1. Detection of HER-2 mRNA and protein expression in breast cancer tissues by using chromogenic in situ hybridization and immunohistochemistry methods

Objective: To detect human epidermal growth factor receptor-2 (HER-2) gene amplification and protein expression by using chromosome in situ hybridization (CISH) method and compare the results with immunohistochemical assays (IHC). Methods: We assessed the amplification status of HER-2 gene with CISH method in 145 cases of breast cancer tissues. Fourteen out of 145 specimens have been tested by fluorescence in situ hybridization (FISH). HER-2 gene amplification and protein expression in the 145 cases of breast cancer tissues were tested by IHC methods. We retrospectively compared the detection results among the three methods and analyzed the correlation of HER-2 expression with various high-risk factors of breast cancer such as pathological grade, lymph node metastasis, menopausal status, and the expression of estrogen receptor (ER)/progesterone receptor (PR). Results: Based on CISH assay, non-amplification of HER-2 gene was observed in 71 cases (50.0%), low-amplification was detected in 11 cases (7.6%), and high-amplification was assessed in 63 cases (43.4%). The concordance rate between FISH (n = 14) and CISH was 100.0% (14/14). The concordance rate between IHC and CISH was 84.8% (n = 145, P < 0.05). In IHC 0/+ and IHC + + + cases, the HER-2 gene amplification status (> 90%) was in agreement with its protein expression. However, the amplification rate of HER-2 gene was only 61.1% IHC + + cases. Both CISH and IHC detection demonstrated that expression of ER/PR was negatively related with the expression of HER-2. The amplification rate of HER-2 gene was significantly higher in ER/PR-negative patients that that in ER/ PR-positive patients (CISH: 68.3% vs 38.8%, P < 0.01; IHC: 71.7% vs 48.2%, P < 0.01). The amplification status of HER-2 gene had no correlation with tumor pathological grade, auxiliary lymph node metastasis, and menopausal status. Conclusions: CISH for HER-2 detection is easy to handle and has high accuracy. It could replace FISH to further confirm the status of HER-2 gene in IHC + + cases. HER-2 is negatively related with ER/PR expression but has no relationship with other risk factors such as pathological grade, auxiliary lymph node metastasis, and menopausal status. HER-2 should be tested independently.

Objective:To compare the biomechanical effects between oblique Ban-pulling manipulation and lumbar erection-rotation manipulation in sitting position in treating lumbar intervertebral disc herniation (LIDH). Methods:A three-dimensional finite element model of L3-S1 was developed to carry out a comparative study between oblique Ban-pulling manipulation and lumbar erection and rotation manipulation in sitting position. The disc protrusion was assumed to be on the rear left of L4 disc, and the manipulations were performed on the right side. The loading process was simulated by two steps. In the first step, only the compression loading was imposed, and in the second step, both the compression loading and axial rotation moment were imposed. The displacement and stress distribution in L4 disc were investigated. Results:The values of stress and displacement in the second step were lower than those in the first step in each manipulation. The stress and displacement differences between the two steps were respectively 1.79 times and 3.03 times larger in oblique Ban-pulling manipulation than those in lumbar erection-rotation manipulation in sitting position. Conclusion: Oblique Ban-pulling manipulation may result in a better biomechanical effect than lumbar erection-rotation manipulation in sitting position for LIDH.

5.Evaluating left ventricular torsion of different right ventricular pacing sites by speckle tracking imaging

