1.Spinal Locomotor Pattern Generator (review)

It is generally accepted that locomotion in mammals, including humans, is based on the activity of neuronal circuits within the spinal cord (the central pattern generator, CPG). Recent studies indicate a plastic behavior of the spinal neuronal circuits following a spinal cord injuries (SCI). Studies have shown that body weight support treadmill training (BWSTT) is effective in restoring locomotor function in cats with a complete spinal lesion. It has become possible to regain some locomotor activity in patients with spinal cord injury through an intense training on BWSTT, as in cat. So, it is a staired problem how to take advantage of the plasticity of spinal locomotor pattern generator and design the gait training program for SCI patients.
3.The effects of 240 g/L EDTA gel on the attachment and proliferation of human periodontal ligment cells onto the root surface of teeth with periodontitis

Objective:To study attachment and proliferation of human periodontal ligment cells(HPDLCs) on demineralized root surface of teeth with periodontitis following EDTA gel application. Methods:44 single-rooted teeth extracted because of severe periodontitis were treated by scaling and root planing(SRP). Then the fragments(5 mm?4 mm) of experimental surfaces were prepared. The fragments were randomly divided into four groups.The root surfaces in group 1,2,3 and 4 were treated by 240 g/L of EDTA gel for 2,3 and 4 min respectively or without treatment(control). Then 1?104 cells were seeded on the surface of each fragment and routinely cultured for 1,3 and 5 days respectively after seeding, the specimens were prepared for microscopic observasion, the cells on the root surfaces were counted. Moreover the other eight fragments were prepared for SEM observasion. Results:HPDLCs attached to and proliferated on all treated surfaces.Group 1, 2 and 3 presented significantly more cells on the root surfaces than the control group did at day 1,3 and 5. The cells on the root surfaces in group 2 and 3 were more than those in group 1 at day 1, 3 and 5. There was no evident difference between group 2 and 3 at any time. The root surfaces in group 2 and 3 had a fibrillar texture associated with an increased diameter of the exposed dentinal tubule openings.Smear layer were observed on root surfaces in group 1. Conclusion:EDTA gel(at 240 g/L) application may enhance attachment and proliferation on root surface of HPDLCs.
4.Evidence-based diagnosis of pulmonary embolism

Objective To search for evidence of diagnosis of pulmonary embolism for guiding clinical practice.Methods We searched PubMed(1970~2005-10)and CBM(1992~2005-10)in Oct.2005 to identify system reviews(SR)and cross-sectional study(CSS)about the diagnosis of pulmonary embolism.Results Sixteen SR and 9 CSS were identified in PubMed,none in CBM.Pulmonary arteriography was the gold standard in the diagnosis of pulmonary embolism.Our result showed that computed tomography,ventilation-perfusion scanning and doppler ultrasound were useful in the diagnosis of pulmonary embolism combind with clinical judgment.Multidetector CT might be another gold standard in the diagnosis of pulmonary embolism.A negative result on quantitative rapid ELISA of D-dimer could exclude the diagnosis of pulmonary embolism.Conclusion Diagnostic tests combind with clinical judgment are useful for diagnosing or excluding pulmonary embolism.
5.Expression of the novel serine protease SNC19 in different kinds of cells lines and its effects on cellular biological behaviors

AIM: To investigate the physiological function of the novel serine protease SNC19 protein and its possible role in cancer invasion and metastasis. METHODS: Monoclonal antibodies directly against SNC19 extracellular domain was prepared. The protein and SNC19 mRNA expression were determined in different kinds of cell lines respectively by Western blot and Northern blot analysis. Cellular migration and adhesion abilities were assayed by monoclonal antibody blocking method. RESULTS: Western blot analysis showed there were two bands of SNC19 protein in BCAP37, COLO205, SW480 cells at about 120 kD and 60 kD while only one band in SW620 cells at 60 kD; Northern blots showed a approximate 3.4-kilobase fragment appearing in most epithelial-derived cell lines with this only form and high levels but no detection was obtained in OV, TCA8113, KB and SGC7901 cells. In antibody blocking experiments, the migration of SW480 cells was significantly inhibited compared with the control and the abilities of (SW480/SW480), SW480/NIH3T3 adhesion increased at the beginning of the experiments, but the difference reduced (along) with the time passed.CONCLUSION: SNC19 protein is closely related with cellular homogeneous and heterogeneous adhesion as well as cellular motility. As a novel serine protease, it may participate both in physiological and pathological processes, such as cell migration, tissue remodeling and cancer invasion and metastasis.
6.Association of rs10004195 single nucleotide polymorphisms in Toll like receptor 10 gene with helicobacter pylori infection and associated diseases

Objective To investigate the association of rs10004195 single nucleotide polymorphisms in Toll like re-ceptor 10(TLR 10) gene with helicobacter pylori infection and associated diseases. Methods A total of 652 pa-tients who has been examined by gastroscopy were obtained, and then peripheral blood samples were collected from all the patients. Immune colloidal gold method was used to test the serological Hp antibody of all the patients. The TLR10 gene rs10004195 polymorphisms were examined by direct DNA sequencing of the PCR products. Results The frequencies of AA,TT and AT genotype on TLR10 rs10004195 were 30. 98%, 20. 71%, 48. 31%;there was significant difference between Hp antibody positive group and Hp antibody negative group in the TT frequencies of TLR10 rs10004195 ( P<0. 05 ) . No significant difference between controls and Hp associated diseases groups in Hp antibody positive group or in Hp antibody negative group were observed. Conclusion There was correlation be-tween the TLR10 rs10004195 loci genotype and the risk of Hp infection, but no correlation with Hp associated dis-eases.
7.Preparation of LEM-GM-CSF and its inhibition effect on HL60/ADM cells in vitro

