1.Analysis on etiology detection results of vaginal secretion in patients with vaginitis

Objective To study the distribution and drug‐resistance of pathogens isolated from vaginal secretion in the patients with vaginitis in Dalian area so as to provide an evidence for clinical diagnosis and treatment .Methods The statistical analysis was performed on the detection results of pathogens isolated from vaginal secretion in 5 308 patients with vaginitis .Results In 5 308 patients with vaginitis ,the routine detection of vaginal secretion found that 810 cases(15 .3% ) were bacterial vaginosis ,1 110 cases (20 .9% ) were candida positive and 60 cases (1 .1% ) were trichomoniasis positive ,in which 3 526 cases were performed the patho‐gen detection ,2 615 strains of pathogen were isolated ,including 1 948 strains (74 .5% ) of mycoplasma ,136 strains (5 .2% ) of Chlamydia ,17 strains (0 .7% ) of Neisseria gonorrhoeae ,276 strains(10 .6% ) of candida ,140 strains (5 .4% ) of Gram‐positive cocci and 98 strains(3 .7% ) of Gram‐negative bacilli .The susceptibility rates of Ureaplasma urealyticum to ofloxacin ,levofloxacin ,spar‐floxacin and roxithromycin were lower ,which were 7 .8% ,18 .2% ,26 .3% and 27 .2% respectively ,the susceptibility rates to clar‐ithromycin ,doxycycline and minocycline were higher ,which were 89 .4% ,99 .8% and 99 .9% respectively .The drug resistance of Mycoplasma humenis was higher .The more sensitive antibacterial drugs were only josamycin ,doxycycline and minocycline ,the sen‐sitivity rates were 75 .3% ,96 .2% and 97 .3% respectively ,which to other antibacterial drugs were less than 7 .0% .Candida albi‐cans was generally sensitive to antifungal drugs .The susceptibility rates of Staphylococcus aureus to commonly used antibacterial drugs were higher .All of 24 strains of Streptococcus agalactiae were sensitive to penicillin .No Gram‐positive coccus resistant to daptomycin ,linezolid and vancomycin was found .No Gram‐negative bacillus resistant to daptomycin ,linezolid and vancomycin was found .No Gram‐negative bacillus resistant to imipenem and meropenem was found .Conclusion The pathogens causing vaginitis are very complex .In the treatment of female vaginitis ,clinicians should rationally use antibacterial drugs according to the results of pathogens detection and drug susceptibility test .
3.Cloning and bioinformatic analysis and expression analysis of beta-glucuronidase in Scutellaria baicalensis.

The β-Glucuronidase gene (sbGUS) cDNA firstly from Scutellari abaicalensis leaf was cloned by RT-PCR, with GenBank accession number KR364726. The full length cDNA of sbGUS was 1 584 bp with an open reading frame (ORF), encoding an unstable protein with 527 amino acids. The bioinformatic analysis showed that the sbGUS encoding protein had isoelectric point (pI) of 5.55 and a calculated molecular weight about 58.724 8 kDa, with a transmembrane regions and signal peptide, had conserved domains of glycoside hydrolase super family and unintegrated trans-glycosidase catalytic structure. In the secondary structure, the percentage of alpha helix, extended strand, β-extended and random coil were 25.62%, 28.84%, 13.28% and 32.26%, respectively. The homologous analysis indicated the nucleotide sequence 98.93% similarity and the amino acid sequence 98.29% similarity with S. baicalensis (BAA97804.1), in the nine positions were different. The expression level of sGUS was the highest in root based on a real-time PCR analysis, followed by flower and stem, and the lowest was in stem. The results provide a foundation for exploring the molecular function of sbGUS involved in baicalcin biosynthesis based on synthetic biology approach in S. baicalensis plants.

4.The research of lymph node tumor diagnosis algorithm for lymphography based on Semi-Naive Bayes Classification model.

