Anorexia is a common pediatric and multiple chronic appetite disorder disease. Long-term anorexia can easily lead to adverse effects on nutritional status, growth, intellectual development and physical and mental health of children. Professor LIU Yi-min, the national famous TCM doctor in Yunnan University of Traditional Chinese Medicine, has much experience in treating this disease, believing that the pathogenesis key is spleen and stomach disharmony and inability to transport. According to clinical experience, it should be treated from indigestion of stomach, spleen dampness, spleen qi deficiency, and spleen weakenness and liver excess. The treatment is to eliminate indigestion, tonify spleen and expel humidity, tonify spleen and nourish qi, and tonify spleen and harmonize liver. The commonly used TCM pairs were Charred Triplet and Galli Gigerii Endothelium Corneum, Plantaginis Herba and Dioscoreae Rhizoma, Hungaian Morningglory Root and Marginate Rockbell Herb and Capsella bursa-pastoris and Chrysanthemi Flos.
2.Effects of progesterone on PMN-mediated inflammatory responses to gonococcal infection

Objective To evaluate the effects of progesterone on polymorphonuclear leukocyte (PMN)-mediated inflammatory responses to gonococcal infection. Methods Peripheral neutrophils were isolated from heparinized peripheral blood obtained from normal individuals, then divided into 4 groups: progesterone group (pretreated with progesterone only), gonococcus group (stimulated with gonococcal suspension), intervention group (pretreated with progesterone followed by stimuation with gonococcal suspension), and control group (receiving no pretreatment or stimulation). Real-time RT-PCR was conducted to detect the mRNA expression levels of inducible nitric oxide synthase (iNOS), tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β)in neutrophils from all groups at 0, 3, 8, 12 and 24 hours after the last treatment, and iNOS protein levels were measured by Western-blot in gonococcus group and intervention group. Results Real-time RT-PCR indicated that the expression levels of iNOS, TNF-α and IL-1β mRNA increased in gonococcus group and intervention group, and reached their peak at 8 hours in gonococcus group, while no significant changes were noted in the above parameters in progesterone group or control group. Also, the level of iNOS, TNF-α and IL-1β mRNA was lower in intervention group than that in the gonococcus group (P < 0.05). Western blot showed an elevation in iNOS protein expression in both gonococcus group and intervention group, and the former group was higher than the latter group in the parameter (P < 0.05). Conclusions Progesterone can downregulate the expressions of iNOS, TNF-α and IL-1 β by PMNs, inhibit the PMN-induced inflammatory responses induced by gonococcal infection, which is likely to be associated with the asymptomatic gonococcal infection in women.
3.Study on Antibacterial Activity and Immunomodulation Effect ofRe-Du-Ning Injection

This study was aimed to discuss the antibacterial and immunomodulation effect ofRe-Du-Ning (RDN) injection. Mice were randomly divided into the normal group, model group,Shuang-Huang-Lian (SHL) group (7.8 mL·kg-1), RDN high dose group (20.30 g·kg-1), middle dose group (10.15 g·kg-1), and low dose group (5.08 g·kg-1), in order to observe the death protective effect of mice with bacterial infection on antibacterial experimentin vivo. Mice were randomly divided into the normal group, model group,Xiang-Gu Duo-Tang (XGDT) group (0.19 mg·kg-1), RDN high dose group (20.30 g·kg-1), middle dose group (10.15 g·kg-1), and low dose group (5.08 g·kg-1). The 2, 4-dinitrochlorobenzene was used to induce delayed hypersensitivity. Immunomodulation was observed by the content of serum hemolysin and the carbon particle clearance index. The results showed that the RDN high dose group and middle dose group had antibacterial effect, which reduced the mortality of mice. The RDN high dose, middle dose and low dose group can enhance the phagocytosis of macrophage in immunosuppressive mice, increase the formation of hemolysin, and strengthen delayed hypersensitivity reaction among immunocompromised mice. It was concluded that RDN injection had antibacterial effect. Its immunomodulation effect was through the enhancing of non-specific immunity, humoral immunity and cellular immunity of mice.
4.Preemptive analgesic effect of administration timing of parecoxib sodium in patients with total hip arthroplasty