Objective To evaluate the left ventricular(LV)torsion in patients with permanent pacemaker implantation by speckle tracking imaging(STI).Methods Twenty-two patients with permanent pacemaker implantation were enrolled.Eleven of them had the pacing electrodes placed in the right ventricular(RV)apex(RVA group)and the others had the pacing electrodes placed in the RV outflow tract (RVOT group).Thirteen healthy subjects were studied as controls.The basal and apical shortaxis views of the left ventricle were acquired and analyzed using EchoPAC 7.0 station.Results ①The curves of LV torsion in the pacing groups were generally lower than those in the control group and notched.The torsion parameters of RVA group and RVOT group were all less than those of control group(Ptw:12.57±4.19°vs 12.43±5.46°vs 18.98±5.73°,all P<0.05),but there were no significant differences between RVA group and RVOT group.The time of torsion was not changed in the pacing groups because there were no significant differences among the three groups.②The values of Ptw-a(peak twist of the apex)were gradually declined among control group,RVA group and RVOT group.There was statistical difference in Ptw-a between RVOT group and control group(7.40±4.27°vs 12.70±6.82°,P=0.0364).③The basal Tp-SD(standard deviation of times to peak twist)of RVOT group was significantly longer than that of control group[14.1(7.97~22.70)%vs 4.87(3.03~7.79)%,P=0.0298].And total Tp-SD of RVA group was significantly longer than that of control group[(7.6±6.5)%vs 2.26(1.62~3.51)%,P=0.0423].④The curves of torsion velocity in the pacing groups were obviously disorganized compared with control group.But there were no statistical differences in Ptv(peak twist velocity),Putv(peak untwisting velocity),Tptv(time to peak twist velocity)and Tutv(time to peak untwisting velocity)among the three groups.Conclusions The RV pacing tends to impair torsional synehrony of LV and depress LV torsion,though it doesn't change the timing of LV torsion.It may impair LV systolic and diastolic function.
6.Evaluation of left ventricular function and dyssynchrony using three-dimensional speckle tracking echocardiography

ObjectiveTo evaluate left ventricular (LV) function and dyssynchrony in patients with dilated cardiomyopathy (DCM) and complete left bundle branch block (CLBBB) by three-dimensional speckle tracking imaging(3D STI).Methods3D STI was performed and analyzed using TomTec 4-D LV analysis 3.0 software in 37 DCM patients with CLBBB and 25 healthy volunteers.The global 3D,longitudinal,circumferential,radial strains were measured.LV dyssynchrony was evaluated by the standard deviation of time to peak from 3D strain of 16 segments related to the heart cycle(3D-SDI).ResultsIn control group,uniformity in the average value of 3D strain was observed between apical,mid-ventricular and basal levels (P > 0.05).Global 3D,longitudinal,radial and circumferential strains had excellent correlations with LV ejection fraction ( r =- 0.92,- 0.84,- 0.78 and 0.81,respectively,P <0.01).Compared with control group,global 3D,longitudinal,radial and circumferential strains were significantly lower in DCM patients ( P <0.01 for all).3D-SDI in DCM patients with CLBBB was significantly longer than that of volunteers ( P <0.01).3D-SDI increased with worsening LV systolic function regardless of QRS duration (P <0.05).ConclusionsWhen image quality is optimal,3D STI represents a promising novel technique for assessment of global LV function and dyssynchrony.
7.Assessment of left ventricular torsion in patients after coronary artery bypass graft by velocity vector imaging

Objective To evaluate left ventricular torsion in patients after coronary artery bypass graft(CABG) surgery by velocity vector imaging(VVI). Methods Twelve patients who were scheduled to have CABG surgery were enrolled. VVI offline software was used to evaluate myocardial rotation in both basal and apical segment before and three months after CABG. The rotation and rotation rate of each a counterclockwise rotation at the apex and a clockwise rotation at the base, and the absolute value of CABG the rotation at the base was significantly improved than that before CABG (P <0.001), and the rotation in the segment of anterior septum, anterior wail and inferior septum at the base was higher than that rotation in the apical and basal segment three months after CABG was also significantly higher than that negative peak rotation rate in the early diastolic period at the apex were significantly higher than those at the improved than that before CABG (P <0.001). And the positive peak rotation of the systolic period in the segment of anterior septum,anterior wall,lateral wall and inferior septum and the negative peak rotation of the diastolic period in the segment of anterior and inferior septum at the base were also higher than those before CABG (P <0.001). Conclusions VVI is a useful method which can quantitatively assess left ventrieular torsion in patients before and after CABG surgery.
8.Cesarean section does not reduce mother-to-infant transmission of hepatitis B virus

