1.Effect of 17β-estradiol on injured dopaminergic neurons induced by 6-hydroxydopamine

Objective To study the effect of 17β-estrogen(17β-E2)on injured dopaminergic (DA) neurons induced by 6-hydroxydopamine (6-OHDA),and explore whether 17β-E2 has neuroprotective effect.Methods The male SD rats aged 2-3 d were decapitated under sterile conditions; the brain slices were induced injury by 6-OHDA(200 μmol/L); cells in the slices were divided into control group and 17β-E2 treatment groups(intervention with 1.0xl0-8 mol/L,1.0xl0-9 mol/L and 1.0×10-10 mol/L of 17β-E2,respectively).The tyrosine hydroxylase(TH)expression in DA neurons was observed by Western blotting at 15,30 and 60 min after the intervention.Results The expression of TH in the 1.0×10-9 mol/L 17β-E2 treatment group was the highest,which was significantly different as compared with that in the control group(P<0.05); increasing or decreasing the concentration of 17β-E2 could not regulate the expression of TH; in the 1.0×10-9 mol/L 17β-E2 treatment group,TH expression at 15 min after intervention enjoyed the highest level; the TH expression could not increase by extending the time.Conclusion The 17β-E2 treatment at certain concentration(1.0×10-9 mol/L)for certain time(15 min)has a protective effect on injuried DA neurons
2.Certain Polymorphisms in SP110 Gene Confer Susceptibility to Tuberculosis: A Comprehensive Review and Updated Meta-Analysis.

PURPOSE: Numerous studies have assessed the association of SP110 gene variants with tuberculosis (TB), but the results were inconsistent. Through a comprehensive review and meta-analysis, our study aimed to clarify the nature of genetic risks contributed by 11 polymorphisms for the development of TB. MATERIALS AND METHODS: Through searching PubMed, web of science, China National Knowledge Infrastructure (CNKI) databases, a total of 11 articles including 13 independent studies were selected. The pooled odd ratios (ORs) along with their corresponding 95% confidence interval (CI) were estimated for allelic comparisons, additive model (homozygote comparisons; heterozygote comparisons), dominant model and recessive model. We also assessed the heterogeneity across the studies and publication bias. RESULTS: The results of combined analysis revealed a significantly increased risk of TB for single nucleotide polymorphism (SNP) rs9061 in all five comparisons (allelic comparisons: OR=1.28, 95% CI=1.14–1.44, p<0.0001; homozygote comparisons: OR=2.84, 95% CI=1.84–4.38, p<0.00001; heterozygote comparisons: OR=1.23, 95% CI=1.05–1.43, p=0.009; dominant model: OR=1.32, 95% CI=1.14–1.53, p=0.0003; recessive model: OR=2.26, 95% CI=1.18–4.34, p=0.01). In subgroup analysis, the risk of TB associated with SNP rs9061 appeared to be increased. Moreover, increased risk of TB was also found in Asian subgroup of SNP rs11556887, while decreased risk of TB appeared in large sample size subgroup of SNP rs1135791. No significant association was observed between other SNPs and the risk of TB. CONCLUSION: Our meta-analysis suggested that the variant of SNP rs9061 might be a risk factor for TB.

3.Association between unhealthy diets and depression in early pregnancy

Objective To explore the association between unhealthy diets and depression in early pregnancy. Methods 7 976 women in early pregnancy were recruited and analyzed in this study from the Chinese Pregnant Women Cohort Study (CPWCS) from July 25th, 2017 to July 24th, 2018. Differences of baseline characteristics between the two groups were conducted by a chi-square test. The qualitative food frequency questionnaire and the edinburgh postnatal depression scale were used to assess depression status and food intake frequency during the first trimester respectively. Log-binomial regression was used to analyze the relationship between unhealthy diets and depression in early pregnancy. Prevalence ratio (PR) and its 95% confidence interval (95% CI) were calculated. Results Eating regularly (PR=0.45, 95% CI:0.38-0.54,P<0.001) and eating breakfast frequently (PR=0.80, 95% CI:0.72-0.90, P<0.001) were related to the low incidence of depression while frequent consumption of fried food (PR=1.25, 95% CI:1.03-1.53, P=0.027), Western-style fast food (PR=1.36, 95% CI:1.06-1.74, P=0.015) and puffed food (PR=1.37, 95% CI:1.11-1.70, P=0.003) as well as drinking sugar-sweetened beverage (PR=1.37, 95% CI:1.17-1.61, P<0.001),wine (PR=1.60, 95% CI:1.26-2.01, P<0.001) and liqueur (PR=1.26, 95% CI:1.00-1.59, P=0.047) were linked with the high incidence of depression. Conclusions There might be an association between unhealthy diets and depression in early pregnancy. Pregnant women should take the initiative to better understand what are healthy diets and reduce the frequency of unhealthy diets consumption to decrease the incidence of depression during pregnancy.
4.Dihydroflavonol glycosides and flavan-3-ols from Bauhinia aurea.

