Background Establising the culture model of Müller cells for obtaining the highly putified target cells is essential for the study about the physiology and pathology of retinal Müller cells. The exsiting purifing method for culturing Müller cells is dissatisfactory. Objective This study was to establish a method to obtain high purifing Müller cells. Methods The retina from 5 clean newborn SD rats were isolated and digested by 0. 01% trypsin and cultured in DMEM containing 10% fetal bovine serum. The cellular suspension was then prepared, and the target cells were screened using flow cytometry based on the size and the quantity of cells. Cultured and passaged cells were identified by transmission electron microscope and light microscope. Immunocytochemistry was used to detecte the expression of GFAP in cultured cells for the determination of type and purity of the cells. Results The cells showed the similar shape to retinal Müller cells after primarily culture with the large volume, and some small other types of cells could been seen. The growth of cells was quickly 3 weeks later. The fibroblasts were removed using sticking-wall by steps, and neurons were eliminated following passage. Aboundent of cellular organs were seen under the transmission electron microscope. The positive response rate of the cells for CFAP was 100%. Conclution Flow cytometry offer a rapid and feasible approach for purifying Muller cell and it builds the foundation for further study about Müller cells.

Objective To analyze the distribution and antimicrobial resistance of pathogens from patients with hospital infection in intensive care unit(ICU),and to discuss the corresponding strategy of prevention and treatment. Methods Clinical data of 891 patients hospitalized in the ICU of our hospital between Jan 2002 and Dec 2005 were collected. All the specimens were analyzed for pathogen distribution and resistance to antibiotics. Results Of all the isolated 1 155 strains,Gram-negative(G-)bacteria were the most popular pathogen(57.84%),whereas Gram-positive(G+) bacteria and fungi accounted for 21.39% and 20.78%,respectively.Most G-bacteria were less sensitive to imipenem(0-10.16%),and Pseudomonas aeruginosa was least sesitive to amikacin(6.89%) and cefoperazone-sulbactam(33.97%).G+ bacteria were most sensitive to vancomycin. Most fungus were Candida albicans(42.92%) and yeast species(22.90%). Conclusion Epidemiological investigation of pathogens in ICU is of great importance to the rational use of anti-biotics and may help to reduce the emergence of resistant strains.

BACKGROUND: Transforming growth factor-β(TGF-β) is an essential factor for pathological scar formation. Smad protein group is the signal protein of lower reaches of TGF-β receptor. Asiaticoside can inhibit proliferation of fibroblasts and synthesis of collagen to reduce TGF-β expression in the scar.OBJECTIVE: To study the mechanism of asiaticoside on proliferation of scar fibroblasts and phosphorylated Smad2 and Smad7.DESIGN: Controlled study with observation, in which cell was taken as the object.SETTING: Department of burn and plastic surgery of a hospital affiliated to a university.PARTICIPANTS: The experiment was performed in Surgical Laboratory in Sun Yat-sen University from April 2002 to March 2003. The specimens were selected from 6 inpatients receiving plastic operation due to hyperplasic scar including 3 male and 3 female cases aged varied from 1 to 35 years. The hyperplasic scar fibroblasts were obtained generated from original culture in laboratory of surgical department.INTERVENTIONS: In the research, the experiment group and the control group were divided. In the experiment group, asiaticoside was applied on fibroblasts; in the control group, asiaticoside was not prescribed. The changes of every index were observed before and after medication.MAIN OUTCOME MEASURES: ① Effect of asiaticoside on expression of phosphorylated Smad2 and Smad7; ② Effect of asiaticoside on cell cycle and apoptosis.RESULTS: Asiaticoside inhibited scar fibroblasts entering M phrase from S phrase and reduced the content of phosphorylated Smad2 in fibroblasts, which did not present significant difference in two groups ( t = 1.53, P =0.08).The content of Smad7 in the cells was (50. 80 ± 22.40)% in the experiment group and (32.18 ± 17.84)% in the control group, which indicated significant difference ( t = 2. 17, P = 0. 024).CONCLUSION: Asiaticoside inhibits scar formation by Smad passage.

Objective To evaluate the effect of remote limb ischemic postconditioning on the level of adiponectin during myocardial ischemia-reperfusion (I/R) in rats. Methods Fifty-seven male Sprague-Dawley rats, aged 8 weeks, weighing 250-350 g,were randomly divided into 3 groups (n =19 each):sham operation group (group S); group I/R; remote limb ischemic postconditioning group (group RLIP).Myocardial ischemia was induced by occlusion of anterior descending branch of left coronary artery for 30 min followed by 180 min reperfusion in urethane-anesthetized rats.In group S,the anterior descending branch was only exposed but not ligated. The animals underwent 10 min ischemia of bilateral hind limbs starting from 20 min of occlusion of the anterior descending branch, followed by reperfusion in RLIP group. The changes in S-T segment were recorded at 30,60,120 and 180 min of reperfusion. At 180 min of reperfusion, the blood samples were taken from the jugular vein for determination of serum levels of creatine kinase MB (CK-MB) and cardiac troponin Ⅰ (cTnⅠ).The animals were then sacrificed and hearts were removed for measurement of myocardial infarct size (IS) and adiponectin contents in myocardial tissues (by ELISA).Results Compared with group S,S-T segment was significantly elevated at each time point, myocardial IS,and serum cTnⅠ and CK-MB levels were increased, and adiponectin contents were decreased in I/R and RLIP groups.Compared with group I/R,elevation of S-T segment was significantly decreased at each time point, myocardial IS,and serum cTnⅠ and CK-MB levels were decreased, and adiponectin contents were increased in RLIP group. Conclusion The mechanism by which remote limb ischemic postconditioning reduces myocardial I/R injury is related to increased level of adiponectin in the myocardium of rats.