Objective To investigate whether cesarean section on pregnant women with chronic hepatitis B virus (HBV) infection may reduce mother-to-infant transmission risk of HBV.Methods Preserved serum samples of pregnant women at 15-20 weeks of gestation which were collected from July 2002 to August 2004 in 14 counties of Jiangsu Province were analyzed retrospectively.These samples were tested for HBV serologic markers with enzyme immunoassay,and 419 cases of positive hepatitis B surface antigen (HBsAg) were found.HBV DNA level of these HBsAg-positive women were further quantitatively measured with real-time polymerase chain reaction assay.Children of these 419 HBsAg-positive mothers were followed up during October 2009 to March 2010 and 298 of them were contactable (71.1%),among which 281 were enrolled in final analysis as they had complete information and had been vaccinated with three regular doses of HBV vaccine during infantile period.The 281 children were tested for serum HBV markers.The HBV infection rate in these children at age 5-7 was compared between those born by cesarean setion (CS group,n =136) and by vaginal delivery (VD group,n=145).T-test,x2 test or Fisher's exact test was used to analyze relevant parameters.Results(1) Maternal HBeAg-positive rates [25.7% (35/136) vs 34.5% (50/145)],HBV DNA levels in HBeAg-positive women [(2.30 × 106) IU/ml vs (2.09× 106) IU/ml],the percentages of HBIG injection after birth [38.2% (52/136) vs 35.9% (52/145)],breastfeeding ratios [82.4% (112/136) vs 75.9% (110/145)],and children' s ages at the follow-up [(5.9 ± 0.8) vs (6.0±0.6)] were comparable between CS group and VD group (all P>0.05).(2) Among these 281 children,272 (96.8%) were HBsAg-negative,9 (3.2%) were HBsAg-positive and 163 (58.0%) were anti-HBs-positive.The prevalence of HBsAg-positive [(2.9% (4/136) vs 3.4% (5/145)],HBsAg negative and anti HBc positive [0.0% (0/136) vs 1.4% (2/145)] and anti-HBs positive [57.4%(78/136) vs 58.6%(85/145)] in CS group were similar to those in VD group.Conclusions Cesarean section does not reduce the risk of mother-to-infant transmission of HBV.In clinical practice,elective cesarean section should not be suggested in order to prevent HBV vertical infection.
9.The Metabolism Yield of Cordycepin and Adenosine in Cordyceps militaris by Liquid Culture

In order to improve the metabolism yield of cordycepin and adenosine,we studied nitrogen sources ,the levels of nitrogen sources and carbon sources,dynamic development in liquid culture ,as well as the total yield of cordycepin and adenosine in culture system through determining the contents of cordycepin and adenosine by HPLC .The results are as follows: not only animal protein but also some plant protein is very good nitrogen sources; in culture solution the suitable levels of nitrogen sources and carbon sources is respectively 3% and 4%; the content of adenosine in culture solution is very low, while it is quite high in mycelia; it is much higher that the total yield of cordycepin in culture solution than in mycelia; when the culture system is oscillated and cultured after 7~9 days, the total yield of cordycepin and adenosine are both high.
10.Waste Air Treatment in a Biotrickling Filter: Biomass Accumulation and Control Measures

Biotrickling filter often offers a cost effective and environmentally friendly alternative to conventional air pollutant control technologies,but major problems with clogging of the filters due to a high biomass accumulation will provent it from the industried uses.In this paper,the effect of the high biomass accumulation in an air pollution treatment with a biotrickling filter is discussed.Two parameters with specific surface area with biofilm growth(?_ f ) and the bed porosity with biofilm(?_ f )are used to analyse its principle of accumulation.Finally,some control measures including chemical methods,physical machine-made methods and other control methods are overviewed.
