1.Effect on proliferation and apoptosis in RPMI-8226 human myeloma cells and regulation on steroid receptor coactivator-3 by deguelin

Objective To investigate the effects of deguelin on the proliferation inhibition and apoptosis induction in RPMI-8226 cells in vitro,as well as the regulation of steroid receptor coactivator-3(SRC-3)to explore the relationship between them.Methods The effect of deguelin on the growth of RPMI-8226 cells was studied by MTT assay.Apoptosis was detected through Hoechst 33258 staining and Annexin-V FITC/PI double-labeled cytometry.RT-PCR Technology was applied to assessment of the mRNA expression of SRC-3,whereas,SRC-3 protein expression and localization were determined by using immunohistochemistry method.Results Deguelin presented striking proliferation inhibition potency on RPMI-8226 cells in vitro and apoptosis induction activity in a time-and dose-dependent manner.The IC50 value for 24 h was(54.55?0.40)nmol/L,(17.04?0.73)% RPMI-8226 cells went apoptotic when treated with 25 nmol/L deguelin for 24 h,with the dose of deguelin increasing to 100 nmol/L,(63.57?1.36)% cells were apoptotic.Over-expression of SRC-3 was found in RPMI-8226 cells,whereas the mRNA and protein expression levels of SRC-3 were significantly downregulated in RPMI-8226 cells induced by deguelin in a dose-dependent manner.The disposition of SRC-3 was situated mainly at the nuclear,occasionally in the cytoplasm.Conclusion Deguelin exhibited potent proliferation inhibition to the human myeloma cell line RPMI-8226 cells,furthermore,deguelin might induce RPMI-8226 cells apoptosis in a dose-dependent manner,which might correspond to the regulation of the expression of SRC-3.
2.Correlations of pulp chamber height and dentine thickness of mandibular first molar with age in Chinese Han population based on cone beam computed tomography

Objective:To investigate the relationship between pulp chamber height and dentine thickness of mandibular first molar on long axis with age in a Chinese Han population by cone-beam computed tomography(CBCT).Methods:CBCT images of 420 Chinese Han subjects(210 males and 210 females) were divided into 6 age groups as follows:15-24,25-34,35-44,45-54,55-64 and 65-74 years(n=70,35 males and 35 females).The height of pulp chamber and dentine thickness of the chamber root and floor of mandibular first permanent molar on long axis was measured by Galileo software of CBCT in vertical direction and then statistical analysis was carried out.Results:On long axis,the height of pulp chamber was (0.85±0.54) mm in males and (0.79±0.51) mm in females(P>0.05);dentine thickness of the pulp chamber roof was (3.60±0.49) mm in males and (3.50±0.49) mm in females(P>0.05);and dentine thickness of the pulp chamber floor was (2.57±0.52) mm in males and (2.49±0.5) mm in females(P>0.05).With aging,the height of pulp chamber was decreased(P<0.05) and dentine thickness of mandibular first permanent molar on long axis was increased(P<0.05).Conclusion:In Chinese Han population,the of pulp chamber height of mandibular first permanent molar on long axis is related to age,The dentine thickness increased with ages,the increase of dentine thickness of pulp chamber floor is more significant than that of the roof.
3.Effects of plant growth regulator uniconazole on plant morphology and biomass allocation of Salvia miltiorrhiza.

In this study, we use pot experiment to evaluate the effect of plant growth regulator on plant morphology and biomass allocation of Salvia miltiorrhiza. Different concentrations of uniconazole were supplied to S. miltioohiza by means of foliar spray. Height, breadth and stem diameter were measured dynamically, the biomass of leaf, stem, flower and fruit, root biomass and biomass ratio were also examined at the harvest time. Owing to the treatment, plant morphology showed significant changes, the height had been greatly reduced and the breadth decreased largely. Meanwhile, the biomass allocation changed too. The biomass ratio of leaf and stem had been notably reduced while the biomass ratio of root had been increased remarkably. It appears that foliar application of uniconazole during vigorous growth period in S. miltioohiza has dramatic effect on dwarfing plant and improving resistant to lodging. This measure could also be applied to condensed cultivation of S. miltioohiza to increase production.

