1.Anti-tumor and immunodulational effect of Phellinus linteus

Objective To study the anti-tumor activity of Phellinus linteus(PL) and its immunomodulational effects.Methods Solid tumor was induced by injecting HepA,MFC,S_(180) cells(3?10~6cells/animal) subcutaneously to the right anterior limb in mice.The tumor weight,thymus weight and spleen weight were measured after mice were executed.The effects of PL on lymphocyte proliferation,mitogen-induced proliferatoin of ICR mouse splenocytes were measured by MTT.Meanwhile,lymphocytes were prepared as described above.SRBC(10%,0.2 mL) was injected to abdominal cavity to elevate the level of antibody production by B cells.Spleen cells suspension(1?10~7?mL~(-1)) was used as the effect cells and L_(929) cells was used as the target cells.L_(929) cells engulfing neutral red determined the cytotoxic activity of NK.Simultaneously,phagocytosis of mice was detected using the method that macrophages engulfed neutral red.The effect of PL on the production of antibody was studied,the lymphocytes from mice were prepared as described above.Results The high-dose(2.0 g?kg~(-1)) and mid-dose(1.0 g?kg~(-1)) of PL could significantly inhibit the growth of HepA,MFC and S_(180)tumor,the inhibitory rates were 38.19% and 37.15%,32.46% and 30.91%,36.0% and 34.16%,respectively(P

3.Detection of genotype and phenotype in 11 vancomycin-resistant enterococci

Objective To detect the phenotype and genotype of 11 vancomycin-resistant enterococci(VRE).Methods Disk diffusion test,microdilution broth method,VITEK-2 and Etest were used to determin the susceptibility of 11 VRE to vancomycin and teicoplanin. MIC to 10 antimicrobial agents were determined with microdiluton broth method. vanA, vanB ,vanC_1, vanC_2 were detected with multiplex PCR and were further confirmed by DNA sequencing. TEM, tetM,ermB,aac(6′)/aph(2′′),ant(6)- I,aph(3′)-Ⅲ were detected with PCR.Results Susceptibilities of 11 VRE to vancomycin and teicoplanin were different in four vitro susceptibility testing methods. VRE showed resistance to erythromycin(9/11),ciprfloxacin(7/11),levofloxacin(7/11),rifampin(8/11) and chloramphenicol(7/11). HLGR and HLSR is 10/11 and 9/11 respectively. 1 VanA,4 VanB,5 VanC_1 and 1 VanC_2 were detected and their phenotypes confirmed to the genotypes. High rate of TEM(8/11), tetM(6/11), ermB(7/11), aac(6′)/aph(2′′)(7/11), ant(6)- I(5/11) and aph(3′)-III(9/11) were detected in 11 VRE.Conclusions VRE screening test and the determination of MIC are reliable in finding VRE. PCR is precise and specific in the testing and confirmation of VRE. The high rates of drug resistance genes showed the complicated mechanisms of drug resistance in VRE.
4.Detection of Human Hepatocarcinoma Cell Line PLC/PRF/5 Genome Hepatitis b Virus DNA Integration with Alu-PCR

Objective In this research with the method of Alu -PCR we investigate the integration of hepatitis B virus ( HBV) DNA in human hepatocarcinoma cell line (PLC/PRF/5) genome DNA.Methods We at first extracted the genome DNA from PLC/PRF/5 cells, and then the potential integration fragments of HBV DNA and human genome DNA were amplified with according to the Alu -PCR after three rounds PCR .The Alu-PCR amplification products were observated with agarose gel electrophoresis , then integration positive electrophoresis bandings were chipped and purified for nucleic acid sequencing .At last he bioinformatics information was acquired by blast online.Results Through agarose gel electrophoresis after Alu -PCR amplification, we got four potential integration bindings , among which we got three integration sequences of HBV DNA in human genome DNA .These integration sequences could be individually located in the human chromosome of 03p21.31, 05p15.33, 12q13 and 12-q14.1.Conclusion With Alu-PCR we can accurately measure the integration of HBV DNA in human genome DNA , and Alu-PCR can be a a convenience and economic method in the study of HBV DNA ′s integration in human genome DNA .
5. The risk factors of primary intracerebral hemorrhage: A comparison between young adults and aged over 45

