2.Population structure and ecological distribution of rhizospheric microorganisms of Angelica sinensis.

OBJECTIVETo study the population structure and ecological distribution of the rhizosperic microorganisms of Angelica sinensis.
METHODThe isolation, culture and identification of microorganisms were studied by the biological methods, and the achieved data were analyzed by the statistical methods for analysis of species diversity.
RESULTWith the growing stages of A. sinensis from Min county of Gansu province, Heqing of Yunnan province and Baoxing of Sichuan province, the quantity of rhizosperic microorganisms increased; and it had reduced since October. In the area of Min county Gansu province, the number ratio of bacteria and fungi was higher than that in the other two areas. In addition, the population diversity and dynamic change were different in three areas. In the area of Min county Gansu province, the number of dominant microbial populations and the population diversities of bacteria, actinomyces and fungi in the rhizosphere were greater than those in the other two areas.
CONCLUSIONThe microecological system and microbial population structure in the rhizosphere of Min county Gansu province were stable. And it was suitable for the growth of A. sinensis in this area.

Salviae Miltiorrhizae Radix et Rhizoma-Carthami Flos is a famous Chinese medicine pair (CMP). Salviae Miltiorrhizae Radix et Rhizoma can promote blood circulation for removing blood stasis, and Carthami Flos can promote blood circulation for removing meridian obstruction and remove blood stasis for relieving pain. The two herbs are important TCMs for activating blood. Danhong injection is the classic application of the two herbs compatibility, which was made from Salviae Miltiorrhizae Radix et Rhizoma and Carthami Flos with scientific formalation by extraction and refining. The CMP is used for treatement of organ flood insufficiency and ischemic infarction diseases. It can obviously relieving symptoms of angina pectoris, improve myocardial ischemia, regress atherosclerosis plaque, and inhibit thrombus. This paper elaberated the bio-active constituents, compatibility effects and action mechanism, and clinical applications of the CMP in order to further upgrade basic research and application levels of the CMP.

It might be an independent formula, and is also usually used in many gynecological formulae and modern TCM prescriptions. This paper mainly analyzed and summarized the compatibility theory, bio-active constituents, compatibility effects and action mechanism, and clinical applications of the CMP, which can provide a basis for the depth research and development of the CMP.

The total effect of Chinese medicine pair (CMP) was not the simply addition of two single herbs, but the interaction of their different components. Therefore, the research on the bio-active components of CMP is the basis of CMP compatibility study, and has important significance for revealing the compatibility effect and action mechanism, and creating traditional Chinese medicine (TCM) new drugs. This paper summed up the latest research progress of CMP on the basis of the bio-active components variation regularity of CMP from chemical solutions and content changes in vitro and the actions of CMP on bodies in vivo, in order to further drive the modern basic and applied research of CMP, and to reveal the scientific essence of CMP compatibility.

7.Comparative analysis of the promoting blood effects of the combination of different proportions of danggui and honghua by the principal component analysis and multi-attribute comprehensive index methods.

The combination of Danggui and Honghua (GH) is a popular herb pair commonly used in clinic for the treatment of blood stasis syndrome in China. To evaluate the activating blood circulation and dissipating blood stasis effects of the combination of different proportions of Danggui and Honghua on acute blood stasis rats, and optimize the proportion of GH to have the best activating blood circulation and dissipating blood stasis effect. Acute blood stasis rat model was induced by subcutaneous injection of adrenaline and ice water bath. The blood stasis rats were administrated intragastrically with GH (1 : 0, 4 : 1, 2 : 1, 3 : 2, 1 : 1, 2 : 3, 1: 2, 1 : 4 and 0 : 1) extracts. The whole blood viscosity (WBV), plasma viscosity (PV), and high shear whole blood relative index (HSWBRI), low shear whole blood relative index (LSWBRI), and erythrocyte aggregation index (EAI) were tested to observe the effects of GH on hemorheology of blood stasis rats. And the maximum aggregation induced by adenosine diphosphate (ADP) was tested to observe the effect of GH on platelet aggregation index of blood stasis rats. In addition, the prothrombin time (PT), thrombin time (TT), activated partial thromboplastin time (APTT) and plasma fibrinogen (FIB) were tested to observe the effects of GH on blood coagulation function of blood stasis rats. Then principal component analysis and multi-attribute comprehensive index methods were both used to comprehensively evaluate the total activating blood circulation and dissipating blood stasis effects of GH. The results showed that the hemorheological indexes and coagulation parameters of model group both had significant differences with normal group. Compared with model group, GH (1 : 0, 4 : 1, 2: 1, 3 : 2, 1 : 1, 2 : 3, 1 : 2, 1 : 4 and 0 : 1) could improve all the blood hemorheology indexes and regulate part indexes of blood coagulation function and platelet aggregation in acute blood stasis rats. Based on principal component analysis and multi-attribute comprehensive index methods, GH 1 : 1 and GH 3 : 2 both had the best effect of blood circulation and dissipating blood stasis, and the effect of GH 1 : 1 was slightly better than GH 3 : 2. These results suggest that GH could obviously ameliorate the abnormality of hemorheology and blood coagulation function in acute blood stasis rats. The optimized proportion of GH was consistent with regulations of medicine usage that GH 1 : 1 had the highest frequency used in traditional Chinese formulae. It could provide scientific basis for more effective application of the compatibility between Danggui and Honghua in modern clinic medicine.

