1.Study on Isolation of Clostridium butyricum from Pits Sludge and Its Characteristics

14 strains were isolated from pits sludge by anaerobic cultivation,two Clostridium butyricum strains were identified by physiological and biochemical experiments and the sequence analysis of 16S rDNA.The physiological characteristics and security of Clostridium butyricum B1 were studied,in vitro research indicated it was tolerant against low pH,bile concentration and antibiotics and has antagonism effects against pathogens.
2.Surveillance of pathogens from patients with hospital infection in intensive care unit

Objective To analyze the distribution and antimicrobial resistance of pathogens from patients with hospital infection in intensive care unit(ICU),and to discuss the corresponding strategy of prevention and treatment. Methods Clinical data of 891 patients hospitalized in the ICU of our hospital between Jan 2002 and Dec 2005 were collected.All the specimens were analyzed for pathogen distribution and resistance to antibiotics. Results Of all the isolated 1 155 strains,Gram-negative(G-)bacteria were the most popular pathogen(57.84%),whereas Gram-positive(G+) bacteria and fungi accounted for 21.39% and 20.78%,respectively.Most G-bacteria were less sensitive to imipenem(0-10.16%),and Pseudomonas aeruginosa was least sesitive to amikacin(6.89%) and cefoperazone-sulbactam(33.97%).G+ bacteria were most sensitive to vancomycin.Most fungus were Candida albicans(42.92%) and yeast species(22.90%). Conclusion Epidemiological investigation of pathogens in ICU is of great importance to the rational use of anti-biotics and may help to reduce the emergence of resistant strains.

4.Blood supply features and interventional therapy of pedunculated hepatocellular carcinoma

Objective To evaluate the blood supply features and effectiveness of arterial chemoembolization for pedunculated hepatocellular carcinoma.Methods Angiography and chemoembolization via supplying blood arteries of tumor were performed in five patients with pedunculated hepatocellular carcinoma.Interventional procedure was carried out with tumor vascular infusion of 350 mg hot elemene emulsion and tumor embolization by cisplantin-lipidol emulsion(cisplantin 60-80 mg+lipidol 8-15 ml)and glutin.Results Ten interventional procedures(TACE)were undertaken in 5 patients.Angiography showed that tumor blood supply mainly coming from collateral circulation adjacent to the tumors,but partially from hepatic artery.Tumor sizes decreased from 30% to 50% in 5 cases,and AFP declined in 4 cases after the treatment. Conclusion Pedunculated hepatocellular carcinoma possessing different blood supply features from intrahepatocellular carcinomas.But transarterial ehemoembolization is still an effective method of choice for this treatment.
5.Therapeutic effect of L‐carnitine combined cyclic adenosine monophosphate and its influence on serum levels of IFN‐γ and IL‐4 in patients with viral myocarditis/

To observe therapeutic effect of L‐carnitine combined cyclic adenosine monophosphate and its influence on serum levels of IFN‐γ and IL‐4 in patients with viral myocarditis .Methods : A total of 132 VMC pa‐tients treated in our hospital from 2017 to 2018 were randomly and equally divided into L‐carnitine group and com‐bined treatment group (received cAMP based on L‐carnitine group ) , both groups were treated for two weeks .Pe‐ripheral serum levels of IFN‐γ , IL‐4 , cardiac troponin T (cTnT) and creatine kinase isoenzyme MB (CK‐MB) be‐fore and two weeks after treatment , therapeutic effect and incidence of adverse drug reactions were observed and compared between two groups .Results : Compared with before treatment , there was significant rise in serum IL‐4 level , and significant reductions in serum levels of IFN‐γ , cTnT and CK‐MB in two groups after two‐week treat‐ment , P=0. 001 all ;compared with L‐carnitine group , there was significant rise in serum IL‐4 level [ (47. 43 ± 9.17) ng/ml vs.(55. 38 ± 10.23 ) ng/ml] , and significant reductions in serum levels of IFN‐γ [ (65.22 ± 11.82 ) ng/ml vs .(52.18 ± 10.06) ng/ml] , cTnT [ (0. 37 ± 0.09) ng/ml vs.(0.18 ± 0.03) ng/ml] and CK‐MB [ (28.56 ± 5. 34) U/L vs.(16. 22 ± 3. 47) U/L] in combined treatment group , P=0.001 all.Total effective rate of com‐bined treatment group was significantly higher than that of L‐carnitine group (92.4% vs.80.3%) , P= 0.042. There was no significant difference in incidence rate of adverse drug reactions between two groups , P=0. 784. Con‐clusion : L‐carnitine combined cAMP can significantly reduce peripheral serum IFN‐γ level and increase IL‐4 level , regulate Th1/Th2 imbalance with significant therapeutic effect in VMC patients .And its safety is good .
7.Influence of stapedectomy on the hearing of guinea pigs.