Objective: To compare the risk factors of intracerebral hemorrhage (ICH) in young adults and aged over 45. Methods: A total of 274 patients who suffered from the first attack of ICH were collected. Among them 34 were attributed to young adults group, and 240 were aged over 45 group. The incidence of various risk factors were calculated, the differences of risk factors, and the influence of the exposure of various risk factors between these two groups were compared. Results: Hypertension was the most prominent risk factor in patients with ICH in this study, and its incidence was 70.1%, the incidence in the young adult group and the aged over 45 group were 55.9% and 72.0% respectively, but there was no significant difference. Then the subsequent orders were hyperhomocysteinemia, history of ischemic stroke, cigarette smoking and alcoholic consumption. The incidence of history of ischemic stroke in the young adult group was 11.8%, and the aged over 45 group was 31.3%, and there was significant difference between these two groups (P > 0.05). There were no significant differences on hyperhomocysteinemia cigarette smoking and alcoholic consumption in both groups. The patients with the suspected inducing factors before the symptom onset in the young adult group and the aged over 45 group were 50.0% and 24.6% (P < 0.01) respectively. The patients with the inducing factors of fatigue or emotional agitation in both groups were 41.2% and 13.8% (P < 0.01) respectively. The symptom onset of the patients during the active state in both groups were 76.5% and 52.9% (P < 0.05) respectively. The risk factors that closely correlated with the onset of ICH were fatigue and emotional agitation before the hemorrhage in the young adult group (OR 4.370, 95% CI 2.012-9. 490, P < 0.01). Conclusion: History of hypertension is the common risk factors of ICH in the young adults and the aged over 45. Patients with history of ischemic stroke in the aged over 45 group is higher than that in the young adult group. Fatigue or emotional agitation before the symptom onset in the young adult group is higher than the aged over 45 group.

BACKGROUND:An amyloid-beta (Aβ) aggregation in the brain can induce nerve cel apoptosis, loss of synapses and functional damage. However, there is stil no effective intervention. Improving the synaptic plasticity provides an important direction for the treatment of early Alzheimer’s disease. OBJECTIVE: To screen the best model of Alzheimer’s disease and to explore the expression of synapse-associated proteins in Aβ25-35-injured PC12 neurons. METHODS:PC12 cels were induced by 50 μg/L nerve growth factor to differentiate into neuronal-like cels. Then, these cels were treated with Aβ25-35 at different concentrations. Consequently, cel survival rate was detected using cel counting kit-8; neurogranin and neuregulin immunofluorescence stainings were used to observe morphological changes of model cels; western blot used to detect the expression level of neurogranin, calmodulin kinase II, postsynaptic density-95 proteins. RESULTS AND CONCLUSION:Over time, the survival rate of PC12 neurons induced by Aβ25-35 was decreased in a dose-dependent manner. Shortened synaptic length, neuronal atrophy and sparsely interconnected neurons were visible. Expression levels of neurogranin, calmodulin kinase II and postsynaptic density-95 proteins were al down-regulated. These findings indicate that to screen the cel model of Alzheimer’s disease, the optimal concentration and interventional time of Aβ25-35are 10 μmol/L and 48 hours, respectively.
7.Tip-apex distance and calcar-referenced tip-apex distance used as a predictor of screws cutout from the femoral head

Intertrochanteric fracture,a common kind of injury in orthopaedic surgery,often requires surgical intervention,especially the unstable ones.Since far back in 1995,Baumgaertner et al.recommended using the distance between the screw tip and femoral head apex (tip-apex distance,TAD) as a predictor of the risk of lag screw cutout,the importance of TAD has been recognized by orthopaedic surgeons.This concept has been widely accepted in practice,and even used as a standard in determining the helical blade position.It is not clear,however,whether the helical blade system is equal to lag screws in terms of TAD.After the limitations of TAD had been reported,Kuzyk et al.proposed a tip-apex distance referenced from the calcar (Cal-TAD) as a predictor of lag screw cutout to improve the concept further.In this article,these concepts and the latest clinical applications of TAD and Cal-TAD are reviewed based on the publications available.
8.Occurrences of depression and anxiety in patients with carcinoma of large intestine and the clinical manifestation

