2.Differential proteomic expression in human liver cells stimulated by hydroquinone.

OBJECTIVETo explore the differential proteomic expression in human liver cells L-02 after exposure to HQ.
METHODSSubcultured L-02 cells were treated by HQ for 24 h at a 1 x 10(-4) mol/L concentration and a blank group was set as the control. Immediately after the treatment, total cellular proteins were extracted and separated by 2-DE, and the images were analyzed by PDQuest software. The experiment was totally repeated 3 times with 3 repetitions for each group every time. The well repeated spots were identified by MALDI-TOF MS and then searched in NCBI human protein database with Mascot.
RESULTSAbout 1,000 spots per gel were found. Compared with the control group, 17, 18 and 24 spots were significantly altered in 3 separate experiments. The 4 well repeated spots were identified by MALDI-TOF MS as Rho GDP dissociation inhibitor GDI alpha, 6-phosphogluconolactonase, erbB3 binding protein EBP1 and lamin A/C, isoform 1 precursor. They were involved in cell skeleton, signal transduction and energy metabolization in functional classification.
CONCLUSIONHydroquinone can change the protein expression in liver cells, which provides clues for exploring the toxic mechanism.

3.Molecular mechanisms involved in the treatment of inflammatory bowel disease by Pulsatilla decoction.

OBJECTIVETo investigate molecular mechanisms underlying in the treatment of inflammatory bowel disease by Pulsatilla decoction.
METHODSWistar male rats were randomly divided into control group, model group, model + positive control group (mesalazine), traditional Chinese medicine treatment group, in addition, the Chinese medical treatment group was divided into middle and high dose group ( n = 8). Intragastric administration was used in the positive control group and traditional Chinese medicine treatment group. The expression of Smad7 and p-Smad3 in the colons were detected by immunohistochemistry and Western blot.
RESULTSCompared with the model group, positive medicine and traditional Chinese medicine group, especially high-dose group, could effectively inhibit the expression of Smad7, while enhancing the p-Smad3 expression.
CONCLUSIONThe activation of TGF-beta1/Smad3 signaling pathway may be the molecular mechanism underlying in the anti-inflammatory effect of inflammatory bowel disease by Pulsatilla decoction.

4.Comparison on predictive capacity of ARIMA model and LSTM model for incidence of hand, foot and mouth disease in Shijiazhuang

Objective To predict the incidence of hand, foot and mouth disease (HFMD) in Shijiazhuang using the multiple seasonal autoregressive integrated moving average model (ARIMA) and long short term memory (LSTM) model, lay theoretical foundation for the prevention and control of HFMD. Methods Multiple seasonal ARIMA model and LSTM model were established separately by using Eviews 8.0 and python 3.7.1 according to the data of monthly incidence of HFMD from January 2013 to May 2018 in Shijiazhuang, and the data from June 2018 to May 2019 were used to verify the prediction precision of model. Finally, the monthly incidence from June to August 2019 was predicted. Results Based on the monthly incidence from January 2013 to May 2018, the optimal models, ARIMA(1,0,0)×(1,1,2)12 and LSTM model were established. Mean absolute percentage of error (MAPE) of ARIMA and LSTM model were 22.14 and 10.03 respectively based on the monthly incidence from June to December 2018, while MAPE of ARIMA and LSTM model were 43.84 and 25.26 respectively based on the monthly incidence from June 2018 to May 2019. These results indicated that LSTM model was superior to ARIMA model in model fitting degree and predicting accuracy, which was relatively consistent with the actual situation. Conclusions LSTM model is able to fit and predict the incidence trend of HFMD well in Shijiazhuang. It can provide guidance to HFMD epidemic prediction and alerting.
5.Effects of different intensity interval exercise of 6 weeks on body composition of obese rats.

