1.Progress of laryngeal cartilage dysplasia

Laryngeal cartilage dysplasia,also known as congenital laryngeal cartilage softening or con-genital laryngeal stridor,a clinical common laryngeal disease in infant. It is often shown as airflow makes special sound through larynx or tracheal stenosis,also known as laryngeal stridor. Obstruction can be caused by airway lumen or external pressure. Airway obstruction position can be located in the nose,pharynx and larynx and tra-chea. Laryngeal stridor can be caused by many diseases,such as congenital laryngeal stridor,acute laryngeal,or acute laryngotracheobronchitis,respiratory tract foreign bodies,congenital laryngeal stenosis and laryngeal web, etc. The pathogenesis,pathology,clinical manifestation,diagnosis and treatment are reviewed.
2.Expression of DNA binding / differentiation inhibitory protein 2 in colon adenocarcinoma and its relationship with cell proliferation

Objective:To detect the expression of inhibitor of DNA binding-2(ID-2) in colorectal adenocarcinoma and analyze its relationship with proliferation.Methods:A total of 67 patients with colorectal adenocarcinoma diagnosed in North China University of Science and Technology Affiliated Hospital were selected from November 2014 to September 2015, tumor tissue as the research objects.All patients underwent radical operation.The tumor tissue was taken as the observation group, and the normal colonic mucosa tissue>3 cm from the edge of the tumor was taken as the control group.Immunohistochemistry was used to detect the expression of ID-2 of the two groups and the Ki67 of cancer tissue.SW480 cell line of ID-2 overexpression was constructed.The expression of proliferating cell nuclear antigen(PCNA)was detected by Western Blot.Cell activity was detected by CCK-8 test.Correlation was analyzed between ID-2 and Ki67 by pearman correlation analysis.Prognostic value of ID-2 was analyzed by Kaplan-Meier survival analysis and Log-rank test in colorectal adenocarcinoma.Results:The positive rate of ID-2 was 49.3%(33/67) in the observation group, which was higher than 9.0%(6/67) in the control group, and the difference between the two groups was was statistically significant(χ 2=23.927, P<0.05). In the observation group, the expression of ID-2 was statistically significant in different invasion depth(serosa and extraserosa was 68.7%(22/32), serosa and extraserosa was 31.4%(11/35)), degree of differentiation(low differentiation was 80.0%(8/10), medium differentiation was 57.9%(11/19), high differentiation was 36.8%(14/38)), clinical stage(Ⅲ and Ⅳ stage were 64.3%(18/28), Ⅰ and Ⅱ stage were 38.5%(15/39)), lymph node metastasis(metastasis was 70.8%(17/24), no metastasis was 37.2%(16/43)) and tumor thrombus(yes was 75.0%(12/16), no was 41.2%(21/51)). The difference was statistically significant (χ 2 value were 6.311, 4.023, 4.349, 6.967 and 5.575, respectively, all P<0.05). Positive correlation was found between ID-2 and Ki67( r=0.65, P<0.05). Survival analysis showed that the expression of ID-2 was related to the prognosis of patients (X2=5.29, P=0.013). Compared with empty vector transfection group and blank control group, the expressions of PCNA and the activity ID-2 overexpression colon cancer cells increased( P<0.05). Conclusion:The higher expression of ID-2 is related to clinicopathological features and prognosis in colorectal adenocarcinoma.The abnormal expression of ID-2 may play a role in regulating the proliferation of colon adenocarcinoma.
3.Longitudinal observations of the prevalence of nutritional risk and undernutrition in patients with head and neck cancer during radiotherapy

Objective To investigate the changes in prevalence of nutritional risk and undemutrition in patients with head and neck cancer during radiotherapy.Methods In this longitudinal observational study,a convenience sampling method was used to recruit patients with head and neck cancer who were receiving radiotherapy in Beijing Cancer Hospital.Nutritional Risk Screening 2002 (NRS 2002) was applied to assess the prevalence of nutritional risk in the patients,and Patient-Generated Subjective Global Assessment (PG-SGA) and body composition test to determine the prevalence of malnutrition (undernutrition) before,during and after radiotherapy.Results 56 patients finished the three follow-up exams.Along with the progress of radiotherapy,the scores of NRS 2002 increased gradually (1.64±1.09 vs.2.30 ±1.06 vs.3.14 ±1.07,x2 =46.639,P<0.001),and the prevalence of nutritional risk also increased gradually (21.43% vs.37.50% vs.71.43%,x2 =29.700,P <0.001);the total scores of PG-SGA [1 (1-13) vs.6 (1-15) vs.12 (1-18),x2 =63.206,P<0.001] and dimensions of weight [0 (0-4) vs.1 (0-4) vs.3 (0-6),x2 =40.798,P<0.001],intake [0 (0-2) vs.1 (0-2) vs.2 (0-4),x2=64.707,P<0.001] and symptoms [0 (0-7) vs.2 (0-10) vs.6 (0-11),x2 =61.562,P < 0.001] all increased gradually with statistical significance.The prevalence of malnutrition in different stage of radiotherapy were significantly different (x2 =64.999,P < 0.001).The body composition analysis in 40 patients showed that all the indicators of body composition decreased significantly along with the progress of radiotherapy.There was a great loss in patients' body weight during radiotherapy,especially the fat-free mass.Conclusions The prevalence of nutritional risk and undernutrition may increase in patients with head and neck cancer during radiotherapy.Lean body mass accounted for most of the weight loss.We should pay more attention to those patients' nutritional status during radiotherapy.
4.Study of Rhizoma Coptidis alkaloids on promoting sleep in mice

