1.The mechanism and correlative factors of SAM syndrome after aortic valve replacement

Objective: To investigate the mechanism and correlative factors of SAM syndrome after aortic valve replacement. Methods: 48 patients with severe aortic valve stenosis were studied, echo-Doppler study was performed one week after aortic valve replacement, and the patients were divided into two groups with or without SAM Syndrome. The left ventricular end-diastolic dimensions, the left ventricular end-Systolic dimensions, the left ventricular outflow tract the thickness of septum and posterior wall, the maximal left ventricular velocities and left intra-ventricular dynamic gradients were recorded and compared. Results: There was no operative mortality. The maximal left ventricular velocities were increased significantly in 9 patients (more than 2。5 m/s) and six of them had SAM syndrome. Objective To study the protective effect of galanin on 2,3-Dimethoxy-1,4-naphthoquinone (DMNQ)-induced oxidative stress and cell damage in HEK-293A cells and its possible mechanisms.Methods The expression of galanin and its three receptors (GalR1-3) in HEK-293A were determined with RTPCR technique.The cultured HEK-293A cells were divided into four groups:Control,DMNQ,DMNQ + GAL and DMNQ + AR-M1896 and the cell viability was measured with CellTiter 96 AQueous One Solution Cell Proliferation Assay (MTS).Results The RT-PCR data revealed the presence of galanin and its receptors in HEK-293A cells.The expression level was in the order of GalR2 =Galanin > GalR3 > GalR1.DMNQ caused oxidative stress and cell damage in HEK-293A cells with a dose-dependent manner with an IC50 =13.4 μM.Application of galanin reduced the DMNQ-induced cellular toxicity in HEK-293A,which increased cell viability by 24.4% and 18.8% in 1 μM and 100 nM,respectively.AR-M1896,an agonist of GalR2 had a similar effect,increased cell viability by 8.7% and 8.9% in 1 μM and 100 nM,respectively.Conclusion These data suggest that galanin has a protective effect on DMNQ-induced oxidative stress and cell damage in HEK-293A cells,probably mediated by GalR2.

5.Study on the remineralization effect of Aominqing Dental desensitizer on initial enamel lesions in vitro

Objective To investigate the remineralization effects of the Aominqing dental desensitizer and the fluoride dentifrice on the demineralized enamels. Methods Sixty-three teeth were randomly divided into three groups after demineralization , then was remineralized for eight days by using Aominqing dental desensitizer, fluoride dentifrice (1.1 g/L), and deionized water, respectively. The thin sections of teeth were analyzed under the con-focal laser scanning microscope (CLSM). The morphology of the surface of teeth was observed under the scanning electron microscope (SEM). Results Under CLSM, the evaluation parameter area of the fluorescent lesion (A,μm2) processed by Aominqing and by fluoride was (3.19 ± 0.19) × 104, (3.61 ± 0.26) × 104 μm2, respectively. The total fluorescence (TF) was (0.61 ± 0.09) × 106, (0.89 ± 0.15) × 106, average fluorescent of the lesion(AF) was (18.98 ± 1.56), (24.65 ± 2.39), and the above parameters were all less than those in the blank control group [A=(4.89 ± 0.24) × 104 μm2,TF=(1.78 ± 0.21) × 106, AF = 36.29 ± 2.57] (P < 0.01). The evaluation parameters in the Aominqing group were less than those in the fluoride dentifrice group(P < 0.05). Under SEM, the surface of the group processed by Aominqing was the smoothest, compared to the fluoride dentifrice group and the blank control group. Conclusions Both Aominqing dental desensitizer and fluoride dentifrice (1.1 g/L) have the remineralization effects on the demineralized enamels, and the former has a stronger effect.

In recent years, with the rapid development of modern disciplines such as omics technology, bioinformatics, high-throughput and high-content screening, and artificial intelligence, the research and development of small-molecule drugs has presented a new paradigm characterized by "integrated medicinal chemistry". This review summarizes new enabling drug discovery technologies, the emergence of new subfields formed through integration innovations and practical chemistry toolbox in the field of medicinal chemistry.
7.Survey on practice of hospice care and influencing factors among health providers in Shanghai