OBJECTIVETo study the chemical constituents of Bauhinia aurea.
METHODThe compounds were isolated by column chromatography over silica gel, reversed-phase RP-18, and Sephadex LH -20. MS and NMR spectroscopic methods were used to determine structures of purified compounds.
RESULTEight compounds were isolated from the ethyl acetate soluble fraction of the ethanolic extract and their structures were elucidated as isoengeletin (1), astilbin (2), neoastilbin (3), isoastilbin (4), neoisoastilbin (5), (+)-catechin (6), (-)-epicatechin (7) and (-)-epicatechin 3-O-gallate (8).
CONCLUSIONFive compounds were isolated from this genus for the first time except for 2, 6 and 8.

5.Determination of trace iodide in drinking water in Shanghai by two columns and gas chromatography with electron capture detection.

OBJECTIVETo modify and optimize gas chromatographic conditions, qualitative and quantitative method on the base of the actual national standard detection method for drinking water on testing iodide with gas chromatography, and detect iodide in drinking water of Shanghai.
METHODSIodobutane derivative was identified by gas chromatography/mass spectrometry, then two capillary columns coupled to electron capture detector were made by qualitative and quantitative analysis. Finally, 100 ml water samples were collected respectively from raw water and finished water of three plants in which water sources were Yangtse river, Huangpu river, and inner river respectively and detected by this developed method.
RESULTSThe results of mass spectrometry showed that iodide would form iodobutane which could generate 1-iodo-2-butanone and 3-iodo-2-butanone isomers by derivatization. The data of qualitative analysis by two capillary columns revealed that iodobutane was separated completely and the total time of chromatogram separation was 19.33 min.3-iodo-2-butanone with the high response value was selected to quantitatively analyse. The linear range was 1 - 100 µg/L, and the coefficient of determination (r(2)) was 0.9997. The limit of detection was 13 ng/L. Recoveries were between 97.68% and 104.37%, and relative standard deviations were between 2.14% and 4.41%. The results of iodide detected by this method in raw water and finished water in three plants in Shanghai showed that the ranking of iodide's concentration in raw water was Huangpu river (15.14 µg/L) > inner river (6.97 µg/L) > Yangtse river (3.55 µg/L). The level of iodide in finished water of plant 1 (3.55 µg/L and 5.92 µg/L for raw and finished water respectively) and 3 (6.97 µg/L and 9.62 µg/L for raw and finished water respectively) increased slightly except for plant 2 (15.14 µg/L and 1.81 µg/L for raw and finished water respectively). The relative standard deviations of all water samples measured in duplicate were less than 9.73%.
CONCLUSIONThe improved method possesses higher degree of sensitivity and accuracy of qualitative and quantitative analysis than actual national standard detection method, and it is fit for trace analysis of iodide in water.

6.Injury characteristics and injury analysis in Beagle dogs after multi-cabin explosion

Objective To investigate the injury characteristics of Beagle dogs after warship multi cabin explosion.Methods Forty-eight adult male Beagle dogs were placed in the simulated blast-cabin and adjacent cabin (24 each),and ammunitions respectively containing 0.75kg TNT and 3.50kg TNT were then detonated in the blast-cabin.The survival situation,fluctuation of vital signs,morphological changes of organs and the incidence of various types of injury of the dogs were observed immediately after the explosion to 24h after injury,and the neurological functions score was performed.Results Twenty dogs died immediately after the explosion,and another 9 dogs died 24h after the explosion.The total mortality was 60.42%(29/48),and the mortalities in blastcabin and adjacent cabin were 79.17%(19/24) and 41.67%(10/24),respectively.The dog's skull was penetrated by bomb fragments,and congestion and bleeding were observed in brain tissue,lung,heart,stomach,bowels,liver and kidneys.Extremities fracture,soft tissue contusions,perforation and rupture were also checked out after explosion.The fatality rate of bomb fragment injury,blast injury and combined bomb fragment-blast injury was 27.59%(8/29),17.24%(5/29) and 55.17%(16/29),respectively.Conclusions The combined bomb fragment-blast injury show high incidence and make high fatality rate and serious injury.Combatants should effectually shield themselves with occluded objects as far as possible to avoid damage and reduce fatality.The key of early treatment is to treat the multiple injuries promptly.Intravenous fluid therapy should be practiced after hemodynamic monitoring.
7. Inhibition of Metformin Combined with Cisplatin on Proliferation of Human Osteosarcoma MG-63 Cells