Acquaintance of the lymph node is very important to evaluate whether the tumor is malignant or benign and therefore to the treatment of the tumors. Lymphography is now considered the gold standard for this purpose. Many computer aided diagnose (CAD) technologies have been developed to help radiologists to diagnose the tumor by the lymphography cases. In this paper, a computer aided diagnose model is constructed by Semi-naive Bayes Classification. The experiments carried out in our laboratory validated the Semi-Naive Bayes Classification on lymphography case. The result of experiments showed that Semi-Naive Bayes Classification could classify lymphography case effectively.

5.Impact of "three-in-one" asthma education and management model on asthma control in adult patients

Objective To evaluate the impact of the asthma education and management model developed by Peking University People's Hospital on the level of asthma control. Methods Trained respiratory physicians from 6 large teaching hospitals in the urban districts of Beijing conducted a successive questionnaire survey in the form of face-to-face interviews with adult asthma patients attending their respective clinics. The results were used to compare the levels of asthma control between patients who were administered "three-in-one" asthma education and management (education group) and those who were not(control group). Results Among the 100 patients in the education group, 85% showed asthma control test(ACT) values ≥20 points. This was significantly higher than that in the 427 control group patients (37%,χ2 =74.345 ,P <0.01 ). During the past 1 year, the rate of hospitalization due to exacerbation of asthma,number of emergency treatments, and missed working days were significantly lower in the education group than in the control group ( 4%, 18%, 20% vs 23%, 32%, 55%, respectively; χ2 = 19.431,7.515 and 17.853 respectively; and P < 0.01 for all). Conclusion The " three-in-one" asthma education and management model can significantly improve asthma control.
6.Inhibitory effects of Radix Tetrastigma Hemsleyani Flavone on growth and invasion of lung carcinoma cells

Aim To study the effect of Radix Tetrastig-ma Hemsleyani Flavone ( RTHF ) on the proliferation and invasion in lung carcinoma A549 cells as well as the possible mechanisms underlying these processes. Methods A549 cells were treated with different con-centrations of RTHF for different time. MTT assay and colone formation assay were used to detect the ability of cell proliferation. Wound healing methods and tran-swell chamber assay were adopted to determine cell mi-gration and invasion. Western blotting assay was used to detect the expression of metastasis-related proteins MMP-2 , MMP-9 , and TIMP-2 . Results RTHF obvi-ously suppressed the proliferation of A549 cells in a dose-and time-dependent manner. Microscope found an apparent decrease of cells in the denude zone of cell migration and transwell testing results show that the treatment of invasion was significantly lower than the proportion in the control group ( P <0. 01 ) . The pro-tein expressions of MMP-2 and MMP-9 were down-reg-ulated and that of TIMP-2 was up-regulated in a dose-dependent manner. Conclusion RTHF inhibits the growth and invasion of lung carcinoma A549 cells, which might be achieved by down-regulating the ex-pressions of MMP-2 and MMP-9 protein.
7.Potassium channel blocker(4-AP)enhances the radiosensitivity of human cervical cancer cell line HeLa to ~(252)Cf neutron ray

Objective To investigate the potential effect of 4-amino-pynidine(4-AP)on human cervical carcinoma cell line HeLa to 252Cf neutron ray.Methods HeLa cells were treated with 4-AP(0,1,5,10,20 mmol/L,respectively)and then exposed to 0.67 Gy 252Cf neutron ray.The effect of 4-AP on proliferation of HeLa cells was assessed by MTT assay.Cell apoptosis were detected by flow cytometry.Reverse transcription polymerase chain reaction(RT-PCR)and Western blotting were used to investigate the levels of HIF-1? mRNA and protein of HeLa cells treated with 4-AP(0,20 mmol/L)and 0.67 Gy 252Cf neutron ray.Results After HeLa cells treated with 0,1,5,10,20 mmol/L 4-AP were exposed to 0.67 Gy 252Cf neutron ray for 48 h,their inhibitory rate was 0,(38.81 ?3.45)%,(46.63?3.88)%,(63.58?6.19)%,(77.51?8.87)%,and their apoptotic rate was(3.15?0.85)%,(8.01?1.19)%,(16.00?1.58)%,(47.20?3.18)%,(62.56?4.27)%,respectively,suggesting 4-AP may inhibit the proliferation of HeLa cells and induce their apoptosis in a dose-dependent manner(P0.05).However,after treated with 20 mmol/L 4-AP and exposed to 0.67 Gy 252Cf neutron ray for 24,48 and 72 h,the optical density ratio of HIF-1? mRNA/?-actin of HeLa cells was(0.401?0.121),(0.364?0.142),(0.257?0.137),(0.165?0.099),while the optical density ratio of HIF-1? protain/?-actin were(0.879?0.117),(0.645?0.115),(0.312?0.114),(0.130?0.118),showing that the levels of HIF-1? mRNA and protein both declined(P
8.Significance of HIF-1? and E-cadherin in cervical carcinoma

