1.The effects of 240 g/L EDTA gel on the attachment and proliferation of human periodontal ligment cells onto the root surface of teeth with periodontitis

Objective:To study attachment and proliferation of human periodontal ligment cells(HPDLCs) on demineralized root surface of teeth with periodontitis following EDTA gel application. Methods:44 single-rooted teeth extracted because of severe periodontitis were treated by scaling and root planing(SRP). Then the fragments(5 mm?4 mm) of experimental surfaces were prepared. The fragments were randomly divided into four groups.The root surfaces in group 1,2,3 and 4 were treated by 240 g/L of EDTA gel for 2,3 and 4 min respectively or without treatment(control). Then 1?104 cells were seeded on the surface of each fragment and routinely cultured for 1,3 and 5 days respectively after seeding, the specimens were prepared for microscopic observasion, the cells on the root surfaces were counted. Moreover the other eight fragments were prepared for SEM observasion. Results:HPDLCs attached to and proliferated on all treated surfaces.Group 1, 2 and 3 presented significantly more cells on the root surfaces than the control group did at day 1,3 and 5. The cells on the root surfaces in group 2 and 3 were more than those in group 1 at day 1, 3 and 5. There was no evident difference between group 2 and 3 at any time. The root surfaces in group 2 and 3 had a fibrillar texture associated with an increased diameter of the exposed dentinal tubule openings.Smear layer were observed on root surfaces in group 1. Conclusion:EDTA gel(at 240 g/L) application may enhance attachment and proliferation on root surface of HPDLCs.
2.Apoptosis in gastric cancer cells induced by trichosanthin is related with down-expression of bcl-2

Objective To investigate the apoptosis in gastric cancer induced by trichosanthin, and the relationship between this apoptosis and expression of bcl2. Methods In in vitro experiments, morphologic test and TUNEL staining method were used to quantitatively and qualitively detect the apoptosis status of gastric adenocarcinoma cell line SGC7901 before and after the trichosanthin treatment. Immunohistochemical staining method and Northern Blot hybridization were used for detecting expression status of apoptosisrelated genes bcl2, before and after trichosanthin treatment. Results When SGC7901 cells were treated with trichosanthin (0.1 ?g/ml, 36 h), they presented some typical apoptotic morphologic changes observed by fluorescent staining. These morphologic changes include nuclear condensation, nucleosomal fragments forming a lunate body under nuclear membrane, etc. When SGC7901 cells were treated with trichosanthin at the concentration 0.1 ?g/ml for 36 h,42 h and 48 h, respectively, TUNEL staining showed a significant increase of apoptotic index (AI), from 3.78%?1.11%, 3.98%?1.12%,3.85%?1.08%, respectively, to 11.30% ? 2.33%, 10.22% ?2.00%,11.18%?1.85%(P
3.Determination of serum insulin-like growth factor and their binding protein-3 in liver cirrhosis and their clinical significances

Objective To investigate the relationship between insulin like growth factor (IGF Ⅰ)、IGF Ⅱ and IGFBP 3 and Child Pugh classification in patients with liver cirrhosis and to determine the potential clinical markers of functional hepatic reserves. Methods Forty four patients with posthepatitic cirrhosis were divided into 3 groups according to disease severity (Child Pugh Score) and 38 healthy subjects severed as controls. Serum levels of IGF Ⅰ, IGF Ⅱ and IGFBP 3 were measured in these groups by immunoradiometric assay. Results Baseline IGF Ⅰ, IGF Ⅱ and IGFBP 3 levels were significantly lower in patients with cirrhosis than in controls, and the serum concentrations of IGF Ⅰ, IGF Ⅱ and IGFBP 3 were associated with the marked changes of liver function due to Child Pugh score. They all showed a significant correlation with the degree of hepatic dysfunction and dropped markedly during the progression of liver failure. 4.Study of digestive diseases diagnosed by M2A capsule endoscopy