OBJECTIVE:
To investigate the effects of different intensity intermittent exercise on the body function of obese rats, and to provide basis for the prevention and treatment of obesity.
METHODS:
Eighty SD rats were randomly divided into normal diet group (n=20) and high-fat diet group (n=60). After adaptive feeding for 8 weeks, 8 normal diet rats and 32 high-fat obese rats were selected for follow-up experiments. The experimental rats were randomly divided into 5 groups (n=8): control diet-sedentary (CS),with ordinary feed and without any exercise; high diet-sedentary (HS), with high-fat feed feeding and without any exercise; high diet-continual exercise(HC), 60 min/day,5 days/week with 6 weeks; high diet-long time-low frequency interval exercise(HLL), 30 min/time,twice/day (intermittent 6 h), 5 days/week with 6 weeks; high diet-short time-high frequency interval exercise(HSH), 20 min/ time, 3 times/d (intermittent 3 h), 5 days/week with 6 weeks. The training intensity of rats in each exercise group was 25 m/min. After 6 weeks, rats in each groups were weighed, and resting metabolic rate(RMR), fasting blood glucose(FBG), triglyceride(TG) and other biochemical indexes were detected, and fat and muscle weight were measured.
RESULTS:
Before experiment, there were no significant differences in RMR, FBG and TG in each groups(P>0.05).The body weight of HSH, HLL, HC and HS groups was higher than that of CS group (P<0.05). After the experiment, RMR of the HSH,HLL and HC groups was significantly higher than that of HS and CS groups (P<0.05), but without significant difference among the HSH,HLL and HC groups (P>0.05).The body weight of HSH, HLL and HC groups was significantly lower than that of HS group (P<0.05), but the three groups was not significant (P>0.05); perirenal fat(PF), idymis fat(EF), perirenal fat/weight(PF/W) and epididymis fat/weight(EF/W) of HSH, HLL, HC group were significantly lower than those of HS group (P<0.01), while there was no statistical difference among the three groups (P>0.05). There was no significant difference in gastrocnemius(GM) and quadricep(QF) of each group (P>0.05), gastrocnemius/weight(GM/W) and quadriceps/weight (QF/W) in HSH,HLL and HC groups were higher than those of HS group (P<0.05),while there was no significant difference among HSH,HLL and HC groups (P>0.05);FEB,TG of HSH,HLL,HC group were lower than those of CS and HS group (P<0.05),but the difference with HS group was more significant (P<0.01),there was no significant difference among training groups(P>0.05).
CONCLUSION
6 weeks of intermittent exercise of different intensity had a good intervention effect on the body composition of obese rats, and high diet-short time-high frequency interval exercise (HSH) may be more effective.

Alzheimer disease (AD) is one of the most common types of dementia.Serotonin (5-HT) plays an important role in the pathogenesis of AD,causing alterations in patients' cognitive abilities,as well as learning and memory capabilities,by regulating the cholinergic and glutamatergic neurotransmission through the signal transduction pathway.Patients suffer from emotional disorders including depression and apathy and clinical manifestations such as unrestrained diet,which are all related to the signaling function of 5-HT receptors.Strengthening the 5-HT receptor-mediated transduction pathway and increasing the concentration of synaptic 5-HT have been used as a novel therapy for delaying the progression of AD.This articles reviews the AD therapies targeting 5-HT transduction pathway.
7.Clinical status of immunocompromised host complicated with pulmonary infection

Pulmonary infections are the most common complications in immunocompromised host (ICH).The immune system of ICH was in inhibitory condition because of long-term use of immunosuppressive agents,such as glucocorticoids,antimetabolite medicine and alkylating agents.Therefore,compared with the immunocompetent host,the condition is more critically serious in ICH with more difficult diagnosis and treatment,and higher motality after pulmonary infections.Early identification of pathogens and early treatment are helpful for improving the prognosis and decreasing mortality.
8.Effects of polychlorinated biphenyl on the expressions of c-fos, c-Myc and beta-catenin in the rat testis.

