Eight-weeks old C57BL/6J male mice were treated with normal or high fat diet for4 weeks, and then bone mass, number and activity of osteoclasts, differentiation of osteoblasts from bone marrow mesenchymal cells and the change in lipid genes expression were studied.Compared with the normal diet treated group, bone mass of femur from high fat diet treated animal was decreased, but there were increased number and activity of osteoclasts and raised lipid genes expression levels.Besides,no obvious change was found in differentiation of osteoblasts from bone marrow mesenchymal cells. The loss of bone mass induced with high fat diet in young mice was mainly due to increased activity of osteoclasts and bone resorption.

Objectives To investigate the value of hepatobiliary scintigraphy in evaluation of the hepatic uptake and excre-tion function in neonatal intrahepatic cholestasis caused by citrin deifciency (NICCD). Methods Hepatobiliary scintigraphy with SPECT was used to detect the hepatic uptake and excretion function of 12 patients with NICCD conifrmed by SLC25A13 gene analysis, and the results were compared with the results of blood biochemical tests. Results The hepatic uptake and excretion function were obviously impaired in all of 12 NICCD patients in the initial scintigraphy. The scintigraphy were performed again in 5 patients in the follow-up after treatment, and showed that the hepatic uptake and excretion function was recovered. It was sug-gested that the hepatic uptake and excretion function was consistent with the level of liver enzymes and the degree of cholestasis. Conclusions Hepatobiliary scintigraphy is of value in evaluation of the hepatic uptake and excretion function in NICCD patients.

Objective To detect the effective strategy for monitoring the early liver leisure. Methods Two grups,in- cluding clinical symptoms with ALT test and clinical symptoms with PA and ALT test, were used to monitor the liver leis- ure. Results There are 74% of cases founded occurring liver leisure during anti-tuberculosis therapy after administe- ring the latter strategy including clinical symptoms with PA and ALT test. The incidence of occurring liver leisure is 51% at one week, none of the defaults occurred after onset of drug induced liver leisure when they were administered the liver protection drugs, and the rate of treatment completion is 100%,only 27% of patients was detected with liver leisure after using the former strategy including clinical symptoms with ALT test. The incidence of liver leisure is 2% at one week, the frequency of discontinued therapy is 16% after on-set of drug induced liver leisure when they took the liver protecting drug, and the full-time therapy rate is 84%.Conclusion The indicator including clinical symptoms,PA and ALT is prospective strategy in monitoring liver leisure, which can be used to detect the early liver leisure effectively induced by anti-tuberculosis drugs. The anti-tuberculosis therapy can be introduced smoothly, and the incidence of treatment fail- ure will be decreased by using liver protection drug after onset of drug induced liver leisure.

Objective Platelet activating factor(PAF),which has been implicated in the pathophysiology of inflammation in asthma,is degraded and inactivated by PAF acetlhydrolase(PAF AH).To investigate the association of PAF AH activity with genotype in asthmatic children. Methods We studied 57 asthmatic children and 30 normal controls. The plasma PAF AH genotype was detected as representative case with 3 different genotypes (Val/Val,Val/Phe and Phe/Phe) by allele specific polymerase chain reaction(AS PCR).The PAF AH activity in plasam was examined by the changes of substrate assay. Results In severe asthmatic individuals plasma PAF AH activities were lower than those of mild or moderate groups and control group, and plasma PAF AH activition was absent 15.4 %.In another three groups plasma PAF AH activation were absent 2 %-3 %.There was significant difference of plasma PAF AH activity among 3 groups of genotype(Val/Val,Val/Phe and Phe/Phe).In the similar genotype, there was no significant difference of plasma PAF AH activity between the groups of control and asthma. Conclusions There was imbalace of PAF/PAF AH in asthmatic children. In severe asthmatic individuals plasma PAF AH activities were lower than those of mild or moderate groups and control group. PAF AH(Val279Phe) gene mutation was related with plasma PAF AH activity.

Silica gel column chromatography and preparative HPLC were applied to isolate the chemical constituents from the roots of Ixeris chinensis. Fifteen compounds were isolated and their structures were elucidated by the physicochemical properties and spectral analysis as chinensioide G(1), chinensioide B(2), 10α-hydroxy-guaia-12,6-lactone-3-keton(3), chinensioide C(4), 10α-hydroxy-11βH-guaia-4(5) -ene-12,6-lactone (5), 3β, 10α-dihydroxy-11βH-guaia-11 (13)-ene-12,6-lactone (6), 3β, 10α-dihydroxy-4βH, 11βH-guaia-12,6-lactone(7), 3β, 10α-dihydroxy-guaia-4 (15), 11 (13) -diene-12, 6-lactone (8), caffeic acid (9), p-hydorxyphenylacetic acid(10), methyl p-hydroxyphenylacetate (11), ethyl p-hydroxyphenylacetate (12), sitosterol (13), daucosterol (14), and ixerin D(15). Compound 1 was new, and 6 and 7 were isolated from I. chinensis for the first time.
