1.Observation of Relationship between Ovarian Arterial Blood Flow Velocity and Follicular Growth by Transvaginal Color Dopper Imaging

The relationship of the flow velocity of ovarian artery to the follicular growth and genital endocrine hormone was observed in 36 normal and 32 infertile women by transvaginal color Doppler imaging(TVCD). The result showed that before and after ovulation, the peak flow velocity occurred in 87 % normal women i the diestolic flow waveforms had high amplitude with low impedance, the diameters of the follicles were large, the blood E2 and LH increased and attained the peak values. However, during the time of early follicular development and late stage of luteum, the diastolic flow waveforms had low amplitude with high impedance and the blood levels of E2 and LH were low. This suggests that a close relationship exists in the flow velocity and waveform of ovarian artery to the follicular development and genital endocrine hormone. In most cases of the infertile group, the diastolic flow of ovarian arteries showed with low amplilude with high impedance > there were no mature follicle with irregular change of ovarian steriods. The study indicats that this technique may forecast the time of ovulation and investigate the causes of infertility.
2.Using humanized nursing for fiver cancer patients by transcatheter hepatic artery chemobolization

Objective In order to know the application effects of transcatheter hepatic artery chemoembolization for patients with liver cancer. Methods Divided 144 liver cancer patients into the experimental group and the control group randomly, there were 82 cases in each group. Routine nursing cares were used in the control group, humanized nursing were used in the experimental group in addition. Compared the curative effect, incidence rate of complication and scores of SAS and SDS between the two groups. Results The curative effect in the experimental group was significant better than in the control, the incidence rate of complications were lower in the experimental group than those of in the control group. Before out of hospital, the SAS and SDS level in the experimental group was better than in the control group. Conclusions Humanized nursing can enhance the curative effec4 avoid disputs between patients and nurses, and then promote nurses' working interesting.
3.The Microsurgical Anatomical Study of transcerebellomedullary fissure approach

Objective To study transcerebellomedullary fissure approach to the fourth ventricle and the lateral brainstem without splitting vermis and depict its adjacent structure, and provide clinicians with anatomical information.Methods Five cadaveric heads fixed with formaldehyde and perfused with color latex in their arteries and veins were dissected with microsurgical anatomical skills. The composition of cerebellomedullary fissure and the distribution of the tela choroida, the inferior medullary velum and the posterior inferior cerebellar artery, and study anatomic base of this approach.Results Cerebellomedullary fissure is a natural anatomical gap which is located between the cerebellar tonsil, biventral lobule and medulla oblongata. The tela choroida and inferior medullary velum do not contain nerve tissue, and thus can be dissected to expose the fourth ventricle. The tonsil- medulla segment and telovelotonsillar segment of the posterior inferior cerebellar artery are the most important blood vessels of the cerebellomedullary fissure approach.Conclusions The transcerebellomedullary fissure approach can reach the fourth ventricle and dorsal brain stem through normal anatomic spaces without splitting the vermis, thus reducing the post-operative complications.
4.Effect of moutan cortex extract on Th17 cells of patients with systemic lupus erythematosus

Objective To study the effect of moutan cortex extract on Th17 cells of systemic lupus erythematosus (SLE) patients, so as to provide reference for clinical practice.Methods A total of 168 SLE patients admitted to our hospital from March 2012 to August 2013 were selected as subjects, andrandomized into the observation group and control group.84 patients in observation group received moutan cortex extract treatment and 84 patients in control group received conventional medical treatment.The Th17 and Th1 cell percentage of two groups before and after treatment were compared, and the incidence of adverse reactions were observed.Results All the indicators before treatment showed no statistically significant differences between two groups.After treatment, the Th17 cell percentage of observation group decreased to (0.27 ±0.15)%, and the Th1 cell percentage increased to (2.53 ± 1.72)%, showing more significant changes than control group (P<0.05).The reduction of IL-6 level, SLEDAI score and ESR after treatment were more significant in observation group than in control group(P<0.05).The incidence of adverse reactions was lower in observation group than in control group ( P <0.05 ) .Conclusion Moutan cortex extract can effectively improve the condition of SLE through reducing the Th17 cell percentage and increasing Th1 cell percentage with less adverse reactions.
5.Ten-year risk of ischemic cardiovascular diseases in middle-aged adults in Henan province

Objective To evaluate 10-year risk of ischemic cardiovascular diseases (ICVD) in middle-aged adults in Henan province to provide evidence for improved health status. Methods The 10- year risk of ICVD in 12 064 middle-aged adults was evaluated by using simplified risk estimation model. Results About 96. 35% of male and 98. 29% of female had a probability of < 10% to develop ICVD over 10 years; 3.65% of male and 1.71% of female had a probability of ≥ 10% to develop ICVD over 10 years; and 0.68% of male and 0.23% of female had a probability of≥20% to develop ICVD over 10 years. The prevalence of risk factors for ICVD in two absolute risk groups ( ≥10% vs < 10% ) was significantly different. Conclusions The risk of ICVD in the middle-aged residents of Henan province may be similar to that in the middle-aged Chinese adults. The risk assessment for ICVD could be used for high- risk populations.
6.Structure and function of a novel thermostable pullulanase.

