1.Anti-atherosclerosis role of N-oleoylethanolamine in CB2.

To observe a PPAR-alpha agonist effect of N-oleoylethanolamine (OEA) on CB2 (cannabinoid receptor 2), an anti-inflammatory receptor in vascular endothelial cell, healthy HUVECs and TNF-alpha induced HUVECs were used to establish a human vascular endothelial cell inflammatory model. Different doses of OEA (10, 50 and 100 micromol x L(-1)) had been given to HUVECs, cultured at 37 degrees C for 7 h and then collected the total protein and total mRNA. CB2 protein expression was detected by Western blotting and CB2 mRNA expression was assayed by real-time PCR. As the results shown, OEA (10 and 50 micromol x L(-1)) could induce the CB2 protein and mRNA expression, but not 100 micromol x L(-1). To detect if anti-inflammation effect of OEA is partly through CB2, CB2 inhibitor AM630 was used to inhibit HUVEC CB2 expression, then the VCAM-1 expression induced by TNF-alpha was detected, or THP-1 adhere to TNF-alpha induced HUVECs was examined. OEA (50 micromol x L(-1)) could inhibit TNF-alpha induced VCAM-1 expression and THP-1 adhere to HUVECs, these effects could be partly inhibited by a CB2 inhibitor AM630. The anti-inflammation effect of OEA is induced by PPAR-alpha and CB2, suggesting that CB2 signaling could be a target for anti-atherosclerosis, OEA have wide effect in anti-inflammation, it may have better therapeutic potential in anti-inflammation in HUVECs, thus achieving anti-atherosclerosis effect.
2.Causes of misdiagnosis and mistreatment of Dravet syndrome with SCN1A mutations

Objective To analyze the causes of misdiagnosis and mistreatment of Dravet syndrome. Methods Patients with Dravet syndrome diagnosed according to clinical features and SCN1A gene mutation detection were recruited within recent 3 years. The patients were grouped into correct diagnosis-treatment group and misdiagnosis-mistreatment group according to whether the patients had ever been misdiagnosed and mistreated by sodium channel blockers. The clinical features were compared between two groups. Results Thirty-five cases with Dravet syndrome were collected and the rate of misdiagnosis reached 40%, Nine cases were misdiagnosed as symptomatic focal epilepsy, 4 as Lennox-Gastaut syndrome and 1 as Doose syndrome. The average age of onset in misdiagnosis-mistreatment group was (5.50 ± 3.56) months,and the age of confirmed diagnosis was (83.57 ± 105.62) months. The percentage of abnormal EEG, onset seizure with partial seizure, the seizure frequency within the first year from onset, onset with afebrile seizure, patients with status epilepticus or cluster seizures was higher in misdiagnosis-mistreatment group but it showed no significant statistical significance when compared with that of correct diagnosis-treatment group. The percentage of patients with mental retardation and focal neurological signs was significantly higher in misdiagnosis-mistreatment group (P=0.005 and 0.002, respectively). Conclusions Dravet syndrome is frequently misdiagnosed as symptomatic focal epilepsy. The appearance of focal neurological signs and mental retardation before confirmed diagnosis are important factors for misdiagnosis. Gene mutation screening will be helpful for differential diagnosis of Dravet syndrome.

4.Studied of dry matter accumulation and echinacoside content of Cistanche tubulosa in Huabei plain.

OBJECTIVETo give some theory support of Cistanche tubulosa cultivation by searching dry matter accumulation and echinacoside content of C. tubulosa.
METHODDry matter accumulation content of C. tubulosa culturing in Huabei plain was analysed in different growth season of C. tubulosa. Echinacoside content was determined by HPLC.
RESULTDry matter accumulation of C. tubulosa showed "S" variation. Dry matter accumulation increased fastest in September among growing seasons. Dry matter amount was 138.58 g after C. tubulosa grew a year. Dry matter amount decreased significantly along with inoculation time retarded. Echinacoside content was 30.59% when C. tubulosa grew in 5 months, decreased guadully after that, and 9.76% in annual.
CONCLUSIONVariation rule of dry matter accumulation and echinacoside content was found in C. tubulosa that grew one year in Huabei plain.

5.Antioxidation activity and protective effection of ginger oil on DNA damage in vitro.