Objective To investigate the clinical preemptive analgesic efficacy of parecoxib sodium(PS) at different administration timing in the patients with total hip arthroplasty(THA).Methods Sixty patients receiving THA were prospectively enrolled and randomized into three groups.The group A started to be intravenously injected by PS 40mg/d on preoperative 3 d until operation day;the group B was intravenously injected by parecoxib sodium 40mg at preoperative 30 min;the group C began to be intravenously injected by the same dosage of normal saline at the same time point as the group A.The rest pain was assessed by using the visual analog scale(VAS) at postoperative 6,24,48,72 h.The duration of patient-controlled intravenous analgesia(PCIA) and total dosage were recorded.The first time unaided ambulation time was observed.Results The VAS scores at various postoperative time points in the group A and B were significantly lower than those in the group C(P<0.05),the VAS scores at postoperative 6,24 h in the group A were remarkably lower than those in the group B.The PCIA duration in the group A,B and C were 25.05±10.32),(36.75± 13.91),(50.40 ± 15.17)h,respectively,the pair-wise comparison of the group A,B and C showed statistical difference(P<0.05).The total dosages of PCIA drug in the group A,B and C were(29.25 ± 4.58),(34.50 ± 5.09),(62.65 ±10.52)tg,respectively,the dosage in the group A and B was significantly lower than that in the group C(P<0.05).The first time unaided ambulation time in the group A,B and C were (2.75 ± 0.81),(3.05 ± 1.08),(4.10 ±-0.92)d respectively,which in the group A and B was earlier than that in the group C (P<0.05).Conclusion Continuously using PS on preoperative 3 d can increase the analgesic effect in THA patients and is conducive to the functional rehabilitation and increase the patient satisfaction.
5.A Feasibility Study of closing the small bowel with high-frequency welding device.

This study aimed to evaluate the feasibility and effectiveness of closing the small bowel in an ex vivo porcine model with high-frequency welding device. A total of 100 porcine small bowels were divided into two groups, and then were closed with two different methods. The fifty small bowels in experimental group were closed by the high-frequency welding device, and the other fifty small bowels in comparison group were hand-sutured. All the small bowels were subjected to leak pressure testing later on. The speed of closure and bursting pressure were compared. The 50 porcine small bowels closed by the high-frequency welding device showed a success rate of 100%. Compared with the hand-sutured group, the bursting pressures of the former were significantly lower (P<0.01) and the closing process was significantly shorter (P<0.01). The pathological changes of the closed ends mainly presented as acute thermal and pressure induced injury. Experimental results show that the high-frequency welding device has higher feasibility in closing the small bowel.

