Objective To analyse the dynamic changes of cardiac diseases over the past 30 years. Method All hospitalized patients of internal medicine department over the past 30 years in the 2nd Xiangya Hospital of Central South University were analyzed in order to understand the dynamic changes of cardiac diseases. Results The results showed the proportion of the heart diseases to all medical diseases was gradually increased during the past three decades. The constitution ratio of the classification of the heart disease was also changing continuously and progressively, with coronary heart disease ranking the first and, in contrast, rheumatic heart disease the second since 1980s. Meanwhile, the number and the proportion of the patients with cardiomyopathy and arrhythmias of unknown causes were increased. Anemic heart disease and cardiovascular syphilis had been wiped out since 1980s and 1990s respectively. Conclusion Classification of cardiac diseases changes gradually and continually during the past 30 years.

Objective To evaluate the performance of Cys C results among two detection system.Methods The particle-enhanced immunonephrometic assay was used in Dade Behring BNII. Immunoturbic assay was used in Hitachi 7170 to evaluate the JING' YUAN reagents.We compared the precison,linearity,interference,correlation,and calibrators agreement with Dade Behring BNII.Results The total CV of the samples that contain 0.6-5.0 mg/L was less than 10%.The Dade Behring and JING'YUAN method showed good linearity.Haemoglobin(10 g/L),Bilirubin(300 mg/L), Vitamin C(5 g/L)in the tested sample had no significant interference in the assay(interference 0.05) between JING' YUAN and Dade Behring reagents.Values were slightly lower than that from the Dade Behring BNII method,the mean bias was-0.16.The bias range was 1.1%-23% between JING'YUAN and Dade Behring for one sample.Conclusions The precision,linearity and interference test were suitable for routine Cys C measurement on automated biochemistry analyzer,but results has bias.

The PET tracer 5-([11C]methyloxy)-L-tryptophan (5-(11)CMTP) was prepared by nucleophilic fluorination and alkylation reaction via a two-step procedure in order to develop specific tumor probe. The biodistribution and microPET imaging of 5-(11)CMTP were executed. The results unveiled that the overall radiochemical yield with no decay correction was (14.6 ±7.2) %, the radiochemical purity was more than 95% and high uptake and long retention time of 5-(11)CMTP in liver, kidney and blood were observed but low uptake in brain and muscle were found, furthermore, high uptake of 5-(11)CMTP in tumor tissue was observed. It seems that 5-(11)CMTP will be a potential amino acid tracer for tumors imaging with PET.

Objective To investigate the assoiation between rheumatoid arthritis(RA)and the pres- ence of the shared epitope(SE)of HLA-DRBI gene in Han nationality of Neimenggu population.Methods The method of DNA amplification with sequence-specific primers(PCR-SSP)was used to determine 17 alleles of HLA-DRB1*01,*04,*10 genotypes in 80 RA patients and 110 healthy controls from the Han nationality population in Neimenggu.Results The frequencies of SE were significantly increased in RA patiens com- pared with controls(48.8%:20%,P＜0.01).Epitope analysis revealed that the most predominant allele subtype of DR4(*0405)was usceptible sequence in Neimenggu patients with RA(28.8%:12%,P＜0.01).No statisticall significant difference of other subtypes of DR1,DR4 nd DR10 was noted including DRB1*0101(2.5%:0.9%), *0102(2.5%:0),*0103(1.25%:0.9%),*0104(2.5%:0),*0401(6.25:1.8%),*0402(3.75%:0.9%),*0403 (1.25%:1.8%),*0404(2.5%:1.8%),*0406(2.5%:2.7%),*0407(1.25%:0.9%),*0408(3.75 %:0.9%),*0409 (1.25%:0),*0410(2.5%:0.9%),*0411(0:0)and *1001(8.75%:4.5%)respectively.Logistic regression analy- sis showed that the disease of patients with SE homozgote was more severe than that of patients with heterozy- gote(P＜0.01).Conclusion The results suggest that there is an association between SE of HLA-DRBI gene and susceptibility and severity of RA,especially,HLA-DR4 subtypes are strongly associated with RA in Han nationality in Neimenggu population.

METHODS To make comparison of the 57 cases of CRS with 423 cases of non-CRS during 1998-2002.First,monovariable chi-square test and then non-condition Logistic regression analysis of the markedly different factors in SPSS10.0 were conducted. Objective To prepare a glycerol reference material.Methods The material was prepared and characterized according to the primary standard substance technological specification(JJG 1006- 1994).Glycerol was dissolved in water containing 0.5% sodium azide and dispersed to glass ampules.The homogeneity and stability of this material were tested with an HPLC method.Glycerol concentration was determined by a titration method as specified in the Pharmacopoeia of China.Results The three time measuring result of glycerol reference material was 1.297 5?0.014 3,1.302 0?0.008 9,1.313 7? 0.007 8,respectively.Statistical analysis showed that this material was homogeneous (F=1.462,P=0.166) .It was stable for at least 4 years at 4℃.The assigned reference value was 0.103 6 g/g and the expanded uncertainty was 0.000 4 g/g.Conclusions This material meet the technical requirements of national primary standard reference material.It is approved as the Certified Reference Material (GBW 09149) by General Administration of Quality Supervision,Inspection,Quarantine of the People's Republic of China in May,2006.

Objective To prepare specific polyclonal antibodies to outer membrane protein (Opr) D_2 of Pseudomonas aeruginosa (PA),and explore the relationship between loss of OprD_2 and imipenem resistance.Methods The genomic DNA of PA was ex- tracted with phenol:chloroform.OprD_2 coding gene was amplified by PCR and prokaryotic expression vector pRSET-OprD_2 was constructed.OprD_2 protein was expressed by IPTG induction in E.coli BL21(DE3),and purified with SDS-PAGE.The new protein band was recovered and used as antigens to subcutaneously immunize two New Zealand rabbits to prepare poly- clonal antibody.The specificity of the antibody was determined by Western blot.The expression of OprD_2 in 32 clinical isolates of PA was detected with the prepared polyclonal antibody by Western blot.Results The vector pRSET-OprD_2 has been success- fully expressed in E.coli BL21 (DE3).The polyclonal anti-OprD_2 antibody with high specificity has been successfully pre- pared.Present results show that of the 27 imipenem-resistant PA clinical isolates,OprD2 protein was low-expressed in 5 iso- lates (18.5%) and normally expressed in 2 isolates (7.4%) but not expressed in 20 isolates (74.1%).Conclusions The loss or low-expression of OprD_2 is one of the essential mechanisms accounting for imipenem resistance in clinical isolates of PA.
