1.Time-course analysis of interleukin 1 alpha and 1 beta gene expression during the skin wound healing in mice

AIM: Studies reveal that, cytokines play an important role in the process of various inflammatory reaction, wound repair and cell mutation, also, cytokines are expected to be a novel indicator of ratiocinating wound time. This study was designed to explore the changes in gene expression of interlukin-1? (IL-1?) and interlukin-1? (IL-1?) during the skin wound healing process in mice at different phases. METHODS: The experiment was carried out in the laboratory of Forensic Medicine Department of China Criminal Police University from March 2006 to December 2007. ①Thirty-five BALB/c inbred male white mice of cleaning grade, aged 25-27 g, were used in this study. ②Skin wound model was induced by injuries in the area of 1.0 cm away from two sides of spinal column, without affection on muscles. Three pairs of injuries were established in each mouse. Five animals were decapitated under anesthesia at 0, 3, 6, 12 hours and 1, 3, 6 days, respectively, whereas those at 0 hour were taken as controls. ③The white blood cells at different phases were observed under microscope after immunochemical staining. Real-time quantitative polymerase chain reaction method was applied to detect gene expression of IL-1? and IL-1?. RESULTS: Thirty-five BALB-c inbred male white mice were all involved in the result analysis. ①Skin wound was weakly positive during 0-5 hours after injury; the positive reaction was attenuated during 6-24 hours, and positive cells increased, of which macrophages were dominant. Infiltration of massive leucocyte neutrophils and a small quantity of mononuclear cells were detectable at 1 day; mononuclear cells increased during 2-4 days, and reached a peak at 3 days. In control group, the epidermis, folliculus pili, glandulae sebaceae and coil gland were all weakly positive. ②IL-1? and IL-1? were expressed at early phase after injury, and two expressing peaks were found at 6 and 72 hours, then decreased to normalized levels at 6 days after injury. CONCLUSION: Genet expression of IL-1? and IL-1? in damaged tissues has a regularity with wound time, and can be used for reference to estimate early injury age in forensic cases.
2.Cloning and Analyzing Biological Activity of A2 Gene in Q? Phage

Objective To construct A2 gene expression vector in Q? phage by gene recombination technology, and then analyze its physiological activities. Methods Amplified A2 gene in Q? genome by PCR, cloned it into pBAD-24 expression vector to construct pBAD A2 recombinant plasmid. The recombinant plasmid was identified by restrictive enzymes digestion and DNA sequencing, then to be transfected into host cell JM109. After induced by Arabinose, the expression level of A2 was detected by SDS-PAGE. The growth curve of E.coli was obtained by phototurbidometry to test the bacteriolysis activity of pBADA2 in various host cells. Results After certified by PCR screening, DNA sequencing and restrictive enzymes digestion, the expression vector of pBADA2 was successfully constructed. The gene expression level is high in JM109 and related with Arabinose concentration, which reach its peak when Arabinose is 0.2%. OD660 value demonstrates that pBADA2 has the function of bacteriolysis, which could dissolve JM109、HB101 and 594 in E.coli rapidly, but not BE110. Conclusion The highly expressed vector pBADA2 was successfully constructed. The protein expressed has the ideal function of bacteriolysis. All of these provide theoretical and practical bases for developing new anti-bacteria drugs.

4.The effect of biomechanis with different bone density by the pedicle screws fixation

Objective To explore the biomechanis with different bone density by the pedicle screws fixation before and after fatigue test.Thus provide biomechanical basis for the clinical work.Methods A total of 27 fresh lumbar vertebrae samples of adult sheep (L1-L5) were equally divided into three groups,such as group A of decalcified with HCL 0 h,group B of decalcified with HCL 2 h and group C of decalcified with HCL 4 h,by using the random number table method,every groups was 9 models.Four screws was performed on the L4-L5 pedicle of vertebral arch of the three groups.Three groups were performed fatigue test of 250 000 times in 4 direction (flexion,extension,left lateral bending,right lateral bending) by (300+ 105) N load.The range of motion(ROM),the axial compressive stiffness were measured and the results in every group were compared before and after fatigue test.Results After fatigue test,ROM and the axial compressive stiffness had no significant change in group A (P>0.05),but ROM of B group and C group were both increased and the axial compressive stiffness were decreased,Significance was found with and between groups.Conclusion The stability of people who are performed pedicle screws simplly may be decrease when the spine of osteopenia.
5.Mechanism underlying intrauterine growth retardation induced by caffeine and its research advance