Research on novel pullulanase has major significance on the domestic industrialization of pullulanase and the breakdown of foreign monopoly. A thermophilic bacteria LM 18-11 producing thermostable pullulanase was isolated from Lunma hot springs of Yunnan province. It was identified as Anoxybacillus sp. by 16S rDNA phylogenetic analysis. Full-length pullulanase gene was cloned from Anoxybacillus sp. LM18-11. The optimum temperature of the pullulanase was between 55 and 60 degrees C with a half-life as long as 48 h at 60 degrees C; and its optimum pH was between 5.6 and 6.4. V(max) and K(m) of the pullulanase was measured as 750 U/mg and 1.47 mg/mL, which is the highest specific activity reported so far. The pullulanase crystals structure showed a typical alpha-amylase family structure. The N-terminal has a special substrate binding domain. Activity and substrate binding were decreased when the domain was deleted, the V(max) and K(m) were 324 U/mg and 1.95 mg/mL, respectively. The pullulanase was highly heterologous expressed in Bacillus subtilis by P43 promoter. The extracellular enzyme activity was 42 U/mL, which increased more than 40 times compared to the initial strain. This pullulanase has good application prospects.

7.Screening of specific microRNAs and bioinformatics analysis on cardiomyocytes in rat with heart failure

Objective To investigate microRNAs ( miRNAs) expression profiling of cardiomyocytes in rats with heart failure, and predict miRNAs-regulated target genes and their functions.Methods Total of 18 male SD rats weighing 200-220 g were randomly divided into 2 groups:the control group ( CON) and the heart failure group (HF).The rats in HF group were injected by adriamycin via tail vein to induce heart failure, meanwhile in CON group, rats were received an equal volume of 0.9% sodium chloride intravenously.The cardiomyocytes isolated from the rat hearts in two groups and cultured overnight.After that, total RNA was extracted and then subjected to miRNA microarray to screen differentially expressed miRNAs.The reults of microarray were further verified by quantitative real-time PCR ( qRT-PCR ) .The target genes regulated by differentially expressed miRNAs were predicted by the software of Targetscan and miRanda.Bioinformatics analysis was performed to predict the miRNAs-regulated target genes and analyze the enriched gene ontology ( GO) and signaling pathway ( KEGG Pathway) .Results The results of miRNA microarray showed that a total of 37 miRNAs were differentially expressed in HF group as compared to CON group, among which 22 miRNAs were up-regulated and 15 miRNAs were down-regulated (P<0.01, FDR<0.05).The expression of miR-133b-5p (t=14.56, P<0.01), miR-6216 (t=9.32, P<0.01) and let-7e-5p (t=13.92, P<0.01) which were detected by qRT-PCR exhibited the similar tendency of up or down regulation to those shown in microarray results.Bioinformatics analysis indicated that miRNAs-regulated target genes were significantly enriched in 31 GOs (P<0.01, FDR<0.05) and 12 signal pathways (P<0.05, FDR<0.05), among which ubiquitin-proteasome system, MAPK signaling pathway and Toll like siganling pathway exhibited a higher enrichment. Conclusion MiRNA expression profile on cardiomyocytes in rat with adriamycin-induced heart failure was significantly changed.These differentially expressed miRNAs might participate in the process of heart failing by regulating their target genes in rat cardiomyocytes.
8.Effect of intrathecal morphine preconditioning on expression of nerve growth factor in dorsal root ganglia in a rat model of myocardial ischemia-reperfusion