6.High glucose on the expression of N -cadherin and fibronectin in retinal pigment epithelium cells

To observe the expression of N - cadherin and fibronectin in retinal pigment epithelium ( RPE) cells in vitro under high glucose conditions, furthermore, to explore the effects of high glucose on epithelial -mesenchymal transition (EMT) in RPE cells.
●METHODS: Human RPE (hRPE) cells were cultured in vitro. Containing a final concentration of 60mmol/ L glucose was used for high glucose treatment. The cells were divided into normal glucose group (5. 5mmol/ L, NG) and high glucose group (24, 48 and 72h) respectively. The expression of N - cadherin and fibronectin in hRPE cells were evaluated by immunofluorescence and real -time PCR.
●RESULTS:RPE cells became disorganized and swollen over time under high glucose conditions, especially in 72h subgroup. lmmunohistochemical analysis revealed that the expression of N - cadherin in RPE cells under high glucose conditions was decreased compared with that in the control group, while the expression of fibronectin was increased. Real - time PCR results showed that the expression of N - cadherin mRNA in high glucose group was decreased at 24h compared with that in the control group, and declined markedly at 72h ( F = 12. 252, P =0. 000). There were no significant differences between the control group and the high glucose group at 24h, while the differences between the control group and the high glucose group (48 and 72h) were significant respectively (P < 0. 05 ). Meanwhile, the expression of fibronectin mRNA in RPE cells was increased in high glucose group at 24h, and reached the peak at 72h (F = 50. 543, P = 0. 000). There were no significant differences between the control group and the high glucose group at 24h. Compared with the control group, the expression of fibronectin mRNA in hRPE cells was increased significantly in high glucose group (48 and 72h) respectively (P= 0. 000, P= 0. 000).
●CONCLUSlON: The expression of epithelium marker N-cadherin is down - regulated under high glucose conditions in hRPE cells in vitro. Meanwhile, the expression of mesenchymal maker fibronectin is induced and appeared to EMT changes. Results of this study will enrich our growing understanding in proliferative diabetic retinopathy and hopefully lead to novel insights for the pathogenesis and therapeutic treatments.
7.The influences of ultrafiltration and alcohol sedimentation on protective effects of Radix Astragali and Radix Hedyseri against rat's cerebral ischemia.

OBJECTIVETo investigate the influences of ultrafiltration and alcohol sedimentation on protective effects of Radix Astragali and Radix Hedyseri against rat's cerebral ischemia.
METHODSUsing dexamethasone (im.) and ligating common carotid artery, the rat stasis model combined transient cerebral ischemia was established to evaluate the effects of the ultrafiltration and alcohol sedimentation through detecting antioxidant system and other indexes in brain tissue.
RESULTSThe results showed that the 6 g/kg water extract(crude drug), ultrafiltration and alcohol sedimentation of Radix Astragali and Radix Hedyseri could upgrade adenosine-triphosphate (ATP), superoxide dismutase (SOD) and catalase (CAT), and degrade malondialdehyde(MDA) and water content of brain tissue in rat stasis model combined transient cerebral ischemia, the water extract and ultrafiltration of them could degrade lactic acid (LD) of brain tissue, and the effects of alcohol sedimentation of Radix Astragali and Radix Hedyseri become weaker than water extract of them.
CONCLUSIONThe water extract, ultrafiltration and alcohol sedimentation of Radix Astragali and Radix Hedyseri have some protective effects on cerebral ischemia in rats, the effective differences of the extract through the same extraction method are not remarkable, and alcohol precipitation method has obvious influences effect on Radix Astragali and Radix Hedyseri.

8.Influence of H102 on the expression of amyloid protein and amyloid precursor protein in the hippocampus of APP695 transgenic mice.

