3.Hyaluronidase with α - lipoic acid and Chinese medicine in the treatment of diabetic vitreous hemorrhage

AIM: To study the effectiveness of hyaluronidase periocular injection combined with α - lipoic acid and Chinese medicine in the treatment of diabetic vitreous hemorrhage.
METHODS: Retrospective analysis of 67 patientswith proliferative diabetic retinophathy ( PDR ) and vitreous hemorrhage, were divided into two groups: treatment group and control group. There were 32 cases in every group, the treatment group were giving hyaluronidase periocular injection combined with α - lipoic acid intravenously and with traditional Chinese medicine in promoting blood circulation to remove blood stasis, control group were only given Chinese medicine treatment in promoting blood circulation to remove blood stasis.
RESULTS: Followed up 2-3mo, the total efficiency of treatment group ( 78%) was better than control group (57%), there was statistically significant difference ( P<0-05 ) . The efficiency of the two groups' patients with vitreous hemorrhage more than 2wk time was 22% and 25% respectively, there was no statistically significant difference in the two groups (P>0. 05).
CONCLUSION: Hyaluronidase periocular injection combined with α-lipoic acid and Chinese medicine in the treatment of PDR combined with vitreous hemorrhageis more effective than only use Chinese medicine. But it only has effect for the early, over 2wk, it has no significant effect.
4.Study of mechanism and the effect of intracerebroventricular microinjection of serotonin on gastric motility in rabbit.

AIMTo explore the effect of central serotonin (5-hydroxytryptamine, 5-HT) on gastric motility in rabbit and to analysis its mechanism.
METHODSIntracerebroventricular (icv) microinjection of drugs were used. The frequency and the amplitude of contractions of gastric motility were recorded.
RESULTS(1) Injection (icv) of 25 microg 5-HT increased the amplitude of contractions and decreased the frequency of gastric motility. (2) The effects of 5-HT were inhibited by injection (icv) of atropine, but intravenous(iv) injection of atropine could only block the increase effect of 5-HT on intragastric pressure. After injection (icv or iv) of phentolamine or propranolol, the effects of 5-HT on gastric motility were not changed markedly. (3) Microinjection (icv) of naloxone or diphenhydramine inhibited the inhibitory effect of 5-HT on the frequency of gastric contractions, and the diphenhydramine reversed the increase effect of 5-HT on intragastric pressure. (4) Vagotomy abolished the effects of 5-HT completely.
CONCLUSIONThere may be different nerve mechanisms in regulation of central 5-HT on the frequency and amplitude of contractions of gastric motility. The increase effect of 5-HT on intragastric pressure is medicated by the cholinergic system in brain, and histamine may be one of the important element on the effect of 5-HT. The cholinergic fibers in vagus nerve transmit the effect. The inhibitory effect of 5-HT on the frequency of contractions is also medicated by activating the cholinergic system in brain, enkephalin and histamine are also involved, and it is transmitted via noncholinergic and nonadrenergic fibers in vagus nerve.

6.Effect of trichostatin A and paclitaxel on the proliferation and apoptosis of lung adenocarcinoma cells.

BACKGROUNDHistone deacetylase inhibitors can regulate gene expression through modulation of the degree of acetylation of histone and non-histone, thus affecting cell proliferation, survival and chemosensitivity. Histone deacetylase inhibitors combined with paclitaxel may enhance the inhibitory effect of drugs on lung cancer cells. This study aimed to observe the effect of trichostatin A (TSA)/paclitaxel on the proliferation and apoptosis in human A549 lung adenocarcinoma cells, and to investigate its mechanism.
METHODSA549 cells were cultured in Dulbecco modified Eagle's medium (DMEM) in the presence of paclitaxel and the histone deacetylase inhibitor TSA, and the growth curve was obtained by trypan-blue exclusion assay and cell count. Apoptosis was assessed using Hoechst 33258 staining and flow cytometry analysis, and cell cycle was detected by flow cytometry analysis. The proteins poly ADP-ribose polymerase (PARP), caspase-3, survivin, and tubulin acetylation were detected by Western blotting.
RESULTSA significant reduction of proliferation was observed in A549 lung adenocarcinoma cells treated by paclitaxel or TSA. Combined treatment with TSA/paclitaxel caused the greatest inhibition of cell proliferation. The combined treatment with TSA and paclitaxel induced more severe apoptosis, and significantly more cells were arrested in G2/M phase (P < 0.05) then with a single drug. Using Western blotting, we demonstrated that treatment with TSA/paclitaxel led to synergistic increase in acetylated tubulin, PARP, caspase-3, and reduced the expression of survivin.
CONCLUSIONTSA and paclitaxel have a synergistic activity that can inhibit cell growth and induce apoptosis.

7.RAPD analysis on different varieties of Rehmannia glutinosa.

