Objective To search for the optimal processing procedure and physico-chemical properties of Endothelium corneum amylase. Methods After dried under 50 ℃ ,amylase was extracted from Endothelium corneum with phosphate-buffered saline. The amylase activity was examined at different temperature,pH and different metal ions,and amylase isozyme was discerned on PAGE. Results The optimal pH,optimal temperature,and optimal substrate concentration for amylase activity were 8.67,50 ℃ and 1.8 g/mL,respectively. Under the experimental conditions,the amylase Km was 0.92 (starch was used as substrate of amylase). Four metal ions tested,including Ca2+,Fe2+,Zn2+,Mn2+,could inhibit the amylase activity to some extent. Furthermore,it showed one amylase isozyme band using PAGE,suggesting only one amylase isozyme existing in the Endothelium corneum tentatively. Conclusion The effects of metal ions should be considered in the process of Endothelium corneum amylase.

Objective To observe the change of umbilical artery S/D value in the third trimester pregnant women with overt hypothyroidism(OH) or subclinical hypothyroidism(SCH).Methods We collect the data of 37-42 weeks pregnancy from January 2014 to December 2015.We devided them into OH group,SCH group and contral group.We compared the umbilical artery S/D value in these three groups and analyzed the relativity between S/D value and thyroid hormone.Results The S/D value was 2.46 ± 0.31 in OH group,2.48 ± 0.27 in SCH group and 2.32 ± 0.17 in control group.The differences were statistically significant between OH group and contral group (P =0.000),SCH group and contral group (P =0.000).There was no statistically significant between OH group and SCH group (P =0.907).The proportion of cases number during different S/D value range in these three groups had difference (P =0.000).The S/D value was positively correlated to TSH in OH group (r =0.186,P =0.037) and SCH group (r =0.146,P =0.015).There was no correlation between FT3 and S/D value(P ＞ 0.05) or FT4 and S/D value(P ＞ 0.05) in both OH and SCH group.There was no correlation between thyroid hormone and S/D value in the contral group either.Conclusion Both OH and SCH can effect umbilical artery S/D value in the third trimester pregnancy.

As one branch of epigenetics, the sirtuins family ( ClassⅢ histone deacetylase) receive much attention in recent years. SIRT1 as the most famous of the sirtuins family members has been verified involved in a variety of age-related diseases. While the SIRT1 formation is paid more and more attention in age-related cataract. Now, we briefly overviewed the research progress on the role of SIRT1 in age-related cataract.

This study was aimed to observe the clinical effect of Pingai Mixture intervention on HIV/AIDS patients in order to clarify the characteristics of Pingai Mixture for AIDS treatment. A total of 41 HIV/AIDS patients who met the inclusion criteria were selected. Pingai Mixture was applied in the treatment. Three months was one treatment course. The treatment was given for two courses. Clinical symptoms, physical signs and changes in CD4+ lymphocytes of HIV/AIDS patients before and after treatment were observed. The SPSS software was used for statistical analysis. The results showed that the posttreatment total score (primary, secondary symptoms and the main physical sign cred-its) was significantly reduced compared with pretreatment (P < 0.01). The posttreatment spontaneous perspiration, night sweats, hair loss and other symptoms of a separate score was significantly reduced compared with pretreatment (P< 0.05). There were no significant changes on other symptoms or physical sign indicators. There was no significant difference in the Karnovsky score after 6-month treatment compared with pretreatment. It was concluded that Pingai Mixture is effective in AIDS treatment. It can improve the clinical symptoms and the administration is safe.