OBJECTIVETo compare the influences of stapedectomy and small fenestra stapedotomy on the hearing of guinea pigs.
METHODSTwenty-four (48 ears) guinea pigs were randomized equally into two groups, and the left ears were subjected to stapedectomy and total stapes replacement with a prosthesis, or sham operation (12 ears) to expose the footplate of the stapes and the round window. Each guinea pig was tested by ABR perioperatively. Four guinea pigs were chosen randomly from each group and decapitated for morphological examination by light microscopy and scanning electron microscopy after ABR test.
RESULTSIn the sham operation group, the post-operative latencies of each wave, the intervals and the hearing threshold exhibited no significant changes other than prolonged latency of wave I. In stapedectomy group, the hearing threshold increased to 23.75-/+3.77 dBSPL 1 h after operation with significantly prolonged post-operative latencies of all the waves and intervals but for III-IV interval, which was shortened. The latencies of each wave (especially waves I and III) in the stapedectomy group were increased by a greater magnitude than those in the sham operation group, but the intervals were comparable between the two groups. No significant difference was noted in the parameters of ABR either 1 h or 1 day after the operation between the two groups, in which the architecture of cochleas remained intact with similar number of spiral ganglion cells. The stereocilia of the outer hearing cells (OHC) were normal in the sham operation group while in stapedectomy group, slight stereocilia disorder occurred but became normal 1 day after operation. No obvious changes were found in the stereocilia of the inner hearing cell (IHC) in either groups.
CONCLUSIONStapedectomy can induce mild hearing loss without seriously damaging the function of the cochlea in guinea pigs.

8.Relationship of Chinese medicine syndrome pattern with heart function and brain natriuretic peptide in patients with heart failure.

OBJECTIVETo observe the relationship of Chinese medicine (CM) syndrome pattern with heart function detected by ultrasonic cardiography and brain natriuretic peptide (BNP) level in patients with heart failure (HF).
METHODSOne hundred and sixteen inpatients of HF hospitalized in Dongfang Hospital from January, 2007 to January, 2009 were assigned to three groups according to their CM syndrome pattern differentiated, the qi-yin deficiency group (QYD), the qi-deficiency and blood-stasis group (QDBS) and the yang-deficiency with water overflowing group (YDWO). Ultrasonic cardiographic (USCG) parameters, including left ventricular end-systolic diameter (LVEDs), left ventricular end-diastolic diameter (LVEDd) and left ventricular ejection fraction (LVEF) were detected, and blood level of BNP was measured.
RESULTSLVEF was decreased while BNP was increased in patients with syndrome patterns in the order of QYD --> QDBS --> YDWO, and showed significant difference between groups (P < 0.01); CM syndrome patterns was related with all the USCG parameters and BNP level, especially the BNP (r = 0.71, P < 0.05) and LVEF (r = -0.34, P < 0.05).
CONCLUSIONLVEF and BNP can reflect the severity of heart failure, and they could be taken as the beneficial objective and quantitative indices for syndrome pattern differentiation.

9.Comparison of two tracing method of transplanted mouse embryonic stem cell.

OBJECTIVETo trace the embryonic stem (ES) cells transplanted into rat brain by labeling the cells with green fluorescent protein (GFP) and by mouse neuronal specific antibody Thy-1 and compare their features.
METHODSFor GFP labeling,transfect pEGFP-N1 plasmid containing GFP and anti-neomycin sequences into embryonic stem cell and add neomycin for more than 10 passages. To test the GFP expression in vivo, the GFP-ES was transplanted into healthy rat brain, and the frozen sectioned slides were observed under fluorescence microscope and laser con-focal microscope 21 days later. For the antibody labeling,embryonic stem cells were directly transplanted into the rat brain. The specific mouse thy-1 antibody was used in immunostaining of transplanted cells. For both of the two labeling method, the slides were also examined by double labeling with the antibodies,neuronal nuclei (NeuN) or glial fibrillary acidic protein (GFAP) to identify the differentiation of transplanted cells.
RESULTSBoth single ES cell and cell pellets expressed bright green fluorescence the day after plasmid transfection, and more than 30% ES cells were labeled. The GFP-labeled cells could still be found gathered around the infusion channel at least 21 days later, but the GFP fluorescent could not be overlapped with NeuN or GFAP staining. On the contrary, Thy-1 antibody overlapped well with NeuN or GFAP staining.
CONCLUSIONSLiposome-helped plasmid GPF transfection is effective in labeling mouse embryonic stem cell in vivo,but is not effective in showing the differentiated cells. On the contrary, Thy-1 antibody can not only show the transplanted cells, but also trace the transplanted cells after their differentiation.