6.Determination of urine iodine by the arsenic cerium catalytic rate method

Objective To establish a arsenic cerium catalytic rate method for determination of urinary iodine,and increase the linear range of urinary iodine determination.Methods Standard series and urine samples after digestion treatment,were tested using dynamics function of spectrophotometer to record the curve of absorbance value (A) change with time (t) during arsenic cerium catalytic reaction for each measurement system,choice (A1,t1) and (A 2,t2) on this curve and calculating the reaction rate (v),v =(lgA1-lgA2)/(t2-t1).Through the determination of the standard series it could calculate regression equation of iodine concentration (C) with X:C =a ± bX,X =1 000 (v-v0),and the v0 is the reaction rate of reagent blank.Results (① C and X were positively correlated.The standard series linear range was 0-1 200 pμg/L and correlation coefficient r was higher than 0.999 1.The minimum detection limit was 3.9 μg/L (0.25 ml urine).②)Precision:5 urine samples (A,B,C,D,E) were selected within the range of 0-1 200 μg/L and the measured value were (72.3 ± 2.7),(148.2 ± 5.2),(210.5 ± 4.4),(562.7 ± 6.8),and (899.3 ± 8.0) μg/L.The relative standard deviation (RSD) was between 0.9%-3.8%.(③)Accuracy:4 samples (A,B,C,D) were measured for standard addition recovery test,recovery was between 94.2%-107.2%;urinary iodine standard material [the given values were (67.9 ± 9.0),(142.0 ± 10.0),(195.0 ± 10.0),(558.0 ± 17.0),(885.0 ± 28.0) μg/L] were determined and the results were in the range of uncertainty of the standard material.④Method contrast:with the national health standard method (method for determination of iodine in urine by arsenic cerium catalytic spectrophotometry) to determinate 120 urine samples,the results showed that there were 60 urine samples within 0-300 μg/L,60 urine samples were more than 300 μg/L.Then rate method was used to test the 120 urine samples.For the 60 samples within the scope of 0-300 μg/L,the determination results of the two methods were positively correlated (r =0.994,P < 0.01);the results of the rate method were lower than those of the standard method and the difference was statistically significant (t =2.047,P < 0.05).But the average deviation was only 2.1 μg/L,for the determination of urine iodine there was no practical significance;for the 60 samples higher than 300 μg/L,the determination results of the two methods were positively correlated (r =0.993,P < 0.01) and the difference was not statistically significant (t =-1.092,P > 0.05).Conclusions Arsenic cerium catalytic rate method has increased the linear range of urinary iodine determination.Using this method,the vast majority samples can be tested directly without dilution,thereby reducing the workload for determination of urine iodine.
7.Analysis of awareness and knowledge about cervical cancer of women in Chengdu

Objective To assess the awareness and knowledge about cervical cancer,cervical cancer screen-ing and human papillomavirus(HPV)of women in Chengdu.Methods A total of 1 077 women aged 19 -71 years old were recruited and assessed in their awareness and knowledge about cervical cancer and its issues.The collection of data was based on the questionnaire items.Results The awareness rates of cervical cancer,cervical cancer screen-ing,and HPV were 71.1%,23.2%,16.0%,respectively.There were significant differences in the awareness and knowledge about cervical cancer,cervical cancer screening and HPV between different ages(χ2 =89.238,16.532, 20.182,all P <0.05).The awareness of people(>40 -50 years old)was highest,the awareness rates of cervical cancer,cervical cancer screening,and HPV were 81.3%,27.7%,20.2% respectively.There were significant differ-ences in the awareness and knowledge about cervical cancer,cervical cancer screening and HPV between different marital status(χ2 =60.899,20.853,16.255,all P <0.05).There were significant differences in the awareness and knowledge about cervical cancer,cervical cancer screening and HPV between different education levels (χ2 =228.026,42.648,32.495,all P <0.05 ).The awareness of people accepted university education was highest,the awareness rates of cervical cancer,cervical cancer screening,and HPV were 87.6%,30.9%,21.6% respectively. There were significant differences in the awareness and knowledge about cervical cancer,cervical cancer screening and HPV between different income levels(χ2 =21.604,8.124,9.224,all P <0.05).Conclusion This study demon-strates a very low level of knowledge about cervical cancer,cervical cancer screening and HPV of women in Chengdu. In order to reduce the incidence rate and mortality of cervical cancer in Chengdu,it is strongly needed to make exten-sive health education to raise awareness of cervical cancer and HPV.
8.Analysis of Myocilin gene regulatory network using a genetic genomics approach