BACKGROUND: Major psychological problems in patients with carcinoma of large intestine(CLI) were depression and anxiety. Are these bad moods correlated with the pathological stages, the pathological classification and the clinical manifestation of CLI?OBJECTIVE: This study was designed to investigate the relationship between the depression and anxiety state and the clinical manifestations in CLI patients.DESIGN: A comparative observation on patients was conducted in this study.SETTING: This study was conducted at the Department of General Surgery, General Hospital of Shenyang Military Area Command(MAC) of Chinese PLA and the Department of General Surgery, the 307 Hospital of the Chinese PLA. PARTICIPANTS: This study was conducted in the General Surgery Department of the General Hospital of Shenyang Military Area Command of Chinese PLA from February 2002 to February 2003. Totally 79 inpatients and outpatients were diagno sed as CLI by pathological examination. Their informed consents were obtained. Among them, 68 were accordant with the inclusion criteria, 51 males and 17 females; 21 with an age ≥55 years and 47 with an age < 55 years; 39 with an average course of disease≥10 weeks and 29 with an average course of disease <10 weeks; 49 in Dukes A-C stages of CLI and 19 in Dukes D stage; 13 with a cancer of anal canal, 55 with a nonanal-canal cancer; 42 with a squamous cell carcinoma and 26 with a nonsquamous carcinoma; 40 with a serum level of carcinoembryonic antigen(CEA) ≥30 μg/L and 28 with it <30μg/L; 44 with a serum level of interleukin-8 (IL-8)≥90ng/L and 24 with it<90 ng/L.METHODS: Sixty-eight patients with CLI received self-rating depression scale(SDS) and self-rating anxiety scale(SAS) testing made by Zung in one week before treatment. Statistical analysis of the scores was performed according to the classifications of age, sex, course of disease, pathological stages, types of carcinoma, and serum level of CEA and IL-8 respectively.MAIN OUTCOME MEASURES: ① The occurrence of depression and anxiety in CLI patients(standard scores of SDS and SAS were ≥50 ). ②The relationship between the standard scores of SDS and SAS and the various variables as age, sex, course of disease, pathological stages, types of carcinoma and serum level of CEA and IL-8.RESULTS: Totally 68 patients with CLI entered the statistical analysis procedure. ① The incidence of depression and anxiety(standard score ≥50) were 48% (33/68) and 43% (29/68) in patients with CLI. ② In patients with an age <55, male patients, and patients with a course of disease ≥10weeks, the standard scores of SDS(46.51 ±9.82, 47.74 ± 14.25, 48.38 ± 11.73) and SAS(44.28 ±11.24, 45.06±13.27, 45.78±12.59) were significantly higher than those in patients with an age ≥55,female, and with a course of disease <10 weeks(SDS: 43.27 ±10.06, 42.63±10.06, 42.19±14.34;SAS: 41.65±12.77, 41.22±10.89, 42.30±10.21) (t=2.192-2. 329, P <0.05). In patients in Dukes D stage of carcinoma, those with a cancer of anal canal and those with a nonsquamous carcinoma, the standard SDS scores(48.04±12.71, 47.23±9.85, 48.06±14.32) and SAS scores (44.96±10.13, 45.17±12.34, 45.28±14.38) were significantly higher than those in patients with Dukes A-C stages of carcinoma, with nonanal canal cancer, and with a squamous cell carcinoma(SDS: 43.42±11.97,44.05±12.14, 44.36±10.57; SAS: 42.23±13.04, 42.39±11.09,43.14±10.60) (t=2.136-2.298, P <0.05).In patients with serum level of CEA ≥30 μg/L and with IL-8 level ≥90 g/L,the standard scores of SDS(48.27±11.69, 49.04±12.24) and SAS(44.94±13.34,45.26±12.70) were significantly higher than those in patients with CEA <30μg/L and IL-8 <90 ng/L( SDS: 43.48±12.76, 44. 15±13.61;SAS: 41.47±10.58, 41.71±14.53) (t =2. 248-2. 314, P<0.05).CONCLUSION: It is showed that those patients with these variables as a longer course of disease, an older age, male, a more advanced Dukes staging,and higher levels of CEA and IL-8 are prone to depression and anxiety.
10.Study on the Relationship of Economic Social Factors and Human Health Status

Objective:To explore the main economic and social factors influencing human health. Methods:By using the partial least squares(PLS) method and the panel data from areas of China in 2011, regression model related with economic social factors and human life expectancy was built. Results: Per capital GDP, the proportion of the tertiary industry, the number of people beyond college diploma per million people and the number of health technical personnel per 1 000 people has inverse relationshop with life expectancy;the proportion of medical costs accounted for fiscal expenditure, the ratio of fiscal deficit, rate of unemployment and Gini coefficient have reverse relationshop with life expectancy. Conclusion: To improve the level of human health, it needs to take comprehensive measures.
