1. Detection of mycoplasma infection in human laryngeal carcinoma

Objective: To detect and analyze the mycoplasma infection in human laryngeal carcinoma and to explore the relationship between the infection with the development, progression and prognosis of laryngeal cancer. Methods: Immunohistochemical methods were employed to detect the mycoplasma infection in 214 specimens of different laryngeal lesions and cervical lymphatic tumors metastasized from laryngeal cancer, including 121 laryngeal cancer specimens, 21 laryngeal precancerous specimens, 17 vocal cord polypus specimens. 14 normal laryngeal tissues adjacent to cancer tissues, 9 normal laryngeal tissues opposite to the cancer, and 32 cervical lymphatic tumor specimens metastasized from laryngeal cancer. The infection results were subjected to statistical analysis. Results: The positive rates of mycoplasma 2G10 antibody in laryngeal carcinoma specimens, cervical lymphatic tumor specimens metastasized from laryngeal cancer, laryngeal precancerous specimens, vocal cord polypus specimens, normal laryngeal tissues adjacent to cancer tissues, and normal laryngeal tissues opposite to the cancer were 44.63% (54/ 121),34.38%(11/32),23.81%(5/21),17.65%(3/17),7.14%(1/14),and 0%(0/9), respectively, with those of laryngeal carcinoma specimens and cervical lymphatic tumor specimens metastasized from laryngeal cancer significantly higher than those of other specimens(P<0.05). The positive rate of early stage laryngeal carcinoma (I-II stage, 16.36%[9 /55]) was significantly lower than that of advanced stage laryngeal carcinoma(III-IV, 68.18%[45/66]), (P<0.01). The positive rate of laryngeal carcinoma with cervical metastasis was higher (62.50%[20/32]) than that without cervical metastasis(35.96%[32/89),(P<0.01). The 3- and 5-year survival rates of III-IV stage patients positive of 2G10 antigen (60.0%[27/45], 58.14%[25/43]) were lower than those of patients negative of 2G10 antigen (85.71%[18/21],84.21%[16/19]),(P<0.05). Conclusion: Laryngeal carcinoma has a high infection rate of mycoplasma, which may be related with the development, progression and prognosis of laryngeal carcinoma.
2.Study on the Drug Utilization for Lower Respiratory Tract Diseases

To study the status and patterns of the drug utilization in the diseases of lower respiratory tract, the drug utilization for 290 inpatients with respiratory tract diseases in a Shanghai hospital druing 1993-1997 was analyzed by using Acute Physiology, Age, Chronic Health Evaluation(APACHE) and index of defined daily dose numbers(DDDs). The relativity between APACHE and DDDs was studied. It was found that most common drugs was anti-infection agents and expectorants, accounting for 39.06% and 38.38 %, respectively. The quantitative relationship between drug consumption and disease severity was not observed. It is concluded that the status of the drug utilization can't be demonstrated by analyzing the frequency of drug use by using DDDs.
3.Noninvasive gastric motility measurement and evaluation by bioimpedance

BACKGROUND:Bioimpedance method can be used to extract physiology and pathology information relative to the gastricmotility states,to investigate the courses of contraction,peristalsis,transmission and emptying of the stomach,to measure andevaluate gastric motility function.However,early studies mainly focused on the emptying of the stomach,and few studies onevaluation of gastric motility by extracting gastric motility information.OBJECTIVE:To understand the gastric motility from electricity-mechanism composite concept and to establish a noninvasive,convenience measurement method of gastric motility impedance.METHODS:Based on the characters of gastric motility impedance signal,a measurement method of impedance gastric motility isintroduced.Some foundation experiments about the gastric emptying measurement,the gastric motility compare between fastingand repast,the gastric motility comparison of healthy volunteers during different periods and some medicine effects on gastricmotility have carried out.The gastric motility measurement of the patients with functional dyspepsia and gastric motility evaluationfor the patients with erosive gastritis had been reported.RESULTS AND CONCLUSION:Using electrical bioimpedance method,according to the electrical property of the stomach tissueor the food in stomach and the change rules of the property,the physiology and pathology information relative to the gastricmotility states can be extracted,the courses of contraction,peristalsis,transmission and emptying of the stomach can beinvestigated and noninvasive measure and evaluation of gastric motility function can be realized.
4.The Curative Effect of MIPPO Technology United LCP Treatment for Humeral Fractures

Objective To discuss the clinical methods and curative effect of minimally invasive percutaneous plate osteosynthesis ( MIPPO) technique applied in the treatment of humeral shaft fractures. Methods A retrospective analysis was applied on 14 patients with humeral fracture underwent the MIPPO operation in our department from April 2012 to March 2013. There were 8 males and 6 females, with their ages ranging from 28 to 64 years. Results Fourteen cases were followed up for 2 months to 12 months (an average of 6 months) . Their incisions got primary healing. The fracture segments got satisfactory reduction with good apposition and alignment radiologically, and the radial never function recovered well. UCLA score concluded as excellent in 13 cases and good in 1 case. Conclusion MIPPO is a safe and effective treatment for the humeral fracture with the benefits of less invasion, fewer complications and higher union rate.
5.Regulatory effect of miR-181 a on breast cancer resistance protein