OBJECTIVETo study the antioxidation activity and protective effect of ginger oil on DNA damage.
METHODChemical light assay was used to detect the oxygen radicals scavenging capacity of ginger oil. The erythrocyte oxidation damage was induced by H2O2. The effect of ginger oil on oxidative erythrocyte was observed by the colorimetric analysis assay, and the content of malondialdehyde (MDA) in rabit hepatocyte was measured. The anaylsis of DNA damage was made with single cell gel electrophoresis(SCGE) technique.
RESULTGinger oil might decrease light value compared with control group and inhibited erythrocyte oxidation damage. Compared with that in control group, the degress of DNA damage reduced significantly in the protected groups. Ginger oil might decrease the content of MDA remarkably and inhitibition rate was 48.16%.
CONCLUSIONGinger oil has dominantive protective effect on DNA damage induced by H2O2. Ginger oil might act as a scavenger of oxygen radical and might be used as an antioxidant.

In this paper, the principle of template imprinting is introduced briefly, the specialty of template imprinting of proteins is analyzed, and the methods of template imprinting of proteins including protein entrapment, microbead surface imprinting, flat surface imprinting as well as the epitope are reviewed in details.

8.Survey on the status of cardiopulmonary resuscitation knowledge master of the masses in Yantian district of Shenzhen city

Objective To analyze the status of first-aid knowledge master of university volunteers and enterprise employees in Yantian district of Shenzhen city so as to provide the basis for the popularity of first-aid knowledge education.Methods Used questionnaire survey method,262 volunteers and 298 enterprise employees who received the university training about first-aid knowledge before and after training were investigated,then contrasted the results.The content validity of the questionnaire was 0.88,the overall Cronbach's α coefficient was 0.865,and the reliability was 0.873.Results Totals of 255 valid questionnaires for volunteers and 288 valid questionnaires for enterprise workers were taken back,with the effective rate of 97.3% and 96.6%.Enterprise workers accounted for 27.78% and volunteers accounted for 40.39% who never training,and the difference was statistically significant (P < 0.05).23.26% enterprise workers and 10.93% volunteers who personally encountered sudden death,and significant difference was found (P < 0.01).The two groups acquired knowledge about unarmed cardiopulmonary resuscitation (CPR) mainly by television,accounting for 39.93% and 37.25%,respectively.Compared with the enterprise employees,14.90% volunteers unwilling give artificial respiration,and the difference was statistically significant (x2 =13.35,P <0.01).Conclusions It is necessary to carry out the universal education about the multifarious first-aid knowledge including CPR,so as to strengthen the attention of public popularity to the training of first-aid knowledge and improve the consciousness and initiative of the public participation in first aid.
9.Clinical anatomy of iliac bone grafts used for mandibular reconstruction and dental implantation

Objective To investigate the anatomical features of iliac bone grafts used for mandibular reconstruction and dental implantation.Methods Sixteen cadavers were dissected.The length,width,height and cortical thickness of the iliac bone were measured with respect to points determined by the relative dimensions of the bone.Results The length of iliac bone graft was (77.2 ± 6.1) mm.The height was from(38.2 ±4.2) mm to (41.9 ±4.7) mm.The width decreased from iliac crest to base line.The least width 10 mm and 15 mm away from iliac crest were (8.4 ±2.2) mm and(6.5 ±2.1) mm respectively.The greatest mean cross-sectional cortical thickness at the intermediate line of the iliac crest was (3.4 ± 0.8) mm.Conclusions Anatomical features of iliac bone are suitable for designing bone grafts for mandibular reconstruction followed by dental implantation.
10.Genetic polymorphism of six short tandem repeat loci in the Han population in Hebei province of China.

OBJECTIVEThis article reports a population genetic study on six short tandem repeat(STR) loci, D7S820, D19S253, D12S391, D5S818, D16S539 and D8S1179, in a sample of unrelated Chinese Han individuals(n=122-173) living in Hebei province.
METHODSDNA extraction from blood samples (200 in number) and multiplex amplification of the above six loci were carried out. Using denaturing polyacrylamide gel electrophoresis and silver stain, the authors investigated the distribution of allele frequencies of the six loci in Han population in Hebei province.
RESULTSThe STR polymorphisms at all of the six loci were observed in Chinese Han population in Hebei province. The observed heterozygosities of D7S820, D19S253, D12S391, D5S818, D16S539 and D8S1179 were 0.828, 0.757, 0.769, 0.837, 0.785 and 0.852, respectively. The measured values of the power of discrimination (PD) were 0.914, 0.919, 0.940, 0.909, 0.917, 0.944; of the mean exclusion chance(MEC) 0.618, 0.740, 0.801, 0.557, 0.655, 0.696 and of the polymorphism information content (PIC) in Chinese 0.771, 0.760, 0.762, 0.708, 0.776 and 0.794, respectively.
CONCLUSIONThe genotype distributions of the six STR were in accordance with Hardy-Weinberg equilibrium. The numerical values of the PD and MEC are relatively high in Hebei province, and thus can be of significant application in population genetics and forensic medicine.
