1.Prognosis of patients with diffuse axonal injuries following continuous lumbar cistern drainage

Objective To observe influence of continuous lumbar cistern drainage on levels of amyloid β-peptide (Aβ) subtype (Aβ1-42) in cerebrospinal fluid and plasma in patients with diffuse axonal injury (DAI) and investigate its clinical significance.Methods Eighty-one DAI patients were enrolled and randomized into treatment group (42 cases) and control group (39 cases).Patients in control group received simple conventional therapy,while the patients in treatment group received not only conventional therapy but 14 days of continuous lumbar cistern drainage.Levels of Aβ1-42 in cerebrospinal fluid and plasma were detected by ELISA assay before therapy and at 1,5,9,and 14 days after therapy.Prognosis was assessed using GOS at 6 months after therapy.Results Levels of Aβ1-42 in cerebrospinal fluid and plasma showed a decrease in the first place and a gradual decrease afterwards in both groups,but a bigger and earlier drop of Aβ1-42 levels was observed in treatment group.Two groups showed significant difference of Aβ1-42 levels at day 14 (P < 0.05).At 6 months after therapy,GOS score between treatment and control groups was (4.1 ± O.5) and (3.4 ± 0.3) points respectively (P <0.05).Conclusion Continuous lumbar cistern drainage improves the prognosis of DAI and this may relates to the decrease of Aβ1-42 levels in cerebrospinal fluid and plasma.
2.Resting state functional magnetic resonance imaging changes in spasmodic torticollis patients based on fractional amplitude of low frequency fluctuation

Objective To observe the differences of cerebral activation pattern with resting state functional magnetic resonance imaging (rs-fMRI) between patients with spasmodic torticollis (ST) and healthy controls,thus to investigate the pathogenesis of ST.Methods Nineteen ST patients and 21 age,sex and education-matched healthy controls,recruited from the Department of Neurology,First Affiliated Hospital of Guangxi Medical University between November 2012 and January 2016,were included in this study.rs-fMRI and factional amplitude of low frequency fluctuation (fALFF) were used to obtain differences between patients with ST and healthy controls,and correlative analysis was made on fALFF values of abnormal brain regions and ST patients' symptom severity (Tsui scores).Results Compared with healthy controls,patients with ST had significantly increased fALFF in the left cerebellum and significantly decreased fALFF in the left posterior cingulate cortex/precuneus,right posterior cingulate cortex/precuneus,left middle temporal gyrus,right angular gyrus,left post-central gyrus,right supplementary motor area (t =-5.714-5.920,P <0.01),and abnormal brain regions' fALFF values had no correlation with patients' age of onset,disease course,symptom severity (P > 0.05).Conclusion Abnormal sensorimotor area,default mode network and cerebellum dysfunction may play a role in the pathophysiology of ST.
3. Clinical efficacy of injected autologous fat and buried polydioxanone threads for nasolabial fold filling

Objective:
To investigate the clinical application of injected autologous fat and buried polydioxanone threads for nasolabial fold filling.
Methods:
From April 2016 to October 2017, 64 cases of mild and moderate nasolabial fold beauty seekers from the First People's Hospital of Jiande were divided into group A and group B with their attitude. Group A (28 cases) were only treated with buried polydioxanone threads for nasolabial fold filling; Group B (36 cases) were treated with injected autologous fat and buried polydioxanone threads for nasolabial fold filling. The surgical effect was recorded for 18 months.
Results:
The most common complications were swelling, bruising and pain immediately after the surgery. Polydioxanone threads exposure occurred in two cases in each group 2 weeks after the surgery. There were satisfactory results in skin color and skin texture in both groups. 9 cases showed inadequate correction in group A while only 2 cases showed inadequate correction in group B in long-term follow-up (>3 months).
Conclusions
Combined treatment of autologous fat and polydioxanone threads is safe and effective for nasolabial fold filling.
4.The therapeutic effects of two different ways of autologous bone marrow mesenchymal stem cells transplantation for treating early spinal cord injury

