AIM: To evaluate the effect of intravitreal bevacizumab (IVB) injection 1 week before pars plana vitrectomy (PPV) in proliferative diabetic retinopathy (PDR) patients. METHODS: A retrospective research was done on 46 PDR patients who were divided into PPV group (n=28) and IVB group (n=18, PPV with preoperative IVB). Bevacizumab was injected 1 week before PPV. Main outcome measures were visual acuity, incidence of iatrogenic retinal breaks, intraoperative and postoperative bleeding. RESULTS: At 1 month after surgery, visual acuity in PPV (82.1%) and IVB group (88.9%) improved significantly (P<0.01) and the difference between the two groups was not significant. Iatrogenic retinal breaks were reported in 18 cases (64.3%) in PPV group and 4 cases (22.2%) in IVB group (P<0.05). Intraoperative bleeding was encountered in all cases in PPV group and 7 cases (39%) in IVB group (P<0.01). Postoperative bleeding was reported in 9 cases (32.1%) in PPV group and none in IVB group (P<0.01). CONCLUSION: IVB injection before PPV is helpful in reducing iatrogenic retinal breaks, intraoperative and postoperative bleeding in PDR patients.

Aldose reductase ( AR ) belonging to nicotinamide-adenine dinucleotide phosphate ( NADPH ) -dependent aldehyde-keto reductase superfamily, is the key rate-limiting enzyme in the polyol pathway which plays an important role in the body’s high-sugar metabolism. AR is widely present in the kidneys, blood vessels, lens, retina, heart, skeletal muscle and other tissues and organs, converts glucose to sorbitol which easy permeability of cell membranes, cause cell swelling, degeneration, necrosis, and have a close relationship with the development of chronic complications of diabetes mellitus. Diabetic retinopathy ( DR ) is a multifactorial disease, the exact cause is currently unknown, but polyol pathway has been demonstrated to play an important role in the pathogenesis of DR. Clinical risk factors such as blood sugar control, blood pressure and other treatments for DR only play a part effect of remission or invalid, if we can find out DR genes associated with the disease, this will contribute to a better understanding of the pathological mechanisms and contribute to the development of new treatments and drugs. The current research progress of AR, AR gene polymorphism, Aldose reductase inhibitors to DR was reviewed in this article.

Objective:To explore the effect of endothelin-1 on atrial ifbrosis in patients with atrial ifbrillation (AF).
Methods: A total of 72 patients with thoracotomy were studied, the patients were divided into 2 groups:AF group, n=39 and Sinus rhythm (SR) group, n=33. The mRNA and protein expressions of endothelin-1 (ET-1), platelet derived growth factor-B (PDGF-B) and collagen I (COL1) in right atrial appendage (RAA) tissue were measured by RT-PCR and Western blot analysis;meanwhile, the impact of ET-1 stimulation and non-selective ET-1 receptor antagonist (sulfafurazole SIZ) on PDGF-B mRNA and protein expressions in H9c2 cells were measured.
Results: ①The RAA tissue mRNA and protein expressions in AF group were higher than those in SR group, as for ET-1 (2.830 ± 2.276) vs (1.220 ± 0.887) and (0.835 ± 0.241) vs (0.286 ± 0.083), both P<0.01;for PDGF-B (2.568 ± 2.348) vs (1.567 ± 0.831) and (0.807±0.241) vs (0.381 ± 0.105), both P<0.05;for COL1α1 (3.376 ± 1.598) vs (1.629 ± 0.833) and (0.652 ± 0.210) vs (0.312 ± 0.12), both P<0.05.②The protein expressions of ET-1 and COL1 had positive correlation (r=0.580, P<0.01).③ET-1 promoted PDGF-B secretion in H9c2 cells in a concentration and time-dependent manner;SIZ could reduce such promotion.
Conclusion: ET-1 plays an important role in AF occurrence which might be related to PDGF-B regulation.

By takung advantage of atomuc force mucroscopes'( AFM) capabuluty of manupulatung and processung materuals on the mucroscale, we duscovered and unvestugated a top-down preparatuon method of buologucal macromolecular nano-fubrul arrays. 50μg/mL solutuon of rat taul type I collagen monomers was used to coat the muca surface to form a proteun membrane. In the AFM Contact Mode, the AFM tup then manupulated the membrane wuth appropruate force between 100 nN and 1 mN, thus producung patterned collagen nano-fubrul arrays wuth specufuc oruentatuons. The nano-fubruls are averagely 2-5 nm un heught and 150-350 nm un wudth. Based on the relatuonshup between the structure of such nano-fubrul arrays and the AFM scannung patterns, we unvestugated and duscussed the formatuon of the proteun fubrul arrays, and verufued the molecular broom mechanusm of the AFM tup. Thus preparatuon method could potentually provude an effucuent technuque to manufacture cell culture vessels, produce buologucal probes wuth hugh specufucutues, and synthesuze novel mucro/nano materuals as well.

