1.Biomechanical study of neotype shape memory alloy cervical hook for atlantoaxial instability

BAcKGROUND:In recent years,the pedicle screws fixation technique,which is used in fixation for atlantoaxial instability associated with trauma,severe degeneration and tumorectomy,has been developed.However,this kind of technique easily causes several complications,including malpositional screws,vascular injuries,and even vertebral artery injury.Based on the biomechanical characteristics of memory alloy and determination of atlantoaxial data,a neotype shape memory alloy cervical hook was designed to treat atlantoaxial instability.OBJECTIVE:To investigate the biomechanieal characteristics of the neotype shape memory alloy cervical hook for atlantoaxial instability.DESIGN,TIME AND SETTING:Repeated measurement analysis of variance test was performed in the Laboratory of Clinical Anatomy and Medical Biomechanics,Southern Medical University between March and April 2008.MATERIALS:Eight fresh adult craniocervical specimens(C0-C4)were provided by Department of Clinical Anatomy,Southern Medical University.Atlantoaxial neotype shape memory alloy cervical hook(50.8%-51.8%nickel and the remaining part was titanium)was fabricated by Shanghai Xinchang Memory Alloy Co.,Ltd.METHODS:The included eight C0-C4 specimens were used to test three-dimension ranges of motion(ROM)by fixation and neotype shape memory alloy cervical hook fixation.Then,the positions of spine varying from no loading to the maximum loading status were scanned and analyzed using image processing software to determine the three-dimensional ROM under different statuses.MAIN OUTCOME MEASURES:Three-dimensional ROM of tested specimens.RESULTS:Neotype shape memory alloy cervical hook fixation and Germany AESCULAP SSE hanger fixation had similar flexion-extension range of motion(P=0.595).Lateral bending three-dimensional ROM was greater in the neotype shape memory alloy cervical hook fixation group than in the Germany AESCULAP AAE hanger fixation(P< 0.05).The rotatory three-dimensional ROM was smaller in the neotype shape memory alloy cervical hook fixation group than in the Germany AESCULAP AAE hanger fixation(P<0.05).CONCLUSION:Neotype shape memory alloy cervical hook fixation had comparative post-surgery immediate stability with the Germany AESCULAP AAE hanger fixation.Neotype shape memory alloy cervical hook fixation produced a little worse biomechanical lateral bending stability and a little better biomechanical rotatory stability than Germany AESCULAP AAE hanger fixation.
2.Studies of cytotoxin from guangxi cobra venom on its purification and cytotoxic effects on the nasopharyngeal and other cancer cells

AIM To study the inhibitory effect of the cytotoxin (CTX) from guangxi cobra venom on human nasopharyngeal cancer cell (CNE) and other tumons cells. METHODS The cytotoxin was isolated and purified from Guangxi cobra venom by successive chromatography on Sephadex G-50 and CM Sepharose CL-6B columns. Its cytotoxic effects and does-effect relationship on human tumors cell lines were examined by MTT assay. RESULTS The inhibitory effects of CTX CM-5 on CNE, human ovarian carcinoma cell (Ho8990), uterus cervical carcinoma cell (HELA) and lymphoma (YAC) cell lines showed a definite does-effect relationship. The IC 50 (48 h in cubation) was 1.84, 2.59, 1.84 and 0.75 mg?L -1 respctively. The inhibitory effects of CTX CM-5 on CNE increased with time. The IC 50 (3 h and 24 h cubation) was 4.78 and 1.04 mg?L -1 respctively. CONCLUSION CTX from Guangxi cobra venom exhibites strong suppressive effect on cultured tumor cells line in vitro.

Metastatic epidural compression of the spinal cord is a significant source of morbidity in patients with systemic cancer. With improvment of oncotheray, survival period in the patients is improving and metastatic cord compression is en- countered increasingly often. Surgical management performed for early circumferential decompression for the spinal cord com- pression with spine instability, and spine reconstruction performed. Patients with radiosensitive tumours without spine instabili- ty, radiotherapy is an effective therapy. Spinal stereotactic radiosurgery and minimally invasive techniques, such as vertebro- plasty and kyphoplasty, percutaneous pedicle screw fixation, radiofrequency ablation are promising options for treatment of cer- tain selected patients with spinal metastases.

