OBJECTIVE: To compare the efficacy, compliance, safety and economics of standard triple therapy and 10-day sequential therapy consisting of rabeprazole, clarithromycin, amoxicillin and tinidazole for Helicobacter pylori eradication. METHODS: A total of 90 pepticulcer patients who were Helicobacter pylori positive proved by C-urea breath test (C-UBT) were randomly divided into two groups. Forty-five patients received rabeprazole-based 10-day sequential therapy and the other forty-five patients received rabe-prazole-based 7-day standard triple therapy. C-UBT was carried out 4weeks after a course of treatment to evaluate the eradication of Helicobacter pylori. The two regimens were evaluated by cost-effectiveness analysis. RESULTS: Eighty-five patients completed the study. The Helicobacter pylori eradication rates by intention-to-treat (ITT) analysis and by per-protocol (PP) analysis in 10-day sequential therapy group were higher than those in 7-day standard triple therapy group (ITT: 91.1% vs. 73.3%, P<0.05; PP: 95.3% vs. 78.6%, P<0.05). The incidences of adverse reactions (PP: 14.0% vs. 11.9%, P<0.05) were similar in the two groups. The cost-effectiveness analysis showed that the C/E value of the 10-day sequential therapy were lower. CONCLUSION: Rabeprazole-based 10-day sequential therapy is more effective for eradication of Helicobacter pylori.

To establish induction and liquid culture system for hairy roots of Danshen (Salvia miltiorrhiza), Agrobacterium rhizogenes A4, LBA9402, 15834 as test bacterium were used to infect aseptic leaves of Danshen. The hairy roots were induced and positive transgenic hairy roots were selected with PCR using rolB and rolC as the target gene. Then hairy roots of S. miltiorrhiza were harvested and salvianolic acids were extracted with 70% methanol containing 1% formic acid. The content of salvianolic acid B (SalB) and rosmarinic acid (RA) were determined by HPLC. According to the above research results, the Danshen hairy roots induced by A. rhizogenes LBA9402 were inoculated into the following group of culture media: MSOH, MS, B5, and 6,7-V liquid media. Then the same methods of extraction and determination for the content of Danshen hairy roots were adopted. Last, the hairy roots of S. miltiorrhiza induced by A. rhizogenes LBA9402 were inoculated into the MSOH liquid media with different pH values. The content of salvianolic acid were extracted with 70% methanol containing 1% formic acid and determined by HPLC. As a result, three kinds of A. rhizogenes A4, LBA9402, 15834 could induce hairy roots and Ri plasmids were integrated into the genome of S. miltiorrhiza by PCR. Danshen hairy roots induced by A. rhizogenes LBA9402 and A4 produced much more salvianolic acid, which were (3.27 ± 0.37)% [including (1.04 ±0.36)% of RA and (2.22 ± 0.29)% of SalB] and (3.17 ± 0.20)% [including (0.92 ± 0.31)% of RA and (2.25 ± 0.26)% of SalB], respectively. Hairy roots induced by A. rhizogenes LBA9402 when they were cultured in MSOH liquid media produced much more salvianolic acid, which was (4.56 ± 0.36)%, including (1.12 ± 0.26)% of RA and (3.44 ± 0.23)% of SalB. Hairy roots induced by A. rhizogenes LBA9402 produced the most salvianolic acid when they were cultured in MSOH liquid media with the pH value 4.81, which was 4.85%, including 1.16% of RA and 3.69% of SalB. So Danshen hairy roots induced by A. rhizogenes LBA9402 and A4 produced much more salvianolic acid when they were cultured in MSOH liquid media with the pH value 4.81. The research had established the foundation on genetic engineering to improve the quality of S. miltiorrhiza.

BACKGROUND:Bioimpedance method can be used to extract physiology and pathology information relative to the gastricmotility states,to investigate the courses of contraction,peristalsis,transmission and emptying of the stomach,to measure andevaluate gastric motility function.However,early studies mainly focused on the emptying of the stomach,and few studies onevaluation of gastric motility by extracting gastric motility information.OBJECTIVE:To understand the gastric motility from electricity-mechanism composite concept and to establish a noninvasive,convenience measurement method of gastric motility impedance.METHODS:Based on the characters of gastric motility impedance signal,a measurement method of impedance gastric motility isintroduced.Some foundation experiments about the gastric emptying measurement,the gastric motility compare between fastingand repast,the gastric motility comparison of healthy volunteers during different periods and some medicine effects on gastricmotility have carried out.The gastric motility measurement of the patients with functional dyspepsia and gastric motility evaluationfor the patients with erosive gastritis had been reported.RESULTS AND CONCLUSION:Using electrical bioimpedance method,according to the electrical property of the stomach tissueor the food in stomach and the change rules of the property,the physiology and pathology information relative to the gastricmotility states can be extracted,the courses of contraction,peristalsis,transmission and emptying of the stomach can beinvestigated and noninvasive measure and evaluation of gastric motility function can be realized.