ObjectiveTo compare the short-term efficacy and safety of autologous bone marrow mesenchymal stem cells transplantation on patients with early spinal cord injury by subarachnoid injection and by intravenous injection.MethodsNinety-six patients with early spinal cord injury were hospitalized and treated from November 2006 to March 2010.Thirty-eight cases (subarachnoid transplantation group) got transplantation by subarachnoid injection,32 cases (intravenous transplantation group) got transplantation by intravenous injection,26 cases (control group) were hospitalized in the same period but not transplanted.The motor and sensory functions of all three groups were evaluated according to the score standard developed by American Spinal Injury Association(ASIA) before treatment and at the first,the third,the sixth month after treatment.Meanwhile,routine blood test,coagulation,biochemical items and tumor markers were also examined in follow-up.ResultsThe motor and sensory function of three groups had different degree of recovery at the first month after treatment,and sensory function recovered muchsignificantly,but the comparison among three groups had no statistical significance.The scores of motor function increased in three groups at the third month after treatment,but still had no statistical significance (P> 0.05).The scores of sensory function of subarachnoid transplantation group[(130.9 ±41.6) scores] and intravenous transplantation group [ (131.2±22.7 ) scores ] increased obviously,and had significant difference compared with control group [ (109.3±36.4) scores] (P < 0.05),but there were nosignificant difference between subarachnoid transplantation group and intravenous transplantation group (P > 0.05).The scores of sensory and motor function of control group didn't increase obviously at the sixth month after treatment,while the scores of subarachnoid transplantation group and intravenous transplantation group gradually improved and had statistical significance compared with control group(P < 0.05).The scores of sensory function was higher in subarachnoid transplantation group[ (151.6±46.9) scores ] than that in intravenous ransplantation group [(134.6 ±40.7) scores] (P <0.05).There were no obvious abnormality in the results of followed-up examination.Conclusions The safety and short-term efficacy of autologous bone marrow mesenchymal stem cells transplantation in treating early spinal cord injury by subarachnoid injection and intravenous injection is certified.The subarachnoid injection is better than intravenous injection,but the long-term efficacy need furter study.
5.Demographic and clinical features of hereditary nonpolyposis colorectal cancer

Objective To investigate demographic and clinical features of hereditary nonpolyposis colorectal cancer (HNPCC) in Chinese patients.Methods Data of 45 HNPCC pedigrees diagnosed in accordance with Armsterdam I and Armsterdam Ⅱ Criteria were collected and analyzed.Results In 264 patients, 305 malignant neoplasms were found.The median age of confirmed diagnosis of HNPCC was 50 years.Out of the 305 primary cancer foci, 180 were colonic cancers (59%), 125 were extracolonic cancer (41%) with carcinoma of the lung, endometrium and stomach accounting for 24.0% (30/125), 14.4% (18/125), and 12.0% (15/125) respectively.Conclusion HNPCC is characterized by early vertical transmission, with extracolonic cancer, and synchronous multiple and/or metachronous multiple primary cancers as common clinical features. Carcinoma of the lung, endometrium and stomach are among the most common extracolonic malignancies.
6.Effects and Its Mechanism of Different Dental Alloys on Proliferation and Apoptosis of Fibroblast

Objective To investigate the effects and its mechanism of different dental alloys on proliferation and apoptosis of fibroblast.Methods Mouse fibroblast L929 cells were divided into negative control group,gold alloy group,Ni Cr alloy group and copper alloy group.Cell proliferation was detected by MTT assay.Cells were cultured for 48h.Cell cycle and apoptosis rate were detected by flow cytometry.The expression of Bcl-2,Bax,cleaved caspase-3 protein were detected by Western blot.Results Cell proliferation rate had no significant difference in gold alloy group and Ni Cr alloy group and negative control group (P > 0.05),while was significantly lower in copper alloy group than the negative control group (P < 0.01).G1 phase,S phase and G2 phase cells between gold alloy group and the negative control group had no significant difference (P > 0.05),while G1 phase cells in Ni Cr alloy and copper alloy group was significantly lower than negative control group.S phase and G2 phase cells were significantly higher than the negative control group (P < 0.05 or P <0.01).Cell apoptosis rate and the expression of Bcl-2,Bax,cleaved caspase-3 protein between gold alloy group and negative control group had no significant difference (P > 0.05).Cell apoptosis rate and the expression of Bax,cleaved caspase-3 protein between Ni Cr alloy and copper alloy group were significantly higher than the negative control group.Bcl-2 protein expression was significantly lower than the negative control group (P < 0.01).Conclusion The gold alloy,Ni Cr alloy and copper alloy can influence proliferation and apoptosis of fibroblast.The gold alloy group is minimum.Different dental alloys leads to the possible mechanism of fibroblast apoptosis was regulation of Bcl-2,Bax,cleaved caspase-3 protein expression.
7.Prevalence of comorbidity of hypertension, diabetes and dyslipidemia and the association between comorbidity and cardiovascular mortality in population aged 40 years and over in Liaoning Province