Objective:To survey the practice of hospice care and influencing factors among health providers in Shanghai.Methods:From November to December in 2019, a sampling survey on the practice of hospice care was conducted among 2 982 health providers from 217 institutions in 16 districts of Shanghai by using the self-designed scale.Results:The overall behavior frequency of hospice care in health providers was between "occasionally" (3 points) and "often" (4 points). The item score was (3.7±0.9), and the total score was (52.1±10.0). In the aspect of building a good relationship with patients and family(77.7%, 2 317/2 982), the participants had the highest work-practice(46.4%, 1 383/2 982). However, they practiced less in recommending the hospice care institutions to terminal patients. In terms of work-practice, the average score in male providers, those with education of undergraduate and above, those being married (including divorced or widowed), medical staff, those with associated senior titles, with witness of dying, or those currently working in the hospice care service department was higher.Conclusions:The hospice care work-practice of health providers in Shanghai is generally satisfactory, however, there is still some discrepancy among health providers with various characteristics. To further improve quality of hospice care, it is necessary to strengthen policy implementation, to encourage multidisciplinary team cooperation and carry out various forms of professional training for health providers.

Virus infection is a serious threat to human health and social development. The increase in pandemics caused by emerging and re-emerging viruses highlights the urgent need for broad-spectrum antivirals. In this perspective, we highlight recent case studies and summarize the universal strategies and methodologies in broad-spectrum antiviral drug discovery from common targets, common steps in viral life cycle, universal strategies, and broad-spectrum molecules, hoping to provide valuable guidance for the current and future development of antiviral drugs.
9.Differentiation of human umbilical cord mesenchymal stem cells into corneal epithelium like cells

Background Corneal blindness caused by ocular surface disease is one of the main reasons for the global blinding corneal diseases.With the development and progress of tissue engineering technology,tissueengineered cornea offers a new approach to the treatment of ocular surface disease.Objective This study was to obscrve the growth and differentiation of human umbilical cord mesenchymal stem cclls (UC-MSCs) on thc corneal stroma of receipts and investigate the feasibility of human UC-MSCs differentiated into corneal epithelium-like cells and the reparation of injury cornea.Methods Human UC-MSCs were isolated from human umbilical cord using collagenase Ⅳ digestion and passaged in DMEM/F12 containing fetal bovine serum in vitro.The immunophenotype of cultured human UC-MSCs was evaluated by flow cytometry.The differentiated osteoblasts from the human UC-MSCs by directional induce was identified.Twenty-four New Zealand albino rabbits were randomly divided into 2 groups.The human UC-MSCs were cultured on porcine corneal matrix without corneal epithelium for 4 days and then transplanted onto the 12 left eyes of 12 New Zealand albino rabbits,and porcine corneal matrix without corneal epithelium was transplanted onto the left eyes of other 12 New Zealand albino rabbits as control group.The rabbits received keratoplasty were examined using in vivo confocal microscope through focusing(CMTF).The eyeballs were taken off after 2,4 and 8 weeks,the growth and differentiation,expression of cytokeratin 3 (CK3),CK12 and ATP-binding cassette superfamily G memben 2 (ABCG2)of human UC-MSCs were observed by histopathology and immunofluorescence staining.This use of the experimental animals complied with ARVO Statement.Results Digestive human UCMSCs formed round in shape and was large in size.The attached cells displayed long-fusiform shape like fibroblasts.The cultured human UC-MSCs phenotype was CD105+/CD29+/CD44+/CD34-/CD45-and could be induced toward osteoblast differentiation under the appropriate experimental conditions.Human UC-MSCs grew well on the porcine corneal matrix.The corneal grafts survived wcll without rejection till the experiment end in experimental eyes,but the rejection of corneal graft occurred in control eyes.Confocal microscope could observe corneal epithelium-like cells.The corneal epithelium cells showed the positive response for CK3 and CK12 and absent response for ABCG2.Conclusions Human UC-MSCs with porcine corneal matrix can survive,proliferate and differentiate into corneal epithelium-like cells after transplanting onto the corneal stroma of rabbits.This result suggests that human UC-MSCs is able to repair and reconstruct the injured corneal surfaces.
10.Bioinorganic chemistry strategies in antiviral drug discovery

Over the course of human civilization, viral infections have been a part of human life and still represent one of the heaviest burdens for human and society, with a huge devastating socioeconomic impact. Inorganic and bioinorganic chemistry have made important contributions to medical science and human health in the past half century. In this paper, we selected the representative cases in recent years, and reviewed the research progress of antiviral drug discovery from the perspective of bioinorganic chemistry.