Intrauterine growth retardation (IUGR) is one of the most commonly encountered developmental toxicity, which could lead to perinatal morbidity and mortality, be also extended from the fetus to adulthood, and seriously affect the quality of the population. Caffeine widely exists in a variety of daily beverages and some drugs. Its consumption is increasing year by year. Caffeine intake during pregnancy is one of the risk factors for IUGR. However, its mechanism of adverse outcome based on embryonic research is still unclear. In this paper, the possible mechanisms of caffeine-induced IUGR focusing on 3 important factors-the mother, placenta and fetus were explored. Caffeine's impact on the mother is the chronic activation of renin-angiotensin system; on the placenta, caffeine induces cell damage or the failure of the cell proliferation/apoptosis balance, leading to blockage of blood supply to the placenta; caffeine is also capable of directly affecting fetal development through interfering its neuroendocrine.

A total of 115 patients with Hoshimoto thyroiditis were classified under 2 groups: one group being treated with selenium and L-throxine (L-T4) and other group with L-T4 alone.All patients were followed up for 3 months.In the selenium treated group thyroid peroxidase antibody level was decreased in 33 patients (56%) and thyroglobulin antibody level was decreased in cases.
7.Detection of 18 Antihypertensive Agents Illegally Added in Traditional Chinese Medicines and Healthy Care Products by HPLC-QTOF/MS

Objective:To establish a rapid, sensitive and accurate HPLC-QTOF/MS determination method for the illegally added antihypertensive drugs in traditional Chinese medicines and healthy care products .Methods:An Agilent Eclipse plus C 18 column ( 50 mm ×2.1 mm,1.8 μm) was adopted with the mobile phase of 0.5%formic acid and acetonitrile with gradient elution .The flow rate was 0.2 ml· min-1 .The electrospray ionization source was applied and operated in a positive ion mode .Results:The detection limit of 18 antihypertensive drugs was within the range of 0.2-2.5 ng· ml-1 .Reserpine was found in one sample .Conclusion:The method is selective and sensitive , which can be used for the detection of 18 chemical medicines illegally added in antihypertensive traditional Chinese medicines and health care products .
8.Prediction of treatment response for cardiac resynchronization therapy by left ventricular twist using three-dimensional speckle tracking imaging in a heart failure canine model

Objective To assess left ventricular (LV) twist in a rapid pacing induced heart failure canine model undergoing cardiac resynchronization therapy (CRT) by three-dimensional speckle tracking imaging (3D-STI).Methods Rapid right ventricular pacing (RVP) was utilized in 22 adult beagle dogs for 3 weeks to induce heart failure.Then 15 dogs received CRT for 2 weeks and others were treated as control.Apical full-volume acquisition of the LV was obtained in conscious animals at baseline,the end of 3-week RVP and the end of 2-week CRT.Peak LV apical (AP-Prot) and basal rotation (MV-Prot) along with peak twist (Ptw) and torsion (Ptor) were automatically calculated by TomTec 4D LV Analysis 3.0 software to identify the ideal parameter in predicting treatment response of CRT.Results After 2 weeks of CRT,LV ejection fraction(LVEF) increased and LV end-systolic volume(LVESV) decreased significantly in dogs with heart failure.CRT treatment response,defined as improvement of LVESV≥15%,was observed in 9 dogs.Significant difference was found in Ptw [(7.43 ± 0.61) vs (6.06 ± 0.89)°,P <0.05] and Ptor [(1.43 ± 0.45) vs (0.67 ± 0.36)°/cm,P <0.05] between responders and nonresponders.Ptw and Ptor predicted CRT response with satisfying sensitivity as 89% and 85%,specificity as 83% and 84%,respectively.Conclusions Peak twist and torsion evaluated by 3D-STI represented overall LV twist and demonstrated potential prediction value for treatment response of CRT.
9.Three-dimensional speckle tracking imaging permits qualification of left ventricular twist in dogs with heart failure