This study aims to construct a eukaryotic expression system for envelope gene of Jaagsiekte sheep retrovirus, observes its localization in 293T cells, and investigates the potential in inducing malignant transformation of NIH3T3 cells. By RT-PCR, the full-length cDNA of envelope gene of Jaagsiekte sheep retrovirus (exJSRV-env) was amplified from the extract of naturally infected sheep lung. The clone of target gene was sub-cloned into eukaryotic expression system pEGFP-C1, and validated by PCR, restriction endonuclease, and sequencing. Bioinformatic analysis concerning biological function and cellular localiza tion of exJSRV-env was also performed. The recombinant clone of exJSRV-env was transfected into 293T cells and NIH3T3 cells by Lipofectamine LTX. The expression and celluar localization in 293T cells were validated by confocal microscopy. Soft agar colony formation assay was employed to test the anchorage-independent growth of NIH3T3. DNA sequencing and restriction enzyme digestion with Kpn I and Hind III indicated the correct construction of the recombinant plasmid, which was named pEGFP-C1/exJSRV-env. Amino acid sequence alignment of exJSRV-env with reference sequences found 85%-100% homogeneity. A YRNM motif was discovered at the cytoplasmic tail of envelope gene, which is exclusively found in exogenous viruses. Phylogenetic tree analysis showed that our clone of exJSRV-env clustered closely with pathogenic exogenous Jaagsiekte sheep retroviruses. Fluorescence microscopy indicated typical membrane localization of exJSRV-env protein. NIH3T3 cells transfected with exJSRV-env lost contact inhibition, and acquired colony forming ability in soft agar. This study indicated that envelope protein of Jaagsiekte sheep retrovirus can induce malignant transformation of mouse fibroblast cell NIH3T3. Discoveries of this study provide a basis for further structural and functional research on Jaagsiekte sheep retrovirus envelope protein.

Objective To observe the levels of serum leptin in Gaves disease(GD)and thyroiditis(HT)Datients and to discuss the immunological role of leptin in the pathogenesis of autoimmune thyroid disease(AITD).Methods 102 newly diagnosed female AITD patients were divided into 3 groups:GD hyperthyroid group,HT hypothyroid group and subclinical hypothyroid group.Age,sex and BMI-matched 27 euthyroid,healthy subjects served as controis.The levels of FT3,FT4 and sTSH were determined by immunofluorometrie assay.ELISA kit was aDplied to measure the levels of serum leptin.Results Serum FT3 and FT4[(19.74±15.39),(78.25±58.68)pmol/L]levels of GD hyperthyroid patients were obviously higher than those of the controls[(4.87±0.25),(15.96±3.15)pmol/L,P＜0.01],but serum sTSH and leptin levels[(0.15±0.08)mU/L,(8.73±1.92)μg/L]were obviously lower than those of the controls[(3.81±0.19)mU/L,(12.38±3.51)μg/L,P＜0.01or＜0.05].Serum FT3 and FT4[(3.36±0.26),(6.95±3.29)pmol/L]levels of HT hypothyroid patients were obviously lower than those of the controls(P＜0.05),but serum sTSH and leptin levels[(45.48±35.83)mU/L,(17.17±3.82)μg/L]were obviously higher than those of the controls(P＜0.01 or＜0.05).Serum FT3 and FT4[(4.67±0.60),(14.87±2.14)pmol/L]levels of subclinical hypothyroid patients had not statistical difference comparing with those of the controls(P＞0.05),but serum sTSH and leptin levels[(13.67±8.66)mU/L,(16.25±3.67)μg/L]were obviously higher than those of the controls(P＜0.01 or＜0.05).Conclusions Leptin might have an immuoregulation role in the pathogenesis of AITD.In addition,serum levels of leptin in AITD is also influenced by many other related hormones.