OBJECTIVETo observe the influence of H102 on the expression of amyloid protein and amyloid precursor protein in the hippocampus of APP695 transgenic mice.
METHODSThe 9-month-old APP695 transgenic mice were randomly divided into the model group and the H102 group; C57BL/6J mice were adopted as normal control group. The H102 group were injected with H102 in a dose of 3 microl/per mouse in lateral ventricle, once a day, for ten days; while the model group and the control group were injected with saline. The hippocampus and temporal cortex of the brain sections from transgenic mice and wild type female mice were subjected to immunohistochemistry and Congo red histological staining, and observed the difference of the protein expression under microscope. The expression of the APP protein was detected by Western blot.
RESULTSAbeta and APP immunohistochemistry showed density of positive cell in the CA1 region of hippocampus of control group were less than model group. H102 peptide reduced the area, and density of positive cells. Congo red staining showed there were lots of amyloid plagues in the brains of model mice but not in the brains of normal control. And the Western blot showed the content of the APP protein of the model group was much higher than the H102 group. H102 significantly decreased the amyloid plagues.
CONCLUSIONThe expression of APP, Abeta are increased in APP695 transgenic mice, and H102 can decrease the level of APP, Abeta in transgenic mice.

9.Changes in peripheral blood T helper 9 cells and interleukin-9 in children in the acute stage of Kawasaki disease.

OBJECTIVETo investigate the changes in the expression levels of peripheral blood T helper 9 (Th9) cells and cytokine interleukin-9 (IL-9) in children in the acute stage of Kawasaki disease (KD) and their clinical significance.
METHODSA total of 45 children in the acute stage of KD who were treated from April 2014 to July 2015 were enrolled, and the children were followed up in the recovery stage. Another 45 healthy children who underwent physical examination were enrolled as the control group. Flow cytometry was used to measure the percentage of peripheral blood Th9 cells, and ELISA was used to measure the serum level of IL-9.
RESULTSThe children in the acute stage of KD showed a significantly higher percentage of Th9 cells and a significantly higher serum level of IL-9 compared with those in the recovery stage and the control group (P<0.05). The percentage of Th9 cells and serum level of IL-9 showed no significant differences between the children in the recovery stage and those in the control group (P>0.05). In the acute stage, the percentage of Th9 cells was positively correlated with the levels of IL-9, C-reactive protein (CRP), procalcitonin (PCT), erythrocyte sedimentation rate (ESR), platelet count (PLT), and globulin (r=0.624, 0.324, 0.402, 0.382, 0.467, and 0.386 respectively, all P<0.05), but negatively correlated with serum albumin (r=-0.306, P<0.05). The serum level of IL-9 was positively correlated with the levels of CRP, PCT, ESR, PLT, and globulin (r=0.365, 0.456, 0.403, 0.423, and 0.453 respectively, all P<0.05), but negatively correlated with serum albumin (r=-0.343, P<0.05).
CONCLUSIONSThe children in the acute stage of KD show significant increases in the percentage of peripheral Th9 cells and serum cytokine IL-9 level, which return to normal in the recovery stage. In the acute stage of KD, the expression levels of Th9 and IL-9 are closely correlated with laboratory markers. The results suggest that Th9 cells and IL-9 play important roles in the pathogenesis and outcome of KD.

10.The number of circulating fibrocytes of skeletal muscle in rats after contusion.

OBJECTION:
To investigate the time-dependent appearance of circulating fibrocytes of skeletal muscle in rats after contusion.
METHODS:
The model of skeletal muscle wound was established in rat. The circulating fibrocytes in contused skeletal muscle were detected by CD45 and procollagen I double immunofluorescence staining method.
RESULTS:
In the control group, CD45- and procollagen I-positive cells were not detected in skeletal muscle. A few CD45 cells were observed aged from 6 h to 1 d after contusion. A few CD45- and procollagen I-positive cells (fibrocytes) initially gathered in injury area 3d after injury. The ratio of positive fibrocytes significantly increased 5 d after injury. The ratio of fibrocytes was highest at 7 d after contusion and then decreased. The volume of fibrocytes showed bigger with injury time increase compared with 3 d group. The expression of procollagen I and CD45 were weakened at 14d after injury.
CONCLUSION
The circulating fibrocytes are detected in contused skeletal muscle in time-dependent pattern. Circulating fibrocytes may be a marker in the wound age determination for contused skeletal muscle.