Background The pathogenesis of primary open angle glaucoma(POAG) and high myopia are very complex.To construct the regulatory network of virulence genes and relevant genes that involved in pathogenicity are helpful for reveal of the pathogenesis.Objective The aim of this study was to investigate myocilin(Myoc),a gene that contributes to POAG and high myopia in eyes of BXD Recombinant Inbred(BXD RI)mice and construct the regulatory network of Myoc.Methods The affymetrix microarray system was used to detect the differential expression of Myoc in the eyes of C57BL/6J(B6),DBA/2J(D2) and BXD RI mice.Expression quantitative trait loci (eQTL) mapping was performed to construct the regulatory network of Myoc gene.Results The average expression level of the Myoc gene in the BXD strains was 10.83,and the gene exhibited expression levels ranging from 8.39 in BXD55 mice tol 1.43 in B6 mice.The eQTL mapping for the Myoc gene showed a significant likelihood ratio statistic (LRS) of 21.78.The QTL was mapped in chromosome 2,and Myoc was located on chromosome 1,indicating that the Myoc gene was a trans-acting QTL.Olfml2a was identified to be a candidate upstream gene of Myoc by analysis of bioinformatics.Genetic regulatory network analysis demonstrated that a series of genes associated with Myoc probably played roles in the pathogenesis and development of POAG and high myopia.Conclusions The genetical genomics approach provides a powerful tool for constructing pathways that contribute to complex traits,such as POAG and high myopia.
9.Effects of guizhi fuling capsule on sex hormone levels and breast issue morphology of mammary gland hyperplasia model rats.

OBJECTIVETo research the effects of Guizhi Fuling capsule on sex hormones levels in blood serum and breast issue morphology of hyperplasia of mammary glands model rats.
METHODThe unpregnancy SD rat models of hyperplasia of mammary glands were established by injecting 0.5 mg x kg(-1) benzoate estradiol. After five weeks doses,the effects of Guizhi Fuling capsule 2.0, 1.0, 0.5 g x kg(-1) and Rupixiao tablet 0.5 g x kg(-1) on the changes of papilla diameter, height and breast issue morphology of the naimal models were explored, and sex hormones levels in blood serum were measured.
RESULTGuizhi Fuling capsule can inhibitnipple swell, improve breast tissue morphology pathological profiles of the animal models, and decrease oestradiol (E2) level and increase progesterone (P) level in blood serum.
CONCLUSIONThese results suggested that Guizhi Fuling capsule could, improve mammary gland pathological profiles. Regulating sex hormone levels may be its important mechanism for treatment of hyperplasia of mammary glands.

10.Comparison of opa typing with Neisseria gonorrhoeae multi-antigen sequence typing for discrimination of Neisseria gonorrhoeae from patients and their sex partners

Objective To compare the potentiality of opa typing versus Neisseria gonorrhoeae multi-antigen sequence typing (Ng-MAST) in discrimination of N. gonorrhoeae isolates, and to investigate the consistency of genotypes of N. gonorrhoeae between patients and their sex partners. Methods N. gonor-rhoeae was isolated from patients and their sex partners who attended sexually transmitted disease clinics of the Affiliated Union Hospital of Tongji Medical College, Huazhong University of Science and Technology from February 2006 to August 2007. All isolates were cultured on Thayer-Martin agar and GC-based choco-late agar, and identified by Gram stain and oxidase reaction. Totally, twenty-four strains of N. gonorrhoeae were isolated form 12 pairs of male-female sex partners. Genomic DNA was isolated from the strains, and subjected to PCR amplification of opa gene as well as pot and tbpB genes, opa typing and Ng-MAST typing were utilized to differentiate the 24 strains respectively. Results The 24 isolates were classified into 10 sequence types (ST) by Ng-MAST typing, and 12 opa types (OT) by opa typing. A novel genotype identi-fied by Ng-MAST (217-86% homologisation 178), was unique in China. Conclusions Both ST and OT of N. gonorrhoeae strains are identical between patients and their corresponding partners except for one pair of partners, 45/46, suggesting that gonorrhea is transmitted to each other by contiguity between sex partners. Opa typing is likely to be more potential than Ng-MAST in genotyping of N.gonorrhoeae strains.
