1.Effects of Compound Malt Pill on Gene Expressions of Androgen Receptor and Insulin Receptor in Polycystic Ovarian Syndrome Rat Models

Objective To explore the effects of Compound Malt Pill on gene expressions of androgen receptor (AR) and insulin receptor (INSR) in polycystic ovarian syndrome (PCOS) rats; To discuss its relevant mechanism of action.Methods Letrozole was used to induce and establish rat models. Rats were randomly divided into normal group, model group, Diane-35 group, low-, medium- and high-dose Compound Malt Pill groups. The normal group and model group received normal saline by gavage; the Diane-35 group was given Diane-35 by gavage; Compound Malt Pill groups were respectively given different concentrations by gavage for 21 d. The levels of serum testosterone (T) and insulin were measured by ELISA, and the gene expressions of AR and INSR were measured by RT-PCR. Results Compared with normal group, the serum T, insulin and the gene expressions of AR and INSR in model group increased significantly (P<0.05,P<0.01); compared with model group, the serum T, insulin and the gene expressions of AR and INSR in low-, medium- and high-dose Compound Malt Pill groups decreased significantly (P<0.05, P<0.01).Conclusion Compound Malt Pill can adjust endocrine-metabolic disorder in PCOS rats.
2.Isokinetic training can improve endurance and walking capacity after incomplete lumbar spinal cord injury

Objective To explore the effects of isokinetic training on muscle endurance and the ambulatory capacity of patients with incomplete lumbar spinal cord injury ( SCI ) . Methods Sixty patients were randomly divided into a control group and a treatment group, each of 30. Both groups were given conventional rehabilitation, while the treatment group was additionally provided with isokinetic muscle strength and muscle endurance training for 3 months. The endurance ratio ( RO) of the quadriceps and hamstrings, the mean power frequency of the rectus femo-ris (RF-MPF) and the biceps flexor cruris (BF-MPF), and distance covered in the 6-minute walk test (6MWT) were evaluated before and after the treatment. An index of functional community ambulation was also compared be-tween the 2 groups. Results After the treatment significant differences were observed in all of the measurements compared to before the treatment for both groups. The average RO improvement of the quadriceps and hamstrings, the BF-MPF, the RF-MPF and the 6MWT distance of the treatment group were all significantly higher compared with the control group. After the treatment, 25 patients of the treatment group could demonstrate functional community ambula-tion, significantly better than that of 20 of the control group. Conclusion Isokinetic training can improve muscle endurance and the walking capacity of persons with an incomplete lumbar spinal cord injury.
3.Protective effects of penehyclidine hydrochloride against acute renal injury induced by hemorrhagic shock and lipopolysaccharides in rats.

OBJECTIVETo investigate the effect of penehyclidine hydrochloride (PHC) in a rat model of renal injury induced by hemorrhagic shock and lipopolysaccharides (LPS).
METHODSForty-five healthy Wistar rats were randomized into sham operated group, model group, and 3 penehyclidine hydrochloride (PHC) dose (1, 2 and 3 mg/kg) groups (PHC1, PHC2, and PHC3 groups, respectively). The arterial blood samples were collected to determine the concentrations of serum tumor necrosis factor-α (TNF-α), interleukin-8 (IL-8), interleukin-1 (IL-1), urine creatinine (Cr) and blood urine nitrogen (BUN), and the renal tissues were collected to measure the expressions of ICAM-1 and nuclear factor-κB (NF-κB) and observe the pathological changes.
RESULTSTNF-α, IL-8, IL-1, Cr, BUN, ICAM-1 and NF-κB in the 3 PHC groups were significantly lower than those in the model group (P<0.05). TNF-α, IL-8, IL-1, Cr and BUN were significantly lower in PHC1 (P<0.05) than in the PHC2 and PHC3 groups, and ICAM-1 and NF-κB were similar between 3 PHC groups (P>0.05). Compared with the model group, the 3 PHC groups showed lessened pathological changes in the renal tubules.
CONCLUSIONPHC has protective effects against renal injury induced by hemorrhagic-endotoxin shock in rats, and treatment with 1 mg/kg PHC produces the most significant protective effect.