Objective To investigate the effect of intrathecal morphine preconditioning on the expression of nerve growth factor (NGF) in the dorsal root ganglia (DRG) in a rat model of myocardial ischemia-reperfusion (I/R).Methods Thirty healthy adult male Sprague-Dawley rats in which intrathecal catheters were successfully placed without complications,weighing 250-350 g,were randomly divided into 5 groups (n =6 each) using a random number table:sham operation group (S group),I/R group,intrathecal morphine preconditioning group (ITMP group),μ receptor antagonist CTOP + intrathecal morphine preconditioning group (CTOP + ITMP group),and CTOP control group (CTOP group).Myocardial ischemia was induced by 30 min of occlusion of the anterior descending branch of the left coronary artery followed by 120 min of reperfusion in all the groups except S group.Intrathecal morphine preconditioning was produced by 3 cycles of 5 min intrathecal injection of morphine 3 μg/kg (10 μl) at 5 min intervals within 30 min before ischemia in ITMP group.In CTOP+ITMP and CTOP groups,1 μg/μ1 CTOP 10 μl was injected intrathecally at 10 min before morphine preconditioning and 40 min before ischemia,respectively.At 120 min of reperfusion,the rats were sacrificed,and myocardial specimens were obtained for determination of myocardial infarct size,and DRGs were removed for determination of the expression of NGF by using immunohistochemistry and Western blot.Results Compared with S group,the myocardial infarct size was significantly increased,and the expression of NGF in DRGs was significantly up-regulated in I/R group (P<0.05).Compared with I/R group,the myocardial infarct size was significantly decreased,and the expression of NGF in DRGs was significantly down-regulated in ITMP group (P< 0.05),and no significant change was found in the parameters mentioned above in CTOP group (P>0.05).Compared with ITMP group,the myocardial infarct size was significantly increased,and the expression of NGF in DRGs was significantly up-regulated in CTOP+ITMP and CTOP groups (P<0.05).Conclusion The mechanism by which intrathecal morphine preconditioning reduces myocardial I/R injury is related to activation of spinal μ receptors,inhibition of NGF expression in DRGs,and reduction of responses to noxious stimulation in the rats.
9.Role of vagus nerve-muscarinic cholinergic receptor pathway in mitigation of myocardial ischemia-reperfusion injury by intrathecal morphine postconditioning in rats

Objective evaluate the role of vagus nerve?muscarinic cholinergic receptor ( M recep?tor) pathway in mitigation of myocardial ischemia?reperfusion (I∕R) injury by intrathecal morphine postcon?ditioning in rats. Methods Seventy adult male Sprague?Dawley rats in which intrathecal catheters were suc?cessfully placed without complications, weighing 250-350 g, were randomly assigned into 7 groups ( n=10 each) using a random number table:sham operation group (Sham group), I∕R group, intrathecal morphine postconditioning group ( MP group) , vagal transection ( VT) group, VT+ intrathecal morphine postcondi?tioning group (VT+MP group), atropine (ATP, M receptor antagonist) + morphine postconditioning group ( ATP+MP group) , and ATP group. Myocardial I∕R was produced by occlusion of the anterior descending branch of left coronary artery for 30 min followed by 2 h of reperfusion. Morphine ( 3μg∕kg, 10μl) was in?trathecally infused over 5 min starting from onset of reperfusion in MP group. Normal saline 10 μl was in?trathecally infused over 5 min starting from onset of reperfusion in NS group. The bilateral vagus nerves were transected at 10 min before reperfusion in VT+MP group. Atropine ( 0?1 mg∕kg, 0?5 ml) was intravenously infused over 5 min starting from 10 min before reperfusion in ATP+MP group. The occurrence of cardiac ar? rhythmia ( premature ventricular contractions ( PVCs) and ventricular tachycardia ( VT)∕ventricular fibrilla?tion ( VF) ) within the first 30 min of reperfusion was recorded. The rats were sacrificed at 120 min of reper?fusion, and myocardial specimens were obtained for determination of myocardial infarct size ( IS) as a per?centage of area at risk (AAR). IS∕AAR ratio was calculated. Results Compared with Sham group, the number of PVCs and VT∕VF and IS∕AAR ratio were significantly increased in the other groups. Compared with I∕R group, the number of PVCs and VT∕VF and IS∕AAR ratio were significantly decreased in MP group. Compared with MP group, the number of PVCs and VT∕VF and IS∕AAR ratio were significantly in?creased in VT+MP and ATP+MP groups. Conclusion Vagus nerve?M receptor pathway is involved in miti?gation of myocardial I∕R injury by intrathecal morphine postconditioning in rats.

ITMP were produced by three cycles of 5 min intrathecal injection of morphine and 5 min intermis-sion before myocardial ischemia, CON were achieved by intrathecal injection of saline in the same way, L-NAME+ITMP, ODQ +ITMP, KT5823 +ITMP were prepared by intrathecally administering L-NAME ( 30 nmol), ODQ(11 nmol) and KT5823(20 pmol) 10 minutes prior to ITMP respectively, L-NAME, ODQ, KT5823 worked as the control of inhibitors themselves respectively without ITMP. Subsequently, all rats were subjected to 30 min of left coronary artery occlusion followed by 2 h of reperfusion except the SHAM group. Myocardial infarct size, as a percentage of the AAR,
was determined by 2 , 3 , 5-triphenyltetrazolium stai-ning. Results Compared with CON, the volumes of IS and IS/AAR were reduced in ITMP ( P <0.01 );the protective effects of ITMP were abolished by pre-treatment with L-NAME, ODQ and KT5823 ( P <0.01 );Conclusions NO/cGMP might be involved in the cardioprotective effect of intrathecal morphine pre-conditioning against myocardial ischemia and reperfu-sion injury in rats.