5.Variations of SPARC in obese patients with type 2 diabetes mellitus

Objective To observe serum secreted protein acidic and rich in cysteine (SPARC) levels in normal subjects,obese subjects,patients with type 2 diabetes mellitus (T2DM),and obese patients with T2DM,as well as the difference of SPARC expression in mesenteric adipose tissue of subjects with and without T2DM.Methods Serum SPARC level was measured with the ELISA.RT-PCR,Western blot,and immunofluorescence were used to examine SPARC mRNA and protein expressions in mesenteric adipose tissue.Results (1) SPARC levels were higher in obesity,T2DM,and T2DM with obesity groups compared with normal group [(1 191.6 ± 718.91,1 223.81 ± 645.96,1 538.01 ± 757.95 vs 851.07 ± 280.21) ng/L,P<0.05].(2) Pearson correlation analysis showed that SPARC level was positively correlated with homeostasis model assessment for insulin resistance index (r =0.205,P< 0.05).(3) The expression levels of SPARC mRNA and protein in mesenteric adipose tissue of T2DM patients were higher than those of control subjects (P < 0.05).Conclusion SPARC is closely related to the development of obesity,insulin resistance,and type 2 diabetes mellitus.
6.Heat shock cognate 71 (HSC71) regulates cellular antiviral response by impairing formation of VISA aggregates.

In response to viral infection, RIG-I-like RNA helicases detect viral RNA and signal through the mitochondrial adapter protein VISA. VISA activation leads to rapid activation of transcription factors IRF3 and NF-κB, which collaborate to induce transcription of type I interferon (IFN) genes and cellular antiviral response. It has been demonstrated that VISA is activated by forming prion-like aggregates. However, how this process is regulated remains unknown. Here we show that overexpression of HSC71 resulted in potent inhibition of virus-triggered transcription of IFNB1 gene and cellular antiviral response. Consistently, knockdown of HSC71 had opposite effects. HSC71 interacted with VISA, and negatively regulated virus-triggered VISA aggregation. These findings suggest that HSC71 functions as a check against VISA-mediated antiviral response.

7.Effects of cluster nursing intervention on dysuria in hospitalized patients undergoing renal biopsy

Objective To study the effects of cluster nursing intervention on dysuria in hospitalized patients after renal biopsy. Methods A total of 106 hospitalized patients undergoing renal biopsy during April. 2016 to September. 2016 were divided into control group (50 cases) and experimental group (56 cases) by random number table method. The control group were implemented with traditional methods of care and the experimental group were implemented with cluster nursing intervention.The incidence of dysuria, first average urination time and post-operative urination pattern were compared between two groups. Results The incidence of dysuria in the experimental group was 10.7%(6/56), which was significantly lower than 28.0% (14/50) of the control group (χ2=5.156, P<0.05).The first average urination time of experimental group was (2.95±1.17) hours, which was lower than (5.04±2.27) hours of the control group (t =5.401, P<0.05). The proportion of patients with post-operative self-urination in the experimental group was significantly higher than that in the control group (χ2=6.152, P<0.05). Conclusions Cluster nursing intervention can reduce the incidence of dysuria, shorten the first average urination time, promote post-operative self-urination of patients after renel biopsy and enhance comfort.
8.The effect of A375 cells corylin melanin synthesis