4.Research progress of ferroptosis and its mechanism of action on intestinal epithelial cells in inflammatory bowel disease

Inflammatory bowel disease(IBD)is a chronic pro-gressive inflammation of the gastrointestinal tract that damages the mucous membrane of the gastrointestinal tract.A growing number of studies have demonstrated that dysfunctional cell death occurs at inflammatory sites,leading to the destruction of the intestinal mechanical barrier and aggravation of inflammatory responses.Ferroptosis is a newly discovered form of programmed cell death mediated by iron-dependent lipid oxidation.There have been many articles that reported intestinal abnormal related factors of ferroptosis in patients with IBD.In view of the unclear current IBD specific pathogenesis and the many limitations in treatment,we summarize the research progress of ferroptosis mechanism in recent years,and expound the potential role of fer-roptosis in the pathogenesis of IBD,aiming to provide direction for the future study of the pathogenesis of IBD and the develop-ment and application of therapeutic drugs.
5.Characteristics of tidal breathing pulmonary function in children with tracheobronchomalacia.

OBJECTIVETo investigate the characteristics of tidal breathing pulmonary function in children with tracheobronchomalacia (TBM).
METHODSIn this study, 30 children who were diagnosed with TBM using electronic bronchoscopy were enrolled in the observation group; 30 healthy children were recruited in the normal control group. For individuals in each group, the assessment of tidal breath pulmonary function was performed at diagnosis and 3, 6, 9, and 12 months after diagnosis.
RESULTSThere were no significant differences in tidal volume, inspiratory time, expiratory time, and inspiratory to expiratory ratio between the two groups (P>0.05). Compared with the control group, the observation group had a significantly higher respiratory rate and significantly lower ratio of time to peak tidal expiratory flow to total expiratory time (TPTEF/TE) and ratio of volume to peak tidal expiratory flow to total expiratory volume (VPTEF/VE). There was a time-dependent increase in TPTEF/TE and VPTEF/VE for TBM children from the time of initial diagnosis to 12 months after diagnosis.
CONCLUSIONSTidal breathing pulmonary function has characteristic changes in children with TBM. Tidal breathing pulmonary function tends to be recovered with increased age in children with TBM.

6.Differential gene expression in nasopharyngeal carcinoma cell with reduced and normal expression of 6A8 alpha-mannosidase.

OBJECTIVETo detect the differential display of mRNA expression between human nasopharyngeal carcinoma cell CNE-2L2 with reduced malignancy caused by transduction of a DNA antisense to 6A8 alpha-mannosidase cDNA (AS cell) and the wild type cell (W cell).
METHODSDifferential display of mRNA expression was analyzed using DNA microarray analysis. The datasets were confirmed by Northern blotting and RT-PCR.
RESULTSOut of the 1069 genes analyzed, 34 genes were up-regulated in AS cells relative to W cells. Conversely, 42 genes were down-regulated. The genes, up-regulation of which might have suppressive effect on tumor malignant behaviors, were P130 mRNA for 130K protein, TGF-betaIIR alpha, GABBR1, TGFBR1, TNFAIP1, STANIN, E-CADHERIN, CTNNA1 and 2, RFX2, TMPO, etc. The genes, down-regulation of which might have suppressive effect on tumor malignant behaviors, were CD44, NDRG1, TGFB1, RPS5, LEGUMAIIN, CBS, CD59, SNRPA1, etc. The microarray datasets were confirmed by Northern blot and RT-PCR analysis.
CONCLUSIONSIn comparison to the W cell, AS cell has up-regulation of 34 genes and down-regulation of 42 genes. Changes of the gene expression may play a role in the malignancy reduction of AS cell.

7.Changes and significance of peripheral blood Th22 level in patients with sepsis

Objective To investigate the change of peripheral blood Th22 cells level in the patients with sepsis and its clinical significance .Methods A total of 180 patients with sepsis in this hospital from April 2014 to April 2017 were selected as the re-search subjects and divided into the common sepsis group (81 cases) ,severe sepsis group (68 cases) and septic shock group (31 ca-ses) according to the sepsis severity .The levels of IL-22 ,IL-6 and PCT in peripheral blood Th22 cells were measured in each group .The Acute Physiology and Chronic Health Evaluation Ⅱ (APACHE Ⅱ) scores were recorded .The efficiency of peripheral blood Th22 cells in the early diagnosis of sepsis was evaluated by adopting the receiver operating characteristic (ROC) curve .Results The levels of peripheral blood Th22 cells and IL-22 in the patients with sepsis were significantly than those in the control group (P<0 .05) .The levels of Th22 cells ,IL-22 and PCT and APACHE Ⅱ scores had statistical differences among the common sepsis group ,severe sepsis group and septic shock group (P<0 .05) .The correlation analysis showed that peripheral blood Th22 cells lev-el was positively correlated with PCT level and APACHE Ⅱ scores respectively (r=0 .80 ,0 .66 ,P<0 .01) .With peripheral blood Th22 cells level=1 .9% as the boundary for early diagnosis of sepsis ,its sensitivity and specificity were 83 .1% and 85 .2% respec-tively ,which was significantly better than the PCT index .Conclusion Peripheral blood Th22 cells and related cytokines have the important clinical application value for the early diagnosis of sepsis and assessment of disease severity .
8.Applicability of three surveillance methods for Aedes albopictus in dengue risk indication