Objective To evaluate the application of M2A capsule endoscopies ( CE) in the diagnosis of digestive tract diseases. Methods Forty - five patients (26 males, 19 females) , suffered from gastrointestinal bleeding, abdominal pain or chronic diarrhea were referred to this study and underwent capsule endoscopies. Their average age was 47. 8 years old (a range of 15 - 78). Capsule endoscopies were performed with the Given M2A video capsule system. The conventional diagnostic procedures (including small bowel radiograph, gastroenteroscopy, colonoscopy, etc. ) were done in 31 patients before or after the CE, thus the results of both procedures were compared. Results Among 45 patients, 44 have completed CE successfully. Any complication never experienced. The quality of photos was excellent. The significant pathological findings were revealed in 37 patients, diagnostic yield was therefore 84. 1% . Patients with esophagogastric diseases, small bowel diseases and colon diseases were found in 5,29 and 3 cases respectively. Thirty -one patients received a conventional diagnostic procedure before or after CE. Twenty one of them suffered from GI bleeding, pathological changes were found in 17 patients by capsule endoscopies, only 4 patients by conventional procedure; five of them suffered from abdominal pain, pathological changes were found in 5 patients by CE, only 2 patients by conventional procedure; 4 of them suffered from chronic diarrhea, pathological changes were found in 4 patients by CE, only 1 patient by conventional procedure. The pathological sites located by CE were quite similar to those by conventional diagnostic work-up. Conclusion CE has the advantages of high safety, excellent visualization and precise localization. Its efficiency in diagnosis is much better than that of conventional procedure, and is indispensable in the diagnosis of digestive tract diseases.
5.Analysis and Evaluation of Sickbed Utilization in the General Rehabilitation Center

ObjectiveTo analyze the sickbed setup and utilization in the general rehabilitation center and offer gists for strengthening hospital optimize project of sickbed setup.MethodsThe factual sickbed demand, standard sickbed demand and the ratio of confessing and requiring were counted and analyzed.ResultsThe sickbed turnover times of the internal medicine and ICU were accord with request, but the rate of sickbed utilization was higher, the sickbed utilization rate of the other section offices were higher. The sickbed turnover times were lower compared with control lever.ConclusionThe hospital manager should take some measures for optimize sickbed setup, and strengthen manage to improve the medical quality.
6.Clinical Features of Spinal Muscular Atrophy Type Ⅱ: 53 Cases Report

Objective To investigate the clinical features of spinal muscular atrophy type Ⅱ.Methods The data of clinical manifestation,laboratory data, onset, diagnosis, and rehabilitation of 53 outpatients suffering from spinal muscular atrophy type Ⅱ (SMA-Ⅱ) were analyzed. Results Among 53 patients with SMA-Ⅱ confirmed by molecular genetic tests, 27 patients were male while 26 were female. The mean age was 3.35 years (ranged 0.75~7.8 years), and the age receiving gene diagnosis was 17.27 months. Only 15% had a family history,and abnormalities were found in 23% patients' mothers during pregnancy. 83.4% of them had water choke cough, while 87% expectoration weakness. None of them had cough assist machines and had regular spirometry monitoring. 53% of the patients took semi-liquid diet, however,none of them used stomach tube. Symmetrical flaccid paralysis was so remarkable, even muscle strength of lower limbs in 87.7% of these patients were only grades 1-2. 92% had scoliosis, while 83% had tendon contracture. EMG showed extensive neurogenic changes. All children did not accept normal pre-school education, and 85% patients did not accept formal rehabilitation. Conclusion Most of phenotype of SMA-Ⅱ for the children was similar and more severe in China. Most of the patient didn't get formal education, rehabilitation and care.
7.Effect of PI3K/Akt inhibitor wortmannin on proliferation and apoptosis of leukemia K562 cells