Objective To verify the effectiveness of electrical impedance tomography (EIT) with multi-layer electrodes in gastric emptying measurement by simulation experiments using EIT with three-layer electrodes.Methods Firstly,EIT system with three-layer electrodes and gastric emptying measurement simulation equipment was designed and constructed to simulate gastric emptying process and obtain EIT reconstruction images.Secondly,the region where the electrical characteristic of gastric changes was selected as region of interest.Finally,the relative impedance change rate in region of interest for every image obtained from all three layers was consecutively recorded,then the gastric emptying curve was formed and gastric emptying time was calculated.Results There are obvious differences among the sharp of impedance change curves of interested region for three layers,and the gastric emptying time and process of each layer is also different from one another.It is suggested that the passing path and emptying manner of food in gastric emptying process reflected by EIT images varied from layer to layer.Therefore,the necessity and feasibility of using EIT with three-layer electrodes in gastric emptying measurement study is verified.Conclusions EIT with three-layer electrodes can provide more abundant information of gastric motility function associated with gastric emptying pathology and physiology state.It is likely to be a noninvasive,convenient and effective imaging method for the detection and evaluation research on gastric emptying and gastric motility.

italic>Salvia miltiorrhiza Bunge is a traditional Chinese medicinal herb widely used to treat cardiovascular and cerebrovascular diseases at clinic. Its main water-soluble components are rosmarinic acid (RA) and salvianolic acid B (SAB), which are produced by phenylpropanoid pathway. 4-Hydroxyphenylpyruvate reductase (HPPR) is a key enzyme in phenylpropanoid metabolism pathway. SmHPPR1 was cloned from S. miltiorrhiza and was constructed into plant expression vector pJR-SmHPPR1. On this basis, SmHPPR1 transgenic Arabidopsis plants were induced and the content of 4-hydroxyphenyllactic acid (pHPL) was determined. SmHPPR1-overexpressing (SmHPPR1-OE) hairy roots of S. miltiorrhiza were obtained and the concentration of active components and transcriptome analysis were performed. The results showed that the concentration of pHPL in SmHPPR1 transgenic Arabidopsis T1 was 0.594 mg·g^-1 dry weight. The concentration of RA, SAB and total salvianolic acid in SmHPPR1-OE-3 hairy roots were 1.09, 1.29, 1.15 times of that in control-3, respectively, and the content of Danshensu was 36.26% of that in control-3. Transcriptomic analysis revealed that overexpression of SmHPPR1 caused the upregulation of other phenylpropanoid pathway genes like SmTAT2. Protein-protein interaction indicated CYT (TR74706_c0_g1), NADP⁺ (TR26565_c0_g1) and NADP⁺ (TR68771_c0_g1) is the central node of the network and participated in metabolic process and cellular process. The tracking work in this study proved that SmHPPR1 could catalyze the reduction of 4-hydroxyphenylpyruvic acid to 4-hydroxyphenyllactic acid in SmHPPR1 transgenic Arabidopsis, and SmHPPR1-overexpressing in hairy roots of S. miltiorrhiza could increase the concentration of salvianolic acids through synergistically regulating other pathway genes.

Three dimensional electrical impedance tomography (3D-EIT) became an important branch of EIT recently. It is important to research imaging and image quality evaluation methods for single targets of different positions and multi-targets in 3D field. Using finite element subdivision method, 3D-EIT field was dispersed into cube unit in the present study for models with single target located in the center of field, middle of field, and near to the edge, respectively. For models with two targets and four targets near to the field edge, Tikhonov-Noser algorithm was adopted in image reconstruction. Imaging error function ER and structure similarity degree function SSIM were introduced to evaluate the reconstructed images. For the models with signal target, with the movement of the target from the center to the edge of the field, the value of ER increased and SSIM decreased, and reconstruction quality decreased. For the models with multi-targets near to the field edge, ER and SSIM increased and decreased respectively with the increase of target number, mage quality also decreased. Tikhonov-Noser algorithm is an effective 3D-EIT algorithm. ER and SSIM are adaptive for the characteristic of 3D-EIT images, and it can quantitatively evaluate the 3D-EIT imaging effect from the two perspective of imaging error and structure quality.