In order to study the effect and its potential mechanism of metformin combined with cisplatin treatment on human osteosarcoma MG-63 cells, MG-63 cells were treated with metformin and cisplatin and the cell proliferation and apoptosis were detected using CCK8 and flow cytometry; Cell clone formation experiment was performed to detect clone formation rate in each group; Trans well experiment was used to detect the migration and invasion ability of osteosarcoma MG-63 cells, and qPCR and Western blot were used to detect the expression of RNA and protein of apoptosis-related genes. The results showed that metformin combined with cisplatin inhibited the proliferation of human osteosarcoma MG-63 cells and promoted their apoptosis significantly (P < 0. 01); Metformin combined with cisplatin inhibited the clone formation (P < 0. 01), and the migration and invasion of human osteosarcoma MG-63 cells (P < 0. 01); Furthermore, metformin combined with cisplatin down-regulated the expression of apoptosis-related genes MCl ̄1and XIAP (P <0. 01), but up-regulated the expression of apoptosis-related genes CASPASE ̄3 and Cyto C (P <0. 01) and migration and invasion related genes MMP ̄2 and MMP ̄9 (P <0. 01). Our study indicated that metformin combined with cisplatin inhibited proliferation, promoted cell apoptosis through MCl ̄1 and XIAP, and inhibited cell migration and invasion by regulating the MMP ̄2 and MMP ̄9 pathways in human osteosarcoma MG-63 cells.

RESULTNine aromatic acid derivatives, two triterpenes and three steroids were isolated from the 90% ethanolic extract and their structures were identified as 3,4-di-hydroxybenzoic acid (1), 4-hydroxybenzoic acid (2), 3-hydroxy-4-methoxy-benzoic acid (3), 4-hydroxy-3-methoxybenzoic acid (4) , gallic acid (5) , methyl gallate (6) , ethyl gallate (7) , lupeol (8) , lupenone (9) , stigmast4-en-3-one (10) beta-sitosterol (11) and daucosterol (12).
CONCLUSIONThis is the first report of chemical constituents from B. aurea. Compounds were isolated from this genus for the first time, except 5,8,11 and 12.

9.Chemical constituents from branch of Macaranga adenantha and their TNF-alpha inhibitory activity.

OBJECTIVETo investigate the chemical constituents of the brach of Macaranga adenantha, and evaluate their TNF-alpha inhibitory activity.
METHODThe chemical conshifuents were isolated and purified by chromatographic methods. Structures of the compounds were identified by spectroscopic methods. The TNF-alpha secretion inhibitory activity of the mouse peritoneal macrophages was evaluated by MTT methods.
RESULTTen compounds were isolated and their structures were identified as: cleomiscosin A (1), cleomiscosin B (2), ellagic acid 4-O-alpha-D-rhamnopyranside (3), ellagic acid 4-O-beta-D-xylopyranoside (4), vanillic acid (5), (24R) -stigmast-4-en-3-one (6), (24R) -stigmast-3, 6-dione (7), (24R) -6beta-hydroxy-stigmast-4-en-3-one (8), daucosterol (9), beta-sitosteryl glucoside-6'-O-heptadecoicate (10). At a concentration of 10 micromol x L(-1), compounds 1, 3 and 4 showed inhibitory activity to TNF-alpha secretion of the mouse peritoneal macrophages with the inhibitory rates of 57.0%, 64.4%, and 57. 4%, respectively.
CONCLUSIONAll compounds were isolated from genus Macaranga for the first time. Compounds 1, 3, and 4 were active against TNF-alpha secretion of the mouse peritoneal macrophages.

10. In Vitro Antitumor Bioactivity and GC-MS Analysis of Petroleum Ether Fraction of Boehmeria nivea Root

Objective:To define the anti-gastric cancer activity in vitro of petroleum ether fraction of Boehmeria nivea root and reveal the material basis of its efficacy, so as to lay the foundation for the development and utilization of B. nivea root. Method:Methyl thiazolyl tetraolium(MTT) method was used to evaluate the inhibitory rate and time-dose relationship of petroleum ether fraction of B. nivea root with different doses and delivery times on human gastric cancer HGC-27 cells. Flow cytometry was used to detect the change of cell apoptosis and cell cycle after petroleum ether fraction of B. nivea root acted on human gastric cancer HGC-27 cells. GC-MS was used to detect the components of petroleum ether fraction of B. nivea root. Result:Experiment data showed significant cell proliferation inhibition in an obvious time-dose-effect manner, with statistically significant differences (P<0.05),after 24,48,72 h of incubation of human gastric cancer HGC-27 cells with different concentrations of petroleum ether fraction of B. nivea root. The effect of petroleum ether fraction of B. nivea root on human gastric cancer HGC-27 cells could induce apoptosis,which affects the normal changes of cell cycle. The percentage of cells was decreased significantly in G0/G1 phase,and that in S phase was significantly increased. GC-MS was used to identify 26 chemical constituents in petroleum ether of B. nivea root,including sitosterol and stigmasterol. Conclusion:Petroleum ether fraction of B. nivea root is the active anti-gastric cancer part,and its main effective component is sterol compounds. This lays the foundation for the rational application of B. nivea root in clinic and the further research in tis anti-tumor effect.