Aim To study the hypnotic effect of five alkaloids extracted from Rhizoma Coptidis (berberine , coptisine,palmatine,epiberberine,jatrorrhizine )in mice,and preliminarily explore its underlying mecha-nism.Methods The experiments of locomotor activity and hypnosis induced by suprathreshold and subthresh-old doses of pentobarbital sodium were used to evaluate the effect of drugs on sleep behavior in mice.Then, HPLC-FLD was used to detect the contents of NE,DA and 5-HT on PCPA mice model.Results Compared with control group,berberine and coptisine notably in-hibited spontaneous activity in behavioral experiments (P <0.05),and increased the sleeping percentage of mice under subthreshold dose of pentobarbital sodium. Berberine and coptisine shortened the period of sleep latency,and prolonged the sustained period of sleeping at suprathreshold dose in mice (P <0.05 or P <0.01 ).Other alkaloids had no significant differences in sleep latency and period of sleep observed in this current experiment.Compared with PCPA mice model group,berberine and coptisine remarkably increased the contents of NE and 5-HT (P <0.01 ),but they had no effects on DA.Conclusions Berberine and coptisine may play a sedative and hypnotic role in PC-PA mice by increasing contents of 5-HT and NE in hy-pothalamus,and the sedative and hypnotic effects of berberine are stronger than those of coptisine.Other alkaloids have no effects on sleeping in mice.
5.Effect Of mechanical ventilation with the least delay on the severe cases of hand-foot-mouth disease

objective To explore the indication of mechanical ventilation and evaluate the effects of mechanical ventilation with the least delay on the Severe cases of hand-foot-mouth disease(HFMD).Methods Retrospective cohort study was conducted among the severe HFMD cases(n=66)admitted to pediatric intensive care unit(PICU)between July 2008 and september 2009.Sixty-six cases were divided into two groups:42 cases(group A,July 5 to October 31,2008)were ventilated with the common mechanical ventilation,24 cases(group B,April 21 to september 30,2009)were ventilated at least delay when the symptoms of neurogenic pulmonary edema appeared.Then the diffcrences on the survival rate and the mortality in the group A and group B were investigated.Results Twenty out of 42 patients died(47.6%),eighteen were cured(42.6%) and four showed improved signs(9.5%)in group A.Twenty-two out of 24 patients were cured(91.7%),two showed improved signs(8.3%)and no death in group B.The clinical effect of group B was much better tllan group A(P<0.001).Conclusion Early mechanical ventilation would improve the survival rate and decrease the mortality of severe hand-foot-mouth disease.
6.Severe hand foot and mouth disease in 68 children with brainstem encephalitis

Objective To explore the clinical features and risk factors of children with severe hand foot and mouth disease (HFMD).Methods The clinical data of 68 children with HFMD associated with brainstem encephalitis were analysed retrospectively from Apr to Sep 2010.Results Among the 68 cases,63cases(92.6% ) were younger than 3 years.The major symptoms and physical signs of 68 cases included rash (68 cases,100% ),fever (68 cases,100% ),fatigue (68 cases,100% ),irritability (65 cases,95.6% ),somnolence (42 cases,61.8% ),vomiting (28 cases,41.2% ),limb tremor (23 cases,33.8% ),tendon reflexe hyperactivity (60 cases,88.2% ),tachypnea or respiratory rhythm abnormality (63 cases,92.6% ),tachycardia (65 cases,95.6% ),and hypertension (54 cases,79.4% ).Twenty-five cases (36.8%,25/68 ) had leucocytosis ( > 12 × 109/L ),and 19 cases ( 27.9%,19/68 ) had hyperglycaemia.X-ray appearances:patchy and pathy shadows in single or bilateral pulmonary were seen in 46 cases( 67.6%,46/68 ).Forty-eight cases were examined by MRI,eight cases displayed ischemic lesions or demyelination.Spinal cord MRI was performed in 3 cases with flaccid paralysis,which showed demyelination.The etiology indicated that 38 cases ( 55.9%,38/68 ) were infected by enterovirus 71,25 cases( 36.8%,25/68 ) were infected by other enterovirus,5 cases (7.3%,5/68) were negative.All the cases were treated by mechanical ventilation with tracheal intubation,in whom 63 cases recovered well,4 cases improved,and 1 case gave up to die.Conclusion Ages < 3 years,enterovirus 71 infection,continual fever,fatigue,somnolence,irdtability,vomiting,limb tremor,tendon reflexes hyperactivity,tachypnea or respiratory rhythm abnormality,tachycardia and hypertension are the high risks of critically ill children associated with severe HFMD.To reduce the fatality rate of HFMD,it is crucial to early judge the high risk factors,and take mechanical ventilations earlier.
7.Establishment of a novel high throughput screening assay for identifying small molecular antagonists of human interleukin-6 receptor