OBJECTIVETo provide molecular evidences for its breeding by studying the genetic relationship among varieties of Rehmannia glutinosa.
METHODNineteen varieties were detected by Randomly Amplified Polymorphic DNA(RAPD) markers.
RESULTThe 20 selected primers produced 163 bands, among which 114(69.9%) were polymorphic. A DNA molecular dendrogram was established based on Hierarchical cluster analysis of 163 DNA bands amplified by 20 primers, which divided the 19 varieties into four groups: Group Beijing, Group 85-5, Group Guolimao and the other Group.
CONCLUSION8 varieties of Group Beijing have a close genetic relationship, and so have varieties of Group 85-5, which provides information for Rehmannia glutinosa's breeding.

8.Construction of cDNA library from NPC tissue and screening of antigenic genes.

To obtain the NPC-associated antigens, a powerful new method, SEREX (serological identification of antigen by recombinant cDNA expression library), was used for identifying the antigens eliciting humoral immune response. Before performing serological analysis, a high quality cDNA library derived from human nasopharyngeal carcinoma (NPC) tissue was constructed. The primary library consisted of 3.64 x 10(6) recombinants and the recombinant rate was 94%. For better preserving the cDNA library, it was amplified. As a result, the titer of the amplified cDNA library was 3.8 x 10(9) pfu/mL. With SEREX method, immunoscreening for the detection of reactive clones in the human NPC tissue cDNA library was performed with autologous serum. As a result, 23 positive clones encoding antigenic genes were obtained after immunoscreening, and the nucleotide sequences of cDNA inserts were determined and analyzed with BLAST software in GenBank. Results showed that the 23 reactive clones were derived from 16 different genes. 10 of 16 genes had high homologous to the genes known in GenBank, such as RPL31, S100 A2, MT2A, etc. However, there were also 6 genes with low homology to the genes known in GenBank. Furthermore, 3 of 6 genes may be novel genes. The associations of these genes to NPC and the roles that they played in the occurrence and development of NPC should be revealed by further research.

9.Conjugated effects of fluorine and aluminum on the sex hormones of male rats

Objective To observe the combined poisonous effects of fluoride and aluminum on sex hormone of male rats.Methods Sixteen weaned SD healthy male rats aged two week were selected and divided into control group,aluminum group,fluoride group,fluorine-aluminum group,four rats in each group.All rats in the experimental groups were fed with corn collected from the prevailng areas containing different fluorine contents respectively for 90 days.Serum testosterone(T)and estradiol(E2)were detected.Results Compared separatelv with the control group[(3.317±0.635)μg/L],serum T level of fluorine-aluminum group[(15.994±6.558)μg/L]was higher(P<0.05),but aluminum[(8.134±3.134)μg/L]and fluorine[(1.868±0.367)μg/L]groups had no significant differences(P>0.05).Compared separately with the control group[(0.319±0.072)nmol/L],E2 level of the fluorine group[(0.172±0.030)nmol/L]being lower(P<0.05),and it was not significant differences(P>0.05)in the control group when compared with aluminum group[(0.282±0.012)nmol/L],and fluorine-aluminum group[(0.265±0.047)nmol/L].Fluorine and aluminum interacted with each other(F=9.82,P<0.05).Conclusion The combined poisonous effects of fluorine and aluminum may influence sex hormone levels of male rats.
10.Correlation between single nucleotide polymorphism H558R in SCN5A gene and chronic Keshan Disease complicated with hypertension, and their electrocardiogram characteristics

Objectives To investigate the relationship between single nucleotide polymorphism (SNP)H558R in SCN5A gene and chronic Keshan disease (KSD) complicated with hypertension,and the relationship between H558R and occurrence of arrythmia in chronic KSD complicated with hypertension.MethodsThirty nine patients with chronic KSD complicated with hypertension and 63 geographical region matched hypertension control subjects were recruited in our study in Fuyu county,Qiqihaer city,Heilongjiang province between 2006 and 2010.H558R polymorphism in case and control groups was genotyped using the polymerase chain reaction single-strand conformation polymorphism(PCR-SSCP) and sequenced,and electrocardiography(ECG) characteristics were examined in the two groups.Case-control study analytical methods were applied to analyze the relationship between H558R and chronic KSD complicated with hypertension,and the relationship between H558R and occurrence of arrythmia in chronic KSD patients complicated with hypertension.Results Subjects of genotype 558 TC in the case group had a decreased risk of chronic KSD complicated with hypertension with odds ratio of 0.288[95% confidence interval (CI):0.104 - 0.794],and subjects of genotype TC in chronic KSD complicated hypertension patients had a decreased risk of QRS prolongation with odds ratio of 0.061 (95%CI:0.006 - 0.612).Conclusions Polymorphism H558R in SCN5A gene may be a predisposition factor of chronic KSD complicated with hypertension and occurrence of arrythmia in chronic KSD complicated with hypertension.