Objective To disscus the applicability of three surveillance methods for Aedes albopictus in dengue risk indication. Methods Larval and adult mosquito surveillance were conducted in two villages of Guangzhou by using three vector monitoring methods. Meteorological data and dengue cases were obtained in the same period. Results The population dynamics of larvae and adult mosquitoes were affected by temperature and their overall trend was consistent, but there were differences between the abundance of larvae and adult mosquitoes. From December to February the next year, Breteau index(BI) ranged from 5.66 to 24.53 with a risk of level 1 to 3 for dengue fever, while the mosquito and oviposition positive index(MOI) and adult-mosquito density index(ADI) were 0 to 4.00 and 0 to 1 per man-hour, respectively, indicating no risk for dengue fever. In March, compared with BI which indicated a risk of level 3, MOI were 2.13 and 3.77, respectively, representing for no risk. ADI were 4 to 6 per man-hour, indicating a risk of level 1 and level 2, respectively. After April, the dengue risk level reflected by each indicator became to be consistent. Conclusions The risk of dengue fever indicated by BI is always higher than or equal to the risk indicated by the other two. Compared with BI, MOI and ADI are more sensitively in reflecting the activity of Ae. albopictus.
9.Analysis of risk factors for anastomotic infectious complications following bowel resection for Crohn disease.

OBJECTIVETo investigate the risk factors for anastomotic infectious complications after bowel resection in patients with Crohn disease.
METHODSClinical data of 124 patients with Crohn disease undergoing bowel resection between January 1990 and October 2012 were analyzed retrospectively. The risk factors were identified by χ(2) test and Logistic regression.
RESULTSFourteen patients (12.3%, 14/114) developed anastomotic infectious complications in the postoperative period, including anastomotic leak (n=7), intra-abdominal abscess (n=6), and enterocutaneous fistula (n=1). Crohn disease activity index (CDAI)>150 (OR=2.185, 95%CI:1.098-6.256, P=0.040), steroid usage (OR=2.674, 95%CI:1.118-8.786, P=0.027), and the presence of preoperative abscess/fistula (OR=3.447, 95%CI:1.254-10.462, P=0.014) were identified as independent risk factors of anastomotic infectious complications. In the absence of these 3 risk factors, the rate of anastomotic infectious complication was 5.7% (3/53), which increased to 11.4% (4/35) when one risk factor was present, 21.1% (4/19) when two risk factors were present, and 42.9% (3/7) when all the 3 risk factors were present.
CONCLUSIONSCDAI>150, steroid usage and preoperative abscess/fistula are associated with higher rates of anastomotic infectious complications following bowel resection for Crohn disease. A prudent management should be carried out if risk factors can not be eliminated preoperatively.

10.Rapid determination of illicit beta2-agonist additives in health foods and traditional Chinese patent medicines with DCBI-MS/MS method.

A novel rapid method for detection of the illicit beta2-agonist additives in health foods and traditional Chinese patent medicines was developed with the desorption corona beam ionization mass spectrometry (DCBI-MS) technique. The DCBI conditions including temperature and sample volume were optimized according to the resulting mass spectra intensity. Matrix effect on 9 beta2-agonists additives was not significant in the proposed rapid determination procedure. All of the 9 target molecules were detected within 1 min. Quantification was achieved based on the typical fragment ion in MS2 spectra of each analyte. The method showed good linear coefficients in the range of 1-100 mg x L(-1) for all analytes. The relative deviation values were between 14.29% and 25.13%. Ten claimed antitussive and antiasthmatic health foods and traditional Chinese patent medicines from local pharmacies were analyzed. All of them were negative with the proposed DCBI-MS method. Without tedious sample pretreatments, the developed DCBI-MS is simple, rapid and sensitive for rapid qualification and semi-quantification of the illicit beta2-agonist additives in health foods and traditional Chinese patent medicines.