Objective To study the influence of low-iodine diet on the expression of homeobox gene nkx2.1 in rat cerebral tissue. Methods Forty female Wistar rats were randomly divided into two groups according to body. quality: low-iodine group and control group,both fed with low-iodine feed at an iodine content of 13.66 μg/kg,respectively given the deionized water and 200 μg/L KIO3 solution. The hormone levels of two group rats were determined with chemiluminescence immunoassay after three months, and then mated with healthy male rats. Cerebral tissues were taken from the fetus of 16-day pregnancy,newborn and 20 days old offspring in low-iodine and control group to detect the content of nkx2.1 mRNA using real-time fluorescence quantitative PCR (FQ-PCR) techniques. Results Serum TT3, TT4, FT3, FT4 level of rats in low-iodine group(0.89±0.20, 0.32±0.16, 3.33± 0.61, 3.28±0.80) was respectively lower than that in the control group(1.04±0.06, 39.42±14.68,4.83±0.33, 26.99±4.48;t = 2.71,6.52,5.70, 12.89, P < 0.05 or < 0.01). The relative nkx2.1 mRNA expression was(5.60± 0.30)×10-3, (1.20 ± 0.29)×10-3, (0.18± 0.06)×10-3 respectively in the fetus of 16-day pregnancy, newborn and 20 days old offspring of control group, while it was (3.00 ± 0.55)×10-3, (1.90 ± 0.21)×10-3,(0.69 ± 0.15)×10-3 in the low-iodine group. The difference of nkx2.1 mRNA expression was significant among fetal and neonatal rats in the control group and low-iodine group(F = 210.07,162.40, both P < 0.01). The nkx2.1 mRNA expression of newborn rats was lower than that of 16-day pregnancy in both groups(P < 0.01), and that of 20 days old rats was lower than that of 16-day pregnant and neonatal rats(P < 0.01). The 16-day pregnant rats of control group had obviously higher level of nkx2.1 expression than those in the low-iodine group(t = 16.073, P< 0.01), while the nkx2.1 of newborn and 20 days old low-iodine rats expressed much higher than healthy rats(t = 7.573,12.221, P < 0.01). Conclusions Brain development retardation caused by low-iodine is closely related to nkx2.1 differential expression in the brain tissue.

Objective Practical electrical impedance tomography (EIT) system with certain measurement resolution (MR) and signal-to-noise ratio (SNR) differs a lot from ideal simulation conditions in EIT methodology research. The aim of this paper was to study the impact of practical system with different MR and SNR on EIT. Methods Based on the ideal simulation boundary voltages of adjacent excitation and adjacent measurement pattern, practical systems whose MR were 0.1 mV and 0.01 mV, SNR ranged from 40-80 dB and no noises were simulated, simulation study for three imaging models A, B and C with different positions in the to-be imaged field under practical system conditions above was carried out using the combined Tikhonov-NOSER regularization algorithm, and error of reconstruction (ER) function and structure similarity (SSIM) function were adopted for quantitative evaluation of image effect. Results There are differences between images obtained under different MR and SNR for different image models. In order to obtain images for three models, SNR of system should be at least 40-50 dB when MR was 0.01 mV. For the goal to obtain high quality images of A, B and C, SNR should be 80, 70 and 60 dB respectively. When MR was 0.1 mV, SNR for obtaining images of A, B, C were 60, 50 and 40 dB. Moreover, the improvement of images with increasing SNR under MR of 0.1 mV was not as obvious as that of MR at 0.01 mV. Conclusions Blind pursuit of single high SNR is of no help to improve system performance and image reconstruction effect except increasing the difficulties in hardware design. The extremity SNR is 50-60 dB for system with MR of 0.01 mV and 60-70 dB for system with MR of 0.1 mV.

To study the protective effect and the mechanism of asiatic acid (AA) from Potentilla chinensis on alcohol hepatic injury in rats. Male Wistar rats were randomly divided into six groups: the normal control group, the AA control group (8 mg · kg(-1) AA), the model group (5.0-9.0 g · kg(-1) alcohol) and high, medium and low-dose AA-treated groups (alcohol + 8, 4, 2 mg · kg(-1) AA). Each group was orally administered with the corresponding drugs once a day for 24 weeks. Approximately 1. 5 hours after the final administration, all rats were killed, and their blood samples and hepatic tissues were collected. The AST and ALT in rat serum and the contents of MPO, TNF-α, IL-1β, SOD, GSH-Px, GSH-Rd and MDA in hepatic tissues were detected. The expressions of NF-κB, TLR4, CD14, MyD88, TRIF and protein expression in hepatic tissues were measured by western blot. The pathological changes in liver tissues were observed by histological examination. The results showed that compared with the model group, the AA-treated groups showed significant decreases in serum ALT, AST and MDA and increases in the activities of SOD, GSH-Px, GSH-Rd and MPO. Moreover, AA markedly inhibited the expressions of TNF-α, IL-1β, TLR4, CD14, MyD88 and NF-κB. The histological examination showed alleviated hepatic issue ijury to varying degrees. In short, asiatic acid (AA) from P. chinensis could protect alcohol-induced hepatic injury in rats. Its mechanism may be related to the inhibition of NF-κB inactivation and the reduction of inflammatory response.