Objective To establish a high throughput screening assay for identifying human small molecular antagonists targeted IL-6R.Methods The full length gene of the human IL-6R extracellular region was amplified by PCR and cloned into a eukaryotic expression vector to construct recombination expression plasmid pABHis -IL6R that was then transfected transiently into HEK293T cells to prepare recombination protein IL-6R.Western blotting assay and receptor-ligand binding experiment were used to analyze the bioactivity of IL-6R.A new screening method based on ELISA was established using the function of IL-6R binding to its ligand and the characteristics of Fc fragment binding to IgG-HRP.Then Z′-factor was calculated and a known antagonist ab 47215 was used to assess the stability and reliability of the new assay .Results Recombination plasmid pABHis-IL6R was constructed and soluble IL-6R was prepared.IL-6R reported herein could be recognized by an anti-IL-6R antibody and specifically bind to its ligand in a dose response manner .A Z′-factor of 0.53 was obtained that could serve high throughput screening assay .Ab47215 , as a known specific antagonist , was able to block rhIL-6 from binding to the receptor in a dose-dependent manner in the new screening assay , the IC50 of which was (0.55 ± 0.11)μg/ml.Conclusion An innovative and easy screening assay for identifying human IL-6R antagonists is established , which might help discover potent and specific antagonists .
8.Study of pro-angiogenesis effect of neuropilin-2 on pancreatic neuroendocrine tumors

Objective To investigate the role and significance of neuropilin-2(NRP2)for regulating the angiogenesis of pancreatic neuroendocrine tumors(PNETs).Methods The NRP2 expression in pancreatic neuroendocrine tumer BON-1 cell line was intevened.The BON-1 cells cultural supernatants in the control group and interference group were used to treat human umbilical vein endothelial cells(HUVEC).CCK-8 was used to detect the cell proliferation,Transwell was used to detected the cell migration and the tubule formation test was used detect the pro-angiogenesis.Results The CCK-8 detection showed that there was no statistically significant difference in the supernatant treated HUVEC proliferations between the interference group and control group medium(P>0.05):the absorbancy in the control group was 0.35±0.04,while which in the interference group was 0.32±0.04.The Transwell test showed that the invasion ability of HUVEC treated with cultural supernatants in the interference group was weakened compared with the control group,the control group was(203±13)/hole,while the interference group was(100±10)/hole(P<0.01);the tubule formation test showed that HUVEC tubular formation treated by cultural supernatant in the interference group was decreased,the control group was 40±5,while the interference group was 24±3(P<0.01).Conclusion Interfering NRP2 expression of BON-1 cells can inhibit the vessel formation ability of co-cultured HUVEC,suggesting that NRP2 may have the pro-angiogenesis effect of PNETs,and may be a potential new target for the treatment of PNETs.
9.Culture and identification of human amniotic mesenchymal stem cells.

OBJECTIVETo establish the method of isolation, purification, and identification of human amniotic mesenchymal stem cells (hAMSCs).
METHODShAMSCs were isolated from human amniotic membrane by trypsin-collagenase digestion, and cultured in Dulbecco's modified Eagle's medium/F12 medium supplemented with 10% fetal bovine serum. Phenotypic characteristics of these cells were analyzed by means of immunocytochemistry and flow cytometry.
RESULTSThe cells successfully isolated from human amniotic membrane expressed representative mesenchymal cell surface markers CD44, CD90, and vimentin, but not CD45.
CONCLUSIONSThis study establishes a potential method for isolation of hAMSCs from human amnion,in vitro culture, and identification. The isolated cells show phenotypic characteristics of mesenchymal stem cells.

10.Effects of paclitaxel loaded-drug micelles on cell proliferation and apoptosis of human lung cancer A549 cells.

This study was conducted to investigate the paclitaxel loaded by hydrazone bonds in poly(ethylene glycol)-poly(caprolactone) micelles (mPEG-PCL-PTX) on proliferation and apoptosis of human lung cancer A549 cells and its possible mechanisms of anti-tumor activity. The cell proliferation was measured with MTT assay. Flow cytometry were used to analyze the cell cycle. The cell apoptosis was analyzed using Hoechst/P staining. The expression levels of apoptotic genes expression in the mitochondrial apoptosis pathway were detected by RT-PCR and Western blotting, respectively. The mPEG-PCL-PTX could inhibit the proliferation of A549 cells and promote the apoptosis. The Bax, caspase-3 protein expression were increased while Bcl-2 protein expression was decreased in A549 cells. Results showed that the polymer containing hydrazone bond is non-toxic in vitro, the mPEG-PCL-PTX micelles can inhibit the proliferation and induce the apoptosis of A549 cells.