Objective To study the significance of the expressions of HIF-1? and E-cadherin in cervical carcinoma. Methods The expressions of HIF-1? and E-cadherin (E-cad) were determined in the clinical specimens of 109 cases of cervical carcinoma, 16 cases of cervical intraepithelial neoplasm and 10 cases of normal cervical tissue with immunohistochemical assay. Results There was nearly no expression of HIF-1? in the normal cervical tissue. The positive expression of HIF-1? and E-cad were 81.4% (70/86) and 28.6% (25/86) respectively in the tissue of invasive cervical carcinoma. Objective To investigate the potential effect of hypoxia on adhersion and invasion of human cervical carcinoma cells HeLa. Methods Reverse transcription-polymerase chain reaction (RT-PCR) was used to investigate the level of HIF-1? mRNA and E-cadherin mRNA of HeLa cells cultured under normoxia, 5%O2, 3%O2, 1%O2. The invasiveness of HeLa cells was investigated by transwell chamber assay. Results Under the condition of normoxia, 5%O2, 3%O2 and 1%O2, the optical density ratio of HIF-1? mRNA/?-actin was (0.423?0.116), (0.764?0.111), (1.687?0.126) and (0.215?0.099), showing that the level of HIF-1? mRNA had a tendency of first inceasing and then decreasing with the decreasing oxygen content. The optical density ratio of E-cadherin/?-actin was (1.045?0.171), (0.719?0.114), (0.386?0.105) and (0.157?0.063), showing a decreasing tendency with the decreasing of oxygen content. The invasiveness of HeLa cells was (112.5?20.7), (254.3?27.5), (333.6?45.2) and (124.5?24.6), showing that the invasiveness of HeLa cells had a tendency of first inceasing and then decreasing with the decreasing oxygen content. Conclusion Moderate hypoxia up-regulates the mRNA levels of HIF-1?, and down-regulates the mRNA levels of E-cadherin, and enhances the tumor invasiveness. The possible mechanism is the increasing mRNA levels of HIF-1?.
10.Observation on therapeutic efficacy of tuina plus cupping for chronic fatigue syndrome

Objective: To observe the therapeutic efficacy of tuina plus cupping on chronic fatigue syndrome (CFS). Methods: A total of 100 CFS patients meeting the inclusion criteria were randomly divided into a treatment group and a control group according to the random number table in the sequence of visit, with 50 patients in each group. The treatment group was treated with tuina plus cupping therapy, 3 times a week. The control group was given oral tablets of wild American ginseng lozenges. After 3 months of treatment, the score of fatigue questionnaire scale (FS-14) was observed and the efficacy was evaluated. Results: The total effective rate was 92.0% in the treatment group, versus 76.0% in the control group and the difference between the two groups was statistically significant (P<0.05). After treatment, the FS-14 scores of the two groups decreased, and the intra-group differences were statistically significant (both P<0.05). The change of FS-14 score after treatment in the treatment group was more significant than that in the control group (P<0.05). Conclusion: Tuina plus cupping is effective for CFS, and can improve fatigue symptoms significantly.