Objective:To investigate the comorbidity status of hypertension, diabetes, and dyslipidemia (the"three diseases") among residents aged≥40 in Liaoning Province, and to explore the correlation between the comorbidity and cardiovascular disease mortality.Methods:This investigation was a prospective cohort study. From February 2017 to March 2019, a multi-stage stratified cluster random sampling method was used to carry out a baseline survey of 18 758 permanent residents aged≥40 years in Liaoning Province. Demographic information and history of hypertension, diabetes, and dyslipidemia were collected and followed up every year. Death was mainly identified by linkage to the Population Death Information Registration Management System. Cox proportional hazard regression model was used to analyze the association between the comorbidity of the "three diseases" and cardiovascular disease mortality risk.Results:A total of 18 758 residents aged≥40 in Liaoning Province were included, with an age of (60.3±9.9) years and 7 325 males (39.1%). The comorbidity rate of hypertension, diabetes, and dyslipidemia was 6.7% (1 256/18 758), and the standardized prevalence rate was 5.4%. The comorbidity rate increased with age (P<0.001), which was higher in women than in men, and more significant in urban areas than in rural areas (all P<0.001). The comorbidity of "three diseases" accounted for 39.3% (1 256/3 198), 18.7% (1 256/6 710), and 11.8% (1 256/10 653) in patients with diabetes, dyslipidemia, and hypertension, respectively. With a follow-up of (4.3±0.6) years, 463 people died of cardiovascular disease. The mortality rate of cardiovascular disease in the comorbidity of hypertension, diabetes, and dyslipidemia was 8.74/1 000 person-years. After adjusting potential confounders, Cox proportional hazard regression model analysis showed that compared with normal individuals, the hazard ratio of cardiovascular disease mortality in patients with the "three diseases" was 2.55 (95% CI: 1.63-3.99). Conclusion:The prevalence of comorbidity of hypertension, diabetes, and dyslipidemia among residents aged≥40 in Liaoning Province was relatively high, and the risk of cardiovascular disease mortality in patients with the "three diseases" was increased.
8.Clinical effects of modified orbital septum fat release on lower blepharoplasty with lacrimal groove deformity

Objective:To observe the clinical effect of modified orbital septum fat release in the treatment of lower eyelid relaxation with lacrimal groove deformity.Methods:From January 2019 to August 2023, 52 patients with lower eyelid relaxation and lacrimal groove deformity were treated with the procedures of modified orbital septum fat release in the First People′s Hospital of Jiande, including 5 males and 47 females. The orbicularis oculi muscle supporting ligament was sutured and fixed at the periosteum of the lower edge of the orbit to achieve anatomical reduction. Suture fixation of the free edge of the orbital septum was performed at the edge of the supporting ligament in the modified orbital septum fat release method. Following-up for 6-12 months after surgery, the complications and satisfactory rate of patients were evaluated.Results:All the incision healed well with improved skin effect, and the incision scar were almost invisible. 52 patients were followed up from 6 to 12 months; 49 patients were satisfied and 2 patients were basically satisfied with their appearance, 1 patient was not satisfied with the result. No complications such as lower eyelid ectropion, lower eyelid retraction, or eyelid ball separation occurred.Conclusions:Modified orbital septum fat release is an effective method for correction of lower eyelid relaxation with lacrimal groove deformity.
9.Effect of TSG-6 mRNA demethylation induced by FTO on keloid

Objective:
To investigate the effect of demethylase obesity-associated protein(FTO) on the expression of tumor necrosis factor-α-stimulating gene-6(TSG-6) mRNA demethylation in keloid tissue and its role in scar formation.
Methods:
Fresh keloid tissue(experimental group) and normal skin tissue(control group) were randomlyselected, and the expression level of TSG-6 in the experimental group and control group was detected by immunofluorescence method. A probe expressing TSG-6 mRNA sense and antisense chain were constructed for RNA pulldown, followed by Western blot detection of samples in each group with N6-methyladenine(m6A) antibody. Real-time fluorescence quantitative polynucleotide chain reaction(qPCR) and Western blot were used to detect the expression of TSG-6 mRNA demethylase and methylase in the experimental group and control group. The expression levels of FTO in experimental group and control group were detected by immunofluorescence assay.
Results:
Immunofluorescence results showed that the expression level of TSG-6 in the experimental group was lower than that in the control group(P<0.01). Pulldown test results of biotin-labeled TSG-6 RNA showed that the m6A expression level of TSG-6 mRNA in the experimental group was lower than that in the control group(P<0.01). The results of qPCR showed that the expression level of FTO mRNA in the experimental group was higher than that in the control group(P<0.01). Western blot analysis showed that FTO protein expression in the experimental group was higher than that in the control group(P<0.001). Immunofluorescence results showed that the expression level of FTO in experimental group was higher than that in control group(P<0.001).
Conclusion
The demethylase FTO catalyzes the demethylation of TSG-6 mRNA and promotes the formation of keloid, which may provide a new idea and direction for clinical treatment of keloid.