AIM: To evaluate the efficacy of implantable collamer lens (ICL)-V4c implantation in the correction for high myopia. METHODS: This retrospective analysis collected twenty-five patients (43 eyes) with high myopia treated with ICL/TICL implantation from September to December 2016 at Department of Ophthalmology, West China Hospital. Routine ophthalmic examinations were performed. The patients were followed 3mo to monitor uncorrected visual acuity (UCVA), best corrected visual acuity (BCVA), refractive error, intraocular pressure, corneal endothelial cell count, contrast sensitivity and scores for Quality of Life lmpact of Refractive Correction Questionnaire ( QIRC ). Statistical analyses were performed using repeated measures ANOVA, LSD-t test and paired-samples T test. RESULTS: Postoperative UCVA and BCVA were significantly higher than before surgery (all P<0. 01). Postoperative spherical mirrors at all time points were lower than before surgery(all P<0.01),and there was no significant difference between the different time point postoperative (all P>0. 05). The safety index in 3mo postoperatively was 1.38士0.24 and the effective index was 1. 22士0. 15. There was no statistical difference in intraocular pressure between preoperative and postoperative time points (F=1.464, P=0.215). There was no statistical difference in endothelial cell density at 3mo after surgery compared to preoperative (t=-0.544, P= 0. 586). The postoperative Log contrast sensitivity (LogCS) of 12.0 cycles per degree (c/d) and 18.0 c/d were significantly higher compared to the preoperative (F=4.347, P=0.010; F=4.539, P=0.005; respectively), but other (the postoperative LogCS of 3.0 c/d and 6.0 c/d) differences were statistically insignificant (F=1.094, P=0.354;F=1 325,P=0.271;respectively). There was no significant difference in the amount of change in the QIRC scores between preoperative and postoperative(F=2.669, P=0 094). CONCLUSION: ICL V4c implantation is a safe and effective method for the correction of high myopia.

The heat shock protein ClpB is a member of the Clp family and functions as molecular chaperones. ClpB is related to the acquired thermotolerance in organisms. A cDNA of 3144 bp was screened out of a tomato cDNA library. The polypeptide deduced from the longest ORF contains 980 amino acid residues, and was classified into HSP100/ClpB family based on the result of molecular phylogenesis analysis. Thus it was named as LeHSP110/ClpB according to its calculated molecular weight. LeHSP110/ClpB was characteristic of heat-inducibility but no constitutive expression, and was demonstrated to locate in chloroplastic stroma. An antisense cDNA fragment of LeHsp110/ClpB under the control of CaMV 35S promoter was introduced into tomato by Agrobacterium tumefactions-mediated method. At high temperature, the mRNA levels of LeHsp110/ClpB in antisense transgenic plants were lower than those in control plants. The PS II of transgenic plants is more sensitive to high temperature than that of control plants according to data of Fv/Fm. These results clearly showed that HSP110/ClpB plays an important role in thermotolerance of high plants.

OBJECTIVETo compare the effects of acupuncture anesthesia (AA) and local anesthesia (LA) on the blood pressure (BP) and the heart rate (HR) in chronic rhinitis patients accompanied inferior turbinate hypertrophy in low temperature radiofrequency ablation (RFA).

METHODSTotally 61 chronic rhinitis patients accompanied inferior turbinate hypertrophy were randomly assigned to the AA group (31 cases) and the control group (30 cases). All patients received RFA respectively under AA and RA. Their heart rate (HR), systolic pressure (SP), diastolic pressure (DP), and mean pressure (MP) were recorded and compared at 10 minutes after their entry into the operating room, immediately before surgery, intraoperation, and 5 min after operation.

RESULTSWhen compared with the control group at the same time points, the SP, DP, MP, and HR all decreased at intraoperation and 5 min after operation in the AA group. There was statistical difference in the SP on the right side at the 2nd melting point and the HR on the left side at the 2nd melting point between the two groups (P < 0.05). Compared with the same group at 10 min after entry into the operating room, the SP on the right side at the 1st melting point, the SP, DP, and MP on the right side at the 2nd melting point, the SP, DP, and MP on the left side at the 1st melting point, and the SP on the left side at the 2nd melting point all obviously increased with statistical difference (P < 0.05, P < 0.01). When compared with the same group at 10 min after entry into the operating room and immediately before surgery, there was no statistical difference in the SP, DP, MP, or HR of the AA group at intraoperation and 5 min after operation (P > 0.05). When compared with the same group at 10 min after entry into the operating room and immediately before surgery, there was no statistical difference in HR of the control group at intraoperation and 5 min after operation (P > 0.05).

CONCLUSIONPatients undergoing AA had less fluctuation of the BP and the HR, indicating AA had better analgesic effects.