Objective To prepare liposome-entrapped mitoxantrone (LEM)-GM-CSF and observe the cytotoxicity of HL-60/ADM cells treated with LEM-GM-CSF, LEM and dihydroxyanthraquinone (DHAQ) in vitro. Methods LEM was prepared by reverse phase evaporation (REV). High speed centrifugation was applied to separate LEM and dissociate DHAQ. Colorimetry was employed to determine encapsulation efficiency. The liposome structure and particle size were determined by transmission electron microscopy. GM-CSF was coupled to LEM by glutaraldehyde method. UV-spectrophotometric analysis was applied to measure the coupled efficiency. Flow cytometry was applied to determine the immunoconjugate retained efficiency. The cytotoxicity of HL60/ADM cells and interdiction efficiency of GM-CSF were investigated by MTT test. Results The encapsulation efficiency of LEM was 80%. Most liposomes were monolayer, and the particle size was 170-220 nm. Its coupled efficiency with GM-CSF was 42.3%, and the immunoconjugate retained efficiency was 74.6%. All LEM-GM-CSF, LEM and DHAQ had cytotoxicity on HL60/ADM, their cytotoxic power in decrement sequence: LEM-GM-CSF, LEM, DHAQ. After treated with LEM-GM-CSF, LEM and DHAQ for 24 h, the IC50 of HL-60/ADM was 8.73, 12.42, 27.31 ?g/ml respectively and for 48 h the IC50 were 0.62, 8.25, 12.44 ?g/ml. The inhibition rate increased in a dose-dependent manner. Conclusion The encapsulation efficiency, the coupled efficiency and the immunoconjugate retained efficiency of LEM-GM-CSF prepared by our method were satisfying. LEM-GM-CSF representing anti-leukaemia efficiency in vitro had cytotoxicity on HL60/ADM cells.
9.Influence of lumbar disc degeneration on the efficacy of lumbar fixed-point rotation manipulation in sitting position:a finite element study

Objective:To investigate the influence of lumbar disc degeneration on the efficacy of lumbar fixed-point rotation manipulation in sitting position in treating lumbar intervertebral disc herniation (LIDH). Methods:We simulated lumbar fixed-point rotation manipulation of sitting position using three finite element models including a normal model, a mild degeneration model and a moderate degeneration model of L3-5, in which the herniated disc was assumed at the left rear of L4 disc and the rotation manipulation was carried out on the right side. The displacement and stress at the left rear of L4 disc of the three models were analyzed. Results:When lumbar fixed-point rotation manipulation in sitting position was carried out, a displacement and stress were generated at the left rear of L4 intervertebral disc of the three models directing forward. The displacement and stress in degeneration models were less than those in the normal model, and the smallest values were found in the moderate degeneration model. From normal model to mild and then to the moderate degeneration model, the displacement decreased by 36% and 59%, and the stress decreased by 22.3% and 45.2%, respectively. Conclusion:The lumbar disc degeneration affects adversely the effectiveness of lumbar fixed-point rotation manipulation in sitting position in the treatment of LIDH. The severer the lumbar degeneration, the greater the influence.
10.The study of human melanoma-specific antigen peptides by HLA-A2 restricted tumor-infiltrating lymphocytes

Objective:To explore the characteristics of human melanoma-specific antigen peptides by HLA-A2 restricted tumor-infiltrating lymphocytes.Methods:The HLA-A2 protein and polypeptides molecules were purified from the three tumor cell lines(624-Mel, Chap-Mel and JY) by immunoaffinity chromatography, after the peptides bound to HLA-A2 protein solution were acidified with acetic acid and boiled by high temperature, and centrifuged through an Ultra-CL filter, then the peptides extracts were fractionated by revered phase high pressure liquid chromatography(RP-HPLC). Individual fractions were assessed for their ability to reconstitute melanoma-specific epitopes by adding to the HLA-A2 Ag-procceing mutant cell, T2. The biological feature of one of three active peptides from RT-HPLC samples was performed by mass spectrometric analysis. The synthetic peptides identical to active peptide sequences were determined in the reconstitute test.Results:Three prominent peaks(P19, P25 and P31) of the fraction from 624-Mel were observed in the reconstitute test, TIL killing rate was 67% for (P31) peptide fraction. The mass spectrometric analysis of one of active peptides (P31) showed that at mass-to-charge ratio(m/z) 948 has been usually nine residues. The sequence is H+ Ala Lue Trp Lue Phe Phe Gly Val Lue OH-. The peptide synthesized comprising epitopes were verified.Conclusion:These results showed the peptides derived from active fractions were related to human melanoma-specific tumor antigen peptides recognized by HLA-A2-restriced TIL. These peptides could develop novel peptide-based an anti-tumor vaccine for immunotherapy of CTL.
