Aim To study the effect of simvastatin on ventricular remodeling in rats after myocardial infarction, and the association between this effect and phosphorylating extracellular signal-regulated kinase1/2(p-ERK1/2).Methods Myocardial infarction(MI)rat models were successfully induced by ligation of anterior descending coronary artery. The treated rats were randomly divided into 4 groups(n=8~10):MI only group,MI plus 20 mg Sim group(20 mg?kg-1?d-1),MI plus 40 mg Sim group(40 mg?kg-1?d-1),and sham-operated group. After 4 weeks, the rats were checked by echocardiography including LVEDd,LVPWd,LVEF,FS,SV and CO.Myocardial p-ERK1/2 was also examined by immunohistochemistry and Western blot. Results Compared with sham-operated group,LVEDd,LVPWd in other groups were significantly increased(P

Objective To investigate the mechanism by which rosiglitazone affects no-reflow following reperfusion after acute myocardial infarction(AMI).Methods Forty-five SD rats were randomly divided into sham-operated group, normal saline treated group and rosiglitazone treated group(at dose of 3 mg?kg-1?d-1,for 14 d),15 rats in each group. To establish no-reflow model, the rats of the latter two groups received coronary artery ligation for 3 h and reopening for 2 h.Then pathological staining for no-reflow area, chemical assay for MPO activity, immunohistochemistry and RT-PCR for protein and mRNA expressions of ICAM-1 and P-selectin in myocardium were performed. Results No-reflow area in rosiglitazone treated group was significantly reduced, compared with that in normal saline treated group(P

Objective To construct the recombinant plasmid pYES6-S and express and purify spike protein of severe acute respiratory syndrome(SANS)coronavirus in Saccharomyces cerevisiae. Methods DNA fragments of SANS coronavirus were obtained by reverse transeription.Four over- lapped fragments of spike protein genes were amplified by polymerase chain reaction(PCR)and ligated into an integral spike protein gene by restriction enzyme digestion. The spike protein gene recombined with pYES6 and cloned into E.coll.The recombinant plasmid pYES6-S was induced and expressed in Saccharomyces cerevisiae(INVScl)by galactose. Results The recombinant plasmid pYES6-S was confirmed that inserted fragment was right in length, direction and base matching by restriction enzyme digestion and sequencing. The purified protein encoded by the whole spike protein gene was about Mr 110?10~3 identified by electrophoresis. Conclusion The whole spike protein gene of SARS coronavirus is cloned into E.coli and the protein is expressed in Saccharomyces cerevisiae successful ly.which can be helpful in SARS vaccine research.

Objective To investigate the influence of rehmannia glutinosa oligosaccharide(RGOs)on apoptosis of human adipose tissue-derived mesenchymal stromal cells(ADMSCs)induced by hydrogen peroxide(H2O2).Methods Cultured human ADMSCs were randomly divided into three groups as the normal group(group N,without any treatment),H2O2 group(group H,treated with 0.1 mmol/L H2O2),and RGOs group(group R,treated with 0.2g/L RGOs plus 0.1 mmol/L H2O2).After 1 h,6 h and 24 h,morphological changes of ADMSCs apoptotic were observed, the apoptotic rate was determined by flow cytometry. Results After 1 h,6 h and 24 h,the apoptotic rate of group H was significantly higher than that of the group N and group R(P<0.05),and the apoptotic rate of group H was significantly higher than that of the group R(P<0.05).Conclusion RGOs can attenuate apoptosis of human ADMSCs induced by H2O2.

Objective To study the effects of cluster nursing intervention on dysuria in hospitalized patients after renal biopsy. Methods A total of 106 hospitalized patients undergoing renal biopsy during April. 2016 to September. 2016 were divided into control group (50 cases) and experimental group (56 cases) by random number table method. The control group were implemented with traditional methods of care and the experimental group were implemented with cluster nursing intervention. The incidence of dysuria, first average urination time and post-operative urination pattern were compared between two groups. Results The incidence of dysuria in the experimental group was 10.7%(6/56), which was significantly lower than 28.0% (14/50) of the control group (χ2=5.156, P<0.05).The first average urination time of experimental group was (2.95±1.17) hours, which was lower than (5.04±2.27) hours of the control group (t =5.401, P<0.05). The proportion of patients with post-operative self-urination in the experimental group was significantly higher than that in the control group (χ2=6.152, P<0.05). Conclusions Cluster nursing intervention can reduce the incidence of dysuria, shorten the first average urination time, promote post-operative self-urination of patients after renel biopsy and enhance comfort.

Chinese Journal of Endocrinology and Metabolism 2016;(1):71-74
[Summary] Graves'ophthalmopathy ( GO) is the most common extrathyroidal manifestation of Graves disease ( GD) . It is an organ-specific autoimmune disorder. The estimated incidence of GO in the general population is 16 women and 3 men per 100,000 population every year. Approximately 3%-5% of patients with GO are exposed to the potential of losing their sight and requiring optimized medical intervention. For the clinicians it is helpful to assess the degree of activity and severity of GO;both are important in deciding whether a patient requires intervention and which type of intervention is indicated. These interventions include management of hyperthyroidism, measures to relieve local symptoms, immunosuppressive therapies for active moderate and severe GO, and finally rehabilitative surgery for inactive patients.

[Objective] To explore the value of the Pressure Ulcer Scale for Healing ( PUSH) tool in evaluating the effect of debridement on pressure ulcers in elder patients . [ Methods] A group of 60 patients with (82 sites) II-IV grade pressure ulcers were recruited in the study .The ulcers were randomly assigned into autolytic debridement group ( n=42 ) and combined debridement group ( n=40 ) .Two groups of wounds were assessed and debrided by consensus procedure porcess respectively .The wounds were ob-served and evaluated by trained nurses at 7,14,21days after debredement , and the results used in PUSH scoring were recorded and subjected to statistical analysis with the SPSS 11.0 software. [Results] The value of pressure ulcer scale for healing ( PUSH) for autolytic、combined debridement group , at 7days( P>0.05) were not significantly different, at 14 days(P<0.05),and at 21days (P<0.01) were significantly different. With wounds in combined debridement group , PUSH scores decreased faster than those in the au-tolytic debridement group . [ Conclusion] The combined debridement method used in elder patients is safe and effective , which can shorten debridement time and improve wounds healing as well .The Pressure Ulcer Scale for Healing is easy to use and its result is reliable , giving a quantitative objective evaluation of debridement effect on pressure ulcers .

AIMTo investigate the effects of pravastatin on endothelin(ET) expression induced by aldosterone in cultured neonatal rat cardiac fibroblasts.
METHODSET concentration in conditioned medium was measured by radioimmunoassay, intracellular ET-1 level was evaluated by flow cytometry, and the expression of preproendothelin-1 (ppET-1) was detected and quantified using reverse transcriptase-polymerase chain reaction (RT-PCR) method.
RESULTSThe cardiac fibroblasts, treated with aldosterone at 107 mol/L, significantly up-regulated ppET-1 mRNA expression, as well as ET-1 synthesis and release. Pravastatin (10(-5), 10(-4), 10(-3) mol/L) dose-dependently blocked these effects. In contrast, pravastatin-induced inhibitory effects were reversed in the presence of mevalonate.
CONCLUSIONPravastatin down-regulated ppET-1 mRNA expression, as well as ET-1 synthesis and release induced by aldosterone in a process specifically related to mevalonate in cardiac fibroblasts.