ObjectiveTo assess left ventricular (LV) twist in Beagle dogs with rapid-pacing induced heart failure by 3-dimensional speckle tracking imaging.MethodsSeventeen adult beagle dogs underwent rapid right ventricular pacing (RVP) to induce heart failure.Right ventricles were paced at 260 beats/min for 3 weeks.Apical full-volume acquisition of the LV was obtained in conscious animals at baseline and the end of 3-week rapid pacing.Peak LV apical rotation(AP-Prot) and basal rotation (MV-Prot) accompanied with peak twist (Ptw) and torsion were automatically calculated by TomTec 4D LV Analysis 3.0 software.The relation between LV twist and QRS duration was further studied.Results After 3 weeks of rapid ventricular pacing,AP-Prot,MV-Prot,Ptw decreased significantly [At-Prot:(13.96 ± 2.00) ° vs (5.85 ± 0.58)°;MV-Prot:(3.34± 0.38)° vs (2.13 ± 0.44)°; Ptw:(16.31 ± 2.01)° vs (7.08 ± 1.16)°,all P <0.05].Dogs with heart failure were divided into two groups according to the QRS duration:pQRSd group with QRS≥100 ms and nQRSd group with QRS<100 ms.No significant difference was found in AP-Prot and MV-Prot between two groups ( P >0.05).In the pQRSd group,the peak of apical rotation occurred earlier than the peak of basal rotation [(162.89 ± 14.33) ms vs (91.43 ± 15.45) ms,P <0.05],which might resulted in further worsening of peak LV twist [pQRSd:(6.02 ± 0.74)° vs nQRSd:(7.91 ± 0.53)°,P <0.05].Conclusions LV twist dynamics was a good indicator of LV systolic function and had the potential to evaluate LV systolic dyssynchrony.
10.The analysis of risk factors about urethral injury secondary to pelvic fracture

Objective To investigate the risk factors of pelvic fracture related urethral injury (PFUI).Methods We retrospectively reviewed 120 patients with pelvic fracture from March 2014 to October 2016 in our hospital.Of all the patients,87 were males and 33 were females.The average age was (42.6 ± 15.4) years old (ranging 1 1-78 years old).The etiology of pelvic fracture included traffic accident in 70 cases,fall injury in 27 case.Heavy crash in 23 cases.43 male patients had an PFUI.Their average age was (40.4 ± 14.5) years old (ranging 14-61 years old).Of PFUI patients,the location of urethral injury included membranous part in 16 cases,bulbomenbranous part in 17 and prostatic part in 10 cases.The average length of stricture was (3.0 ± 1.9) centimeter.One patient suffered the stricture,combined with bladder rupture.One patient had urethro-rectal fistula.Two patients had urethrocutaneous fistula.The pelvic fractures were categorized using Tile classification,including type A1 fractures in 10 cases,A2 in 10 cases,B1 in 14 cases,B2 in 31 cases,B3 in 42 cases,C1 in 2 cases and C2 in 11 cases.The patients' number of urethral injury in each type was 1,2,2,7,29,0,2,respectively.Hazard degree analysis and chisquare test were used to evaluate the risk factors.Variables with an association of P < 0.05 were then tested using binary logistic regression and the area under the curve(AUC).Results On hazard degree analysis,female(x2 =25.419,P < 0.05,OR =0.571) and type B3 fracture (x2 =31.002,P < 0.05,OR =10.198)have a significant impact on the occurrence of PFUI.On binary logistic regression,female(Walds =16.528,P < 0.05,adjusted OR =0.003)is the protective factor of PFUI while type B3 fracture (Walds =10.853,P <0.05,adjusted OR =28.537) is the risk factor.The area under the curve (AUC =0.891,95% CI0.826-0.956) illustrates the regression model has a high credibility.Conclusions Pelvic fracture urethral injury is much less likely to occur in females than men.Type B3 fracture is more likely to cause an PFUI than the others type.