8.Seed germinating characteristics of endangered plant Magnolia officinalis.

OBJECTIVETo find out the optimal condition for the germination and cultivation condition on Magnolia officinalis, and offer the basis for standardized culture of M. officinalis.
METHODThe study was carried out with in-door experiment, using light, temperature, soil water content and seed soaking in different water temperature treatments, the effects of different treatments on seed germination percentage of M. officinalis were investigated.
RESULT AND CONCLUSIONThe germination percentage between light and dark treatments was significantly difference, it is indicated that the seed is light-sensitive. M. officinalis can germinate in a wide range of temperature, from 20 degrees C to 35 degrees C, and the suitable temperature was 25-30 degrees C, at which germination percentages were 59.2% and 54.6%. Germination percentage at 20/30 degrees C was 65%. The optimum soil water content was 25% for seed germination, at which germination percentages were 66.7%. The suitable soil water content was 20%-25%. In order to find out optimal temperature for soaking, seeds were soaked in water at the temperatures of 40, 60, 80, 90 degrees C for 10 minutes. The optimum water temperature was 60 degrees C for seed germination, and hard coated seed percentage was reduced. Seed germination rate can be inhibited under too high water temperature. Seed germination percentage was significantly difference in different germinating bed.

9.The expression and biological significance of interleukin-1 receptor I,interleukin-lreceptorⅡand interleukin-1 receptor accessory protein in synovium of osteoarthritis

Objective To investigate the expression of the interleukin-1 receptor(IL-1R)Ⅰ,IL-1RⅡand IL-1R accessory protein(IL-1RAcP)in osteoarthritis and analyse their biological significance.Methods Immunohistochemistry and reverse transcription-polymerase chain raction(RT-PCR)were adopted to detect the expression of IL-1RⅠ,IL-1RⅡand IL-1RAcP on the synovium of 107 OA patients.Results Immunohis- tochemistry showed strong positive expression of IL-1RⅠand IL-1RAcP,and positive expression of IL-1RⅡ. The expression was distributed in lining cells,monocyts and vascular endothelial cells of the sublining area, but all of them were negative or weak positive in normal synoviums.RT-PCR showed the expression of IL-1RⅠ,IL-1RⅡand IL-1RAcP in OA synoviums was significantly enhanced than normal synoviums (P<0.05),and the expression of IL-1RⅠwas significantly enhanced than IL-1RⅡ(P<0.05),but no sig- nificant difference with IL-1RAcP(P>0.05).In stageⅡandⅢOA synoviums,the expression of IL-1RⅠand IL-1 RAcP had no significant difference with normal synoviums(P>0.05).The expression of IL-1RⅡin stageⅢOA synoviums was significantly enhanced than normal(P<0.05).Conclusion IL-1RⅠ,IL-1RⅡand IL-1RAcP play significant roles in the pathogenesis of OA,especially IL-1RⅠand IL-1RAcP.But their increase is only observed in the early stage of OA.These suggest that they may have no association with the development of OA and have no direct association with the severity of OA.OA can be cured by interrupting the signal transduction path in which IL-1 has played biological roles.
10.Primary study of arginine as feed attractants on Whitmania pigra.

The effects of the feed attractants on Whitmania pigra were studied. The average weight of Wh. pigra were 5.0 g. Arginine was selected as feed attractants, xanthan gum was selected as feed substrate. The times of Wh. pigra going into the inducing room were recorded. The water temperature was 22-25 degrees C during the whole experiment. Arginine that had better inducing effect was chosen to carry on in the gradient experiment. The results showed that the best inducing effect was found when the added amount of arginine was 0.3%, which was close to the arginine content of the natural body fluid of Wh. Pigra and Bellamya purificata, 2.97 mg x g(-1).
