be elevated through improving the method of primary culture. The ultrastructures of endometriotic glandular and stromal cells are obviously different from those of eutopic endometrial glandular and stromal cells from endometriosis-free women and patients with endometriosis.

Objective To study the changes of negative symptoms in PCP-induced schizophrenia rat model.Methods Thirty newborn female SD rats randomly divided into control group,PCP-week 6 group and PCP-week 10 group( n=10 in each group).Perinatal rat treated with PCP ( 10 mg/kg) on postnatal days 7,9 and 11(10 mg/kg,ip),and sucorse intalce test(SIT),forced swimming test(FST) and resident-intruder test(RIT) were used to test the emotional and negative symptoms.Results In the SIT,there was no difference between control and PCP groups (con:(28.24 ±0.86) ml/kg; week 6:(26.57 ± 1.01 ) ml/kg; week 10:(27.98 ±0.99) ml/kg,F =12.35,P ＞ 0.05 ).In the FST,PCP model rats showed longer still time ( con:(39.32 ± 1.98 ) s ; week 6:(52.39 ± 1.66)s,week 10:(55.56 ± 1.49)s,F=3.99,P＜ 0.05 ).In the RIT,PCP models rats showed less explore time ( (40.31 ± 13.56)s vs (63.90 ± 13.12)s,(43.65 ±12.86 )s vs (65.18 ± 15.12)s,P ＜ 0.05 ) and more escape time ((19.33±2.26) s vs (9.26 ± 1.32) s,(17.79 ±2.99) s vs (9.38 ± 1.36) s,P＜ 0.05).Conclusion Perinatal PCP injection can induce the long-lasting negative-symptoms changes.

Objective To investigate the mechanism underlying the WNK4 kinasemediated inhibitory effect on BK channel. Methods Cos-7 cells were cotransfected with BK in combination with either CD4 (control group) or wild type WNK4 (WNK4-WT).Immunostaining and confocal microscopy,chemiluminescence,Western blotting analysis were then employed to determine the BK localization in cells,BK surface expression and total protein level,respectively.To further investigate whether the reduction of BK protein expression is due to an increase in degradation through a lysosomal pathway,BK protein level was determined after treated with bafilomycin A1(Baf A1),a proton pump inhibitor affecting lysosomal degradation. Results Immunostaining and confocal microscopic study showed that BK was localized both in plasma membrane and cytosol in the control group.After cells transfected with WNK4-WT,BK expression was markedly reduced.Chemiluminescent assay found that BK surface expression level was 299.9±18.6 in the control group,whereas it was significantly reduced (148.4±13.7,P＜0.01) in the WNK4-WT group.Western blotting analysis showed that total BK protein level was markedly reduced in the presence of WNK4-WT compared to the control group.WNK4-WT was shown to significantly reduce the BK total protein level (42.3％±15.2％) compared to the control group (100％) (P＜0.01).When the cells was treated with Bafilomycin A1 (Baf A1,0.5 μmol/L),WNK4-mediated reduction in BK protein was reversed (82.2％±12.1％,P＜0.05). Conclusions WNK4 inhibits total and surface protein expression of BK in Cos-7 cells whick is likely due to an increase in BK degradation through a lysosomal pathway.

Background and purpose: Multidrug resistance (MDR) is the dominating obstacle to the chemotherapy. There is strong evidence that the phosphoinositide 3-kinases (PI3Ks) signaling pathway is involved in MDR phenotype, however, the mechanism of MDR occurrence is still unknown. This study tended to investigate the regulating effect of PI3K/Akt signaling pathway and its downstream target genes in P-glycoprotein (P-gp) (ABCB1 gene encoding)-mediated MDR in human colon carcinoma HCT-116/L-OHP cells. Methods:Pretreatment with PI3K selective inhibitor LY294002 (20μmol/L) for 2 h, the sensitivity of L-OHP was evaluated by the CCK-8 (cell counting kit-8) assay in HCT-116/L-OHP cells, and the expressions of P-gp, LRP, MRP-2, Akt, p-Akt, IκB and p-IκB were evaluated by Western blot. The activity of ABCB1 promoter was evaluated by chromatin immunoprecipitation analysis (CHIP). Results: After inhibiting the activity of PI3K/Akt signaling pathway, the IC50 value of L-OHP decreased from(157.48±16.73)μg/mL to (53.68±3.18)μg/mL in HCT-116/L-OHP cells, and the reversal index was 2.93 (P<0.01). The expressions of P-gp, p-Akt and p-IκB were down-regulation compared with the concrol group (P<0.01), but the expressions of LRP, MRP-2, Akt and IκB didn't change signiifcantly. CHIP result has conifrmed that NF-κB protein could bind to the region of ABCB1 gene promoter in HCT116/L-OHP cells. Conclusion:Blocking of PI3K/Akt/NF-kB signal pathway could increase the drug sensitivity to MDR cells, inhibit the phosphorylation of p-Akt and p-IκB, and reversing ABCB1/P-glycoprotein-mediated multidrug resistance in colon carcinoma cells.