The solid-phase method to synthesis HCV polypetides was used. The connecting rate and the polypeptides' productivity and purity were compared when Wang resin and 2-chlorotrity choloride resin acting as vectors.By the identification of mass spectrum, the products' molecular weight was analysed.By ninhydrin method we compared to the connecting rate between two resins and amino acid.By RP-HPLC we carried out the analysis and purity of hetero peptides. The result showed the first amino acid's connecting rate along with the purity and productivity which we need when 2-chlorotrityl choloride resin acted as vector was notably higher than those when Wang resin acted as vector. So 2-chlorotrityl choloride as vector is used to synthesize HCV polypeptides, which adapts to the demand of big scale's industrialization production.

Journal of Forensic Medicine 2014;30(4):276-278
In the criminal cases of driving under the influence (DUI), DNA evidence can be collected from the deployed airbag of the motor vehicle and submitted to the crime lab for touch DNA analysis. The evidence can be acquired when the skin cells are observed on the surface of the airbag in a traffic accident. However, the low quantity or quality of the evidence collected from a crime scene prevents further identification analysis in many cases. In the current study, we reported a case of identifying touch DNA extraction from the shed skin cells from the deployed airbag of a motor vehicle. We managed to collect DNA evidence from the shed skin cells in an airbag using a proper approach of collection and extraction. The 5.87 ng of extracted DNA was sufficient for genotyping and forensic identification, which helped to identify the driver of the car in collision with a pier in the street. In DUI cases and other traffic accidents, therefore, the amount of touch DNA extracted from the deployed airbag can be sufficient for DNA marker genotyping and further analysis.

Objective To evaluate the value of ice-compression therapy in mice skeletal muscle after acute crush injuries and correlate treatment effect with different compression time by MR DTI. Methods Forty Weistar mice were randomly divided into 4 groups by random number table method: control group (A), 5 min compression time group( B), 15 min compression time group(C) and 30 min compression time group(D). Diffusion tensor imaging examinations were performed before, immediately after, 24, 48 and 72 hours after injuries. ADC and FA values were calculated by fiber tracking tool. The morphological changes were confirmed by histopathology, and immunohistochemical methods were used for the assessment of Desmin expression with mean of A value. Statistical analysis by LSD-t test and Spearman rank correlation. Results (1) For every group before injuries, ADC valueswere (1.38±0.04) ×10-3,(1.38±0.08) ×10-3, ( 1.34 ± 0. 05 ) × 10 -3, ( 1.36 ± 0. 09 ) × 10 -3 mm2/s respectively, FA value were 0. 46 ± 0. 05,0. 45 ±0. 03,0. 45 ± 0. 05,0. 48 ± 0. 04 respectively. ADC values increased significantly and FA values reduced in each group immediately after injuries compared with pre-injury values. ADC values were ( 1.84 ±0. 10) × 10-3, ( 1.79 ±0. 09) × 10-3, ( 1.55 ±0. 07) × 10-3, ( 1.57 ±0. 04) × 10 -3mm2/s respectively,FA value were 0. 21 ±0. 04, 0. 26 ±0. 03, 0. 31 ±0. 02, 0. 30 ±0. 04 respectively. ADC values were still higher and FA values lower than pre-injury values at 24 hours after injury in A, B groups. ADC values were (1.54±0.13) ×10-3, (1.57±0.13) × 10-3mm2/s, FA value were 0.25 ±0.03, 0.26±0.02. (2)DTT showed fibers distorted and the number of fiber bundles reduced, some separation and displacement in each group immediately after injury. C, D groups improved more than A, B groups over time. (3) The disorder arrangement of skeletal muscle cells with edema and filaments separation were found in HE staining after injury, but the degree mitigated in C, D groups. Desmin staining became lighter with fuzzy edge immediately and 24 hours after injury, and changed more than 72 hours after injury. (4) The correlation coefficients of ADC, FA values and A value were respectively - 0. 789 and 0. 763 ( P < 0. 05 ). Conclusions DTI can non-invasively reflect the pathological changes after acute crush injuries of muscles of mice and ice compression therapy. It is a useful method to guide ice compression treatment after acute crush injuries.
