Objective:To study attachment and proliferation of human periodontal ligment cells(HPDLCs) on demineralized root surface of teeth with periodontitis following EDTA gel application. Methods:44 single-rooted teeth extracted because of severe periodontitis were treated by scaling and root planing(SRP). Then the fragments(5 mm?4 mm) of experimental surfaces were prepared. The fragments were randomly divided into four groups. The root surfaces in group 1,2,3 and 4 were treated by 240 g/L of EDTA gel for 2,3 and 4 min respectively or without treatment(control). Then 1?104 cells were seeded on the surface of each fragment and routinely cultured for 1,3 and 5 days respectively after seeding, the specimens were prepared for microscopic observasion, the cells on the root surfaces were counted. Moreover the other eight fragments were prepared for SEM observasion. Results:HPDLCs attached to and proliferated on all treated surfaces.Group 1, 2 and 3 presented significantly more cells on the root surfaces than the control group did at day 1,3 and 5. The cells on the root surfaces in group 2 and 3 were more than those in group 1 at day 1, 3 and 5. There was no evident difference between group 2 and 3 at any time. The root surfaces in group 2 and 3 had a fibrillar texture associated with an increased diameter of the exposed dentinal tubule openings.Smear layer were observed on root surfaces in group 1. Conclusion:EDTA gel(at 240 g/L) application may enhance attachment and proliferation on root surface of HPDLCs.

Objective:To investigate the influence of lumbar disc degeneration on the efficacy of lumbar fixed-point rotation manipulation in sitting position in treating lumbar intervertebral disc herniation (LIDH). Methods:We simulated lumbar fixed-point rotation manipulation of sitting position using three finite element models including a normal model, a mild degeneration model and a moderate degeneration model of L3-5, in which the herniated disc was assumed at the left rear of L4 disc and the rotation manipulation was carried out on the right side. The displacement and stress at the left rear of L4 disc of the three models were analyzed. Results:When lumbar fixed-point rotation manipulation in sitting position was carried out, a displacement and stress were generated at the left rear of L4 intervertebral disc of the three models directing forward. The displacement and stress in degeneration models were less than those in the normal model, and the smallest values were found in the moderate degeneration model. From normal model to mild and then to the moderate degeneration model, the displacement decreased by 36% and 59%, and the stress decreased by 22.3% and 45.2%, respectively. Conclusion:The lumbar disc degeneration affects adversely the effectiveness of lumbar fixed-point rotation manipulation in sitting position in the treatment of LIDH. The severer the lumbar degeneration, the greater the influence.

OBJECTIVETo compare the proteomic profiles of whole unstimulated saliva (WUS) of subjects with chronic periodontitis (CP) with that of healthy volunteers, and to identify proteins.
METHODSWUS was obtained from five subjects with CP and five healthy subjects, and proteins were separated using two-dimensional gel electrophoresis (2-DE). Proteins, the level of which was significantly different among the two groups, were identified by computer image analyses and subsequent matrix-assisted laser desorption/ionization tandem time-of-flight mass spectrometry (MALDI-TOF-TOF MS).
RESULTSSeventeen differentially expressed points in the 2-DE gel between the two samples were found by professional software. And protein types of thirteen points were then identified. Among them, the point 737, 725, 692, 986 was the same protein, as well as the point 716, 535. They were respectively serum albumin; beta-fibrin; chain B, human serum transferrin, recombinant N-terminal lobe, Apo form, crystal form 2; Ig alpha-1 chain C region; the GA module complexed with human serum albumin (HAB-GA); chain B, crystal structure of a Rca-Rhogdi complexes; PRO2044; hypothetical protein; unnamed protein. Compared with whole saliva of healthy control subjects, the levels of them were increased in whole unstimulated saliva of CP subjects.
CONCLUSIONComparison of the proteomic profiles of WUS of CP subjects with that of healthy control subjects reveals at least 9 differential proteins. The approach might be helpful to better understand to the etiology of CP.

Objective To investigate the correlation between the hemodynamic characteristics of time intensity curve(TIC) by contrast enhanced ultrasound(CEUS) and the expression of antigen Ki-67 of the 4 different clinic stages (T0 andTis,T1 ,T2,T3) in ductal carcinoma in order to predict the grade of the expression of antigen Ki-67 and clinic stages. Methods Masses founded in 80 cases proved by pathological test after resection or needle biopsy were divided into 4 groups base on clinic stages:group 1(T0 andTis) , group 2(T1),group 3(T2),and group 4(T3).Some parameters gained after analyzing TIC of CEUS would be used to make a rank correlation analysis with the expression grade of antigen Ki-67 through immunohistochemical diagnosis.A regression equation would be gained from those parameters which had a correlation with the expression grade of antigen Ki-67 (P < 0.05). Results Most TIC of ductal carcinomas were possessed of the characteristics of fast in and slow-out.Compared to the other three groups, group 4(T3) had significant differences in arrive time, time to peak, arrive intensity, peak intensity and area under curve.Compared to group 1(T0 andTis),group 4(T3) had significant differences in continuous time and rising curve's slope and group 3(T2) has significant differences in arrive intensity and area under curve. Parameters entered the regression equation through mult-variable linear regression analysis were arrive time, continuous time, peak intensity and rising curve's slope.R square was 0.83 1 and adjust R square was 0.822.Through the regression equation verification,5 cases accorded with the clinic stages standard of ductal carcinoma. Conclusions The hemodynamic characteristics of time intensity curve (TIC) by contrast enhanced ultrasound had a good correlation with the expression grade of antigen Ki-67 of the 4 different clinic stages in ductal carcinoma. The regression equation had a good explanatory and degree of fitting,it can be used to predict the expression grade of antigen Ki-67 and indirectly forecasts clinic stages of ductal carcinoma.

A cellulase high-yield strain was identified and named as Trichoderma longibrachiatum SSL by ITS sequence identification. The endoglucanase1 gene (eg1) encoding endo-l,4-?-D-glucanase I was ampli-fied by RT-PCR method, which including 1386 bp and encoding 461 amino acid. Sequence analysis showed that: This gene has a more 90% homology with the T. longibrachiatum eg1 gene. The eg1 gene encoding the mature peptide was inserted into the Pichia pastoris expression vector pPIC9K, which resulted in construc-tion of the recombinant expression plasmid, pPIC9k-eg1. The pPIC9k-eg1 was then introduced into the host Pichia pastoris GS115. After the induction of methanol, extracellular recombinant endoglucanase I from the supernatant of the recombinant Pichia pastoris strain reached 73 U/mL. A clear strengthening of the protein bands, whose molecular weight is about 58 kD, appeared in the SDS-PAGE.