Objective: To study the effect of wortmannin (WM), a PI3K/Akt inhibitor, on the proliferation and apoptosis of leukemia cells and the possible mechanism. Methods: Human leukemia cell line K562 was treated with different concentrations of WM. The proliferation of K562 cells was examined by MTT assay. DNA damage in K562 cells was examined by single cell gel electrophoresis assay, and apoptosis of K562 cells was detected by Annexin V-FITC/PI double-staining. The expressions of total Akt, phosphorate-Akt (p-Akt), and NF-κB p65 mRNA and protein were detected by RT-PCR and Western blotting, respectively. Results: WM inhibited the proliferation of K562 cells in a dose- and time-dependent manner, with the IC((50) value of 24 h being 25 nmol/L. WM also induced apoptosis of K562 cells in a dose-dependent manner. DNA damage in K562 cells was demonstrated by appearance of comet tail after treatment with WM, with the rate of DNA tail and the tail length being significantly higher than those in the control group (P<0.01). WM dose-dependently inhibited P-Akt and NF-κB p65, but not the total Akt, mRNA and protein expressions. Conclusion: WM can inhibit proliferation and induce apoptosis of K562 cells in a dose- and time-dependent manner, probably through down-regulation of phosphorate PI3K/Akt signal pathway and NF-κB expression.
9.Cerebrovascular reactivity by using arterial spin-labeling magnetic resonance imaging in subcortical ischemic vascular disease

Objective To assess regional cerebrovascular reactivity (CVR) and cognition impairment of subcortical ischemic vascular disease (SIVD). To show whether CVR affects cognitive impairment of SIVD patients. Methods Arterial spin-labeling (ASL) by MR image was applied in measuring regional cerebral blood flow (rCBF) of frontal lobe, temporal lobe, parietal lobe, occipital lobes after 5% CO2 inhalation, and the CVR was demonstrated by increase rate of rCBF in all the subject areas. Results (1)The patients with SIVD had reduced CVR in cortex of frontal lobes, white matter of frontal lobes, temporal lobes and occipital lobes(P < 0.05). (2) CVR in the SIVD patients with cognitive impairment decreased in frontal cortex and white matter when compared with the patients without cognitive impairment(P < 0.01). Conclusion These results showed that CVR decreased significantly in cortical gray matter and white matter in elderly patients with SIVD. Also the reduction of CVR in the frontal cortex and white matter was associated with cognitive impairment.
10.Relationship Between High-sensitivity C-reactive Protein and P Wave Dispersion in Patients With Lone Paroxysmal Atrial Fibrillation

Objective: To analyze the relationship between high-sensitivity C-reactive protein (hs-CRP) and P wave dispersion (Pd) in patients with lone atrial ifbrillation (AF), and to explore the effect of inlfammation on atrial electrophysiological remodeling.
Methods: Our research included 2 groups. AF group, containing 71 consecutive paroxysmal lone AF patients, and Control group, containing 71 paroxysmal supra ventricular tachycardia patients with the matched age and gender. The clinical characteristics, electrocardiographic Pd assessment and plasma hs-CRP levels were compared between 2 groups. The relationship between hs-CRP and Pd was studied by linear and multi linear regression analysis.
Results: Compared with Control group, AF group showed increased left atrial diameter, Pd and hs-CRP, all P<0.05. multilinear regression analysis indicated that hs-CRP was positively related to Pd in both groups, P<0.01. Multiple logistic regression analysis revealed that hs-CRP was an independent risk factor for AF occurrence (HR=15.430, 95%CI 6.031-39.476, P<0.001), with adjusted Pd, multiple logistic regression analysis presented that both Pd and hs-CRP were the independent risk factors for AF occurrence, both P<0.001, while the HR for hs-CRP predicting AF occurrence was attenuated from 15.430 to 6.246.
Conclusion: Plasma hs-CRP level and electrocardiographic Pd were the important risk factors for paroxysmal lone AF, the interaction between hs-CRP and AF occurrence could be mediated by Pd, suggesting that inlfammation might be involved in atrial electrophysiological remodeling.