Aim ToinvestigatetheeffectsofmiRNA-181 a on breast cancer resistance protein ( BCRP ) . Methods Bioinformaticspredictedbindingsitesof BCRP mRNA-3ˊUTR region and miR-181 a;further lu-ciferase reporter gene analysis confirmed that miR-181 a could combine with BCRP mRNA-3ˊUTR; qRT-PCR and Western blot detected related mRNA and protein expressionlevels.Results Comparedwithnegative transfection group, after the miR-181a mimic and PGL3-BCRP 3ˊUTR were co-transfected, luciferase ac-tivity was significantly decreased ( P <0 . 05 ) . After miR-181a mimic transfected MCF7/MX cells for 48h, compared to the negative group, the expression of miR-181 a in MCF-7/MX cells was increased ( P<0. 05 ) , BCRP mRNA, BCRP protein expression were signifi-cantly decreased ( P<0 . 05 ) , while mRNA expression and protein levels of MRP, P-gp, LRP did not change significantly ( P >0. 05 ); after transfecting miR-181 a inhibitor for 48h, compared to the negative group, the expression of miR-181 a in MCF-7 cells was reduced (P<0. 05). Meanwhile,BCRP mRNA expression and BCRP protein expression were also significantly in-creased ( P<0. 05 ) . The mRNA expression and pro-tein levels of MRP, P-gp, LRP did not change signifi-cantly(P>0.05).Conclusion miR-181acanregu-late BCRP expression by targeting the BCRP mRNA-3ˊUTR region.
6.Protective effect of asiatic acid from Potentilla chinensis on alcohol hepatic injury in rats.

To study the protective effect and the mechanism of asiatic acid (AA) from Potentilla chinensis on alcohol hepatic injury in rats. Male Wistar rats were randomly divided into six groups: the normal control group, the AA control group (8 mg · kg(-1) AA), the model group (5.0-9.0 g · kg(-1) alcohol) and high, medium and low-dose AA-treated groups (alcohol + 8, 4, 2 mg · kg(-1) AA). Each group was orally administered with the corresponding drugs once a day for 24 weeks. Approximately 1. 5 hours after the final administration, all rats were killed, and their blood samples and hepatic tissues were collected. The AST and ALT in rat serum and the contents of MPO, TNF-α, IL-1β, SOD, GSH-Px, GSH-Rd and MDA in hepatic tissues were detected. The expressions of NF-κB, TLR4, CD14, MyD88, TRIF and protein expression in hepatic tissues were measured by western blot. The pathological changes in liver tissues were observed by histological examination. The results showed that compared with the model group, the AA-treated groups showed significant decreases in serum ALT, AST and MDA and increases in the activities of SOD, GSH-Px, GSH-Rd and MPO. Moreover, AA markedly inhibited the expressions of TNF-α, IL-1β, TLR4, CD14, MyD88 and NF-κB. The histological examination showed alleviated hepatic issue ijury to varying degrees. In short, asiatic acid (AA) from P. chinensis could protect alcohol-induced hepatic injury in rats. Its mechanism may be related to the inhibition of NF-κB inactivation and the reduction of inflammatory response.

7.R & D of medicine and vaccine for Ebola hemorrhagic fever

The outbreak of Ebola hemorrhagic fever in West Africa in February 2014 is the worst one in the history of the disease.Medicines and vaccines for Ebola have attracted special attention .This study utilizes Cortellis database of Thom-son Reuters and knowledge about Ebola virus to analyze current R&D of medicines and vaccines for Ebola in order to offer references for research and development .Study findings shows that there are mainly 5 important categories:vaccines, neu-tralizing antibodies , small-molecule anti-Ebola drugs , RNAi drugs and nucleotide drugs .In each category , there are prom-ising products under development that are worthy of attention .

10.Inhibiting PI3K/Akt pathway increases DNA damage of cervical carcinoma HeLa cells by drug radiosensitization.

This study examined the role of PI3K/Akt pathway in radiosensitization of DNA damage of cervical carcinoma cells. The 50% inhibition concentration (IC(50)) of cisplatin and docetaxel in HeLa cells was detected by Mono-nuclear cell direct cytotoxicity assay (MTT) in vitro. HeLa cells were treated by cisplatin/docetaxel of 10 percent of IC(20) alone or combined with LY294002 for 24 h, and then radiated by different doses of X-ray. The cell survival ratio was obtained by means of clone formation. One-hit multi-target model was fitted to the cell survival curve to calculate dose quasithreshold (Dq), mean lethal dose (D(0)), 2Gy survival fraction (SF(2)) and sensitization enhancement ratio (SER). The pAkt and total Akt expression was detected by Western blotting and DNA damage by neutro-comet electrophoresis. The HeLa cells were randomly divided into 7 groups in terms of different treatments: Control; radiation treatment (RT) group; LY294002+RT group; cisplatin+RT group; docetaxel+RT group; LY294002+cisplatin+RT group; LY294002+docetaxel+RT group. The apoptosis ratio of each group was measured by flow cytometry. The results showed that docetaxel and cisplatin significantly enhanced the phosphorylation of Akt in radiation-treated HeLa cells. The Dq, D(0) and SF2 in LY294002-contained groups were lower than those in docetaxel or cisplatin+RT group. The SER in the LY294002+docetaxel+RT group was 1.35 times that of the docetaxel+RT group, and that in the LY294002+cisplatin+RT group 1.26 times that of the cisplatin+RT group. The Comet electrophoresis showed that tail distance in the LY294002+cisplatin+RT group or LY294002+docetaxel+ RT group was longer than in the cisplatin+RT group or docetaxel+RT group. The apoptosis ratio in the LY294002+cisplatin+RT group or LY294002+docetaxel +RT group was higher than in the cisplatin+RT group or docetaxel+RT group. It was concluded that inhibiting PI3K/Akt pathway can increase the effect of docetaxel and cisplatin on the radiosensitivity of HeLa cells and DNA damage resulted from radiation.
