2.Down-regulation of leucine-rich repeats and immunoglobulin-like domain proteins (LRIG1-3) in HP75 pituitary adenoma cell line.

Three human leucine-rich repeats and immunoglobulin-like domains (LRIG) genes and proteins, named LRIG1-3, has been previously characterized and it was proposed that they may act as suppressors of tumor growth. The LRIG1 protein can inhibit the growth of tumors of glial cells and the down-regulation of the LRIG1 gene may be involved in the development and progression of the tumor. Real-time reverse transcription-polymerase chain reaction (RT-PCR) is a recently developed technique for quantitative assessment of specific RNA levels. In the current study, it was demonstrated that LRIG1-3 and EGFR mRNA was detected in human pituitary adenoma cell lines and a normal pituitary sample, with differences in the expression levels. Compared to the normal pituitary samples, the expression of LRIG1-3 in HP75 cell line was lower, but the expression of EGFR in HP75 cell line was higher. The results are consistent with LRIG1-3 being tumour suppressor genes, and LRIG genes decreasing the expression of EGFR. The ratio of EGFR/LRIG1 was increased at least 13-fold in HP75 cells compared with the normal pituitary cells, which was also the case for the ratio of EGFR/LRIG2 (14-fold increase in HP75) and EGFR/LRIG3 (11-fold increase in HP75). Further studies were needed to elucidate the explicit role of LRIG genes as negative regulators of oncogenesis in human pituitary adenoma.

OBJECTIVE:To evaluate the research and application situation of Sunshine Medicine Electronic Monitoring Data Analysis System. METHODS:The Sunshine Medicine Electronic Monitoring data Analysis System based on business intelligence technology was introduced in respects of design process,development,implementation and application example. RESULTS:The whole architecture of the system mainly includes hospital business platform,data integration platform,information processing plat-form and application service platform;the functions of the system include medicine homepage show,single species analysis,antimi-crobial agent analysis,national essential medicine analysis,injection analysis and sunshine medicine analysis. It can monitor the drug utilization in multi-angle and multi-level manners by building data center and creating multidimensional models. Besides,the sys-tem could solve thedrugs unified coding and information controlproblems,data collection and information integrationprob-lems in different hospital,andthe efficient calculation and analysis of a large number of drug use data. It can realize drug analy-sis and monitoring in the hospital,analysis and online monitoring of drugs prescribed by the doctor,finding,warning and evaluat-ing abnormal phenomenon of drug use in the medical institutions,so that it is better for the supervisors to monitor the usage of drugs. CONCLUSIONS:The system with easy operation,flexible monitoring,rich chart shows,comprehensive monitor index has a positive effect on rational medication level.

To investigate the anticancer effects of ring C in 18β-glycyrrhetinic acid (GA), a series of GA derivatives featured with 9(11)-ene moiety in ring C were designed and synthesized. The structures were confirmed by IR, LC-MS and 1H NMR. Their inhibitory effects towards human prostate cancer PC-3 and leukemia HL-60 cell lines were determined. Most of the derivatives displayed stronger antiproliferative activities than GA. Particularly, compound 14 showed promising anticancer activity with the GI50 values of 4.48 µmol · L(-1) and 1.2 µmol · L(-1) against PC-3 and HL-60 cells respectively, which is worth further study.

The paper is intended to study characteristics of different courses,to find out the correlation between courses and Chinese Nursing License Test so as to improve teaching methodology and rate of passing test by means of the factor analysis of 156 students' academic performance who passed nursing license test.On the basis of the results of factor analysis,a regression prediction model between courses and marks of nursing license test has been set up and this model has been proved to have higher prediction with lower mistake.
8.Positron emission tomography imaging of cell transplantation in a rat model of Alzheimer's disease.

OBJECTIVETo explore the value of positron emission tomography (PET) in the Alzheimer's disease (AD) rat model verification and in monitoring the therapeutic effectiveness of cell transplantation.
METHODSA beta(1-40) hippocampus injected rat model was successfully established and neural stem cells were injected into hippocampus. Results of behavior tests and histological examinations were compared between model group and graft group, and then the N-methyl-[(11)C]2-(4 methylaminophenyl)-6-hydroxybenzothiazole ((11)C-PIB) and (18)F-fluorodeoxyglucose ((18)F-FDG) imaging were performed to observe whether the result of imaging was matched with behavior test and histological examination.
RESULTSThe Morris water maze showed that the latent period of the escape was significantly longer in model group than in control group (P<0.01). In histological examinations, the neuron loss and A beta deposition were found in hippocampus CA1 and dentate gyrus of rat model. (11)C-PIB imaging showed increased uptake in model rat hippocampus district (P<0.05), while (18)F-FDG imaging showed that the uptake in the injected side of hippocampus in model group was significantly lower than that in the same side in control group (P<0.001). After cell transplantation, the latent period of the escape was significantly shorter in graft group than in model group (P<0.01). Histological examinations showed that there was no obvious changes in A beta deposition; in addition, the neural stem cells differentiated and expressed neuronal nuclei-positive cells, and continuously expressed 5-bromodeoxyuridine-positive cells for six weeks. (11)C-PIB imaging and (18)F-FDG imaging showed the uptakes were not significantly different between between model group and transplantation group(P>0.05).
CONCLUSION(11)C-PIB imaging is useful in diagnosing AD and monitoring the pathological change of AD model in vivo, while (18)F-FDG imaging provides useful visual information for monitoring short-term therapeutic effectiveness of stem cell transplantation.

9.The clinical features and strategies in the treatment of brain tumor in the elderly

Objective To study the clinical features and to improve the treatment on the elderly patients with brain tumor.Methods Retrospective analysis of 163 cases with brain tumor which had been confirmed by CT,MRA or pathology.Results Of all the 163 cases,121 were located in supratentorium,most of which were meningiomas and gliomas.Most patients(129 cases)had comorbidity.After operation,symptoms disappeared or obviously improved in 126 cases,moderately improved in 19 cases,and did not changed in 6 patients.Twelve cases died after operation in a month, in which 9 patients were over 75 years old.The death rate of operation was 6.1%.Conclusions It is important to know the atypical manifestation of brain tumor in the elderly,which may prevent clinical misdiagnosis and mistherapy.The perioperative management is indispensable to the prognosis of the patients.The choice of operation and medication should be in individualized.