OBJECTIVETo study the effects of polychlorinated biphenyl (PCB) on the phenotype of the testis tissue and the testis tissue and the expression c-fos, c-Myc and beta-catenin in the rat testis.
METHODSForty-five Wistar male rats were divided into a control and three perimental groups, the former fed normally, and the latter with PCB at 0.1, 1 and 10 mg/kg respectively for 90 days. Then the effects of PCB on the phenotype of the testis tissue and the expressions of c-fos, c-Myc and p-catenin were determined by histopathology and immunohistochemistry.
RESULTSHistopathological examinations revealed testis edema, damage of the mesenchymal phenotype, morphological changes of the contorted seminiferous tubules, absence of stromal cells, spermiocytes and prespermatids, and decreased number of sperm. The expressions of c-fos and c-Myc were significantly higher in the 1 and 10 mg/kg PCB groups than in the control and 0.1 mg/kg PCB groups (P < 0.01). The expression of beta-catenin was downregulated in the 0.1 mg/kg PCB group, with significant differences from the other groups (P < 0.01), but it was higher in the 1 mg/kg PCB than in the control and 10 mg/kg PCB groups (P < 0.01).
CONCLUSIONPCB causes changes in the phenotype of the testis tissue, and the abnormal expressions of c-fos, c-Myc and beta-catenin are closely related to the PCB-induced testis injury.

9.Effects of pravastatin on the proliferation and invasion of human hepatocarcinoma HepG2 cell line.

OBJECTIVETo observe the effects of pravastatin on the proliferation and invasion of human hepatocarcinoma HepG2 cell line.
METHODSThe effects of pravastatin on the proliferation, migration and invasion of HepG2 cells was observed by MTT assay, Boyden chamber assay and motility assay. p38 activity was measured, and the expression of p-p38, MKP-1, RhoC and MMP-2 was analyzed by Western blot.
RESULTSPravastatin inhibited the proliferation of HepG2 cells. The intracellular p38 activity and expressions of p-p38, RhoC and MMP-2 were decreased, while MKP-1 expression was elevated in pravastatin treated cells. In addition, pravastatin inhibited the invasion and motility.
CONCLUSIONPravastatin can inhibit the proliferation and invasion of HepG2 cells.

10.A Matched Comparison Study of Uniportal Versus Triportal Thoracoscopic Lobectomy and Sublobectomy for Early-stage Nonsmall Cell Lung Cancer.

BACKGROUNDBoth uniportal and triportal thoracoscopic lobectomy and sublobectomy are feasible for early-stage non-small cell lung cancer (NSCLC). The aim of this study was to compare the perioperative outcomes of uniportal and triportal thoracoscopic lobectomy and sublobectomy for early-stage NSCLC.
METHODSA total of 405 patients with lung lesions underwent thoracoscopic lobectomy or sublobectomy through a uniportal or triportal procedure in approximately 7-month period (From November 2014 to May 2015). A propensity-matched analysis, incorporating preoperative variables, was used to compare the short-term outcomes of patients who received uniportal or triportal thoracoscopic lobectomy and sublobectomy.
RESULTSFifty-eight patients underwent uniportal and 347 patients underwent triportal pulmonary resection. The conversion rate for uniportal and triportal procedure was 3.4% (2/58) and 2.3% (8/347), respectively. The complication rate for uniportal and triportal procedure was 10.3% and 9.5%, respectively. There was no perioperative death in either group. Most patients had early-stage NSCLC in both groups (uniportal: 45/47, 96%; triportal: 313/343, 91%). Propensity score-matching analysis demonstrated no significant differences in operation time, intraoperative blood loss, numbers of dissected lymph nodes, number of stations of lymph node dissected, duration of chest tube, and complication rate between uniportal and triportal group for early-stage NSCLC. However, the duration of postoperative hospitalization was longer in the uniportal group (6.83 ± 4.17 vs. 5.42 ± 1.86 d, P = 0.036) compared with the triportal group.
CONCLUSIONSUniportal thoracoscopic lobectomy and sublobectomy is safe and feasible, with comparable short-term outcomes with triportal thoracoscopic pulmonary resection. Uniportal lobectomy and sublobectomy lead to similar cure rate as triportal lobectomy and sublobectomy for early NSCLC.