Objective To study the effect of Corylin on A375 cells melanin synthesis,and explore its mechanism.Methods The cells were randomly divided into the control group, the estradiol group, and corylin group including 10-3μmol/L, 10-2μmol/L, 10-1μmol/L, 1μmol/L, 10μmol/L, 100μmol/L. Estradiol estradiol intervention group were given 10-3 mol/L. Corylin group (10-3μmol/L, 10-2μmol/L, 10-1μmol/L, 1μmol/L, 10μmol/L,100μmol/L) were given 10-3μmol/L, 10-2μmol/L, 10-1μmol/L, 1μmol/L, 10μmol/L, 100μmol/L corylin intervention. The activity of proliferation were detected by MTT, NaOH method, dopa oxidation , both melanin content and tyrosinase activity (tyrosinase, TYR). TYR, yrosinase related protein (tyrosinase related protein, TRP)-1 and TRP-2 expression levels of mRNA were determined by RT-PCR.Results Compared with the control group, 10, 100μmol/L of Corylin group cell proliferation rate significantly decreased (P<0.01). The 1μmol/L, 10-1μmol/L, 10-2μmol/L of Corylin group cell melanin content, TYR significantly decreased (P<0.01 or P<0.05). The 1μmol/L corylin group TYR (0.303 ± 0.003vs. 0.628 ± 0.012), TRP-1 (0.313 ± 0.008 vs. 0.677 ± 0.022), TRP-2 (0.456 ± 0.028vs. 0.687 ± 0.020) mRNA expression level significantly decrease (P<0.01).Conclusions The results showed that Corylin could inhibit melanin synthesis, which worked probably through inhibiting the activity of TYR and cutting the mRNA expressions of TYR,TRP-1/2.
9.Investigation of microRNA-373 function and mechanism in HepG2 cell growth suppression induced by 5-fluorouracil

Objective To investigate the inhibitory effect of 5-fluorouracil (5-FU) on hepatocellular carcinoma (HCC) cell growth and to elucidate its potential molecular mechanism.Methods Real-time PCR analysis was conducted to determine the expression of miR-373 in HCC tissue specimens and HCC cell lines.The expression of miR-373 was also evaluated in HepG2 cells after 5-FU treatment.Western blot analysis was performed to detect the protein levels of PPP6C,a verified target of miR-373,with transfection of miR-373 mimics or 5-FU treatment.A rescue assay was conducted to investigate the cell growth in HepG2 cells by using CCK-8.Results miR-373 expression was up-regulated in both HCC tissues and cell lines.miR-373 expression depicted about 2.94-fold augment in HepG2 cells as compared to normal liver cells control (P <0.01).5-FU treatment led to a significant decrease of miR-373 levels (approximately 50%,P <0.01,48 h) and resulted in a marked increase of PPP6C protein (approximately 2.1-fold,48 h) in HepG2 cells.The overexpression of miR-373 could prevent the impact of 5-FU treatment on cell growth in HepG2 cells and CCK-8 assay showed that HepG2 cell growth was rescued approximately 81% and 84% at 24 h (P < 0.05) and 48 h (P < 0.01),respectively.Conclusion 5-FU can repress endogenous miR-373 level,which activates the expression of downstream targeted gene PPP6C,thereby exerting an inhibitory effect on HepG2 cells.
10.Effects of 5 classical recipes on anti-oxidative stress in rat liver with cirrhosis.

OBJECTIVETo compare the anti-oxidative stress effects of 5 classical traditional Chinese herbal recipes in rats with dimethylnitrosamine (DMN)-induced cirrhosis.
METHODSDMN was injected intraperitoneally three consecutive days a week to male Wistar rats for 4 weeks to develop cirrhosis model, and the model cirrhotic rats were randomly grouped and administered with 5 traditional Chinese classic recipes, i. e. Yiguanjian; Huangqi Decoction; Yinchenhao Decoction; Xiayuxue Decoction and Xiaochaihu Decoction, by gastrogavage, respectively, for 2 weeks. Changes in activity of total superoxide dismutase (T-SOD), glutathione peroxidase (GSH-Px), glutathione-S-transferase (GST) and malondialdehyde (MDA) content in liver tissue, and serum parameters of liver function were determined.
RESULTSGST activity and MDA content increased significantly in model rats, peaked at week 2 (liver fibrosis formation period) and week 4 (cirrhosis formation period), respectively, while activity of T-SOD and GSH-Px decreased markedly. As compared with the model control group at week 6, the degree of cirrhosis was evidently milder with lower level of MDA and GST and higher T-SOD activity in rats treated by Huangqi Decoction and Yinchenhao Decoction.
CONCLUSIONAnti-oxidative stress action should be one important mechanism for Yinchenhao Decoction and Huangqi Decoction in reversing liver cirrhosis of rats. The action of Yinchenhao Decoction focuses on eliminating hepatic lipid peroxide and that of Huangqi Decoction emphasizes for enhancing anti-oxidation ability of the organism.
