1.Risk factors of myasthenic crisis after thymectomy: a meta analysis

Objective To evaluate the risk factors of myasthenic crisis after thymectomy for myasthenia gravis.Methods Meta-analysis was performed to screen risk factors of myasthenic crisis after thymectomy for myasthenia gravis based on 21 associated articles published and carried out using Stata12.0.Results Twenty one studies were identified, including 3 092 patients and 698 patients developed myasthenic crisis after thymectomy.Complicated with thymoma(OR =3.93, 95% CI 2.37-6.50, P =0.000), Ossermen classification (OR =0.12, 95 % CI 0.09-0.17 , P =0.000) , history of preoperative myasthenic crisis (OR =5.36, 95 % CI 3.82-7.51, P =0.000) , use steroid hormonea (OR =1.75,95 % CI 1.26-2.43, P =0.001) , postopertive lung infection(OR =4.27,95% CI 1.63-11.18, P =0.003) , operation methods(OR =0.34,95% CI 0.21-0.56, P =0.000) , the dose of preoperative anticholinesterase drags (OR =0.35, 95 % CI 0.18-0.66, P =0.000) ,pulmonary function(OR =0.18, 95% CI 0.06-0.57, P =0.004), preoperative serum level of anti-acetylcholine receptor antibody(OR =0.06, 95% CI0.03-0.13, P =0.000)were associated with myasthenic crisis after thymectomy.Gender, course of disease were not associated with myasthenic crisis after thymectomy.Conclusion With thymoma, Ossermen classification, history of preoperative myasthenic crisis, use steroid hormonea, operation methods, the high dose of preoperative anticholinesterase drugs, poor pulmonary function, postopertive lung infection, high preoperative serum level of anti-acetylcholine receptor antibody are risk factors affecting myasthenic crisis after thymectomy.
2.Application of ROC curve analysis in evaluating the virtual screening of ?-secretase inhibitors

Objective To explore the application of ROC curve analysis in evaluating the virtual screening of ?-secretase (BACE-1) inhibitors.Methods Virtual screening were performed for BACE-1 inhibitors using different scoring functions (Surflex-score,D-score,G-score,PMF-score,ChemScore),the screening library was composed of 50 BACE-1 inhibitors and 9 950 inactive compounds.Five scoring functions were used for ranking,and application of ROC curve analysis in evaluating the results of ranking.Results Using D-score to rank the database,the area under the curve (AUC) was 0.935.If specificity was considered primarily,more active compound were obtained by Surflex-score;if sensitivity was considered primarily,more active compounds were obtained by D-score.Conclusions ROC curve consists of sensitivity and specificity together,it can be used to set the right threshold after virtual screening to obtain active compounds as many as possible.
3.Effect of Some Amino Acids in Gentamicin Production

Gentamicin production at home and abroad was fermentation cycle long,production rate low,production costs high and short of competitiveness.Added a little quantity of lysine,glycine,tyrosine,methionine into the medium of gentamicin producing Micromonospora purpurea during inoculation could improve the metabolic capability of microbe,shorten fermentation cycle and increase production rate of gentamicin.The new method was successfully developed in shaking flask test and applied to 5L glass fermentor for one month.Average data of one month showed that the fermentation time per cycle was shortened about 40% and the yield of gentamicin per fermentor increased about 14% in comparison with the conventional technique.The production rate of gentamicin was increased 30% ~95%(according to the capability of strain).The quality of the end product gentamicin conformed to CP2000,BP2000 and USP26 pharmacopoeia.Therefore,the new method may reduce production costs of gentamicin remarkably,it shows strong competitiveness in the marketplace.
4.Expressions of Transferrin Receptor mRNA,Ferritin Receptor mRNA and Iron Regulatory Protein 1 mRNA in the Placentas Complicated with Fetal Iron Deficiency

Objective To investigate the expressions of transferrin receptor (TfR) mRNA, ferritin(Fn) mRNA and iron regulatory proteinl (IRP-1) mRNA in the placentas complicated with fetal iron deficiency(ID). Methods Depending on the cord SF,the subjects were divided to fetal ID group and fetal iron sufficient( IS) group. TfR mRNA, Fn mRNA and IRP - 1 mRNA in placentas were measured by RT - PCR. Results 1. The expression of TfR mRNA in ID group was 1.10 ? 0. 26, it was significantly higher than that in IS group (t=0.028 P0.05) ;4. TfR mRNA and Fn mRNA correlated with fetal IS respectively. Conclusion Fetal ID induces highly expressed TfR mRNA and lower level of Fn mRNA to furthest satisfy the iron need for fetus.
5.Effect of leptin on proliferation and c-myc gene expression of human colon carcinoma HT-29 cell line

Objective To detect the effect of leptin on proliferation and c.Myc mRNA expression of human colon carcinoma HT-29 cell line and investigate the role of Leptin in the development of the HT-29 cell line.Methods Human colon carcinoma HT-29 cell line was cultured in vitro.After treatment with various concentration of Leptin for 72h.MTr was used to detect the proliferative and inhibitive status.And c-myc mRNA-expression Was detected by RT-PCR.Results After treatment with various concentration of Leptin.The cell pmlifemtion and c-myc mRNA expression Wag obviously promoted,compared with the control group.The effect wag in a time-dose-dependent manner within a certain range.Conclusion Leptin can improve cell proliferation and c-myc gene expression level in HT-29 cell line.Promoting the c-myc gene expression level may be one of the reasons that Leptin improves the proliferation of the human colon carcinoma HT-29 cell line.
6.Diagnostic and prognostic application of proteomic patterns in breast cancer

AIM: To detect the serum proteomic patterns in patients of breast cancer by the method of SELDI-TOF-MS and CM10 ProteinChip, and to screen the biomarker candidates, build and validate the diagnostic models, and evaluate its clinical value in surveillance and follow-up after operation. METHODS: The SELDI-TOF-MS technology and CM10 ProteinChip were used to detect the proteomic patterns of serum from 63 breast cancer patients and 40 healthy women. The biomarker candidates were screened and the diagnostic models were constructed by ZJU-PDAS software. Meanwhile, the model was blind-validated in another 23 patients and 20 healthy women. At the same time, 16 serum samples were detected to evaluate its value in surveillance and follow-up after operation. RESULTS: The best model was composed by two protein peaks (BC1/3.9 kD and BC2/5.6 kD) with its sensitivity and specificity of 87.30% (55/63) and 95.00% (38/40), respectively. The sensitivity and specificity in the blind-validation of new cases were 95.65% (22/23) and 85.00% (17/20), respectively. The diagnostic efficacies were the same to the patients of different stages (P>0.05). The expression of BC1 increased while BC2 decreased after operation. The expression of BC2 in the patients with recurrence or metastasis was higher than that in the tumor-free survivors (P<0.05). CONCLUSION: This method shows its potential in detection, surveillance and follow-up after operation. The method is also useful for screening the novel and better biomarkers in breast cancer.

MicroRNAs(miRNAs)are a class of small RNA molecules which play a pivotal role in the regulation of genes involved in diverse processes.Recently,many viral-encoded miRNAs have been discovered,which suggests that viruses also use this fundamental mode of gene regulation.Although the functions of most viral- encoded miRNAs are unknown,some of them are involved in evading CTL,mediating latent infection,apoptosis suppression,etc.Uncovering the role of viral miRNAs in the pathopoiesis offers an immense opportunity not only to develope effective antiviral therapies,but also to identifying novel molecular targets for developing antiviral reagents.Therefore,recent progress on vmiRNAs was reviewed.
